Developing efficient and sensitive MOF-based luminescence sensors for bioactive molecule detection is of great significance and remains a challenge. Benefiting from favorable chemical and thermal ...stability, as well as excellent luminescence performance, a porous Zn(II)Ho(III) heterometallic–organic framework (ZnHoMOF) was selected here as a bifunctional luminescence sensor for the early diagnosis of a toluene exposure biomarker of hippuric acid (HA) through “turn-on” luminescence enhancing response and the daily monitoring of NFT/NFZ antibiotics through “turn-off” quenching effects in aqueous media with high sensitivity, acceptable selectivity, good anti-interference, exceptional recyclability performance, and low detection limits (LODs) of 0.7 ppm for HA, 0.04 ppm for NFT, and 0.05 ppm for NFZ. Moreover, the developed sensor was employed to quantify HA in diluted urine samples and NFT/NFZ in natural river water with satisfactory results. In addition, the sensing mechanisms of ZnHoMOF as a dual-response chemosensor in efficient detection of HA and NFT/NFZ antibiotics were conducted from the view of photo-induced electron transfer (PET), as well as inner filter effects (IFEs), with the help of time-dependent density functional theory (TD-DFT) and spectral overlap experiments.
Rapid detection of the 3-nitrotyrosine (3-NT) biomarker in serum is of great significance in clinical diagnosis and daily disease monitoring. In this work, two chemorobust 3D metal–organic frameworks ...(MOFs) of Co(DBrTPA)(NMP) n (CoMOF-1) and Co5(DBrTPA)4(DMF)6(HCOO)2 n (CoMOF-2) were fabricated from a halogen-modified ligand of 2,5-dibromoterephthalic (H2DBrTPA) and cobalt(II) clusters under solvothermal conditions. The introduction of halogens can improve the stability of the overall framework in different media and pH environments, which also endow two CoMOFs with great potential as luminescent sensors with high sensitivity and selectivity, fast response, good anti-interference, as well as recyclable performance in detecting 3-NT in a PBS buffer solution through quenching effects, with the K sv values being 9.19 × 104 M–1 for CoMOF-1 and 1.05 × 105 M–1 for CoMOF-2 and the LODs as low as 23.6 ng·mL–1 for CoMOF-1 and 20.1 ng·mL–1 for CoMOF-2. Furthermore, the developed sensors were employed to quantify 3-NT in real serum samples, with satisfactory results. In addition, the possible sensing mechanisms of CoMOFs toward the 3-NT biomarker were also discussed from photoinduced electron transfer and spectral overlaps. This work demonstrated the excellent potential of MOF-based sensors for biomarker detection in serum and provided an outlook to design MOF-based sensors with the help of artificial intelligence and machine learning.
Abstract Based on the mixed ligands of 1,4‐bis(2,4,6‐tricarboxylpyrid‐5‐yl)benzene (H 6 BTPB) and 1,4‐bis(imidazol‐1‐yl)benzene (bib), a new cadmium(II) coordination polymer with the molecular ...formula of {Cd(bib)( μ 2 ‐H 2 O)(H 2 O) 2 (H 4 BTPB) 0.5 (bib) 0.5 } n (CdCP) was constructed under solvothermal condition, and displayed a 3D supramolecular structure expanded by the hydrogen bonds between the 1D Cd(bib)( μ 2 ‐H 2 O)(H 2 O) 2 n ladder chain and the guests. Profiting from its superior stability and inherent luminescence, the prepared CdCP dispalyed great potential as a highly sensitive and selective luminescent sensor in trace detection of 3‐NT biomarker in both water and real bodily fluids through the luminescence quenching effects, with the K sv and LODs being 2.408×10 5 M −1 and 30.3 nM in water, 2.230×10 5 M −1 and 44.7 nM in diluted urine, 2.249×10 5 M −1 and 43.9 nM in diluted serum, respectively. Subsequent mechanism studies indicated the synergistic effect of internal filtering effect, photo‐induced electron transfer, and fluorescence resonance energy transfer determining the luminescence quenching response of CdCP towards 3‐NT biomarker.
AIM: To explore the expression of heparanase mRNA and point mutation in hepatocellular carcinoma (HCC). METHODS: Reverse transcription polymerase chain reaction was used to measure the expression of ...heparanase mRNA in the primary tumor tissues and surrounding liver tissues of 33 HCC patients. T-A cloning and sequencing were used to detect whether there was any mutation in the amplified PCR products. RESULTS: The expression of heparanase mRNA waspositive in 16 primary tumor tissues of HCC, and the positive rate was 48.5%, which was significantly higher than that in the surrounding liver parenchyma (P<0.01). The positive rate for heparanase gene in high-tendency to metastatic recurrence group (71.4%, 10/14) was obviously higher than that in low-tendency to metastatic recurrence group (31.6%, 6/19) (P= 0.023). The positive rate for heparanase gene in paltients with metastatic recurrence during postoperative follow-up (78.6%, 11/14) was also significantly higher than that in those without metastatic recurrence (21.4%, 3/14) (P = 0.003). Sequence analysis of the HPA PCR products was made in 7 patients, and 2-point mutations were found in 4 patients, one of which was sense mutation, neither base insertion nor deletion was detected. The mutation rate was 57.1% (4/7). CONCLUSION- The expression rate of heparanase mRNA increases in HCC, and HPA mRNA may be one of the reliable markers for the metastatic activity gained by the liver tumor cells and could be used clinically in predicting metastatic recurrence of HCC. Point mutation may be one of the causes for enhanced heparanase mRNA expression.
Cervical cancer (CC) is one of the most common malignancies affecting female worldwide. The main objective of this study was to explore the functions and mechanism of LINC00885 in CC.
RT-qPCR and western blot were used to detect RNA and protein levels. Functional and mechanism assays were respectively done for the analysis of cell behaviors and molecular interplays.
Long intergenic non-coding RNA 885 (LINC00885) was discovered to be upregulated in CC tissues and cell lines through bioinformatics analysis and RT-qPCR. Overexpression of LINC00885 promoted proliferation and inhibited apoptosis, whereas its silence exerted opposite effects. The cytoplasmic localization of LINC00885 was ascertained and furthermore, LINC00885 competitively bound with miR-3150b-3p to upregulate BAZ2A expression in CC cells. Rescue assays confirmed that LINC00885 regulated CC proliferation and apoptosis through miR-3150b-3p/BAZ2A axis. Finally, we confirmed that LINC00885 aggravated tumor growth through animal experiments.
LINC00885 exerted oncogenic function in CC via regulating miR-3150b-3p/BAZ2A axis. These findings suggested LINC00885 might serve as a potential promising therapeutic target for CC patients.

In our previous study, two crude polysaccharides from red kidney bean and small black soybean (RK, SB) have shown the alleviative effect on type 2 diabetic mice. Meanwhile, hepatic dysfunction usually accompanied with type 2 diabetes mellitus (T2DM), and closely related to glucose and lipid homeostasis. Therefore, this time we further investigated the protective effect on hepatic dysfunction of RK and SB. Results found that both crude polysaccharides had the protective effects. In addition, investigation on purified polysaccharides identified that the polysaccharide was the biofunctional component basis in crude RK and SB. Subsequently, further research investigated the regulating mechanism of two pure polysaccharides (RKPH and SBPH) on hepatic metabolism and lipid metabolism. The results showed the improved different metabolites of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) by RKPH and SBPH to affect glycerophospholipid metabolism pathway might be involved in the improvement of the glucose, lipids homeostasis and liver function in T2DM.
Due to various reasons, such as limitations in data collection and interruptions in network transmission, gathered data often contain missing values. Existing state-of-the-art generative adversarial ...imputation methods face three main issues: limited applicability, neglect of latent categorical information that could reflect relationships among samples, and an inability to balance local and global information. We propose a novel generative adversarial model named DTAE-CGAN that incorporates detracking autoencoding and conditional labels to address these issues. This enhances the network's ability to learn inter-sample correlations and makes full use of all data information in incomplete datasets, rather than learning random noise. We conducted experiments on six real datasets of varying sizes, comparing our method with four classic imputation baselines. The results demonstrate that our proposed model consistently exhibited superior imputation accuracy.
