We use an ensemble of surface (EPA CSN, IMPROVE, SEARCH, AERONET), aircraft (SEAC4RS), and satellite (MODIS, MISR) observations over the southeast US during the summer-fall of 2013 to better ...understand aerosol sources in the region and the relationship between surface particulate matter (PM) and aerosol optical depth (AOD). The GEOS-Chem global chemical transport model (CTM) with 25 25 km2 resolution over North America is used as a common platform to interpret measurements of different aerosol variables made at different times and locations. Sulfate and organic aerosol (OA) are the main contributors to surface PM2.5 (mass concentration of PM finer than 2.5 mu m aerodynamic diameter) and AOD over the southeast US. OA is simulated successfully with a simple parameterization, assuming irreversible uptake of low-volatility products of hydrocarbon oxidation. Biogenic isoprene and monoterpenes account for 60 % of OA, anthropogenic sources for 30 %, and open fires for 10 %. 60 % of total aerosol mass is in the mixed layer below 1.5 km, 25 % in the cloud convective layer at 1.5-3 km, and 15 % in the free troposphere above 3 km. This vertical profile is well captured by GEOS-Chem, arguing against a high-altitude source of OA. The extent of sulfate neutralization (f = NH4+/(2SO42- + NO3-) is only 0.5-0.7 mol mol-1 in the observations, despite an excess of ammonia present, which could reflect suppression of ammonia uptake by OA. This would explain the long-term decline of ammonium aerosol in the southeast US, paralleling that of sulfate. The vertical profile of aerosol extinction over the southeast US follows closely that of aerosol mass. GEOS-Chem reproduces observed total column aerosol mass over the southeast US within 6 %, column aerosol extinction within 16 %, and space-based AOD within 8-28 % (consistently biased low). The large AOD decline observed from summer to winter is driven by sharp declines in both sulfate and OA from August to October. These declines are due to shutdowns in both biogenic emissions and UV-driven photochemistry. Surface PM2.5 shows far less summer-to-winter decrease than AOD and we attribute this in part to the offsetting effect of weaker boundary layer ventilation. The SEAC4RS aircraft data demonstrate that AODs measured from space are consistent with surface PM2.5. This implies that satellites can be used reliably to infer surface PM2.5 over monthly timescales if a good CTM representation of the aerosol vertical profile is available.
We report observations of superradiance for atoms trapped in the near field of a photonic crystal waveguide (PCW). By fabricating the PCW with a band edge near the D(1) transition of atomic cesium, ...strong interaction is achieved between trapped atoms and guided-mode photons. Following short-pulse excitation, we record the decay of guided-mode emission and find a superradiant emission rate scaling as Γ̅(SR)∝N̅Γ(1D) for average atom number 0.19≲N̅≲2.6 atoms, where Γ(1D)/Γ'=1.0±0.1 is the peak single-atom radiative decay rate into the PCW guided mode, and Γ' is the radiative decay rate into all the other channels. These advances provide new tools for investigations of photon-mediated atom-atom interactions in the many-body regime.
In 2010 there were more than 200 million cases of malaria, and at least 655,000 deaths. The World Health Organization has recommended artemisinin-based combination therapies (ACTs) for the treatment ...of uncomplicated malaria caused by the parasite Plasmodium falciparum. Artemisinin is a sesquiterpene endoperoxide with potent antimalarial properties, produced by the plant Artemisia annua. However, the supply of plant-derived artemisinin is unstable, resulting in shortages and price fluctuations, complicating production planning by ACT manufacturers. A stable source of affordable artemisinin is required. Here we use synthetic biology to develop strains of Saccharomyces cerevisiae (baker's yeast) for high-yielding biological production of artemisinic acid, a precursor of artemisinin. Previous attempts to produce commercially relevant concentrations of artemisinic acid were unsuccessful, allowing production of only 1.6 grams per litre of artemisinic acid. Here we demonstrate the complete biosynthetic pathway, including the discovery of a plant dehydrogenase and a second cytochrome that provide an efficient biosynthetic route to artemisinic acid, with fermentation titres of 25 grams per litre of artemisinic acid. Furthermore, we have developed a practical, efficient and scalable chemical process for the conversion of artemisinic acid to artemisinin using a chemical source of singlet oxygen, thus avoiding the need for specialized photochemical equipment. The strains and processes described here form the basis of a viable industrial process for the production of semi-synthetic artemisinin to stabilize the supply of artemisinin for derivatization into active pharmaceutical ingredients (for example, artesunate) for incorporation into ACTs. Because all intellectual property rights have been provided free of charge, this technology has the potential to increase provision of first-line antimalarial treatments to the developing world at a reduced average annual price.

We report the first measurement of coherent elastic neutrino-nucleus scattering (CEvNS) on argon using a liquid argon detector at the Oak Ridge National Laboratory Spallation Neutron Source. Two ...independent analyses prefer CEvNS over the background-only null hypothesis with greater than 3σ significance. The measured cross section, averaged over the incident neutrino flux, is (2.2±0.7)×10^{-39} cm^{2}-consistent with the standard model prediction. The neutron-number dependence of this result, together with that from our previous measurement on CsI, confirms the existence of the CEvNS process and provides improved constraints on nonstandard neutrino interactions.
Epigenetic deregulation of gene expression has a role in the initiation and progression of prostate cancer (PCa). The histone methyltransferase MMSET/WHSC1 (Multiple Myeloma SET domain) is ...overexpressed in a number of metastatic tumors, but its mechanism of action has not been defined. In this work, we found that PCa cell lines expressed significantly higher levels of MMSET compared with immortalized, non-transformed prostate cells. Knockdown experiments showed that, in metastatic PCa cell lines, dimethylation of lysine 36 and trimethylation of lysine 27 on histone H3 (H3K36me2 and H3K27me3, respectively) depended on MMSET expression, whereas depletion of MMSET in benign prostatic cells did not affect chromatin modifications. Knockdown of MMSET in DU145 and PC-3 tumor cells decreased cell proliferation, colony formation in soft agar and strikingly diminished cell migration and invasion. Conversely, overexpression of MMSET in immortalized, non-transformed RWPE-1 cells promoted cell migration and invasion, accompanied by an epithelial-mesenchymal transition (EMT). Among a panel of EMT-promoting genes analyzed, TWIST1 expression was strongly activated in response to MMSET. Chromatin immunoprecipitation analysis demonstrated that MMSET binds to the TWIST1 locus and leads to an increase in H3K36me2, suggesting a direct role of MMSET in the regulation of this gene. Depletion of TWIST1 in MMSET-overexpressing RWPE-1 cells blocked cell invasion and EMT, indicating that TWIST1 was a critical target of MMSET, responsible for the acquisition of an invasive phenotype. Collectively, these data suggest that MMSET has a role in PCa pathogenesis and progression through epigenetic regulation of metastasis-related genes.

. Here we report observations of 15 bursts from FRB 180301 and find various polarization angle swings in seven of them. The diversity of the polarization angle features of these bursts is consistent with a magnetospheric origin of the radio emission, and disfavours the radiation models invoking relativistic shocks.
Coulomb collisions provide plasma resistivity and diffusion but in many low-density astrophysical plasmas such collisions between particles are extremely rare. Scattering of particles by ...electromagnetic waves can lower the plasma conductivity. Such anomalous resistivity due to wave-particle interactions could be crucial to many processes, including magnetic reconnection. It has been suggested that waves provide both diffusion and resistivity, which can support the reconnection electric field, but this requires direct observation to confirm. Here, we directly quantify anomalous resistivity, viscosity, and cross-field electron diffusion associated with lower hybrid waves using measurements from the four Magnetospheric Multiscale (MMS) spacecraft. We show that anomalous resistivity is approximately balanced by anomalous viscosity, and thus the waves do not contribute to the reconnection electric field. However, the waves do produce an anomalous electron drift and diffusion across the current layer associated with magnetic reconnection. This leads to relaxation of density gradients at timescales of order the ion cyclotron period, and hence modifies the reconnection process.
Abstract Observations reveal that surface and body modes exist in solar pores under photospheric conditions. While the effects of resonant absorption on photospheric surface modes are well ...established, its effect on body modes is not known yet. In this paper, we investigate resonant absorption for the body modes under photospheric conditions. We numerically solve the dispersion relation induced by thin boundary approximation and obtain the wave dispersion curves and damping rates of three arbitrarily chosen body modes for sausage and kink waves, respectively. The results show that resonant absorption for the body modes is weaker than for the slow surface modes in both cusp and Alfvén continua. The damping behavior of body modes is similar to slow surface modes while the higher body mode has stronger resonant absorption.
Abstract
The shear flow influences the stability of magnetohydrodynamic (MHD) waves. In the presence of a dissipation mechanism, flow shear may induce an MHD wave instability below the threshold of ...the Kelvin–Helmholtz instability, which is called dissipative instability. This phenomenon is also called negative energy wave instability because it is closely related to the backward wave, which has negative wave energy. Considering viscosity as a dissipation mechanism, we derive an analytical dispersion relation for the slow sausage modes in a straight cylinder with a discontinuous boundary. It is assumed that the steady flow is inside and dynamic and bulk viscosities are outside the circular flux tube under photospheric condition. When the two viscosities are weak, it is found that for the slow surface mode, the growth rate is proportional to the axial wavenumber and flow shear, consistent within the incompressible limit. For a slow body mode, the growth rate has a peak at a certain axial wavenumber, and its order of magnitude is similar to surface mode. The linear relationship between the growth rate and the dynamic viscosity established in the incompressible limit develops nonlinearly when the flow shear and/or the two viscosities are sufficiently strong.
Aim
Claudin‐2 is a tight junction protein typically located in ‘leaky’ epithelia exhibiting large paracellular permeabilities like small intestine and proximal kidney tubule. Former studies revealed ...that claudin‐2 forms paracellular channels for small cations like sodium and potassium and also paracellular channels for water. This study analyses whether the diffusive transport of sodium and water occurs through a common pore of the claudin‐2 channel.
Methods
Wild‐type claudin‐2 and different claudin‐2 mutants were expressed in MDCK I kidney tubule cells using an inducible system. Ion and water permeability and the effect of blocking reagents on both were investigated on different clones of the mutants.
Results
Neutralization of a negatively charged cation interaction site in the pore with the mutation, D65N, decreased both sodium permeability and water permeability. Claudin‐2 mutants (I66C and S68C) with substitution of the pore‐lining amino acids with cysteine were used to test the effect of steric blocking of the claudin‐2 pore by thiol‐reactive reagents. Addition of thiol‐reactive reagents to these mutants simultaneously decreased conductance and water permeability. Remarkably, all experimental perturbations caused parallel changes in ion conductance and water permeability, disproving different or independent passage pathways.
Conclusion
Our results indicate that claudin‐2‐mediated cation and water transport are frictionally coupled and share a common pore. This pore is lined and determined in permeability by amino acid residues of the first extracellular loop of claudin‐2.
