Improving passive design methods are important for achieving zero-carbon building (ZCB) targets. Data-driven design approaches are more accurate than conventional methods for multiple-objective ...optimization (MOO) targets such as carbon emissions (CE), economics, and thermal comfort. In this study, a comprehensive data-driven-based PSO-SVM-NSGA-III method that assists in optimizing passive design parameters are proposed. First, the simulation results for the objectives of CE, incremental cost (IC), and time not comfort (TNC) are performed for an office building in a cold region in China using EnergyPlus and jEPlus software. Then, key influencing factors are chosen via a sensitivity analysis. Second, nonlinear mapping relationships between the passive design parameters and objectives are established with the PSO-SVM model. Compared to the BPNN, SVM, and PSO-BPNN algorithms, the PSO-SVM model exhibits superior prediction performance, with R2 values of 0.977, 0.925, and 0.903 for CE, IC, and TNC, respectively. Third, the nonlinear mapping relationships are used in the established objective functions of NSGA-II and NSGA-III. NSGA-III performed 100 iterations within 21 min and exhibited robust diversity within its population, with a hypervolume value of 0.738. Finally, the Pareto-optimal solution set is formed with the PSO-SVM-NSGA-III framework. This data-driven method offers an efficient approach for improving building performance.

Interleukin-17 (IL-17) plays an important role in inflammation and host defense in mammals. In this study, we identified two duplicated IL-17A/F2 genes in the common carp (Cyprinus carpio) ...(ccIL-17A/F2a and ccIL-17A/F2b), putative encoded proteins contain 140 amino acids (aa) with conserved IL-17 family motifs. Expression analysis revealed high constitutive expression of ccIL-17A/F2s in mucosal tissues, including gill, skin and intestine, their expression could be induced by Aeromonas hydrophila, suggesting a potential role in mucosal immunity. Recombinant ccIL-17A/F2a protein (rccIL-17A/F2a) produced in Escherichia coli could induce the expression of proinflammatory cytokines (IL-1β) and the antimicrobial peptides S100A1, S100A10a and S100A10b in the primary kidney in a dose- and time-dependent manner. Above findings suggest that ccIL-17A/F2 plays an important role in both proinflammatory and innate immunity. Two duplicated ccIL-17A/F2s showed different expression level with ccIL-17A/F2a higher than b, comparison of two 5′ regulatory regions indicated the length from anticipated promoter to transcriptional start site (TSS) and putative transcription factor binding site (TFBS) were different. Promoter activity of ccIL-17A/F2a was 2.5 times of ccIL-17A/F2b which consistent with expression results of two genes. These suggest mutations in 5'regulatory region contributed to the differentiation of duplicated genes. To our knowledge, this is the first report to analyze 5'regulatory region of piscine IL-17 family genes.

Covalent organic frameworks (COFs) are a class of crystalline porous organic polymers with periodic networks that are constructed from small molecular units via covalent bonds, which have low ...densities, high porosity, large specific surface area, and ease of functionalization. The one-dimension nanochannels in COFs offer an effective means of transporting lithium ions while maintaining a stable structure over a wide range of temperatures. As a new category of ionic conductors, COFs exhibit unparalleled application potential in solid-state electrolytes. Here, we provide a comprehensive summary of recent applications and research progress for COFs in solid-state electrolytes of lithium metal batteries and discuss the possible development directions in the future. This review is expected to provide theoretical guidance for the design of high-performance solid-state electrolytes.
Bladder cancer (BCa) threatens human health due to the high occurrence and mortality. Nowadays, more and more researchers focussed on the molecular mechanisms and biological functions of miRNAs in ...human cancers. The present study aims to study the biological role of miR-454-3p and miR-374b-5p in BCa. The expression levels of miR-454-3p and miR-374b-5p were detected in BCa tissues and cell lines by qRT-PCR analysis. Kaplan-Meier analysis revealed that the expression levels of miR-454-3p and miR-374b-5p were positively correlated with the overall survival (OS) rate of BCa patients. Gain-of-function assays were conducted to demonstrate the inhibitory effects of miR-454-3p and miR-374b-5p on the invasion, migration, and epithelial-mesenchymal transition (EMT) of BCa cells. Mechanically, ZEB2 was found to be a target of both miR-454-3p and miR-374b-5p. Rescue assays revealed that ZEB2 reversed the inhibitory effects of miR-454-3p and miR-374b-5p on the invasion and migration of BCa cell lines. In summary, miR-454-3p and miR-374b-5p negatively regulated invasion and migration of BCa cell lines via targetting ZEB2.

We aimed to explore the expression of miRNAs and their potential roles in the DNA damage response (DDR) induced by Cr (VI) exposure in human B lymphoblast cells (HMy2.CIR cells) and in a population of Cr (VI)-exposed humans. Methods: Differentially expressed miRNAs were found by a combination of miRNA sequencing and RT-qPCR validation in HMy2.CIR cells treated with K2Cr2O7. Differentially expressed miRNAs related to DDR were selected for functional study. The expression levels of differential miRNAs were also investigated in chromate workers. Results: A total of 214 differentially expressed miRNAs were identified by sequencing, and the expression of 5 miRNAs among 25 associated with DDR was found to be consistent between sequencing and validation studies.Functional studies showed that miR-148a-3p, miR-21-5p, and miR-424-3p might be related to Cr (VI)-induced cell apoptosis, and miR-221-3p might participate in Cr (VI)-induced DDR. We also found that the expression of miR-21-5p and miR-424-3p was upregulated in chromate workers. Conclusions: Cr (VI) exposure could significantly impact miRNAs expression in vitro and in chromate workers. Functional studies showed that miR-148a-3p, miR-21-5p and miR-221-3p might take a crucial role in the cellular DDR induced by Cr (VI) exposure.
With semiconductor technology gradually approaching its physical and thermal limits, recent supercomputers have adopted major architectural changes to continue increasing the performance through more ...power-efficient heterogeneous many-core systems. Examples include Sunway TaihuLight that has four management processing elements (MPEs) and 256 computing processing elements (CPEs) inside one processor and Summit that has two central processing units (CPUs) and six graphics processing units (GPUs) inside one node. Meanwhile, current high-resolution Earth system models that desperately require more computing power generally consist of millions of lines of legacy code developed for traditional homogeneous multicore processors and cannot automatically benefit from the advancement of supercomputer hardware. As a result, refactoring and optimizing the legacy models for new architectures become key challenges along the road of taking advantage of greener and faster supercomputers, providing better support for the global climate research community and contributing to the long-lasting societal task of addressing long-term climate change. This article reports the efforts of a large group in the International Laboratory for High-Resolution Earth System Prediction (iHESP) that was established by the cooperation of Qingdao Pilot National Laboratory for Marine Science and Technology (QNLM), Texas A&M University (TAMU), and the National Center for Atmospheric Research (NCAR), with the goal of enabling highly efficient simulations of the high-resolution (25 km atmosphere and 10 km ocean) Community Earth System Model (CESM-HR) on Sunway TaihuLight. The refactoring and optimizing efforts have improved the simulation speed of CESM-HR from 1 SYPD (simulation years per day) to 3.4 SYPD (with output disabled) and supported several hundred years of pre-industrial control simulations. With further strategies on deeper refactoring and optimizing for remaining computing hotspots, as well as redesigning architecture-oriented algorithms, we expect an equivalent or even better efficiency to be gained on the new platform than traditional homogeneous CPU platforms. The refactoring and optimizing processes detailed in this paper on the Sunway system should have implications for similar efforts on other heterogeneous many-core systems such as GPU-based high-performance computing (HPC) systems.

The ribosomal DNA (rDNA) can act as a sensor and responder of cancer-associated stress. Here we investigated rDNA copy number in gastric cancers and its association with existing biomarkers and metals exposure. This study was performed on paired tumor and adjacent normal tissues obtained from 65 gastric cancer patients who underwent gastrectomy. Immunohistochemistry was used to assess HER-2, E-cadherin, EGFR, CK (pan), CK20, CK7, TopoⅡ, CAM5.2, P53, and Ki-67 expression. Inductively coupled plasma mass spectrometry (ICP-MS) was used to detect the concentrations of 17 metals in gastric tissues. rDNA copy number was detected by qPCR in genomic DNA isolated from tissue samples. Associations between the expression of existing markers, metal concentrations, and rDNA copy number were evaluated. Within patients with gastric cancer, the copy number of the 45S rDNA components (18S, 5.8S, 28S) and the 5S rDNA in tumor tissues were significantly higher than those in adjacent normal tissues, whereas mitochondrial DNA (mtDNA) copy number was significantly lower in tumor tissues than that in adjacent normal tissues. Further analysis revealed that the increase in 18S, 5.8S, and 28S rDNA copy number in tumor tissues was diminished in the context of EGFR and P53 loss. Moreover, analysis of metals revealed particularly high concentrations of As, Cd, Cr, Cu and Fe in the gastric tissues of these patients. Intriguingly, rDNA copy number variation across individuals was correlated with the concentrations of some metals. The rDNA was amplified in tumor tissues of gastric cancer patients, and its amplification may be associated with metals exposure. The expression of EGFR and P53 may influence rDNA copy number, with diminished amplification of the rDNA in cancers that were negative for these biomarkers. Our observation further our understanding of rDNA copy number in gastric cancer and its potential as a simple and useful marker in gastric cancer monitoring.

has been shown to possess immunomodulatory potential. In this study, we aimed to investigate whether the combination treatment of ginseng polysaccharides (GPs) and αPD-1 monoclonal antibody (mAb) could sensitise the response by modulating gut microbiota.
Syngeneic mouse models were administered GPs and αPD-1 mAb, the sensitising antitumour effects of the combination therapy on gut microbiota were assessed by faecal microbiota transplantation (FMT) and 16S PacBio single-molecule real-time (SMRT) sequencing. To assess the immune-related metabolites, metabolomics analysis of the plasma samples was performed.
We found GPs increased the antitumour response to αPD-1 mAb by increasing the microbial metabolites valeric acid and decreasing L-kynurenine, as well as the ratio of Kyn/Trp, which contributed to the suppression of regulatory T cells and induction of T
cells after combination treatment. Besides, the microbial analysis indicated that the abundance of

was higher in responders to anti-PD-1 blockade than non-responders in the clinic. Furthermore, the combination therapy sensitised the response to PD-1 inhibitor in the mice receiving microbes by FMT from six non-responders by reshaping the gut microbiota from non-responders towards that of responders.
Our results demonstrate that GPs combined with αPD-1 mAb may be a new strategy to sensitise non-small cell lung cancer patients to anti-PD-1 immunotherapy. The gut microbiota can be used as a novel biomarker to predict the response to anti-PD-1 immunotherapy.
Nonequilibrium molecular dynamics (NEMD) simulations were performed to investigate the tribology behaviors of Al.sub.2O.sub.3 and MoS.sub.2 nanoparticles confined between iron (Fe) slabs. Results ...indicated that the combined use of these two nanoparticles yielded the lowest and most stable friction force, normal force, interface temperature and wear rate, which exhibited a significant synergistic lubrication effect. A novel parameter the rolling/sliding motion coefficient (K.sub.rs) was proposed to evaluate the motion pattern of spherical Al.sub.2O.sub.3. There were 51% rolling + 49% sliding motion when used alone and 91% rolling + 9% sliding in the existence of MoS.sub.2. Similarly, about 72.3% of the friction was shared by interlayer sliding of MoS.sub.2 monolayers in the presence of Al.sub.2O.sub.3, which was higher than used alone (54.8%). Then, the diffusion of atoms at the friction interface was explored to reveal the synergistic lubrication mechanism. The tribofilm formed by the diffusion of Fe and S atoms could protect the metal surfaces from further wear. The adsorption of S atoms to Al.sub.2O.sub.3 nanoparticle could promote its rolling effect and prevent it from embedding into iron matrix. Besides, Al.sub.2O.sub.3 could also facilitate the separation of MoS.sub.2 monolayers to enhance their interlayer sliding effect.
Following chicken domestication, diversified chicken breeds were developed by both natural and artificial selection, which led to the accumulation of abundant genetic and phenotypic variations, ...making chickens an ideal genetic research model. To better understand the genetic structure of chicken breeds under different selection pressures, we genotyped various chicken populations with specific selection targets, including indigenous, commercial, gamecock, and wild ancestral chickens, using the 600K SNP array. We analyzed the population structure, genetic relationships, run of homozygosity (ROH), effective population number (Ne), and other genetic parameters. The wild ancestral population, red junglefowl (RJF), possessed the highest diversity, in comparison with all other domesticated populations, which was supported by linkage disequilibrium decay (LD), effective population number, and ROH analyses. The gamecock breeds, which were subjected to stronger male-biased selection for fighting-related traits, also presented higher variation than the commercial and indigenous breeds. Admixture analysis also indicated that game breed is a relatively independent branch of Chinese local breeds. Following intense selection for reproductive and productive traits, the commercial lines showed the least diversity. We also observed that the European local chickens had lower genetic variation than the Chinese local breeds, which could be attributed to the shorter history of the European breed. ROH were present in a breed specific manner and 191 ROH island were detected on four groups (commercial, local, game and wild chickens). These ROH islands were involved in egg production, growth and silky feathers and other traits. Moreover, we estimated the effective sex ratio of these breeds to demonstrate the change in the ratio of the two sexes. We found that commercial chickens had a greater sex imbalance between females and males. The commercial lines showed the highest female-to-male ratios. Interestingly, RJF comprised a greater proportion of males than females. Our results show the population genetics of chickens under selection pressures, and can aid in the development of better conservation strategies for different chicken breeds.
