Hantavirus pulmonary syndrome (HPS) is an increasing health problem in Brazil because of encroachment of sprawling urban, agricultural, and cattle-raising areas into habitats of subfamily ...Sigmodontinae rodents, which serve as hantavirus reservoirs. From 1993 through June 2007, a total of 884 cases of HPS were reported in Brazil (case-fatality rate 39%). To better understand this emerging disease, we collected 89 human serum samples and 68 rodent lung samples containing antibodies to hantavirus from a 2,500-km-wide area in Brazil. RNA was isolated from human samples and rodent tissues and subjected to reverse transcription-PCR. Partial sequences of nucleocapsid protein and glycoprotein genes from 22 human and 16 rodent sources indicated only Araraquara virus and Juquitiba virus lineages. The case-fatality rate of HPS was higher in the area with Araraquara virus. This virus, which may be the most virulent hantavirus in Brazil, was associated with areas that have had greater anthropogenic changes.

Researchers have investigated several therapeutic approaches to treat non-union fractures. Among these, bioactive glasses and glass ceramics have been widely used as grafts. This class of biomaterial ...has the ability to integrate with living bone. Nevertheless, bioglass and bioactive materials have been used mainly as powder and blocks, compromising the filling of irregular bone defects. Considering this matter, our research group has developed a new bioactive glass composition that can originate malleable fibers, which can offer a more suitable material to be used as bone graft substitutes. Thus, the aim of this study was to assess the morphological structure (via scanning electron microscope) of these fibers upon incubation in phosphate buffered saline (PBS) after 1, 7 and 14 days and, also, evaluate the in vivo tissue response to the new biomaterial using implantation in rat tibial defects. The histopathological, immunohistochemistry and biomechanical analyzes after 15, 30 and 60 days of implantation were performed to investigate the effects of the material on bone repair. The PBS incubation indicated that the fibers of the glassy scaffold degraded over time. The histological analysis revealed a progressive degradation of the material with increasing implantation time and also its substitution by granulation tissue and woven bone. Histomorphometry showed a higher amount of newly formed bone area in the control group (CG) compared to the biomaterial group (BG) 15 days post-surgery. After 30 and 60 days, CG and BG showed a similar amount of newly formed bone. The novel biomaterial enhanced the expression of RUNX-2 and RANK-L, and also improved the mechanical properties of the tibial callus at day 15 after surgery. These results indicated a promising use of the new biomaterial for bone engineering. However, further long-term studies should be carried out to provide additional information concerning the material degradation in the later stages and the bone regeneration induced by the fibrous material.
Thiazolidinediones (TZDs) enhanced body weight (BW) partially by increased adipogenesis and hyperphagia. Neuronal PPARγ knockout mice on high-fat diet (HFD) are leaner because of enhanced leptin ...response, although it could be secondary to their leanness. Thus, it still is an open question how TZDs may alter energy balance. Multiple factors regulate food intake (FI) and energy expenditure (EE), including anorexigenic hormones as insulin and leptin. Nonetheless, elevated hypothalamic AMPK activity increases FI and TZDs increase AMPK activity in muscle cells. Thus, the aim of the present study was to investigate whether Pioglitazone (PIO) treatment alters hypothalamic insulin and leptin action/signaling, AMPK phosphorylation, and whether these alterations may be implicated in the regulation of FI and EE.
Swiss mice on HFD (2 months) received PIO (25 mg kg(-1) per day-gavage) or vehicle for 14 days. AMPK and AdipoR1 were inhibited via Intracerebroventricular injections using Compound C (CompC) and small interference RNA (siRNA), respectively. Western blot, real-time PCR and CLAMS were done.
PIO treatment increased BW, adiposity, FI, NPY mRNA and decreased POMC mRNA expression and EE in HFD mice. Despite higher adiposity, PIO treatment improved insulin sensitivity, glucose tolerance, decreased insulin and increased adiponectin serum levels. This result was associated with, improved insulin and leptin action/signaling, decreased α2AMPK(Ser491) phosphorylation and elevated Acetyl-CoA carboxylase and AMPK(Thr172) phosphorylation in hypothalamus. The inhibition of hypothalamic AMPK with CompC was associated with decreased adiposity, FI, NPY mRNA and EE in PIO-treated mice. The reduced expression of hypothalamic AdipoR1 with siRNA concomitantly with PIO treatment reverted PIO induced obesity development, suggesting that adiponectin may be involved in this effect.
These results demonstrated that PIO, despite improving insulin/leptin action in hypothalamus, increases FI and decreases EE, partially, by activating hypothalamic adiponectin/AdipoR1/AMPK axis. Suggesting a novel mechanism in the hypothalamus by which TZDs increase BW.
Abstract
Three experiments were conducted with the aim of developing and validating an equation to predict the ME of corn for pigs from its chemical composition, physical characteristics and particle ...size. Exp. 1: Eight lots of corn were ground in a hammer mill, using 5 sieves with different screen opening sizes, generating 40 batches of ground corn. The chemical composition (DM, CP, ether extract, crude fiber, ADF, NDF, and ash) and physical characteristics (bulk density– BD and 1,000-kernel weight- TKW) were determined in the 8 lots and geometric mean diameter (GMD) and N-corrected ME (AMEn) were determined in the 40 batches of corn. The AMEn values were determined in 16 metabolism assays with pigs. Mathematical models were adjusted by regression analysis, based on the Akaike Information Criterion. Based on statistical parameters (R2 = 0.76 and prediction error = 1.05%), number of predictor variables, and easiness of measurements, an equation with 2 segments was chosen: y = 2845.41 + 0.9385 × BD – 20.8784 × CP, if GMD ≤ 522.98 and y = 3105.75 – 0.4978 × GMD + 0.9385 × BD – 20.8784 × CP, if GMD > 522.98. Exp. 2 and 3: Sixty four gilts (Exp. 1; 29.5 ± 3.8 kg) and 64 barrows (Exp. 2; 29.3 ± 3.6 kg), 1 lot of corn, and 3 particle sizes (GMD = 483, 632, and 904 µm) were used in a 3 × 2 factorial arrangement with 2 methods of diet formulation, differing in ME value of corn: “FIX” (value from nutrient composition table) vs. “ESTIMATED” (estimated for each particle size using the equation developed in Exp. 1). In Exp. 2, ADFI was greater (P < 0.05) and feed efficiency was lower (P < 0.05) in the diet with GMD of 904 µm compared to the diets with GMD of 632 or 483 µm, but only for diet formulation by the FIX method. In the treatments with GMD of 483 µm, gilts fed with the diet formulated by the ESTIMATED method had greater (P < 0.10) ADFI, backfat, fat area, and fat:meat ratio than gilts fed the diet formulated by the FIX method. In Exp. 3, particle size and formulation method did not affect (P > 0.10) growth performance and carcass traits of barrows. The equation developed was effective to adjust the ME value of corn, considering particle size variation. However, improvement to the proposed equation is necessary to achieve greater precision for predicting corn ME. Validation of the equation with more lots of corn of different chemical compositions and densities appears necessary to assess the efficacy of the equation regarding the variation of the other predicting variables.

Infection is the most relevant surgical complication in implant or grafting procedures. Osteomyelitis and other chronic conditions pose a constant challenge in current medical practice. In this context, a grafting biomaterial that possesses antibacterial properties combined with bioactivity could have great clinical impact. Researchers at the Vitreous Materials Laboratory (LaMaV–UFSCar) recently developed a glass composition, named F18, that presents an improved workability range combined with high bioactivity. With F18, one can easily manufacture complex shapes, such as scaffolds, continuous fibres and coat implants. This biomaterial has proven to be a viable alternative for bone and skin regeneration in in vivo tests, however its antimicrobial properties have not been explored. Hence, the purpose of this study was to systematically investigate the antibacterial activity of F18 in powder and fibre forms according to the JIS Z 2801:2010 standard. Whether incorporation of silver into F18 glass could impact its antimicrobial activity was also evaluated. Four clinically relevant Gram-positive and Gram-negative pathogenic bacterial strains (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli and Pseudomonas aeruginosa) were used in this study. In both powder and fibre forms, F18 presented extremely efficient bactericidal activity against all strains tested, eliminating virtually 100% of the bacterial cells after 24 h. Kinetic tests showed that silver doping further increased the bactericidal activity, leading to S. aureus eradication in only 30 min after incubation. Both doped and non-doped glasses demonstrated very high bactericidal activity, making F18 a promising infection-preventing alternative for bone and wound regeneration in clinical practice.
Aim
To evaluate the biocompatibility, induction of mineralization and antimicrobial activity of experimental intracanal pastes based on two glass and glass‐ceramic materials. Calcium hydroxide ...(Ca(OH)2) paste was used as the positive control.
Methodology
The glass‐ceramic powder two‐phased Biosilicate (BS‐2P) and F18 bioactive glass were mixed with distilled water (ratio 2 : 1), inserted in polyethylene tubes and implanted in the subcutaneous tissues of 16 rats. Empty tubes were used as negative control. After 7 and 30 days (n = 8), the rats were euthanized for haematoxylin–eosin, von Kossa, polarized light and osteopontin (OPN) immunolabeling analysis. Direct contact tests using a suspension of each paste were performed with Enterococcus faecalis planktonic cells to evaluate antimicrobial activity (24 h of contact), in a pilot study. The number of CFU mL−1 was calculated for each group. The antimicrobial analysis data were submitted to one‐way anova and Tukey tests, whilst biocompatibility and immunohistochemical data were submitted to the Kruskal–Wallis and Dunn tests (P < 0.05).
Results
Most specimens of the control, BS‐2P and Ca(OH)2 groups were associated with moderate inflammation seven days following implantation, whilst F18 was associated with moderate to severe inflammation, without differences amongst the groups (P > 0.05). At 30 days, most specimens of control, F18 and BS‐2P groups had mild inflammation, whilst Ca(OH)2 had mild to moderate inflammation; however, no differences were determined amongst the groups (P > 0.05). The fibrous capsule was thick at 7 days, becoming thin at 30 days. All pastes induced von Kossa‐positive structures and were birefringent to polarized light. At seven days, the BS‐2P group had significantly more OPN immunolabeling compared to the control and Ca(OH)2 groups (P < 0.05). At 30 days, the F18 group had significantly more OPN immunolabeling compared to the control and Ca(OH)2 groups (P < 0.05). All pastes reduced the total number of E. faecalis; however, the reduction was only significant when comparing BS‐2P and Ca(OH)2 groups to the control (P < 0.05). Only calcium hydroxide eliminated E. faecalis.
Conclusions
Experimental BS‐2P and F18 pastes were biocompatible, stimulated biomineralization and induced significant OPN immunolabeling compared to Ca(OH)2. Only the BS‐2P paste demonstrated antimicrobial activity comparable to Ca(OH)2.

Hypothalamic CLK2
phosphorylation, which induces CLK2 activity, is regulated in vivo by refeeding, insulin and leptin, in a PI3K (phosphoinositide 3-kinase)-dependent manner. The reduction of CLK2 expression in the hypothalamus, by chronic pharmacological inhibition with TG003 or by chronic knockdown with small interfering RNA was sufficient to abolish the anorexigenic effect of insulin and leptin, to increase body weight, fat mass, food intake and to decrease energy expenditure in mice on chow. In contrast, CLK2
phosphorylation in the hypothalamus in response to insulin, leptin or refeeding was impaired in mice on HFD or in db/db mice. Chronic CLK2 inhibition in the hypothalamus was associated with a slight increase in the fasting blood glucose levels, reduction in PEPCK (phosphoenolpyruvate carboxykinase) expression in the liver and enhanced glucose production from pyruvate, suggesting a regulation of hepatic glucose production. Further, overexpressing CLK2 in the mediobasal hypothalami of mice on HFD or in db/db mice by adenovirus partially reversed the obese phenotype.
Thus, our results suggest that protein CLK2 integrates some important hypothalamic pathways, and may be a promising molecule for new therapeutic approaches for obesity and diabetes.
Bioactive glasses are able to chemically bond to hard and soft tissues and have been proposed and used for tissue regeneration in several dentistry and medical applications. However, the majority of ...bioactive glass compositions do not support prolonged or repeated heat treatments, since these procedures often result in uncontrolled crystallization, which usually degrade their mechanical properties and, in most instances, substantially diminish their bioactivity. Therefore, the manufacturing of 3D devices, fibers or scaffolds, which aim to expand the usage of these materials, is a challenging task. To overcome this phenomenon, a new bioactive glass composition was recently developed at the Vitreous Materials Laboratory (LaMaV-UFSCar, Brazil) and licensed to the start-up company VETRA. This new bioactive glass composition shows high stability against crystallization coupled with high bioactivity, which allows the development of bioactive fibers, meshes and other complex 3D shapes. In addition, this bioactive glass has an elevated bioactivity, is bioresorbable and flexible (in fiber form), which makes this glass a potential alternative for soft and hard tissue regeneration. In this article, we discuss this recent development and summarize the latest advances in testing the effectiveness of this new material in in vitro and in vivo tests. To date, the results indicate that this new glass composition presents a larger workability window, which allows the development of numerous medical devices. This feature combined with the high bioactivity of this new glass delivers a promising broad spectrum of applications as a material for tissue engineering.
