Drug resistance and frequent tumor relapses are the major obstacles in glioblastoma therapy, and recurrent tumors are practically incurable. We previously reported that insulin receptor substrate 1 ...(IRS-1), which is a typical signaling molecule for insulin and insulin-like growth factor 1 receptors, can translocate to nucleus, and that nuclear IRS-1 (nIRS-1) was found in different tumor cells, including glioblastomas. To unravel its function, we employed glioblastoma cell culture, animal models, and clinical samples. Using confocal imaging, molecular cloning, subcellular fractionation, mass spectrometry, gene expression analysis, and different approaches to verify protein-protein interactions, we demonstrate for the first time that nIRS-1 can form complex nuclear structures in a restricted number of cancer cells in glioblastoma biopsies and in intracranial glioblastoma xenografts. We also demonstrated the formation of highly organized ring-like structures in several cell lines, following ectopic expression of IRS-1 cloned in frame with nuclear localization signal (NLS-IRS-1). In these nuclear structures IRS-1 localizes at the periphery, and the core of the structure harbors a key autophagy protein, LC3; however, other autophagy proteins or biological membranes were not detected. In living cells expressing NLS-IRS-1-GFP fusion protein, IRS-1LC3 structures are highly dynamic. They rapidly exchange IRS-1 molecules with nucleoplasm and interact with other nuclear complexes including BMI1-positive Polycomb bodies, PML bodies and Cajal bodies. Importantly, clones and mixed populations of cells expressing the NLS-IRS-1 and capable of forming the IRS-1LC3 ring-like structures undergo extensive remodeling of gene expression, which suggests a transition to stem-like phenotype and associated resistance to several different anticancer drugs, including temozolomide. This is the first demonstration of IRS-1LC3 nuclear complexes, which are highly dynamic and may play a role in epigenetic remodeling of glioblastoma cells towards stemness. Further studies are required to determine detailed molecular composition and to explain how these new nuclear structures function.

Polillas tigre. Polillas avispas. Polillas de líquenes. Lista de especies. The arctiid moths are macrolepidopteran of different sizes and nocturnal habits mostly. The richness of this group is poorly known in Argentina, particularly in Cordoba. The Serrano Forest is one of the highest biodiversity areas in Cordoba province. The aim of this study was to update the richness of arctiid moths in the Serrano Forest. Thirty-eight samplings were performed in different areas of Sierras Chicas using light traps, from December 2006 to April 2011, with an unequal frequency between the different sampling sites. A total of 26 species, belonging to the tribes Arctiini and Lithosiini, were found. The genus Agylla Walker is a new citation for Argentina and 21 species are new records for this environment. This study increased to 28 the arctiid species attracted by light traps that live in the Serrano Forest. KEY WORDS. Tiger moths. Wasp moths. Lichen moths. Checklist.
Abstract
Glioblastoma multiforme is the most common and aggressive primary brain tumor in adults. It is characterized by a rapid cell growth, CNS infiltration and is resistant to all known anticancer ...regimens. Therefore, development of effective therapies against these malignant brain tumors represents challenge for contemporary medicine. Anti-neoplastic potential of a common lipid-lowering drug, fenofibrate, is gaining a lot of attention, however, the precise mechanism of fenofibrate anti-cancer action remain to be elucidated. Fenofibric acid, which is an active metabolite of fenofibrate, is a potent agonist of peroxisome proliferator activated receptor alpha (PPARα), which transcriptional activity is expected to shift energy metabolism from glycolysis to fatty acid β-oxidation, therefore, it may be toxic to glycolytic tumor cells (Warburg effect). In parallel, to PPARα- dependent action, we have observed a strong PPARα- independent effect of fenofibrate on cancer cell energy metabolism, which was specific only to fenofibrate but not to fenofibric acid or to other PPARα agonists. We detected fenofibrate in mitochondrial membrane fractions of LN-229 cells treated with fenofibrate by using HPLC and subcellular fractionation. The accumulation of fenofibrate in mitochondrial membrane was accompanied by significant change in mitochondrial function, including loss of mitochondrial membrane potential, dramatic loss of oxygen consumption and lack of respiratory capacity measured as oxygen consumption rate (OCR). At the same time, fenofibrate increased the level of glycolysis (extracellular acidification rate, ECAR), possibly as compensation for impaired oxidative phosphorylation. In addition, fenofibrate-induced reduction of ATP levels triggered phosphorylation of AMPK and activation of autophagy.
In conclusion, our results demonstrate two seemingly independent mechanisms of fenofibrate anticancer action: PPARα-dependent, which shifts cell metabolism from glycolysis to fatty acid β-oxidation and PPARα-independent, which causes accumulation of fenofibrate in cell membranes and inhibition of oxidative phosphorylation. Both mechanisms counteract to make this drug very effective against Glioblastoma tumors causing inhibition of cell growth, survival and finally leading to massive apoptosis of these practically incurable tumors.
