Microplastic pollution in farmlands has become a source of major concern, but few previous studies have focused on the effect of microplastics on higher plants. In this study, the distribution of ...polystyrene nanoplastics (PSNPs) of four different particle sizes (100, 300, 500, and 700 nm) was investigated in cucumber plants, and their influence on physiological indexes of the root system and fruit quality was determined. The results showed that PSNPs initially accumulated in the root system before being transported to the aboveground parts of the plant. Finally, they were distributed in the leaves, flowers, and fruits, through the stems. The 300-nm plastic microspheres significantly increased root activity and malondialdehyde (MDA) and proline content of the roots. The results demonstrated that the environmental pressures caused by PSNPs of different particle sizes were different. The amount of soluble protein in cucumber fruits was significantly increased, and the levels of Mg, Ca, and Fe were significantly decreased by PSNPs of different particle sizes. Our findings provide a scientific basis for risk assessment of PSNP exposure in the soil–plant systems.
Changes in the types and contents of metabolites in plants can occur in response to environmental stress. In this study, pumpkin seeds were cultivated in a cadmium ion solution (cadmium sulfate) for ...7 days, and growth parameters, antioxidant enzyme activities, and metabolites in the root, stem, and leaf were analyzed. The results showed that cadmium accumulation characteristics were in the order of root > stem > leaf. Cadmium restrained root growth and promoted superoxide dismutase, peroxidase, catalase activities in the root, but inhibited their activities in the leaf. Cadmium did not change the total biomass of pumpkin seedlings. Orthogonal partial least squares (OPLS) analyses were conducted to detect the relationships between fresh weight and metabolites. These analyses revealed that maltose had significantly positive relationships with the fresh weight of the root, stem, and leaf. Cadmium influenced glyoxylate and dicarboxylate metabolism, aminoacyl-tRNA biosynthesis, sulfur metabolism, butanoate metabolism, alanine, aspartate and glutamate metabolism, glutathione metabolism, glycine, serine and threonine metabolism in the root; glycolysis/gluconeogenesis in the stem; and biosynthesis of unsaturated fatty acids, galactose metabolism, cutin, suberine and wax biosynthesis in the leaf. It is important that cadmium inhibited root growth by inhibiting carbohydrate transport from the leaf to the root and promoted leaf growth by the accumulation of carbohydrates in the leaf. Furthermore, cadmium also restrained amino acid metabolism in the root of pumpkin seedlings. These results provide new information about how pumpkin seedlings respond to cadmium stress.

Glucocorticoids are a vital class of endogenous steroid hormones that regulate essential biological processes including growth, development, metabolism, behavior and apoptosis. Most, if not all, of ...these actions are thought to be mediated through the glucocorticoid receptor. The exact mechanisms of how one hormone, via one receptor, modulates such diverse biological functions are largely unknown. However, recent studies from our lab and others have suggested that a contribution for the diversity results from multiple isoforms of the glucocorticoid receptor that result from alternative RNA splicing and translation initiation of the glucocorticoid receptor mRNA. Additionally, each isoform is subject to several post-translational modifications, including phosphorylation, ubiquitination and sumoylation, which have been shown to modulate the receptor protein stability and/or function. Together these data provide potentially diverse mechanisms to establish cell type specific regulation of gene expression by a single transcription factor. Here, we summarize the recent advances and processes that generate these receptor isoforms and these post-translational modifications. We speculate that the composition and proportion of individual isoforms expressed in particular cellular contexts account for the diverse effects of glucocorticoid hormones.
Metabolomics is an implement for testing the toxicity of antibiotics, and provides a comprehensive view of the overall response to stress; however, the connections between metabolites and biologic ...endpoints keep unclear in response to antibiotics. In this study, wheat seeds were exposed to tetracycline for 5 days. The results proved that tetracycline restrained growth, reduced chlorophyl and carotinoid contents and cell permeability, and increased reactive oxygen species (ROS) levels and malondialdehyde (MDA) content. Orthogonal partial least squares (OPLS) was used to analyze the connections between metabolites and biologic endpoints, which discovered that 11 metabolic pathways were significantly affected by tetracycline, and amino acid metabolism could largely apply to root growth and ROS accumulation, while carbohydrate metabolism could have a ruling effect on tetracycline-induced cell permeability. 13 metabolites all played active roles in mediating tetracycline’s effects on root length, root fresh weight and cell permeability but had no significant effects on ROS levels. The majority of metabolites with passive effects on root length, root fresh weight and cell permeability had active effects on ROS levels. These results offer a view about stress reaction of wheat to tetracycline.

The miR395 plays an indispensable role in biochemical processes by regulating their target genes. However, little is known about the roles of miR395 in cucumber fruit expansion and response to ...abiotic stresses. Here, 4 Csa-miR395s and 8 corresponding target genes were identified in the cucumber genome. Csa-miR395s were all located on the same chromosome (Chr 5). Csa-miR395a/b/c and Csa-miR395d were distributed in different branches without a closer genetic relationship. Massive cis-acting elements, including light, phytohormone, and stress response elements, were detected in the promoter regions of Csa-MIR395s, indicating that Csa-miR395s might be involved in complex regulatory networks to control cucumber growth and development and stress response. In addition, Csa-miR395a/b/c shared the same target genes, and Csa-miR395d had its specific target genes. Tissue-specific expression analysis showed that Csa-miR395a/b/c were all expressed in the leaf, root, ovary, and expanded fruit of cucumber and highly expressed in the expanded fruits compared to the ovary, while Csa2G215520 and Csa1G502860 (target genes of Csa-miR395a/b/c) presented a downregulated trend in the expanded fruit compared to the ovary. Meanwhile, the protein co-expression network revealed that these target genes had interactions in sulfur metabolism. These results suggested that Csa-miR395a/b/c targeting Csa2G215520 and Csa1G502860 might promote cucumber fruit expansion by affecting sulfur metabolism. Additionally, Quantitative Real-time PCR analysis validated that Csa-miR395s could be regulated by NaCl stress, and Csa-miR395a/b/c could respond to PEG stress, which further confirmed the reliability of cis-acting elements data. Taken together, our results could be helpful for further exploration of the functions of miR395s in cucumber fruit expansion and response to abiotic stresses.

The number and proportion of female flowers per plant can directly influence the yield and economic benefits of cucurbit crops. Ethephon is often used to induce female flowers in cucurbits. However, ...the mechanism through which it affects floral sex differentiation in pumpkin is unknown. We found that the application of ethephon on shoot apical meristem of pumpkin at seedling stage significantly increased the number of female flowers and expedited the appearance of the first female flower. These effects were further investigated by transcriptome and hormone analyses of plants sprayed with ethephon. A total of 647 differentially expressed genes (DEGs) were identified, among which 522 were upregulated and 125 were downregulated. Gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) analysis indicated that these genes were mainly enriched in plant hormone signal transduction and 1-aminocyclopropane-1-carboxylate oxidase (ACO). The results suggests that ethylene is a trigger for multiple hormone signaling, with approximately 4.2% of the identified DEGs involved in ethylene synthesis and multiple hormone signaling. Moreover, ethephon significantly reduced the levels of jasmonic acid (JA), jasmonoyl-L-isoleucine (JA-ILE), and para-topolin riboside (pTR) but increased the levels of 3-indoleacetamide (IAM). The accumulation of carotenoids in plants is a key nutritional quality in many horticultural crops. Although the structural genes encoding the biosynthetic enzymes are well-characterized, little is ...known regarding photoperiod-mediated carotenoid accumulation in the fruits of some horticultural crops. Herein, we performed physiological and transcriptomic analyses using two cucumber genotypes, SWCC8 (XIS-orange-fleshed and photoperiod-sensitive) and CC3 (white-fleshed and photoperiod-non-sensitive), established under two photoperiod conditions (8L/16D vs. 12L/12D) at four fruit developmental stages. Day-neutral treatments significantly increased fruit β-carotene content by 42.1% compared to short day (SD) treatments in SWCC8 at 40 DAP with no significant changes in CC3. Day-neutral condition elevated sugar levels of fruits compared to short-day treatments. According to GO and KEGG analyses, the predominantly expressed genes were related to photosynthesis, carotenoid biosynthesis, plant hormone signaling, circadian rhythms, and carbohydrates. Consistent with β-carotene accumulation in SWCC8, the day-neutral condition elevated the expression of key carotenoid biosynthesis genes such as PSY1, PDS, ZDS1, LYCB, and CHYB1 during later stages between 30 to 40 days of fruit development. Compared to SWCC8, CC3 showed an expression of DEGs related to carotenoid cleavage and oxidative stresses, signifying reduced β-carotene levels in CC3 cucumber. Further, a WGCNA analysis revealed co-expression between carbohydrate-related genes (pentose-phosphatase synthase, β-glucosidase, and trehalose-6-phosphatase), photoperiod-signaling genes (LHY, APRR7/5, FKF1, PIF3, COP1, GIGANTEA, and CK2) and carotenoid-biosynthetic genes, thus suggesting that a cross-talk mechanism between carbohydrates and light-related genes induces β-carotene accumulation. The results highlighted herein provide a framework for future gene functional analyses and molecular breeding towards enhanced carotenoid accumulation in edible plant organs.
Abstract
Cucumis hystrix Chakr. (2n = 2x = 24) is a wild species that can hybridize with cultivated cucumber (C. sativus L., 2n = 2x = 14), a globally important vegetable crop. However, cucumber ...breeding is hindered by its narrow genetic base. Therefore, introgression from C. hystrix has been anticipated to bring a breakthrough in cucumber improvement. Here, we report the chromosome-scale assembly of C. hystrix genome (289 Mb). Scaffold N50 reached 14.1 Mb. Over 90% of the sequences were anchored onto 12 chromosomes. A total of 23,864 genes were annotated using a hybrid method. Further, we conducted a comprehensive comparative genomic analysis of cucumber, C. hystrix, and melon (C. melo L., 2n = 2x = 24). Whole-genome comparisons revealed that C. hystrix is phylogenetically closer to cucumber than to melon, providing a molecular basis for the success of its hybridization with cucumber. Moreover, expanded gene families of C. hystrix were significantly enriched in “defense response,” and C. hystrix harbored 104 nucleotide-binding site–encoding disease resistance gene analogs. Furthermore, 121 genes were positively selected, and 12 (9.9%) of these were involved in responses to biotic stimuli, which might explain the high disease resistance of C. hystrix. The alignment of whole C. hystrix genome with cucumber genome and self-alignment revealed 45,417 chromosome-specific sequences evenly distributed on C. hystrix chromosomes. Finally, we developed four cucumber–C. hystrix alien addition lines and identified the exact introgressed chromosome using molecular and cytological methods. The assembled C. hystrix genome can serve as a valuable resource for studies on Cucumis evolution and interspecific introgression breeding of cucumber.
Microplastics pollution in farmlands has become a major concern. However, few studies have assessed the effects of microplastics on higher plants. In this study, we investigated the influence of ...polystyrene nanoplastics (PSNPs, 50 mg L−1), with four different particle sizes (100, 300, 500, and 700 nm), on the physiological and biochemical indexes of cucumber leaves. The biomass of cucumber plants significantly decreased after exposure to 300 nm PSNPs. Similarly, the chlorophyll a, chlorophyll b, soluble sugar, carotenoid, and proline content, as well as the fluorescence of cucumber leaves were significantly reduced by 100 nm PSNPs. Malondialdehyde, proline, peroxidase gene expression and enzyme activity, and hydrogen peroxide content significantly increased in cucumber leaves exposed to 700 nm PSNPs. In addition, increasing PSNPs particle size led to decreased relative expression levels and activities of the major antioxidant enzymes superoxide dismutase and catalase, while vitamin C and soluble protein content significantly increased. Overall, our results indicated that PSNPs affect the photosynthetic, antioxidant, and sugar metabolism systems of cucumber leaves, with the latter clearly affecting the total biomass of cucumber plants. The benzene ring resulting from the degradation of PSNPs in cucumber leaves may be the main factor affecting chlorophyll metabolism and sugar metabolism. Our findings provide a scientific basis for the risk assessment of PSNPs exposure in soil-plant systems.
