The SARS-CoV-2 vaccine NVX-CoV2373 is a protein-based vaccine that might circumvent the difficulties in distributing mRNA vaccines to regions with limited access to cold-chain and refrigeration. ...However, the NVX-CoV2373-induced T cell and antibody responses remain poorly understood. In this issue of the JCI, Moderbacher et al. characterized SARS-CoV-2-specific CD4.sup.+ and CD8.sup.+ T cell responses elicited by one or two doses of NVX-CoV2373 in individuals enrolled in a phase I/IIa trial. Substantially increased spike-specific CD4.sup.+ and T follicular helper cells were found after the first or second vaccine dose, with some individuals developing a modest spike-specific CD8.sup.+ T cell response. Correlation analysis revealed an association between spike-specific CD4.sup.+ T cells and neutralizing antibody titers. Notably, preexisting T cell immunity showed negligible effects on NVX-CoV2373-induced T cell responses. These findings indicate that the protein-based vaccine NVX-CoV2373 induces robust T cell immunity capable of recognizing SARS-CoV-2 antigens and supporting humoral immune responses.
Fluorescent calcium indicators are a popular means for observing the spiking activity of large neuronal populations, but extracting the activity of each neuron from raw fluorescence calcium imaging ...data is a nontrivial problem. We present a fast online active set method to solve this sparse non-negative deconvolution problem. Importantly, the algorithm 3progresses through each time series sequentially from beginning to end, thus enabling real-time online estimation of neural activity during the imaging session. Our algorithm is a generalization of the pool adjacent violators algorithm (PAVA) for isotonic regression and inherits its linear-time computational complexity. We gain remarkable increases in processing speed: more than one order of magnitude compared to currently employed state of the art convex solvers relying on interior point methods. Unlike these approaches, our method can exploit warm starts; therefore optimizing model hyperparameters only requires a handful of passes through the data. A minor modification can further improve the quality of activity inference by imposing a constraint on the minimum spike size. The algorithm enables real-time simultaneous deconvolution of O(105) traces of whole-brain larval zebrafish imaging data on a laptop.

Titanium oxide (TiO2) has been commonly used as an electron transport layer (ETL) of regular‐structure perovskite solar cells (PSCs), and so far the reported PSC devices with power conversion ...efficiencies (PCEs) over 21% are mostly based on mesoporous structures containing an indispensable mesoporous TiO2 layer. However, a high temperature annealing (over 450 °C) treatment is mandatory, which is incompatible with low‐cost fabrication and flexible devices. Herein, a facile one‐step, low‐temperature, nonhydrolytic approach to in situ synthesizing amino‐functionalized TiO2 nanoparticles (abbreviated as NH2‐TiO2 NPs) is developed by chemical bonding of amino (‐NH2) groups, via TiN bonds, onto the surface of TiO2 NPs. NH2‐TiO2 NPs are then incorporated as an efficient ETL in n‐i‐p planar heterojunction (PHJ) PSCs, affording PCE over 21%. Cs0.05FA0.83MA0.12PbI2.55Br0.45 (abbreviated as CsFAMA) PHJ PSC devices based on NH2‐TiO2 ETL exhibit the best PCE of 21.33%, which is significantly higher than that of the devices based on the pristine TiO2 ETL (19.82%) and is close to the record PCE for devices with similar structures and fabrication procedures. Besides, due to the passivation of the surface trap states of perovskite film, the hysteresis of current–voltage response is significantly suppressed, and the ambient stability of devices is improved upon amino functionalization.
Amino‐functionalized TiO2 nanoparticles are synthesized in situ by a facile onestep, low‐temperature, nonhydrolytic approach, and are applied as the electrontransport layer of regular‐structure planar heterojunction perovskite solar cells, offering a dramatic performance increase due to the passivation of the surface trap states of the perovskite film.
Hepatocellular carcinoma (HCC) is one of the world's most lethal malignant tumors with a poor prognosis. Growing evidence has been demonstrating that immune-related long non-coding RNAs (lncRNAs) are ...relevant to the tumor microenvironment (TME) and can help assess the effects of immunotherapy and evaluate one's prognosis. This study aims to identify an immune-related lncRNA signature for the prospective assessment of the immunotherapy and prognosis in HCC.
We downloaded HCC RNA-seq data and clinical information from The Cancer Genome Atlas (TCGA) project database. We first used ESTIMATE to evaluate the TME. Then, we conducted a cox regression analysis to construct a prognostic signature and the riskScore. We then applied the univariate Cox regression, multivariate Cox regression, principal components analysis (PCA), receiver operating characteristic (ROC) curve, and stratification analyses to confirm our previous assessments. Afterward, we employed a gene set enrichment analysis (GSEA) to explore the biological processes and pathways. Besides, we used CIBERSORT to estimate the abundance of tumor-infiltrating immune cells (TIICs). Furthermore, we investigated the relationship between the immune-related lncRNA signature and immune checkpoint genes. Finally, we used the quantitative real-time polymerase chain reaction (qRT-PCR) assays to demonstrate the expression of the six lncRNAs.
We identified six immune-related lncRNAs - MSC-AS1, AC145207.5, SNHG3, AL365203.2, AL031985.3, NRAV - which show the ability to stratify patients into high-risk and low-risk groups with significantly different survival rates. The univariate Cox regression, multivariate Cox regression, ROC, and stratification analyses confirmed that the immune-related six-lncRNA signature was a novel independent prognostic factor in HCC patients. The high-risk group and low-risk group illustrated contrasting distributions in PCA. The GSEA suggested that the immune-related six-lncRNA signature was involved in the immune-related biological processes and pathways. Besides, the immune-related six-lncRNA signature was associated with the infiltration of immune cells. Furthermore, it was linked with the expression of critical immune genes and could predict immunotherapy's clinical response. Finally, the qRT-PCR demonstrated that the six lncRNAs were significantly differentially expressed in HCC cell lines and normal hepatic cell lines.
In summary, we identified an immune-related six-lncRNA signature that can predict the outcomes, immune cell infiltration, and immunotherapy response in patients with hepatocellular carcinoma.
Activity in striatal direct- and indirect-pathway spiny projection neurons (SPNs) is critical for proper movement. However, little is known about the spatiotemporal organization of this activity. We ...investigated the spatiotemporal organization of SPN ensemble activity in mice during self-paced, natural movements using microendoscopic imaging. Activity in both pathways showed predominantly local but also some long-range correlations. Using a novel approach to cluster and quantify behaviors based on continuous accelerometer and video data, we found that SPN ensembles active during specific actions were spatially closer and more correlated overall. Furthermore, similarity between different actions corresponded to the similarity between SPN ensemble patterns, irrespective of movement speed. Consistently, the accuracy of decoding behavior from SPN ensemble patterns was directly related to the dissimilarity between behavioral clusters. These results identify a predominantly local, but not spatially compact, organization of direct- and indirect-pathway SPN activity that maps action space independently of movement speed.

The interfaces between perovskite layer and electrodes play a crucial role on efficient charge transport and extraction in perovskite solar cells (PSCs). Herein, for the first time we applied a ...low-cost nonconjugated polymer poly(vinylpyrrolidone) (PVP) as a new interlayer between PCBM electron transport layer (ETL) and Ag cathode for high-performance inverted planar heterojunction perovskite solar cells (iPSCs), leading to a dramatic efficiency enhancement. The CH3NH3PbI3–x Cl x -based iPSC device incorporating the PVP interlayer exhibited a power conversion efficiency (PCE) of 12.55%, which is enhanced by ∼15.9% relative to that of the control device without PVP interlayer (10.83%). The mechanistic investigations based on morphological, optical, and impedance spectroscopic characterizations reveal that incorporation of PVP interlayer promotes electron transport across the CH3NH3PbI3–x Cl x perovskite/Ag interface via PCBM ETL. Besides, PVP incorporation induces the formation of a dipole layer, which may enhance the built-in potential across the device, conjunctly promoting electron transport from PCBM to Ag cathode and consequently leading to significantly improved fill factor (FF) from 58.98 to 66.13%.
Metformin has garnered more interest as a chemo-preventive agent given the increased liver cancer risk in diabetic patients. This work was undertaken to better understand the effect of metformin use ...on liver cancer risk in diabetic patients.A comprehensive literature search was performed in PubMed, Embase, BIOSIS Previews, Web of Science, and Cochrane Library through July 30, 2016. Meta-analyses were performed using Stata version 12.0, with odds ratio (ORs) and 95% confidence intervals (CIs) as effect measures.Twenty-three studies were included. Meta-analysis of 19 studies involving 550,882 diabetic subjects suggested that metformin use reduced the ratio of liver cancer by 48% (OR = 0.52; 95% CI, 0.40-0.68) compared with nonusers. The protective effect was validated in all the exploratory subgroup analyses, except that pooled result of post hoc analyses of 2 randomized controlled trials found no significant difference between subjects with metformin and those without, with OR being 0.84 (95% CI, 0.10-6.83). After adjusting for hepatitis B/C virus infection, cirrhosis, obesity, behavioral factors, and time-related bias, the association was stable, pooled OR ranged from 0.42 to 0.75.A protective effect for liver cancer was found in diabetic metformin users. However, more randomized clinical evidence is still needed to verify the results.
Advances in fluorescence microscopy enable monitoring larger brain areas in-vivo with finer time resolution. The resulting data rates require reproducible analysis pipelines that are reliable, fully ...automated, and scalable to datasets generated over the course of months. We present CaImAn, an open-source library for calcium imaging data analysis. CaImAn provides automatic and scalable methods to address problems common to pre-processing, including motion correction, neural activity identification, and registration across different sessions of data collection. It does this while requiring minimal user intervention, with good scalability on computers ranging from laptops to high-performance computing clusters. CaImAn is suitable for two-photon and one-photon imaging, and also enables real-time analysis on streaming data. To benchmark the performance of CaImAn we collected and combined a corpus of manual annotations from multiple labelers on nine mouse two-photon datasets. We demonstrate that CaImAn achieves near-human performance in detecting locations of active neurons.

Kesterite-structured quaternary semiconductor Cu2ZnSnS4 (CZTS) has been commonly used as light absorber in thin film solar cells on the basis of its optimal bandgap of 1.5 eV, high absorption ...coefficient, and earth-abundant elemental constituents. Herein we applied CZTS nanoparticles as a novel inorganic hole transporting material (HTM) for organo-lead halide perovskite solar cells (PSCs) for the first time, achieving a power conversion efficiency (PCE) of 12.75%, which is the highest PCE for PSCs with Cu-based inorganic HTMs reported up to now, and quite comparable to that obtained for PSCs based on commonly used organic HTM such as 2,2′,7,7′-tetrakis(N,N-di-p-methoxyphenylamine)-9,9′-spirobifluorene (spiro-MeOTAD). The size of CZTS nanoparticles and its incorporation condition as HTM were optimized, and the effects of CZTS HTM on the optical absorption, crystallinity, morphology of the perovskite film and the interface between the perovskite layer and the Au electrode were investigated and compared with the case of spiro-MeOTAD HTM, revealing the role of CZTS in efficient hole transporting from the perovskite layer to the top Au electrode as confirmed by the prohibited charge recombination at the perovskite/Au electrode interface. On the basis of the effectiveness of CZTS as a low-cost HTM competitive to spiro-MeOTAD in PSCs, we demonstrate the new role of CZTS in photovoltaics as a hole conductor beyond the traditional light absorber.
