Long-term measurements of aerosol optical depths (AODs) at 440 nm and Aangstrom exponents (AE) between 440 and 870 nm made for CARSNET were compiled into a climatology of aerosol optical properties ...for China. Quality-assured monthly mean AODs are presented for 50 sites representing remote, rural, and urban areas. AODs were 0.14, 0.34, 0.42, 0.54, and 0.74 at remote stations, rural/desert regions, the Loess Plateau, central and eastern China, and urban sites, respectively, and the corresponding AE values were 0.97, 0.55, 0.82, 1.19, and 1.05. AODs increased from north to south, with low values (< 0.20) over the Tibetan Plateau and northwestern China and high AODs (> 0.60) in central and eastern China where industrial emissions and anthropogenic activities were likely sources. AODs were 0.20-0.40 in semi-arid and arid regions and some background areas in northern and northeastern China. AEs were > 1.20 over the southern reaches of the Yangtze River and at clean sites in northeastern China. In the northwestern deserts and industrial parts of northeast China, AEs were lower (< 0.80) compared with central and eastern regions. Dust events in spring, hygroscopic particle growth during summer, and biomass burning contribute the high AODs, especially in northern and eastern China. The AODs show decreasing trends from 2006 to 2009 but increased ~ 0.03 per year from 2009 to 2013.
With the aim of gathering temporal trends on bacterial epidemiology and resistance from multiple laboratories in China, the CHINET surveillance system was organized in 2005. Antimicrobial ...susceptibility testing was carried out according to a unified protocol using the Kirby-Bauer method or automated systems. Results were analyzed according to Clinical and Laboratory Standards Institute (CLSI) 2014 definitions. Between 2005 and 2014, the number of bacterial isolates ranged between 22 774 and 84 572 annually. Rates of extended-spectrum β-lactamase production among Escherichia coli isolates were stable, between 51.7 and 55.8%. Resistance of E. coli and Klebsiella pneumoniae to amikacin, ciprofloxacin, piperacillin/tazobactam and cefoperazone/sulbactam decreased with time. Carbapenem resistance among K. pneumoniae isolates increased from 2.4 to 13.4%. Resistance of Pseudomonas aeruginosa strains against all of antimicrobial agents tested including imipenem and meropenem decreased with time. On the contrary, resistance of Acinetobacter baumannii strains to carbapenems increased from 31 to 66.7%. A marked decrease of methicillin resistance from 69% in 2005 to 44.6% in 2014 was observed for Staphylococcus aureus. Carbapenem resistance rates in K. pneumoniae and A. baumannii in China are high. Our results indicate the importance of bacterial surveillance studies.
A regulator of the protein phosphatase 2A (PP2A), α4, has been implicated in a variety of functions that regulate many cellular processes. To explore the role of α4 in human cell transformation and ...tumorigenesis, we show that α4 is highly expressed in human cells transformed by chemical carcinogens including benzo(a)pyrene, aflatoxin B(1), N-methyl-N'-nitro-N-nitrosoguanidine, nickel sulfate and in several hepatic and lung cancer cell lines. In addition, overexpression of α4 was detected in 87.5% (74/80) of primary hepatocellular carcinomas, 84.0% (21/25) of primary lung cancers and 81.8% (9/11) of primary breast cancers, indicating that α4 is ubiquitously highly expressed in human cancer. Functional studies revealed that elevated α4 expression results in an increase in cell proliferation, promotion of cell survival and decreased PP2A-attributable activity. Importantly, ectopic expression of α4 permits non-transformed human embryonic kidney cells (HEKTER) and L02R cells to form tumors in immunodeficient mice. Furthermore, we show that the highly expressed α4 in transformed cells or human tumors is not regulated by DNA hypomethylation. A microRNA, miR-34b, that suppresses the expression of α4 through specific binding to the 3'-untranslated region of α4 is downregulated in transformed or human lung tumors. Taken together, these observations identify that α4 possesses an oncogenic function. Reduction of PP2A activity due to an enhanced α4-PP2A interaction contributes directly to chemical carcinogen-induced tumorigenesis.
Cell-free DNA (cf-DNA)-based liquid biopsy is emerging as a revolutionary new method in individualized cancer treatment and prognosis monitoring, although detecting early-stage cancers using cf-DNA ...remains challenging, partially because of the undefined biological background of cf-DNA.
We investigated somatic mutations in the cf-DNA of 259 cancer-free individuals with a median age of 47 years using an endogenous barcoding duplex method with an ultralow base error rate (2 × 10−7) and compared the variant allele frequencies (VAFs) of these mutations between the cf-DNA and the corresponding blood cell DNA.
Sixty percent (155/259) of the samples showed at least one nonsynonymous mutation on either of two similar target panels covering 508 and 559 cancer-related genes. For individuals older than 50 years of age, the positive rate increased to 76%. Most cf-DNA mutations were also present at similar VAFs in the paired blood cell DNA. The most frequently mutated genes were driver genes of hematologic malignancies, including DNMT3A, TET2, AXSL1, and JAK2. However, the other 58.4% (192/329) of the mutations were likely ‘passenger mutations’ of clonal hematopoiesis, including mutations in NOTCH2, FAT3, EXT2, ERBB4, and ARID2, which are driver genes of solid tumors.
Hematopoietic clone-derived mutations, including ‘driver mutations’ and ‘passenger mutations’, are prevalent in the cf-DNA of both healthy individuals and cancer patients and may be a potential source of false positives in the liquid biopsy. Our results also suggest the ineffectiveness for distinguishing clonal hematopoietic mutations of low VAF (≤0.1%) from tumor-derived mutations using conventional next-generation sequencing of blood cell DNA. However, an error correction model with an ultralow error rate and high coverage depth is required for blood cell DNA sequencing, which is difficult and costly to achieve with current technologies.
The CDEX-1 experiment conducted a search of low-mass (< 10GeV/c super(2)) weakly interacting massive particles dark matter at the China Jinping Underground Laboratory using a p-type point-contact ...germanium detector with a fiducial mass of 915 g at a physics analysis threshold of 475 eVee. We report the hardware setup, detector characterization, data acquisition, and analysis procedures of this experiment. No excess of unidentified events is observed after the subtraction of the known background. Using 335.6 kg-days of data, exclusion constraints on the weakly interacting massive particle-nucleon spin-independent and spin-dependent couplings are derived.
► Long-term fertilization increased POM fraction, but did not change silt and clay sized fraction. ► A logarithmic relation was found between C input and C sequestration. ► C sequestration efficiency ...was greater in paddy soil than in upland soil. ► The C stabilization mechanisms are different in the two agroecosystems.
Soil organic carbon (SOC) stock can be improved through the return of crop residues. However, the efficiency of C sequestered in soil (i.e., ΔSOC/ΔC input) might differ among crop systems. In this paper, we investigated the C input and SOC stabilization in paddy soil and upland soil under different long-term fertilization practices. Our objectives were to determine (i) the response of SOC stock to C input under different fertilization practices, and (ii) C sequestration efficiency in the two contrasting agroecosystems. The long-term fertilization experiment in paddy soil started in 1981, while the adjacent upland soil experiment commenced in 1986. Each experiment consists of 9 treatments: CK (no fertilization), N, P, K, NP, NK, NPK, 2NPK (double dose), and NPKOM (NPK plus organic manure). Physical SOC fractions (cPOM, silt+clay_f, fPOM, iPOM_m, silt+clay_m) were isolated by sieving, dispersion, and density flotation. Fertilization increased crop yield and C input, but it did not change the quality of SOC as revealed by CPMAS-13C NMR spectra. During the period of the experiment, SOC stock was improved by 6.7–15.3Mgha−1 in paddy soil for all fertilization practices, while in upland soil the CK, N, P, K, and NP fertilizations reduced the SOC by 1.2–3.8MgCha−1 and the other four fertilizations increased it by 0.5–7.4MgCha−1. The change in SOC was mainly ascribed to the POM fraction, whereas the two silt+clay sized fractions were independent of fertilization practice except the NPKOM treatment. At a given C input, the C sequestration efficiency was greater in paddy soil than in upland soil, which may be attributed to lower microbial activity but greater chemical (i.e., oxalate-soluble Fe) and physical stabilizations (i.e., soil structure) in paddy field. Our results indicate that paddy soil may sequester more SOC, with higher efficiency, than upland soil does.
We report the first results of a light weakly interacting massive particles (WIMPs) search from the CDEX-10 experiment with a 10 kg germanium detector array immersed in liquid nitrogen at the China ...Jinping Underground Laboratory with a physics data size of 102.8 kg day. At an analysis threshold of 160 eVee, improved limits of 8×10^{-42} and 3×10^{-36} cm^{2} at a 90% confidence level on spin-independent and spin-dependent WIMP-nucleon cross sections, respectively, at a WIMP mass (m_{χ}) of 5 GeV/c^{2} are achieved. The lower reach of m_{χ} is extended to 2 GeV/c^{2}.
In March 2020, widespread anthracnose was observed on soybean (Glycine max) in southeastern Jiangsu (Nantong municipality; 120.53° E, 31.58° N) in China. Plants exhibited irregular brown necrotic ...lesions in stem and leaves, and pronounced wilting. The symptoms were detected in one soybean field, 0.42 ha, surrounded by healthy wheat fields. Approximately 65% of the soybean plants showed the disease symptoms, and crop yield was reduced by 28-35% with respect the yield achieved in previous years, when no symptoms were observed. The symptoms were consistent with those previously reported for anthracnose on soybean caused by Colletotrichum chlorophyti, C. cliviae and C. gloeosporioides (Barbieri et al. 2017; Mahmodi et al. 2013; Yang et al. 2012). Diseased, 3-week old plants were collected. Small pieces, approximately 1 cm2 in size, of symptomatic tissue were surface sterilized in 1.5% NaOCl for 1 min, and washed twice with sterile ddH2O. The pathogen was isolated and cultured on potato dextrose agar (Song et al. 2020), containing chloramphenicol (50 µg/mL), under darkness at 28 °C for 3 days. Sequence of internal transcribed spacer (ITS), actin (ACT), β-tubulin (TUB2) and glyceraldehyde 3-phosphate dehydrogenase (GAP/span>DH) genes was performed as reported by Yang et al. (2015). Sequences were submitted to GenBank under accession numbers MT361074 (ITS) and MT415548-MT415550 (ACT, TUB2 and GAPDH). Blast search revealed that the amplified sequences had 100% (ITS; C. brevisporum TCHD, MH883805), 97.66% (ACT; C. brevisporum S38, KY986905), 99.06% (TUB2; C. brevisporum PF-2, KY705061) and 100% (GAPDH; C. brevisporum LJTJ27, KP823797) matches to multiple C. brevisporum strains, whereas all reported C. chlorophyti, C. cliviae and C. gloeosporioides strains showed no similarity to at least 2 of the studied genes. Molecular phylogenetic tree constructed using MEGA7 confirmed the identity of the pathogen. ACT and ITS sequences were blasted separately in Muscle (https://www.ebi.ac.uk/Tools/msa/muscle/) and then combined together to make the phylogenetic tree. The evolutionary history was inferred by using the Maximum Likelihood method based on the Tamura 3-parameter model, and the tree with the highest log likelihood (-1749.2186) is shown in Figure 1. The Colletotrichum strains previously found causing anthracnoseon soybean, and other relevant strains used in taxonomic analyses were included in the phylogenetic tree. Microscope observations showed the presence of 15-µm-long cylindrical conidia and septate mycelium, and agree with those reported for the morphology of C. brevisporum by Damm et al. (2019). To confirm pathogenicity, the mycelia from a 2 day-old culture on PDA was collected and suspended in sterile ddH2O (≈ 106 cells/mL) to prepare the inoculum. The pathogen was sprayed-inoculated on stem and leaves of healthy soybean plants. In control plants, sterile ddH2O was used. Inoculated plants were maintained in growth chamber at 28 °C and 50% relative humidity. Typical anthracnose symptoms were obsered 20 days after inoculation (Figure 2). C. brevisporum was reported to produce anthracnose on pumpkin, papaya, mulberry, coffee, passion fruit and pepper in China (Liu et al. 2017; Liu et al. 2019; Xue et al. 2019). Here, we report for the first time C. brevisporum causing anthracnose on soybean, an economically-relevant crop in China.
Magnetization reversals through the formation of a vortex state and the rotation of an onion state are two processes with comparable probabilities for symmetric magnetic nanorings with a radius of ...about 50 nanometers. This magnetic bistability is the manifestation of the competition between the exchange energy and the magnetostatic energy in nanomagnets. The relative probability of the two processes in symmetric nanorings is dictated by the ring geometry and cannot be altered after fabrication. In this work, we report a novel type of nanorings--asymmetric nanorings. By tuning the asymmetry, we can control the fraction of the vortex formation process from about 40% to nearly 100% by utilizing the direction of the external magnetic field. The observed results have been accounted for by the dependence of the domain-wall energy on the local cross-section area for which we have provided theoretical calculations.
