The probiotic strains Lactobacillus brevis CCMA1284 and Lactobacillus plantarum CCMA0359 were microencapsulated by spray drying using different matrices – whey powder (W), whey powder with ...inulin (WI) and whey powder with maltodextrin (WM). Viability of the microencapsulated strains in acid and bile juices and during 90 days of storage (seven and 25 °C) was evaluated. The two strains exhibited high encapsulation efficiency (> 86%) by spray drying. The different matrices maintained L. plantarum viability above six log CFU g−1 at 7 °C for 90 days, whereas similar results for L. brevis were observed only for W. The use of inulin as matrix of encapsulation did not enhance bacterial viability in the evaluated conditions. In general, the use of W and WM as matrices was effective for L. plantarum viability. However, only W was effective for L. brevis in the evaluated conditions. The spray drying technique was successfully adopted for the encapsulation of L. plantarum CCMA0359 and L. brevis CCMA1284 strains.

CCMA 0243 showed better results of γ-decalactone production. This yeast species can be considered an alternative producer of γ-decalactone in biotechnological processes.
The total antioxidant capacity (TAC) and total phenolic compounds (TPC) of cocoa beans and chocolate produced from spontaneous and inoculated fermentations of different cocoa varieties were ...evaluated. Fourier transform infrared spectroscopy (FTIR), as well as conventional methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), was used to determine TAC and TPC. Chocolate showed higher (p<0.05) TPC (47.17–57.16mgGAE/g) and TAC (1.66–2.33mMTE/g and 8.86–11.35mMTE/g as measured by DPPH and ABTS, respectively) than cocoa beans (6.30–26.05mgGAE/g, 0.24–1.17mMTE/g and 1.29–4.83mMTE/g for TPC, DPPH and ABTS, respectively). Partial least square (PLS) model for infrared data showed a good calibration coefficient (R2cal>0.94), indicating that the FTIR technique represents a fast and reliable tool to evaluate TPC and TAC in cocoa beans and chocolate.

Microencapsulation appears to be a promising technique for protecting probiotics against conditions found in gastrointestinal digestion. Therefore, the survival of free and microencapsulated ...Lactiplantibacillus plantarum CCMA 0359, a potential probiotic strain, into cream cheese during storage and simulated gastrointestinal digestion was evaluated. The influence of the microencapsulated probiotic on the physicochemical and sensory properties of the cream cheese was also evaluated. Microencapsulation significantly enhanced the protection of cells against simulated gastrointestinal digestion, regardless of the storage time, and the cell count was slightly higher (8.31 log cfu g−1) compared with the free cell count (8.07 log cfu g−1). The addition of the probiotic (notably microencapsulated cells) increased the acidity and decreased the pH of cream cheese during storage and did not affect consumer acceptance. Findings indicate cream cheese as an alternative vehicle for the potential probiotic L. plantarum CCMA 0359, without affecting the acceptance of the product.

Svrha je ovoga rada bila ispitati različite uvjete ...proizvodnje γ-dekalaktona s pomoću tropskih sojeva kvasca Yarrowia lipolytica CCMA 0242 i Lindnera saturnus CCMA 0243. Rast kvasca Y. lipolytica CCMA 0242 i proizvodnja γ-dekalaktona bili su izraženiji u podlozi s ricinusovim uljem nego s glicerolom. Koncentracija γ-dekalaktona dobivena šaržnom fermentacijom nije se bitno razlikovala od one dobivene fermentacijom s pritokom supstrata. S pomoću kvasca L. saturnus CCMA 0243 proizvedeno je više γ-dekalaktona pri pH=5, a s pomoću soja Y. lipolytica CCMA 0242 pri pH=6. Soj L. saturnus CCMA 0243 proizveo je više γ-dekalaktona od soja Y. lipolytica CCMA 0242 pri istim uvjetima fermentacije. Sirovi glicerol nije dobar supstrat za proizvodnju γ-dekalaktona s pomoću soja Y. lipolytica CCMA 0242, jer su bolji rezultati postignuti u podlozi s 30 % ricinusovog ulja. Proizvodnja je γ-dekalaktona bila bolja s pomoću soja L. saturnus CCMA 0243, pa se taj kvasac može smatrati alternativnim proizvođačem γ-dekalaktona u biotehnološkim procesima.
Human pluripotent stem cells (PSC) have been used for disease modelling, after differentiation into the desired cell type. Electrophysiologic properties of cardiomyocytes derived from pluripotent ...stem cells are extensively used to model cardiac arrhythmias, in cardiomyopathies and channelopathies. This requires strict control of the multiple variables that can influence the electrical properties of these cells. In this article, we report the action potential variability of 780 cardiomyocytes derived from pluripotent stem cells obtained from six healthy donors. We analyze the overall distribution of action potential (AP) data, the distribution of action potential data per cell line, per differentiation protocol and batch. This analysis indicates that even using the same cell line and differentiation protocol, the differentiation batch still affects the results. This variability has important implications in modeling arrhythmias and imputing pathogenicity to variants encountered in patients with arrhythmic diseases. We conclude that even when using isogenic cell lines to ascertain pathogenicity to variants associated to arrythmias one should use cardiomyocytes derived from pluripotent stem cells using the same differentiation protocol and batch and pace the cells or use only cells that have very similar spontaneous beat rates. Otherwise, one may find phenotypic variability that is not attributable to pathogenic variants.
