Comme tous les ciliés, P. tetraurelia réarrange son génome à chaque génération sexuelle pendant le développement de son macronoyau somatique ¿ partir du micronoyau germinal. Les réarrangements ...incluent l’excision précise de courtes séquences dérivant de transposons et appelés IES (Internal Eliminated Sequences) dont la majorité sont intragéniques. L’excision d’une fraction d’entre elles dépend de petits ARN maternels (appelés scnARN) qui sont produits à partir de tout le génome germinal pendant la méiose. Ce mécanisme pose un problème lors d’une conjugaison entre deux souches présentant des polymorphismes d’insertion : une cellule sera théoriquement incapable d’exciser une IES portée par l’allèle paternel reçu si cette IES est absente de l’allèle maternel ou si la séquence est trop divergente. Mes résultats montrent cependant que les allèles paternels divergents sont correctement excisés en utilisant les scnARN produit par la cellule paternelle. Dans le cas d’un polymorphisme absence/présence, l’IES que j’ai étudié est excisée chez 70 % des hétérozygotes F1, également via les scnARN paternels. Nous avons exploré deux hypothèses pour expliquer comment ils pouvaient agir. Il pourrait s’agir d’une programmation précoce des noyaux gamétiques ou alors d’un échange cytoplasmique des scnARN. Finalement, j’ai montré qu’un défaut de scnARN maternels n’est pas une cause possible de dysgénésie hybride. Cependant, 30 % des hétérozygotes F1 présentent une rétention variable de l’IES étudié via un mécanisme inconnu. Si cela est généralisable à toutes les IES homozygotes, alors ce mécanisme aurait un effet délétère sérieux sur les F1 et pourrait contribuer à l’isolement reproductif.
Like all ciliates, P. tetraurelia entirely rearranges its genome during development of the somatic macronucleus from the germline micronucleus, in each sexual generation. Rearrangements include the precise excision of IESs (Internal Eliminated Sequences), single-copy intervening sequences likely derived from transposon insertions. At least for a fraction of IESs, correct excision, which is required to reconstitute functional genes in the macronucleus, is thought to depend on their recognition by Piwi-bound small RNAs (called scnRNAs) produced from the maternal germline genome during meiosis. This raises a problem during conjugation between strains presenting insertion polymorphisms: a cell will be theoretically unable to excise an IES from the incoming (paternal) allele if that IES is absent from the maternal allele, or if its sequence is too divergent. Our results, however, indicate that divergent paternal alleles are correctly rearranged, using scnRNAs produced by the paternal cell. In the case of an absence/presence polymorphism, the IES we studied is excised in 70% of heterozygotes, also using paternal scnRNAs. We explored two hypotheses to explain how they can act. It could be either an early programming of the gametic nuclei or through cytoplasmic exchange of scnRNAs. My results seem to favor the latter. Overall, I showed that the lack of maternal scnRNAs is not a possible cause of hybrid dysgenesis. However, 30% of heterozygous F1 display a variable retention of the IES through an unknown mechanism. If this is true for all hemizygous IESs then it will have a strong deleterious effect on hybrid F1s and may contribute to reproductive isolation.
Le cil primaire, véritable organite sensoriel cellulaire est présent à la surface de la plupart des cellules de mammifères en quiescence. Truffé de récepteurs à sa membrane, le cil capte les signaux ...mécaniques et chimiques, jouant ainsi un rôle clé dans de nombreux processus développementaux et physiologiques. Un défaut de structure et/ou de fonction du cil est à l'origine de cancérogénèse et de pathologies humaines appelées ciliopathies.Le cil primaire est ancré à la membrane plasmique grâce au corps basal, structure dérivée du centriole père et connectée aux trois réseaux du cytosquelette. La formation du cil primaire nécessite une succession d'étapes cytoplasmiques hautement régulées. Elle débute par la maturation du centriole père en corps basal. Cette étape nécessite le recrutement de protéines spécifiques au centriole père permettant l'association avec une vésicule ciliaire à l'extrémité distale du centriole père. Ce complexe migre et vient s'ancrer à la membrane apicale déclenchant la nucléation de microtubules pour la formation de la partie externe du cil, ou axonème. En parallèle, la ciliogénèse nécessite un remodelage important du cytosquelette d'actine ainsi qu'un trafic de vésicules orienté vers la base du cil. Si la plupart des étapes sont bien caractérisées, celle concernant la migration du corps basal ainsi que la contribution du cytosquelette reste mal comprise.Afin de mieux appréhender les mécanismes impliqués dans la migration du corps basal lors de la ciliogénèse, nous avons développé un système expérimental basé sur l'utilisation de micro-patrons adhésifs recouverts de fibronectine. Cette technologie comporte de nombreux avantages. Elle permet le contrôle de l'étalement de la cellule inhérent à la surface imposée par la matrice extracellulaire régulant ainsi l'organisation du cytosquelette ainsi que le positionnement des organelles subcellulaires. Par ailleurs, le volume cellulaire induit par le confinement spatial facilite l'observation de la position du centrosome en z au cours du temps, indispensable pour l'étude de chaque étape de la ciliogénèse cytoplasmique.Dans un premier temps, nous avons démontré que la forme et l'architecture du cytosquelette d'actine qui en dépend sont des régulateurs majeurs du processus ciliogénique. Les cellules confinées spatialement et sevrées 24h sur des petits disques développent un réseau branché au niveau de leur surface apicale nécessaire à la croissance du cil primaire. A l'inverse, les cellules étalées sur des grands disques sont beaucoup plus contractées. Elles développent d'importantes fibres de stress sur leur surface ventrale. Le centrosome reste sous le noyau et le niveau de contraction empêche l'assemblage du cil. Le niveau de contractilité module donc la formation du réseau d'actine apicale qui contrôle en retour le mouvement du corps basal et l'élongation du cil.Dans un deuxième temps, nous avons étudié la dynamique du cytosquelette d'actine et de microtubules durant l'étape de migration du corps basal c'est à dire juste après la privation de sérum. Nos résultats indiquent que la migration nécessite une augmentation transitoire de la stabilité des microtubules concomitante avec une augmentation de la contractilité des filaments d'actine. Un crible basé sur l'ARN interférence nous a permis d'identifier des gènes impliqués dans le processus de migration dont CEP164, contribuant à l'ancrage du centriole père à la vésicule ciliaire. Les cellules déficientes en CEP164 montrent un défaut de réorganisation du cytosquelette expliquant l'inhibition du transport du corps basal vers la membrane apicale.L'ensemble des résultats nous permet d'avancer dans la compréhension des conditions requises pour le mouvement du corps basal vers la membrane apicale. Celui-ci nécessite à la fois un remodelage significatif du cytosquelette en constant dialogue et en interaction avec certains composants ciliaires nécessaires à la formation du cil primaire.
The primary cilium is a sensory organelle present on the surface of most quiescent cells. It possesses numerous receptors on its surface and is responsible for transducing biochemical and mechanical signals to the interior of the cell and playsimportant roles during development and in homeostasis. Defects in primary cilium assembly are the underlying cause of a group of pleiotropic diseases referred to as ciliopathies.The primary cilium is anchored to the plasma membrane through the basal body which is derived from the mother centriole and is connected to three networks of the cytoskeleton. Primary cilium formation is a highly regulated and multi-step process that begins with the maturation of the centriole mother into basal body in the cytoplasm of the cell. One of the first steps of primary cilium assembly is the recruitment of specific proteins to the mother centriole to initiate the formation of a ciliary vesicle at the distal end of the mother centriole. Once formed, the mother centriole migrates to and is anchored to the apical membrane, triggering the elongation of microtubules from the distal end of the mother centriole to form the outer part of primary cilium, or axoneme. In order for this to occur, significant remodeling of the actin cytoskeleton and directe-trafficking of vesicles to the base of the cilium is required. While much progress has been made in characterizing the initial steps of primary ciliogenesis, how the basal body migrates to the plasma membrane is not fully understood.To gain a better understanding of the mechanisms involved in the migration of basal body during ciliogenesis, we developed an experimental system based on the use of adhesive micro-patterns coated with fibronectin. This technology has many advantages. It enables the control of the cell spreading which is imposed by the size of the adhesive area and, in turn, the regulation of cytoskeletal organization and the positioning of subcellular organelles. Furthermore, this technique enables the cell volume induced by the spatial confinement, to be controlled, facilitating the observation and measurement of the centrosome's position in z throughout the primary ciliogenesis process.First, we demonstrated that the shape and architecture of the actin cytoskeleton are major regulators of primary ciliogenesis. Cells spatially confined and starved for 24h on small discoidal micropattern develop an apical web like actin network necessary for the primary cilium growth. In contrast, cells plated on large discs are much more contracted and they develop significant stress fibers on their ventral surface. In this situation, the centrosome remains below the nucleus and the level of contraction prevents the assembly of a primary cilium. The level of contractility therefore modulates the formation of apical actin network that in turn controls the movement of the basal body and the cilium elongation.Secondly, we studied actin cytoskeleton and microtubule reorganization during the basal body migration step that occured just after serum starvation. Our results indicate that migration requires a transient increase in the stability of microtubules, concomitant with an increase in contractility of actin filaments. By RNA interference screening, we have identified genes involved in the migration process including CEP164, which has previously been shown to participate in the anchoring of the ciliary vesicle to the mother centriole. CEP164-deficient cells were found to have defects in cytoskeletal reorganization thereby explaining why basal body transport to the plasma membrane was blocked in these cells.Altogether, these results enable our understanding of how basal body movement to the apical membrane is driven. This requires both significant remodeling and crosstalk between the actin and microtubule cytoskeleton and interaction with ciliary components necessary for the formation of a primary cilium.
Les forces mécaniques dirigeant la valvulogénèse sont mal connues. Chez le poisson zèbre, le flux induit l’expression du facteur de transcription klf2a de façon endothéliale spécifique afin d’initier ...ce processus. Nous nous sommes intéressés aux mécanismes moléculaires activés par le flux et aboutissant à la formation des valves cardiaques. En altérant et modifiant le pattern fluidique, nous avons observé le rôle des forces engendrées par le flux pour le contrôle de l’expression de klf2a ainsi que du nombre de cellules endocardiques. Nous avons ensuite regardé les divers mécanosenseurs pouvant intervenir lors de ce processus. Ainsi nous avons mis à jour la présence de cils dans l’endocarde et fait la relation entre les canaux membranaires TRPP2 et TRPV4, la présence de calcium intracellulaire dans les cellules endocardiques et la voie moléculaire PKC-PKD2-HDAC5, composants nécessaires à l’expression du gène klf2a en réponse au flux. Ces données nous permettent de suggérer le rôle de TRPP2, TRPV4 et la voie moléculaire récemment découverte, PKC-PKD2-HDAC5 dans la formation valvulaire dépendante de klf2a.
Mechanical forces that dictate valve formation are not well understood. In zebrafish, blood flow induces expression of the transcription factor klf2a in a specific subset of endocardial cells that will go on to form functional valves. We aimed to identify the molecular mechanisms activating this flow-induced valve formation. By altering and modifying blood flow patterns, we observed that flow-mediated forces are necessary to control early klf2a expression and endocardial cell numbers. We then looked at different mechanosensors operating at early stages of valve development. Using in vivo labelling, we identified primary cilia in the endocardium and showed that the membrane channels TRPP2 and TRPV4 increase intracellular calcium which activates a PKC-PKD2-HDAC5 pathway necessary for klf2a expression in response to flow. Together these data suggest a role for TRPV4, TRPP2 and the recently described PKC-PKD2-HDAC5 signalling pathway in klf2a-mediated valvulogenesis.
La clairance mucociliaire est un processus physico-chimique qui sert à transporter et éliminer le mucus bronchique. Pour cela, des milliards d'appendices de taille micrométrique, que l'on nomme cils, ...recouvrent l'épithélium respiratoire. Ces cils propulsent le mucus en suivant un motif périodique comprenant une phase de poussée où leur pointe peut pénétrer dans le mucus, et une phase de récupération où ils sont totalement immergés dans le fluide périciliaire. Un dysfonctionnement de ce processus peut engendrer de nombreux problèmes de santé. Il a été observé expérimentalement que les cils ne battent pas aléatoirement, mais synchronisent leurs battements avec leurs voisins, formant ainsi des ondes métachronales. Toutefois, les observations in vivo sont extrêmement difficiles à réaliser, et les propriétés de ces ondes restent mal connues. Dans cette thèse, nous utilisons un solveur Lattice Boltzmann - Frontière Immergée afin de reproduire un épithélium bronchique et étudier l'émergence, ainsi que les capacités de transports et de mélanges, de ces ondes
The mucociliary clearance process is a physico-chemical process which aims is to transport and eliminate bronchial mucus. To do so, billions of micro-sized appendages, called cilia, cover the respiratory epithelium. These cilia propel the mucus by performing a periodical pattern composed of a stroke phase where their tips can enter the mucus layer, and a recovery phase where the cilia are completely immersed in the periciliary liquid layer. A failure of this process may induce numerous health problems. It has been experimentally observed that cilia do not beat randomly, but instead adapt their beatings accordingly to their neighbours, forming metachronal waves. However, in vivo observations are extremely difficult to perfom, and the properties of these waves remain poorly understood. In this thesis, we use a Lattice Boltzmann - Immersed Boundary solver to reproduce a bronchial epithelium and study the emergence, as well as the transport and mixing capacities, of these waves
La kinase Aurora A est une protéine essentielle au cycle cellulaire et plus particulièrement lors de la mitose. En effet, Aurora A est nécessaire à l'entrée en mitose et joue un rôle dans la ...maturation des centrosomes. Elle participe à l'assemblage du fuseau mitotique et est nécessaire à la réussite de la cytodiérèse. Elle est également nécessaire à l'égale répartition des mitochondries dans les cellules filles et joue un rôle dans l'épissage alternatif des ARNm de facteurs apoptotiques. Au-delà de ses fonctions mitotiques, plusieurs études récentes indiquent qu'Aurora A présente des fonctions supplémentaires dans les cellules en interphase. Elle est notamment essentielle au désassemblage du cil primaire et joue un rôle dans la dynamique des microtubules et la migration cellulaire. Enfin, une dérégulation de son expression, de sa stabilité et/ou de son activité perturbe le déroulement du cycle cellulaire ce qui conduit à la transformation des cellules et favorise l'apparition de cancers. Ses fonctions normales ainsi que ses fonctions lors de la carcinogenèse sont conduites à travers les nombreux partenaires protéiques qui entrent en interaction avec elle. Ils modulent son activité, sa localisation et sa stabilité. En retour Aurora A phosphoryle un bon nombre d'entre eux régulant ainsi leur activité, localisation et stabilité. Cependant, l'analyse des interactions déjà connues d'Aurora A ne permet pas d'expliquer tous les phénotypes observés lors de sa dérégulation. Afin de mieux comprendre les fonctions d'Aurora A, les mécanismes qui la régulent et mettre en évidence ses multiples rôles au sein de la cellule, j'ai construit puis analysé un interactome d'Aurora A généré à partir d'une méthode de purification d'affinité couplée à la spectrométrie de masse en tandem. J'ai identifié 477 partenaires potentiels dont 180 présentant une forte probabilité d'être des partenaires directs de la kinase. L'analyse bioinformatique approfondie de cet interactome a permis de révéler les partenaires associés à des mécanismes liés à la mitochondrie et l'épissage des ARN messagers mettant en évidence une implication potentielle d'Aurora A dans ces mécanismes. Pour valider cet interactome, j'ai choisi d'étudier plus précisément deux partenaires identifiés dans cette étude : les protéines WDR62 et CEP97. J'ai montré que ces deux partenaires co-localisent avec Aurora A et sont phosphorylés par la kinase. Ainsi, ce travail de thèse a permis de mettre en évidence un nombre important de nouveaux partenaires d'Aurora A associés à de nouvelles fonctions. L'étude de ces nouvelles fonctions liées aux mitochondries et à l'épissage des ARN, constitue deux nouveaux projets actuellement menés par des collaborateurs au sein de notre institut.
The kinase Aurora A is an essential mitotic cell cycle protein. Aurora A is necessary for mitotic entry and for the maturation and separation of centrosomes. It participates in mitotic spindle assembly and chromosome biorientation, and it is essential for the completion of cytokinesis. Furthermore, Aurora A activity is necessary for the equal distribution of mitochondria to daughter cells and, through its role in the alternative splicing of mRNA of apoptotic factors, it provides a link between cell cycle control and apoptosis. Beyond its mitotic functions, several recent studies suggest that Aurora A is also important during interphase. Notably, it influences microtubule dynamics, promotes cell migration and polarity control and is essential for primary cilia disassembly. Reflecting the fact that Aurora A is found to be up-regulated in many cancers, deregulation of Aurora A activity can result in an aberrant cell cycle, ultimately leading to malignant transformation of cells. The crucial regulation of Aurora A’s numerous functions is achieved through its interaction with several protein partners, which modulate its activity, localisation and stability. Aurora A in turn phosporylates a number of them, thus regulating their activity, localisation and stability. However, the known interactions of Aurora A cannot explain all the phenotypes that have been described of its deregulation.To better understand the functions of Aurora A, the regulation mechanisms governing it, and to expose its multiple roles in the cell, I have built and analysed an Aurora A interactome using tandem affinity purification coupled with mass spectrometry. This resulted in the identification of 477 potential interacting partners, of which, 180 were determined to have a high probability of interacting directly with the kinase.In-depth bioinformatic analysis of this interactome has revealed the associated partners to be related to mitochondria and mRNA splicing, highlighting the potential involvement of Aurora A in these mechanisms. To validate the interactome, two of the proteins identified in this study, WDR62 and CEP97, were examined in detail. Here I show that these two proteins colocalise with Aurora A, and are phosphorylated by the kinase.WDR62 is implicated in microcephaly and is deregulated in certain cancers. I have shown that Aurora A phosphorylates WDR62 during mitosis, and that this phosphorylation is necessary for its localisation to the centrosomes. CEP97 is a poorly charactarised protein of the primary cilium, abnormalities of which are associated with ciliopathies. I have shown that Aurora A phosphorylates CEP97 in vitro, and that the inhibition of Aurora A activity in vivo perturbs the localisation of CEP97 to cilia and centrosomes.This study has identified a number of new Aurora A-interacting proteins, implicating the kinase with novel functions. These functions, related to mitochondria and mRNA splicing have opened up a new area for further investigation.
Les cils et les flagelles sont des organites cellulaires très conservés qui assurent des fonctions essentielles. Chez l'Homme, les défauts d'assemblage des cils et des flagelles conduisent à de ...multiples pathologies, les ciliopathies. Afin de comprendre comment se forment et fonctionnent les cils, j'ai analysé la fonction de deux nouveaux gènes identifiés comme cible des facteurs de transcription de ciliogenèse RFX. Tout d'abord je me suis focalisée sur le gène CCDC151, évolutivement conservé dans les espèces possédant des cils motiles. J'ai pu montrer que CCDC151 est impliquée dans le transport dépendant de l'IFT des bras de dynéine chez les animaux et qu'elle est nécessaire à la perception sensorielle chez la drosophile. Par ailleurs, j'ai également montré que cette protéine possède des fonctions cellulaires additionnelles puisqu'elle est requise pour l'orientation correcte des plans de division cellulaire et qu'elle est impliquée dans la régulation de la taille du cil primaire chez les mammifères. Je me suis ensuite intéressée au gène LRRC48 également conservée dans les espèces possédant des cils motiles. Cette protéine est nécessaire à la motilité des flagelles de spermatozoïdes et des cils des neurones sensoriels en 9+0 et dans la réponse auditive chez la drosophile. De plus LRRC48 est indispensable au développement des vertébrés puisque son absence chez le poisson zèbre conduit à l'hydrocéphalie, des kystes rénaux et des défauts de motilité des cils. Elle est également essentielle à la biogenèse de l'oreille dans cet organisme.En conclusion, il s'agit de deux nouveaux acteurs de la ciliogenèse potentiellement impliqués dans les pathologies ciliaires chez l'Homme
Cilia are highly conserved structures found from protozoa to mammals where they play essential physiological and developmental functions and cilia dysfunction leads to various syndromes in humans known as ciliopathies. To understand cilia formation and function, I performed functional analysis of two new target genes of the RFX ciliogenic transcription factors. First, I focused on CCDC151 that is evolutionary conserved in motile ciliated species. I showed that CCDC151 is involved in the control of IFT-dependent dynein arm assembly in animals and required for geotaxis behavior of adult flies. In zebrafish, depletion of Ccdc151 leads to left-right asymmetry defects and kidney cysts, two phenotypes resulting from impaired ciliary beating. However, I also showed that CCDC151 is also implicated in other cellular functions in vertebrates as it is involved in proper orientation of cell divisions and implicated in the regulation of primary cilium length in mammalian cells. In a second part, I studied LRRC48 that is also conserved in species with motile cilia. I showed that this protein is essential for motility of flagellar spermatozoids and for motility of the 9+0 sensory cilia as well as in the auditory response in drosophila. In zebrafish, morpholinos induced depletion of this protein leads to hydrocephaly, kidney cysts, inner ear abnormalities and cilia motility defects. Moreover this protein is also required for inner ear biogenesis in the model. In conclusion, these two genes are essential for ciliogenesis and they are new candidate genes potentially implicated in human ciliary diseases
Il s’agit de l’étude des successions écologiques du phytoplancton dans la lagune de Ghar El Melh au niveau de 5 stations pendant (i) un suivi bimensuel allant du janvier 2011 à janvier 2012 et (ii) ...un suivi saisonnier à partir du novembre 2012 à mars 2013. L’étude écologique a été consacrée au microphytoplancton en relation avec les facteurs abiotiques (température, salinité et nutriments) et biotiques (microalgues épiphytes, kystes des dinoflagellés, ultraplancton et ciliés). L’analyse hydrologique du milieu a montré une variation principalement saisonnière de la plupart des paramètres avec des valeurs élevées de la température et de la salinité en été et des concentrations exceptionnelles des nutriments en automne et en hiver. Le phytoplancton de la lagune de Ghar El Melh a été principalement dominé par les dinoflagellés et les diatomées avec une manifestation alarmante des espèces nuisibles atteignant plus que 70 % du peuplement. Ces proliférations ont été remarquées durant toute l’année avec une manifestation plus marquante dans les régions internes de la lagune ce qui est principalement tributaire à la température de l’eau, l’azote total, le nitrogène et l’orthophosphate. Une importante contribution des espèces épiphytes au phytoplancton a été signalée notamment celle du dinoflagellé toxique Prorocentrum lima. L’inventaire spécifique du phytoplancton dominé par l’espèce Prorocentrum micans (> 28 %) et d’autres espèces ne produisant pas de kyste a été en concordance avec les faibles valeurs enregistrées pour les kystes de dinoflagellés dans le sédiment où le maximum enregistré a été de l’ordre de 229 kystes g-1. Une part importante de l’ultraplancton a été représentée par le nanophytoplancton et les hétérotrophes procaryotes, dépassant largement les concentrations microphytoplanctoniques, ce qui est un indice de l’eutrophisation du milieu. Des corrélations hautement significatives ont été trouvées entre le microphytoplancton et les organismes microzooplanctoniques évoquant un probable rôle de ces derniers dans le contrôle des proliférations microalguales nuisibles.
We explored the distribution of the microphytoplankton community in relation to both abiotic (temperature, salinity and nutrients) and biotic factors (epiphytes microalgae, dinoflagellate cysts, ultraplankton and ciliates) in Ghar al Melh Lagoon at 5 stations during (i) a biweekly sampling from January 2011 to January 2012 and (ii) a seasonal sampling from november 2012 to march 2013. The results pointed out a seasonal variation of all parameters with high water temperature and salinity in summer and exceptional concentrations of nutrients in autumn and winter. Microphytoplankton was mainly dominated by dinoflagellates and diatoms with sequences of harmful species proliferation attaining more than 70 % of total phytoplankton. These sporadic proliferations occurred throughout the year with exacerbation in the inner parts of the lagoon mainly related to temperature and total nitrogen, nitrogen and orthophosphate. An important contribution of epiphytic species to phytoplankton abundance was recorded with especially the presence of the toxic dinoflagellate Prorocentrum lima. Microphytoplankton was dominated by Prorocentrum micans (> 28 %) and other non-cyst former species consistent with the low dinoflagellate cysts found at sediment surface (maximum = 229 cysts g-1 DS). Urtraplankton was chiefly represented by nanophytoplankton and heterotrophic prokaryotes, with low microphytoplanktonic abundance. This indicates that the lagoon is undergoing eutrophication. Significant correlations were reported between phytoplankton and microzooplankton suggesting a potential role of the former in controlling harmful microalgae proliferations.

Abstract Bardet-Biedl syndrome (BBS) is a ciliopathy causing multivisceral abnormalities. Its prevalence in Europe is from 1/125,000 to 1/175,000. This disorder is defined by a combination of ...clinical signs: obesity, pigmentary retinopathy, post-axial polydactyly, polycystic kidneys, hypogenitalism, and learning disabilities, many of which appearing after several years of evolution. Individual clinical phenotype is highly variable. Most signs are present in a majority of patients but only pigmentary retinopathy is constant after infancy. There are many other associated minor clinical signs including diabetes, blood hypertension, congenital cardiopathy or Hirschsprung disease. This broad clinical spectrum is associated to a great genetic heterogeneity, with mainly an autosomal recessive transmission and, sometimes cases of oligogenism. To date, mutations in 12 different genes ( BBS1 to BBS12 ) are responsible for this phenotype. These genes code for proteins involved in the development and function of primary cilia. Absent or non functional BBS proteins affect cilia in certain organs such as kidney or eye. However, some symptoms are still not clearly related to cilia dysfunction. BB syndrome has to be recognized because a molecular diagnosis is possible and will lead to familial genetic counseling and possibly prenatal diagnosis. Patients with BBS will need a multidisciplinary medical care. The renal abnormalities are the main life-threatening features because they can lead to end-stage renal failure and renal transplantation. Retinal dystrophy leading to progressive vision loss, moderate mental retardation, and obesity will affect social life of these patients.
