Ole Hendricks was an immigrant both representative and
exceptional-a true artistic talent who nevertheless lived a
familiar immigrant experience. By day, he was a farmer. But at
night, his fiddle lit ...up dance halls, bringing together all manner
of neighbors in rural Minnesota. Each tune in his repertoire of
waltzes, reels, polkas, quadrilles, and more were copied neatly
into his commonplace book.
Such tunebooks, popular during the nineteenth century, rarely
survive and are often overlooked by folk scholars in favor of
commercially produced recordings, published sheet music, or oral
tradition. Based on extensive historical and genealogical research,
Amy Shaw presents a grounded picture of a musician, his family, and
his community in the Upper Midwest, revealing much about music and
dance in the area. This notable contribution to regional music and
folklore includes more than one hundred of Ole's dance tunes,
transcribed into modern musical notation for the first time.
Ole Hendricks and His Tunebook will be valuable to readers
and scholars interested in ethnomusicology and the Norwegian
American immigrant experience.

Tujuan untuk mencerahkan masyarakat tentang pentingnya penanaman dan penataan pohon rempah secara artistik di ...sekitar Air Terjun Tumburano. Metode penyuluhan terhadap 20 orang warga desa (kepala desa dan perangkatnya, anggota kelompok tani, dan ibu-ibu PKK). Hasilnya menunjukkan adanya transfer pengetahuan baru bagi peserta mengenai pohon rempah sebagai tanaman leluhur di kebun pribadi, berubah menjadi tanaman yang menarik di lahan pemerintah sebagai milik semua warga di sekitarnya, sehingga harus dikelola dengan baik agar lestari dan berkelanjutan. Pemerintah segera membuat kerjasama dengan perguruan tinggi dan pengusaha dalam pengelolaan destinasi wisata ini serta segera menyiapkan insfrastruktur jalan, penyediaan bibit tanaman rempah dan ruang terbuka hijau (RTH) di sekitarnya. Penyuluhan ini dapat ditindaklanjuti dengan jenis pengabdian lain seperti: pelatihan mengolah bahan-bahan lokal yang bisa dinikamti di Air Terjun Tumburano dan menjadi ole-ole, serta pembuatan souvenir khas Kabupaten Konawe Kepulauan.
Cadmium (Cd) is a well-known industrial and environmental pollutant having hazardous and poisonous properties in various forms of kidney impairment. Consequently, this study aimed to evaluate the ...nephroprotective effect of palm dates extract (PDE) and olive leaves extract (OLE) against cadmium chloride (CdCl2)-induced nephrotoxicity. A total of 36 adult male albino rats were divided into six equal groups. Group 1: control group injected with physiological saline, group 2: oral gavage with PDE, group 3: orally administrated with OLE, group 4: injected daily with CdCl2 (3 mg/kg, i.p.). Groups 5 and 6: were orally treated with either PDE or OLE, respectively one hour prior to ip administration of CdCl2. After one-week samples were collected from all groups for serum biochemical analysis of kidney function as well as investigation of malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx). Kidney tissue specimens were obtained for histopathological examination. CdCl2 exposure caused a significant elevation (P<0.05) in serum creatinine, urea, uric acid and MDA levels with marked reduction in CAT, SOD and GPx indicating renal damage. CdCl2 induced severe congestion and vasculitis with marked perivascular edema, inflammation, hemorrhage, glomerular shrinkage, massive degeneration, necrosis and apoptosis in the renal tubular epithelium. PDE and OLE treatment resulted in significant improvement in kidney function and oxidative markers with a marked reduction of MDA level. Histopathological changes were also ameliorated. In conclusion, PDE or OLE treatment significantly reduced the deleterious effects of acute CdCl2 renal damage by reducing oxidative stress as a protective mechanism.

Olive pollen is one of the most important causes of seasonal respiratory allergy in Mediterranean countries, where this tree is intensely cultivated. Among the high number of protein allergens ...detected in this pollen, 8 - Ole e 1 to Ole e 8 - have been isolated and characterized. Ole e 1 is the most frequent sensitizing agent, affecting more than 70% of the patients suffering of olive pollinosis, although others, such as Ole e 4 and Ole e 7, have also been shown to be major allergens. In this context, the prevalence of many olive pollen allergens seems to be dependent on the geographical area where the sensitized patients live. Some of the olive allergens have been revealed as members of known protein families: profilin (Ole e 2), Ca(2+)-binding proteins (Ole e 3 and Ole e 8), superoxide dismutase (Ole e 5) and lipid transfer protein (Ole e 7). No biological function has been demonstrated for Ole e 1, whereas Ole e 4 and Ole e 6 are new proteins without homology to known sequences from databases. cDNAs encoding for Ole e 1, Ole e 3 and Ole e 8 have been overproduced in heterologous systems. The recombinant products were correctly folded and exhibited the functional activities of the natural allergens. In addition to the Oleaceae family, other species, such as Gramineae or Betulaceae, contain pollen allergens structurally or immunologically related to those of the olive tree. This fact allows to detect and evaluate antigenic cross-reactivities involving olive allergens. The aim of this research is the development of new diagnostic tools for olive pollinosis and new approaches to improve the classical immunotherapy.
Triacylglycerols (TAG), accumulate within lipid droplets (LD), predominantly surrounded by OLEOSINs (OLE), that protect TAG from hydrolysis. We tested the hypothesis that identifying and removing ...degradation signals from OLE would promote its abundance, preventing TAG degradation and enhancing TAG accumulation. We tested whether mutating potential ubiquitin-conjugation sites in a previously reported improved Sesamum indicum OLE (SiO) variant, o3-3 Cys-OLE (SiCO herein), would stabilize it and increase its lipogenic potential. SiCOv1 was created by replacing all five lysines in SiCO with arginines. Separately, six cysteine residues within SiCO were deleted to create SiCOv2. SiCOv1 and SiCOv2 mutations were combined to create SiCOv3. Transient expression of SiCOv3 in Nicotiana benthamiana increased TAG by two-fold relative to SiCO. Constitutive expression of SiCOv3 or SiCOv5, containing the five predominant TAG-increasing mutations from SiCOv3, in Arabidopsis along with mouse DGAT2 (mD) increased TAG accumulation by 54% in leaves and 13% in seeds compared with control lines coexpressing SiCO and mD. Lipid synthesis rates increased, consistent with an increase in lipid sink strength that sequesters newly synthesized TAG, thereby relieving the constitutive BADC-dependent inhibition of ACCase reported for WT Arabidopsis. These OLE variants represent novel factors for potentially increasing TAG accumulation in a variety of oil crops.
Noncoding RNAs (ncRNAs) longer than 200 nucleotides are rare in bacteria, likely because bacterial genomes are under strong evolutionary pressures to maintain a small genome size. Of the long ncRNAs ...unique to bacteria, the OLE (ornate, large, extremophilic) RNA class is among the largest and most structurally complex. OLE RNAs form a ribonucleoprotein (RNP) complex by partnering with at least two proteins, OapA and OapB, that directly bind OLE RNA. The biochemical functions of the OLE RNP complex remain unknown, but are required for proper adaptation to certain environmental stresses, such as cold temperatures, short chain alcohols, and high magnesium concentrations. In the current study, we used electrophoretic mobility shift assays to examine the binding of OLE RNA fragments by OapB and found that OapB recognizes a small subregion of OLE RNA, including stem P13, with a dissociation constant (KD) of ∼700 pm. Analyses with mutated RNA constructs, and the application of in vitro selection, revealed that strong binding of OLE RNA by OapB requires a stem containing a precisely located single-nucleotide bulge and a GNRA tetraloop. Although the vast majority of bacteria with the ole gene also have the oapB gene, there are many whose genomes contain oapB but lack ole, suggesting that OapB has other RNA partners in some species that might exhibit similar structural features.
Objective: As there is a drastic change in the nature of written literacy, both tertiary level students and educators are expected to maximize the opportunities offered by digital tools like ...Classkick application when giving and receiving constructive feedback about their language lessons. The purpose of this study was to get feedback from the students, especially about their perceptions of Student Interaction, Student Engagement, Instructor Presence, Student Satisfaction & Perceived Learning in using ‘Classkick’ App for writing lessons as well as to get the learners’ suggestions onto how do we use ‘Classkick’ App to create more interesting lessons. Methodology: The methods used were an adapted questionnaire named Student Learning and Satisfaction in Online Learning Environments Instrument (SLS-OLE) questionnaire and two semi-formal interview questions. 96 students from the Law Faculty at Universiti Teknologi MARA (UiTM) Kampus Dengkil located in Malaysia were chosen as an informant group. Findings: The results indicate that the most common condition concerning students’ readiness in getting constructive feedback is that students get to interact with others using this application in order for them to be engaged with the lecturer’s presence so that it will lead to students’ satisfaction in enabling the construction of their own perceived learning in online classrooms. Implications: The main conclusion was that more constructive feedback received by the students will lead to more motivating factors to learn writing digitally.
