We describe the metagenomics-derived feline enteric virome in the faeces of 25 cats from a single shelter in California. More than 90 % of the recognizable viral reads were related to mammalian ...viruses and the rest to bacterial viruses. Eight viral families were detected: Astroviridae, Coronaviridae, Parvoviridae, Circoviridae, Herpesviridae, Anelloviridae, Caliciviridae and Picobirnaviridae. Six previously known viruses were also identified: feline coronavirus type 1, felid herpes 1, feline calicivirus, feline norovirus, feline panleukopenia virus and picobirnavirus. Novel species of astroviruses and bocaviruses, and the first genome of a cyclovirus in a feline were characterized. The RNA-dependent RNA polymerase region from four highly divergent partial viral genomes in the order Picornavirales were sequenced. The detection of such a diverse collection of viruses shed within a single shelter suggested that such animals experience robust viral exposures. This study increases our understanding of the viral diversity in cats, facilitating future evaluation of their pathogenic and zoonotic potentials.
Swimming in ocean water, including ocean water at beaches not impacted by known point sources of pollution, is an increasing health concern. This study was an initial evaluation of the presence of ...indicator microbes and pathogens and the association among the indicator microbes, pathogens, and environmental conditions at a subtropical, recreational marine beach in south Florida impacted by non-point sources of pollution. Twelve water and eight sand samples were collected during four sampling events at high or low tide under elevated or reduced solar insolation conditions. The analyses performed included analyses of fecal indicator bacteria (FIB) (fecal coliforms, Escherichia coli, enterococci, and Clostridium perfringens), human-associated microbial source tracking (MST) markers (human polyomaviruses HPyVs and Enterococcus faecium esp gene), and pathogens (Vibrio vulnificus, Staphylococcus aureus, enterovirus, norovirus, hepatitis A virus, Cryptosporidium spp., and Giardia spp.). The enterococcus concentrations in water and sand determined by quantitative PCR were greater than the concentrations determined by membrane filtration measurement. The FIB concentrations in water were below the recreational water quality standards for three of the four sampling events, when pathogens and MST markers were also generally undetectable. The FIB levels exceeded regulatory guidelines during one event, and this was accompanied by detection of HPyVs and pathogens, including detection of the autochthonous bacterium V. vulnificus in sand and water, detection of the allochthonous protozoans Giardia spp. in water, and detection of Cryptosporidium spp. in sand samples. The elevated microbial levels were detected at high tide and under low-solar-insolation conditions. Additional sampling should be conducted to further explore the relationships between tidal and solar insolation conditions and between indicator microbes and pathogens in subtropical recreational marine waters impacted by non-point source pollution.

Heavy metal contamination has become a major problem that causes severe environmental and health issues due to their biosorption, bioaccumulation, and toxicity. This study was designed to evaluate heavy metal chelating abilities of alginic acid (AA) extracted from the brown seaweed Ecklonia cava and two of its derivatives prepared by the partial oxidation of the 2° OH groups (OAA) and partial carboxylation of the monomeric units (CAA) upon reducing the heavy metal biosorption in zebrafish (Danio rerio) modal. Metal ions were quantified using ICP-OES and biopolymers were characterized by FTIR and XRD analysis. All investigated biopolymers indicated potential ability for chelating Pb2+, Cu2+, Cd2+, As3+, and Ag+. The sorption capacities were in the order of CAA>OAA>AA. All biopolymers indicated a comparatively higher chelation towards Pb2+. AA, OAA, and CAA could effectively reduce Pb2+ induced toxicity and Pb2+ stress-induced ROS production in zebrafish embryos. Besides, they could reduce the biosorption of Pb2+ in adult zebrafish which could lead to bioaccumulation. Since alginic acid purified from E. cava and its derivatives could be utilized as seaweed derived biopolymers to purify heavy metals contaminated water and as a dietary supplement to reduce heavy metal biosorption in organisms.
Agar, alginate, and carrageenans are high-value seaweed hydrocolloids, which are used as gelation and thickening agents in different food, pharmaceutical, and biotechnological applications. The ...annual global production of these hydrocolloids has recently reached 100,000 tons with a gross market value just above US$ 1.1 billion. The techno-functional properties of the seaweed polysaccharides depend strictly on their unique structural make-up, notably degree and position of sulfation and presence of anhydro-bridges. Classical extraction techniques include hot alkali treatments, but recent research has shown promising results with enzymes. Current methods mainly involve use of commercially available enzyme mixtures developed for terrestrial plant material processing. Application of seaweed polysaccharide targeted enzymes allows for selective extraction at mild conditions as well as tailor-made modifications of the hydrocolloids to obtain specific functionalities. This review provides an update of the detailed structural features of κ-, ι-, λ-carrageenans, agars, and alginate, and a thorough discussion of enzyme assisted extraction and processing techniques for these hydrocolloids.

The fate and removal of 48 pharmaceuticals and personal care products (PPCPs) during anaerobic digestion of sewage sludge were investigated in four full-scale sewage treatment plants (STPs). We measured concentrations in both the liquid and solid phases of the sludge to compare the distribution ratios (Kd) between phases before and after digestion. The results showed changes in Kd values of PPCPs with carboxyl or amino functional groups, probably due to a shift of dissociation equilibrium with the increase in pH. Sulfamethoxazole and trimethoprim were almost completely degraded (>90%); triclosan, triclocarban, and ofloxacin were moderately degraded (around 30–50%); but carbamazepine was not eliminated. To our knowledge, this is the first report that shows (i) the occurrence and removal of several tens of PPCPs by anaerobic sludge digestion in full-scale municipal STPs and (ii) the change of distribution between the liquid and solid phases during digestion.
To discover and develop novel natural compounds, active ingredients, single herbs and combination formulas or prescriptions in traditional Chinese medicine (TCM) with therapeutic selectivity that can ...preferentially kill cancer cells and inhibit the amplification of cancer without significant toxicity is an important area in cancer therapy. A lot of valuable TCMs were applied as alternative or complementary medicines in the United States and Europe. But these TCMs, as one of the main natural resources, were widely used to research and develop new drugs in Asia. In TCMs, some specific herbs, animals, minerals and combination formulas were recorded and exploited due to their active ingredients and specific natural compounds with antitumor activities. The article focused on the antitumor properties of natural compounds and combination formulas or prescriptions in TCMs, described its influence on tumor progression, angiogenesis, metastasis, and revealed its mechanisms of antitumor and inhibitory action. Among the nature compounds, triptolide, berberine, matrine, oxymatrine, kurarinone and deoxypodophyllotoxin (DPT) with specific molecular structures have been separated, purified, and evaluated their antitumor properties in vitro and in vivo. Cancer is a multifactorial and multistep disease, so the treatment effect of combination formulas and prescriptions in TCMs involving multi-targets and multi-signal pathways on tumor may be superior than that of agents targeting a single molecular target alone. Shi Quan Da Bu Tang and Yanshu injection, as well known combination formulas and prescriptions in TCMs, have shown an excellent therapeutic effect on cancer.
Diarrheal diseases caused by intestinal protozoan parasites are a major food-borne public health problem across the world. Vegetables and fruits provide important nutrients and minerals, but are also ...common sources of some food-borne human pathogenic microorganisms. The contamination of raw vegetables and fruits with human pathogenic parasites are now a global public health threat, despite the health benefits of these foods in non-pharmacological prophylaxes against diseases. A large number of reports have documented the contamination of vegetables or fruits with human pathogenic microorganisms. In this paper, we reviewed the contamination and detection methods of human pathogenic intestinal protozoans that are frequently recovered from raw vegetables and fruits. The protozoan parasites include Cryptosporidium spp., Giardia duodenalis, Cyclospora cayetanensis, Entamoeba spp., Toxoplasma gondii, Balantioides coli, Blastocystis sp., Cystoisospora belli and Enterocytozoon bieneusi. The risk factors involved in the contamination of vegetables and fruits with parasites are also assessed.
Propolis is a resinous substance collected by bees from exudates of different plants that is rich in well-known health-relevant phenolic compounds such as flavonoids and phenolic acids. Propolis ...extracts are very complex matrices difficult to study. Different analytical methods are usable to analyse propolis extracts and to obtain chemical fingerprint but to our knowledge NMR has not previously been used for this purpose.
This study aims to demonstrate that it is possible to use ¹H-NMR for the simultaneous recognition of phenolic compounds in complex matrices, such as propolis extracts, using appropriate tools for spectra pre-treatment and analysis.
In this work 12 typical phenolic propolis compounds (apigenin, chrysin, galangin, kaempferol, quercetin, naringenin, pinocembrin, pinostrobin, caffeic acid, cinnamic acid, p-coumaric acid and ferulic acid) were considered as reference compounds and their presence in samples was verified by HPLC-MS. A simple ¹H-NMR sequence was used to obtain spectra of samples. Spectra were pre-treated by using an appropriate tool for spectra alignment and analysed by using software for the study of spectra originated from complex matrices. Sixty-five propolis samples were used to test the proposed identification procedure.
Ten out of 12 considered compounds were identified as statistically significant in most of the samples.
This work suggests that it is possible to efficiently use ¹H-NMR, coupled with appropriate spectral analytical tools, for the simultaneous detection of phenolic compounds in complex matrices.
In the present work, the usefulness of ultrasonic power as a dispersion and mixing tool to accelerate the adsorption of Safranin O (SO), methylene blue (MB), Pb(2+) ions and Cr(3+) ions onto the ...novel composite Fe3O4-NPs-AC adsorbent was investigated. This new material was extensively characterized and analyzed by different techniques such as XRD, FESEM, Raman spectroscopy and FT-IR. Central composite design (CCD) based on designed runs revealed that adsorbent mass, sonication time, MB concentration, SO concentration, Pb(2+) ion and Cr(3+) ion concentration and some of their interactions have significant contributions to the target compounds removal percentages. A combination of response surface methodology and Design-Expert software was used to qualify and estimate the influence and magnitude of each terms contribution to the response. An optimization study using the following investigated increments of the effective variables, adsorbent mass (0.01-0.03 g), sonication time (2-6 min), initial dye concentration (5-25 mg L(-1)), and initial metal ion concentration (20-60 mg L(-1)), revealed that fixing the experimental variables at 0.025 g of Mn-Fe3O4-NPs-AC, with a 3 min sonication time, and 20 mg L(-1) of MB, 10 mg L(-1) of SO, 38 mg L(-1) of Pb(2+) ions and 42 mg L(-1) of Cr(3+) ions at room temperature lead to the achievement of the best characteristics and performance. Conduction of 32 experiments according to the limitations of CCD and a subsequent analysis of variance (ANOVA) gave useful information about the significant and also approximate contributions of each term (main and interaction of variables) in an empirical equation for the expected response. The results indicate that the R(2) values are more than 0.988 and the adjusted R(2) values are in reasonable agreement with R(2). Under the optimal conditions, the MB, SO, Pb(2+) ion and Cr(3+) ion removal efficiencies reached 99.54%, 98.87%, 80.25% and 99.54% after 3 min, while their equilibrium data with high performance can be represented by Langmuir isotherms and a pseudo second-order kinetic model. The maximum adsorption capacities for the single component system, 229.4 mg g(-1) for MB, 159.7 mg g(-1) for SO, 139.5 mg g(-1) for Pb(2+) ions and 267.4 mg g(-1) for Cr(3+) ions, support the high efficiency of Mn-Fe3O4-NPs-AC as a new adsorbent.
Abstract Introduction Chronic apical periodontitis (CAP) is a frequent condition that has a considerable effect on a patient's quality of life. We aimed to reveal root canal microbial communities in ...antibiotic-naive patients by applying Illumina sequencing (Illumina Inc, San Diego, CA). Methods Samples were collected under strict aseptic conditions from 12 teeth (5 with primary CAP, 3 with secondary CAP, and 4 with a periapical abscess PA) and characterized by profiling the microbial community on the basis of the V6 hypervariable region of the 16S ribosomal RNA gene by using Illumina HiSeq2000 sequencing combinatorial sequence-tagged polymerase chain reaction products. Results Root canal specimens displayed highly polymicrobial communities in all 3 patient groups. One sample contained 5–8 (mean = 6.5) phyla of bacteria. The most numerous were Firmicutes and Bacteroidetes , but Actinobacteria , Fusobacteria , Proteobacteria , Spirochaetes , Tenericutes , and Synergistetes were also present in most of the patients. One sample contained 30–70 different operational taxonomic units; the mean (± standard deviation) was lower in the primary CAP group (36 ± 4) than in the PA (45 ± 4) and secondary CAP (43 ± 13) groups ( P < .05). The communities were individually different, but anaerobic bacteria predominated as the rule. Enterococcus faecalis was found only in patients with secondary CAP. One PA sample displayed a significantly high proportion (47%) of Proteobacteria , mainly at the expense of Janthinobacterium lividum. Conclusions This study provided an in-depth characterization of the microbiota of periapical tissues, revealing highly polymicrobial communities and minor differences between the study groups. A full understanding of the etiology of periodontal disease will only be possible through further in-depth systems-level analyses of the host-microbiome interaction.
