Evolutionary adaptations for maintaining beneficial microbes are hallmarks of mutualistic evolution. Fungus-farming “attine” ant species have complex cuticular modifications and specialized glands ...that house and nourish antibiotic-producing Actinobacteria symbionts, which in turn protect their hosts’ fungus gardens from pathogens. Here we reconstruct ant–Actinobacteria evolutionary history across the full range of variation within subtribe Attina by combining dated phylogenomic and ultramorphological analyses. Ancestral-state analyses indicate the ant–Actinobacteria symbiosis arose early in attineant evolution, a conclusion consistent with direct observations of Actinobacteria on fossil ants in Oligo-Miocene amber. qPCR indicates that the dominant ant-associated Actinobacteria belong to the genus Pseudonocardia. Tracing the evolutionary trajectories of Pseudonocardia-maintaining mechanisms across attine ants reveals a continuum of adaptations. In Myrmicocrypta species, which retain many ancestral morphological and behavioral traits, Pseudonocardia occur in specific locations on the legs and antennae, unassociated with any specialized structures. In contrast, specialized cuticular structures, including crypts and tubercles, evolved at least three times in derived attine-ant lineages. Conspicuous caste differences in Pseudonocardia-maintaining structures, in which specialized structures are present in worker ants and queens but reduced or lost in males, are consistent with vertical Pseudonocardia transmission. Although the majority of attine ants are associated with Pseudonocardia, there have been multiple losses of bacterial symbionts and bacteria-maintaining structures in different lineages over evolutionary time. The early origin of ant–Pseudonocardia mutualism and the multiple evolutionary convergences on strikingly similar anatomical adaptations for maintaining bacterial symbionts indicate that Pseudonocardia have played a critical role in the evolution of ant fungiculture.
Since morels were first successfully cultivated commercially in Sichuan in 2012, morel cultivation has expanded to more than 20 provinces in China. The highest yield currently reaches 15,000 kg/ha. ...Morel cultivation is characterized by its environmental friendliness, short cycle length, and high profit. However, the yield obtained is unstable which makes morel cultivation a high-risk industry. Although 10 production cycles have passed, there is still a gap between morel cultivation practice and our basic knowledge of morel biology. This mini-review concentrates on the development needs of morel cultivation. We illustrate the key techniques used in the large-scale commercial cultivation of morels and their relevant studies, including nutritional requirements, mechanisms of nutrient bag, soil type, vegetative and reproductive growth conditions, and disease control. This review will be a useful practical reference for the commercial artificial cultivation of morels and promoting the vital technologies required.

Spent mushroom substrate is made from the waste remaining after the harvest of mushrooms. Here, we evaluated the potential of five spent edible fungi (Auricularia cornea, Lentinus edodes, Pleurotus ...eryngii, P. citrinopileatus and P. ostreatus) substrates as feed sources for Tenebrio molitor larvae. Young larvae did not survive on any substrate except the spent L. edodes substrate (36.7%). The survival rates in young larvae were similar among the different diets in which wheat bran or rice bran was replaced with 0, 20, 30, 40, 50, or 60% spent L. edodes substrate. The weights of the surviving larvae were decreased only when 70% of wheat bran and > 40% of rice bran was replaced with spent L. edodes substrate. In addition, the middle-aged larvae fed wheat bran only were significantly larger than those fed diets with 30~60% spent L. edodes substrate in dry feed, but the larvae of all treatments failed to pupate. Whereas the green feed was added in dry feed, there were no significant differences in pupal weight, pupation rate, pupal duration, adult emergence, or deformed adults among the three treatments in middle-aged larvae that were fed on diets containing 0, 30, or 40% spent L. edodes substrate. Collectively, these results suggest that spent L. edodes substrate has considerable potential to be used as a partial replacement (< 40%) of conventional feed for T. molitor, and spent mushroom substrate waste may be recycled as feed material for resource insects.

Some lineages of ants, termites, and beetles independently evolved a symbiotic association with lignocellulolytic fungi cultivated for food, in a lifestyle known as fungiculture. Fungus-growing ...insects' symbiosis also hosts a bacterial community thought to integrate their physiology. Similarities in taxonomic composition support the microbiota of fungus-growing insects as convergent, despite differences in fungus-rearing by these insects. Here, by comparing fungus-growing insects to several hosts ranging diverse dietary patterns, we investigate whether the microbiota taxonomic and functional profiles are characteristic of the fungiculture environment. Compared to other hosts, the microbiota associated with fungus-growing insects presents a distinctive taxonomic profile, dominated by Gammaproteobacteria at class level and by Pseudomonas at genera level. Even with a functional profile presenting similarities with the gut microbiota of herbivorous and omnivorous hosts, some differentially abundant features codified by the microbiota of fungus-growing insects suggest these communities occupying microhabitats that are characteristic of fungiculture. These features include metabolic pathways involved in lignocellulose breakdown, detoxification of plant secondary metabolites, metabolism of simple sugars, fungal cell wall deconstruction, biofilm formation, antimicrobials biosynthesis, and metabolism of diverse nutrients. Our results suggest that the microbiota could be functionally adapted to the fungiculture environment, codifying metabolic pathways potentially relevant to the fungus-growing insects' ecosystems functioning.
Leafcutter ants propagate co‐evolving fungi for food. The nearly 50 species of leafcutter ants (Atta, Acromyrmex) range from Argentina to the United States, with the greatest species diversity in ...southern South America. We elucidate the biogeography of fungi cultivated by leafcutter ants using DNA sequence and microsatellite‐marker analyses of 474 cultivars collected across the leafcutter range. Fungal cultivars belong to two clades (Clade‐A and Clade‐B). The dominant and widespread Clade‐A cultivars form three genotype clusters, with their relative prevalence corresponding to southern South America, northern South America, Central and North America. Admixture between Clade‐A populations supports genetic exchange within a single species, Leucocoprinus gongylophorus. Some leafcutter species that cut grass as fungicultural substrate are specialized to cultivate Clade‐B fungi, whereas leafcutters preferring dicot plants appear specialized on Clade‐A fungi. Cultivar sharing between sympatric leafcutter species occurs frequently such that cultivars of Atta are not distinct from those of Acromyrmex. Leafcutters specialized on Clade‐B fungi occur only in South America. Diversity of Clade‐A fungi is greatest in South America, but minimal in Central and North America. Maximum cultivar diversity in South America is predicted by the Kusnezov–Fowler hypothesis that leafcutter ants originated in subtropical South America and only dicot‐specialized leafcutter ants migrated out of South America, but the cultivar diversity becomes also compatible with a recently proposed hypothesis of a Central American origin by postulating that leafcutter ants acquired novel cultivars many times from other nonleafcutter fungus‐growing ants during their migrations from Central America across South America. We evaluate these biogeographic hypotheses in the light of estimated dates for the origins of leafcutter ants and their cultivars.
Mushroom cultivation produces a significant amount of wastewater containing high levels of both organic and inorganic contaminants. In this study, mushroom farm wastewater (MFW) was treated ...separately by aquatic macrophytes (Ceratophyllum demersum L.) and algae (Chlorella vulgaris). The laboratory experiments consisted of a constructed reactor planted with selected aquatic plants and a microalgal culture and operated for 16 days. The pollutant removal efficiency was evaluated using different experimental combinations such as control 1 (C. demersum using borewell water), control 2 (C. vulgaris using borewell water), T1 (C. demersum using MFW), and T2 (C. vulgaris using MFW), respectively. The results showed that the T1 treatment had the highest significant (p < 0.05) removal efficiency of selected pollutant parameters (total dissolved solids: 86.00%; biochemical oxygen demand: 83.10%; chemical oxygen demand: 86.60%; total nitrogen: 84.30%; total phosphorus: 75.60%). The kinetic studies using the first-order reaction model showed a good fit (Rsup.2 > 0.8317) and the maximum rate constant (k) of pollutant reduction in T1 treatment. In addition, the growth, biochemical, and proximate parameters of both C. demersum and C. vulgaris were highest in the same treatment. Therefore, the proposed experiment offers a promising approach for the efficient and environmentally friendly treatment of MFW.
A number of experiments were done to further our understanding of the substrate utilization in button mushroom crops (Agaricus bisporus). An analysis of the degradation of dry matter of the substrate ...during a crop cycle revealed that for pin formation the upper 1/3rd layer is used, for the production of flush one all layers are involved and for flush two mainly the lower 1/3 layer is used. A reduction in substrate depth leads to a decrease in yield/m.sup.2 but an apparent increase in yield per tonne of substrate with a lower mushroom quality. A short daily interruption of the connection between the casing soil with the substrate results in a delay of the first flush. Interruptions with only part of the substrate did not lead to delay in production. Daily interruption of the connection with all or only part of the substrate leads to a shift in yield from flush one to flush two but the total yield remains unchanged. The mycelial biomass in the substrate increases from filling up to pinning, has a steeper increase during flush one, and is levelling off during flush two, indicating that in the period of venting and up to/including flush one, enzymes are secreted by growing hyphae generating nutrients to feed a fixed amount of mushroom biomass for two flushes. A sidewise extension of the substrate (without casing soil, thus not producing mushrooms) showed that the substrate at a distance more than somewhere between 20-50 cm away from the casing soil does not contribute to feeding mushrooms in the first two flushes. The observations are discussed with respect to relevant previous research.

Thirty unique non-host RNAs were sequenced in the cultivated fungus, Agaricus bisporus, comprising 18 viruses each encoding an RdRp domain with an additional 8 ORFans (non-host RNAs with no ...similarity to known sequences). Two viruses were multipartite with component RNAs showing correlative abundances and common 3' motifs. The viruses, all positive sense single-stranded, were classified into diverse orders/families. Multiple infections of Agaricus may represent a diverse, dynamic and interactive viral ecosystem with sequence variability ranging over 2 orders of magnitude and evidence of recombination, horizontal gene transfer and variable fragment numbers. Large numbers of viral RNAs were detected in multiple Agaricus samples; up to 24 in samples symptomatic for disease and 8-17 in asymptomatic samples, suggesting adaptive strategies for co-existence. The viral composition of growing cultures was dynamic, with evidence of gains and losses depending on the environment and included new hypothetical viruses when compared with the current transcriptome and EST databases. As the non-cellular transmission of mycoviruses is rare, the founding infections may be ancient, preserved in wild Agaricus populations, which act as reservoirs for subsequent cell-to-cell infection when host populations are expanded massively through fungiculture.

Spent mushroom substrate (SMS) of Pleurotus ostreatus was supplemented with wheat bran and soybean flour and used as substrate for a new cultivation cycle of the oyster mushrooms Pleurotus ...ostreatus and Pleurotus pulmonarius. The bioconversion efficiency of mushrooms produced over substrate (BE%) used and the chemical composition of sporophores were evaluated. The concentration of mycelial mass, crude exopolysaccharide content and laccase enzyme activity were also determined at the supplemented SMS before inoculation, at 50% and 100% of colonization stages in the new cultivation and in the final re-utilized SMS. The laccase enzyme was extracted to examine SMS potential for the dephenolization of olive mill and winery wastewaters. Results showed that both Pleurotus species exhibited BE over 185%, demonstrating this bioprocess could represent a promising strategy to convert SMS into nutritional food. Data also indicate the strong positive impact that SMS could have in the solid wastes’ management and agribusiness enhancement.

Agro-wastes, such as crop residues, leaf litter, and sawdust, are major contributors to global greenhouse gas emissions, and consequently a major concern for climate change. Nowadays, mushroom ...cultivation has appeared as an emerging agribusiness that helps in the sustainable management of agro-wastes. However, partial utilization of agro-wastes by mushrooms results in the generation of a significant quantity of spent mushroom substrates (SMS) that have continued to become an environmental problem. In particular, Shiitake (Lentinula edodes Berk.) mushrooms can be grown on different types of agro-wastes and also generate a considerable amount of SMS. Therefore, this study investigates the biotransformation of SMS obtained after Shiitake mushroom cultivation into biogas and attendant utilization of slurry digestate (SD) in tomato (Solanum lycopersicum L.) crop fertilization. Biogas production experiments were conducted anaerobically using four treatments of SMS, i.e., 0% (control), 25, 50, and 75% inoculated with a proportional amount of cow dung (CD) as inoculum. The results on biogas production revealed that SMS 50% treatment yielded the highest biogas volume (8834 mL or 11.93 mL/g of organic carbon) and methane contents (61%) along with maximum reduction of physicochemical and proximate parameters of slurry. Furthermore, the biogas digestate from 50% treatment further helped to increase the seed germination (93.25%), seedling length (9.2 cm), seedling root length (4.19 cm), plant height (53.10 cm), chlorophyll content (3.38 mg/g), total yield (1.86 kg/plant), flavonoids (5.06 mg/g), phenolics (2.78 mg/g), and tannin (3.40 mg/g) contents of tomato significantly (p < 0.05) in the 10% loading rate. The findings of this study suggest sustainable upcycling of SMS inspired by a circular economy approach through synergistic production of bioenergy and secondary fruit crops, which could potentially contribute to minimize the carbon footprints of the mushroom production sector.
