Oocyte and embryo developmental competence are shaped by multiple extrinsic and intrinsic factors. One of the most extensive research areas in the last decade is the regulation of lipid metabolism in ...oocytes and embryos of different species. We hypothesized that differences in developmental competence of oocytes and embryos between prepubertal and cyclic gilts may arise due to distinct fatty acid profiles in follicular fluid. We found that supplementation of oocyte maturation media with follicular fluid from prepubertal pigs affected quality and development of embryos from prepubertal pigs while embryos of cyclic pigs were not affected. PLIN2, SCD and ACACA transcripts involved in lipid metabolism were upregulated in embryos originating from oocytes of prepubertal pigs matured with autologous follicular fluid. The surface occupied by lipid droplets tend to increase in oocytes matured with follicular fluid from prepubertal pigs regardless oocyte origin. The change into follicular fluid of cyclic pigs increased the efficiency of embryo culture and improved quality, while gene expression was similar to embryos obtained from cyclic gilts. We assume that the follicular fluids of prepubertal and cyclic pigs influenced the quality of oocytes and embryos obtained from prepubertal pigs which are more susceptible to suboptimal in vitro culture conditions.
Locomotion is one of the most important ecological functions in animals. Precocial animals, such as pigs, are capable of independent locomotion shortly after birth. This raises the question whether ...coordinated movement patterns and the underlying muscular control in these animals is fully innate or whether there still exists a rapid maturation. We addressed this question by studying gait development in neonatal pigs through the analysis of spatio-temporal gait characteristics during locomotion at self-selected speed. To this end, we made video recordings of piglets walking along a corridor at several time points (from 0 h to 96 h). After digitization of the footfalls, we analysed self-selected speed and spatio-temporal characteristics (e.g. stride and step lengths, stride frequency and duty factor) to study dynamic similarity, intralimb coordination and interlimb coordination. To assess the variability of the gait pattern, left-right asymmetry was studied. To distinguish neuromotor maturation from effects caused by growth, both absolute and normalized data (according to the dynamic similarity concept) were included in the analysis. All normalized spatio-temporal variables reached stable values within 4 h of birth, with most of them showing little change after the age of 2 h. Most asymmetry indices showed stable values, hovering around 10%, within 8 h of birth. These results indicate that coordinated movement patterns are not entirely innate, but that a rapid neuromotor maturation, potentially also the result of the rearrangement or recombination of existing motor modules, takes place in these precocial animals.
Chronic subclinical levels of disease occur frequently in intensive swine production and compromise nutrient use efficiency. Feeding additional Met plus Cys (M+C) has been implicated in improving the ...response of the animal to immune system stimulation (ISS) because they can serve as substrates for generating compounds involved in the immune response, such as glutathione and acute phase proteins. A N-balance study was conducted to assess the optimal dietary Met to Met plus Cys ratio (M:M+C) during ISS in 20-kg pigs. Thirty-six pigs were fed 800 g/d of 1 of 5 M+C-limiting diets, containing graded levels of M:M+C (0.42, 0.47, 0.52, 0.57, and 0.62) and supplying 2.5 g/d of M+C. After adaptation, N balances were determined sequentially during a 5-d prechallenge period and 2 ISS periods of 3 and 4 d, respectively. To induce ISS, pigs were injected intramuscularly with repeated and increasing doses of Escherichia coli lipopolysaccharide. Eye temperature and blood profile confirmed effective ISS. During ISS period 1, ISS reduced the mean N balance more severely than ISS period 2 (8.7 ± 0.3 vs. 9.6 ± 0.4 g/d; P < 0.001) and was less than the prechallenge period (10.0 ± 0.2 g/d; P < 0.001). An interactive effect of ISS and diet on N balance was observed (P < 0.001). Based on quadratic-plateau regression analysis, the optimal dietary M:M+C was 0.57 ± 0.03 and 0.59 ± 0.02 for the prechallenge period and ISS period 2, respectively. The optimal dietary M:M+C for ISS period 1 was found to be greater than 0.62, indicating that the optimal M:M+C is greater during ISS. It is suggested that this is the result of preferential use of Met during ISS. In conclusion, ISS results in an increase in the optimal dietary M:M+C in growing pigs.
The present study investigated the effect of feeding bovine colostrum (BC) to piglets in comparison with feeding a milk replacer (MR) and conventional rearing by the sow on the intestinal immune ...system and number of enterotoxigenic Escherichia coli (ETEC) colonising the intestinal tissue. Piglets (23-d-old) were allocated to one of the following four groups: (1) killed at the beginning of the experiment (Base); (2) separated from the sow and fed BC (BC-fed); (3) separated from the sow and fed a MR (MR-fed); (4) kept with the sow (Sow-Milk). Blood was sampled on days 1 and 8, and faecal samples were collected on days 1, 3, 5 and 8. On day 8, piglets were killed and gastrointestinal digesta and intestinal segments were collected. The frequency of diarrhoea was found to be higher (P≤ 0·019) in MR-fed piglets than in BC-fed and Sow-Milk piglets. Piglets from the MR-fed group had the lowest lactic acid bacteria:haemolytic E. coli ratio (P
treat= 0·064) in the faeces. The number of E. coli colonising the intestinal tissue was higher (P< 0·001) in piglets from the MR-fed group than in those from the BC-fed and Sow-Milk groups. Piglets from the Sow-Milk group had a higher (P= 0·020) mucosal IgG concentration than those from the MR-fed group, but did not exhibit any difference when compared with piglets from the Base and BC-fed groups. Piglets from the BC-fed group exhibited a reduced (P≤ 0·037) expression level of Toll-like receptor-4 in the intestinal mucosa when compared with those from the MR-fed and Sow-Milk groups. The expression level of IL-2 was higher (P≤ 0·051) in piglets from the MR-fed group than in those from the other treatment groups. In conclusion, feeding BC rather than MR to the piglets reduced the colonisation of intestine by ETEC and modulated the intestinal immune system, whereas no differences were observed in piglets fed BC and conventionally reared by the sows.

This study investigated the effects of feeding Hermetia illucens larvae (0, 4, and 8%; HI0, HI4, and HI8 groups, respectively) on growth performance, carcass traits, and meat quality of finishing ...pigs. Results showed that the HI4 diet increased (P < .05) final body weight and average daily gain and decreased (P < .05) feed to gain ratio compared with HI0 and HI8 group. HI4 and HI8 diets increased (P < .05) loin-eye area, marbling scores, and inosine monophosphate content of longissimus thoracis (LT) compared with HI0 diet. The intramuscular fat content was greater (P < .05) in HI4 group than in the HI0 group. Furthermore, HI4 diet up-regulated (P < .05) lipogenic genes and MyHC-IIa mRNA levels in LT compared with HI0 diet. Our results indicated that dietary inclusion of H. illucens larvae has a beneficial impact on growth performance and meat quality, and the underlying mechanism may be due to the altered lipogenic potential induced by H. illucens larvae.

Genome‐wide association study results are presented for intramuscular fat in Italian Large White pig breed. A total of 886 individuals were genotyped with PorcineSNP60 BeadChip. After quality ...control performed with plink software and in R environment, 49 208 markers remained for the association analysis. The genome‐wide association studies was conducted using linear mixed model implemented in GenABEL. We detected seven new SNPs of genes till now not found associated to intramuscular fat (IMF). Three markers map in a wide intergenic region rich of QTL linked to fat traits, one map 388 kb upstream the gene SDK1, one map inside PPP3CA gene, one inside SCPEP1 gene and the last is not mapped in the porcine genome yet. Associations here presented indicate a moderate effect of these genes on IMF. In particular, PPP3CA, that is involved in the oxidative metabolism of skeletal muscle, could be considerated as an interesting candidate gene for IMF content in pigs. However, further studies are needed to clarify the role of these genes on the physiological processes involved in IMF regulation. These results may be useful to control this trait that is important in terms of nutritional, technological and organoleptic characteristics of fresh meat and processed products.
Individuals can increase inclusive fitness benefits through a complex network of social interactions directed towards kin. Preferential relationships with relatives lead to the emergence of kin ...structures in the social system. Cohesive social groups of related individuals and female philopatry of wild boar create conditions for cooperation through kin selection and make the species a good biological model for studying kin structures. Yet, the role of kinship in shaping the social structure of wild boar populations is still poorly understood. In the present study, we investigated spatio-temporal patterns of associations and the social network structure of the wild boar Sus scrofa population in Białowieża National Park, Poland, which offered a unique opportunity to understand wild boar social interactions away from anthropogenic factors. We used a combination of telemetry data and genetic information to examine the impact of kinship on network cohesion and the strength of social bonds. Relatedness and spatial proximity between individuals were positively related to the strength of social bond. Consequently, the social network was spatially and genetically structured with well-defined and cohesive social units. However, spatial proximity between individuals could not entirely explain the association patterns and network structure. Genuine, kin-targeted, and temporarily stable relationships of females extended beyond spatial proximity between individuals while males interactions were short-lived and not shaped by relatedness. The findings of this study confirm the matrilineal nature of wild boar social structure and show how social preferences of individuals translate into an emergent socio-genetic population structure.

Adenosine-to-inosine (A-to-I) RNA editing meditated by adenosine deaminases acting on RNA (ADARs) enzymes is a widespread post-transcriptional event in mammals. However, A-to-I editing in skeletal ...muscle remains poorly understood. By integrating strand-specific RNA-seq, whole genome bisulphite sequencing, and genome sequencing data, we comprehensively profiled the A-to-I editome in developing skeletal muscles across 27 prenatal and postnatal stages in pig, an important farm animal and biomedical model. We detected 198,892 A-to-I editing sites and found that they occurred more frequently at prenatal stages and showed low conservation among pig, human, and mouse. Both the editing level and frequency decreased during development and were positively correlated with ADAR enzymes expression. The hyper-edited genes were functionally related to the cell cycle and cell division. A co-editing module associated with myogenesis was identified. The developmentally differential editing sites were functionally enriched in genes associated with muscle development, their editing levels were highly correlated with expression of their host mRNAs, and they potentially influenced the gain/loss of miRNA binding sites. Finally, we developed a database to visualize the Sus scrofa RNA editome. Our study presents the first profile of the dynamic A-to-I editome in developing animal skeletal muscle and provides evidences that RNA editing is a vital regulator of myogenesis.
Current evidence suggests that pigs were first domesticated in Eastern Anatolia during the ninth millennium cal BC before dispersing into Europe with Early Neolithic farmers from the beginning of the ...seventh millennium. Recent ancient DNA (aDNA) research also indicates the incorporation of European wild boar into domestic stock during the Neolithization process. In order to establish the timing of the arrival of domestic pigs into Europe, and to test hypotheses regarding the role European wild boar played in the domestication process, we combined a geometric morphometric analysis (allowing us to combine tooth size and shape) of 449 Romanian ancient teeth with aDNA analysis. Our results firstly substantiate claims that the first domestic pigs in Romania possessed the same mtDNA signatures found in Neolithic pigs in west and central Anatolia. Second, we identified a significant proportion of individuals with large molars whose tooth shape matched that of archaeological (likely) domestic pigs. These large ‘domestic shape’ specimens were present from the outset of the Romanian Neolithic (6100–5500 cal BC) through to later prehistory, suggesting a long history of admixture between introduced domestic pigs and local wild boar. Finally, we confirmed a turnover in mitochondrial lineages found in domestic pigs, possibly coincident with human migration into Anatolia and the Levant that occurred in later prehistory.
