SUMMARY
We report an intracranial stenting procedure complicated by active bleeding from the femoral puncture site because of high arterial puncture. The patient was treated by placement of two ...PTFE‐covered self‐expanding coronary stent‐grafts. To our knowledge, there have been very few reports on stent‐grafting of femoral artery in the literature. The low profile and flexibility of the coronary stent‐graft enabled treatment via a six French sheath and 12th month patency is demonstrated with CT angiography.

Evolutionary studies suggest that 200-250 million years ago an aphid ancestor was infected with a free-living eubacterium. The latter became established within aphid cells. Host and endosymbiont ...(genus Buchnera) became interdependent and unable to survive without each other. The growth of Buchnera became integrated with that of the aphids, which acquired the endosymbionts from their mothers before birth. Speciation of host lineages was paralleled by divergence of associated endosymbiont lineages, resulting in parallel evolution of Buchnera and aphids. Present day Buchnera retains many of the properties of its free-living ancestor, containing genes for proteins involved in DNA replication, transcription, and translation, as well as chaperonins and proteins involved in secretion, energy-yielding metabolism, and amino acid biosynthesis. Some of these processes are also observed in isolated endosymbiont cells. Genetic and physiological studies indicate that Buchnera can synthesize methionine, cysteine, and tryptophan and supply these amino acids to the aphid host. In the case of some fast-growing species of aphids, the overproduction of tryptophan by Buchnera involves plasmid-amplification of the gene coding for anthranilate synthase, the first enzyme of the tryptophan biosynthetic pathway. These recent studies provide a beginning in our understanding of Buchnera and its role in the endosymbiosis with aphids.

La surexploitation des ressources naturelles, consécutive à un besoin accru en terres arables des populations locales, entraîne de graves dysfonctionnements dans le bio-fonctionnement du sol ...(carences minérales, stress salins, etc.). Ce dysfonctionnement, aggravé par les aléas climatiques (sécheresse), nécessite la mise en œuvre de stratégies d’ingénierie écologique permettant une réhabilitation des milieux dégradés via la restauration de services écologiques essentiels. Les premiers symptômes du processus d’altération de la qualité des sols des milieux tropicaux et méditerranéens se traduisent par une altération de la structure de la strate épigée avec, en particulier, une paupérisation de la diversité et de l’abondance des espèces végétales. Cette dégradation est accompagnée par une fragilisation des sols et une augmentation de l’impact de l’érosion sur la couche superficielle, entraînant une baisse de la fertilité des sols au niveau de leurs caractéristiques physico-chimiques, mais aussi biologiques (par exemple, la microflore tellurique). Parmi les composantes microbiennes particulièrement sensibles à ces phénomènes d’érosion, les microorganismes symbiotiques (rhizobia, champignons mycorhiziens) sont connus pour être des composantes clés dans le fonctionnement des principaux cycles biogéochimiques telluriques (C, N et P). De nombreux travaux ont montré l’importance de la gestion de ces microorganismes symbiotiques dans les stratégies de réhabilitation et de revégétalisation des milieux dégradés, mais également pour améliorer la productivité des agro-systèmes. En particulier, la sélection de symbiotes et leur inoculation en masse dans le sol de plantation ou de culture ont été fortement encouragées au cours de ces dernières décennies. Ces inoculants étaient sélectionnés, non seulement pour leur impact sur la plante, mais également pour leur capacité à persister dans le sol, au détriment de la microflore résiduelle native. La performance de cette technique était donc évaluée au niveau du couvert végétal, mais les répercussions au niveau des caractéristiques de la microflore tellurique étaient généralement ignorées. Le rôle de la diversité microbienne sur la productivité et la stabilité (résistance, capacité de résilience, etc.) des éco- et agro-systèmes a été mis en évidence relativement récemment et a conduit à une remise en cause des bases conceptuelles de l’inoculation contrôlée dans la gestion durable des terres. Il a été notamment suggéré que les caractéristiques du milieu à réhabiliter devaient être prises en compte, et plus particulièrement son niveau de dégradation par rapport au seuil de résilience écologique. Cette prise en compte devrait permettre d’optimiser les pratiques culturales pour soit (i) redonner à l’écosystème ses propriétés originelles dans le cas de milieux peu dégradés ou (ii) transformer un écosystème dans le cas de sols hautement dégradés (par exemple, les sols miniers). Dans ce chapitre, nous évoquerons, à travers différents exemples d’expérimentations réalisées en milieu tropical et méditerranéen, les performances des différentes stratégies de gestion du potentiel microbien des sols (inoculation d’espèces exotiques vs des espèces natives, inoculation contrôlée vs la gestion du potentiel microbien via la strate végétale épigée, etc.) suivant le niveau de dégradation du milieu.
The overexploitation of natural resources, resulting in an increased need for arable lands by local populations, causes a serious dysfunction in the soil's biological functioning (mineral deficiency, salt stress, etc.). This dysfunction, worsened by the climatic conditions (drought), requires the implementation of ecological engineering strategies allowing the rehabilitation of degraded areas through the restoration of essential ecological services. The first symptoms of weathering processes of soil quality in tropical and Mediterranean environments result in an alteration of the plant cover structure with, in particular, the pauperization of plant species diversity and abundance. This degradation is accompanied by a weakening of soils and an increase of the impact of erosion on the surface layer resulting in reduced fertility of soils in terms of their physicochemical characteristics as well as their biological ones (e.g., soil microbes). Among the microbial components particularly sensitive to erosion, symbiotic microorganisms (rhizobia, Frankia, mycorrhizal fungi) are known to be key components in the main terrestrial biogeochemical cycles (C, N and P). Many studies have shown the importance of the management of these symbiotic microorganisms in rehabilitation and revegetation strategies of degraded environments, but also in improving the productivity of agrosystems. In particular, the selection of symbionts and their inoculation into the soil were strongly encouraged in recent decades. These inoculants were selected not only for their impact on the plant, but also for their ability to persist in the soil at the expense of the residual native microflora. The performance of this technique was thus evaluated on the plant cover, but its impact on soil microbial characteristics was totally ignored. The role of microbial diversity on productivity and stability (resistance, resilience, etc.) of eco- and agrosystems has been identified relatively recently and has led to a questioning of the conceptual bases of controlled inoculation in sustainable land management. It has been suggested that the environmental characteristics of the area to rehabilitate should be taken into account, and more particularly its degradation level in relation to the threshold of ecological resilience. This consideration should lead to the optimization of the cultural practices to either (i) restore the original properties of an ecosystem in case of slightly degraded environments or (ii) transform an ecosystem in case of highly degraded soils (e.g., mine soils). In this chapter, we discuss, through various examples of experiments conducted in tropical and Mediterranean areas, the performance of different strategies to manage the microbial potential in soils (inoculation of exotic vs. native species, inoculation or controlled management potential microbial stratum via aboveground vegetation, etc.) based on the level of environmental degradation.
L’interaction symbiotique légumineuses-rhizobium nécessite l'infection des racines de la plante par les bactéries et l’initiation de divisions cellulaires dans le cortex racinaire.Les cytokinines ...sont des hormones végétales agissant via une signalisation par phosphotransfert qui conduit à l’activation de Régulateurs de Réponse de type B (RRBs), des facteurs de transcription régulant l'expression des gènes de réponse primaire aux cytokinines. Une étude phylogénétique menée sur plusieurs espèces de légumineuses a révélé une expansion génique de la famille des RRBs et l’apparition de formes non-canoniques de ces facteurs de transcription. Chez Medicago truncatula,MtRRB3 est le RRB le plus fortement exprimé dans les racines et les nodosités et est impliqué dans la nodulation. En effet, les plantes dont l’expression de MtRRB3 a été réduite par ARNi ainsi que des mutants rrb3 présentent une diminution significative du nombre de nodosités formées. De plus, l’expression de gènes associés à la nodulation, tels que "Nodulation Signalling Pathway 2" (MtNSP2) et "Cell Cycle Switch 52A"(MtCCS52A), est réduite en réponse aux cytokinines dans ces mutants. Des fusions transcriptionnelles avec le rapporteur GUS montrent que MtRRB3, MtNSP2 et MtCCS52Aprésentent un profil d’expression spatiale largement chevauchant dans les racines et lesnodosités. Des expériences de ChIP-qPCR et de trans-activation en protoplastes indiquent par ailleurs que MtRRB3 peut respectivement interagir avec et activer les promoteurs des gènesMtNSP2 et MtCCS52A. Cette thèse a donc permis d’établir des mécanismes moléculaires impliqués dans les régulations transcriptionnelles médiées par les cytokinines lors de la mise en place des nodosités symbiotiques fixatrices d’azote.
The legume-rhizobium interaction requires the infection of plant roots by rhizobia and the initiation of cell divisions in the root cortex. Cytokinins, a class of plant hormones acts trough a phosphotranfert signalling leading to the activation of Type-B Response Regulators(RRBs) which are transcription factors regulating the expression of cytokinins primary response genes. Phylogenetic analyses carried out indifferent legume species genomes showed anexpansion of the RRB genes family associated toan increase in non-canonical RRBs. In Medicago truncatula nodules, MtRRB3 is the most expressed RRB in roots and nodules. MtRRB3 islinked to nodulation as MtRRB3 RNAi silencedplants as well as rrb3 mutants display asignificant decrease of nodule number. Inaddition, the expression of the nodulation related genes Nodulation Signalling Pathway 2(MtNSP2) and Cell Cycle Switch 52A(MtCCS52A) is reduced in response to cytokininsin rrb3 mutants. The expression pattern of apMtRRB3-GUS fusion overlaps with thepMtNSP2-GUS and pMtCCS52A-GUS fusions in roots and nodules. Finally, ChIP-qPCR and protoplast trans-activation experiments showed that MtRRB3 can respectively interacts with and activate MtNSP2 and MtCCS52A promoters. This thesis have thus established molecular mechanisms associated to transcriptional regulations mediated by cytokinins during the legume symbiotic nitrogen-fixing nodulation.
En cas de carence azotée, les légumineuses sont capables de mettre en place une symbiose avec des bactéries du sol fixatrices d’azote appelées rhizobia. Cette symbiose a lieu dans un organe appelé ...nodosité où les bactéries sont endocytées et appelées bactéroïdes. Certains clades de légumineuses imposent un processus de différenciation à leurs bactéroïdes qui agrandissent considérablement et deviennent polyploïdes, menant à des morphotypes bactériens allongés ou sphériques. Au cours de cette thèse, j’ai étudié la différenciation des bactéroïdes de Bradyrhizobium spp. en association avec Aeschynomene spp.. Les bactéroïdes de ces plantes présentent des degrés de différenciation distincts qui dépendent de l’espèce hôte. Mes données suggèrent que les bactéroïdes les plus différenciés sont aussi les plus efficaces. J’ai cherché à savoir quels facteurs procaryotes pourraient être impliqués dans les adaptations des bactéroïdes au processus de différenciation et à leurs divers hôtes, le tout en lien avec cette différence d’efficacité symbiotique au travers d’approches globales sans a priori de type -omiques. Les conditions considérées sont des bactéroïdes de différents morphotypes et des cultures libres de référence. Les fonctions activées en conditions symbiotiques ont été identifiées, ainsi que les gènes spécifiques d’un hôte donné. Des analyses fonctionnelles des gènes d’intérêt ont également été menées. Les mutants bactériens n’ont toutefois pas présenté de phénotype symbiotique drastique, montrant ainsi l’existence de réseaux de gènes complexes menant à la résilience des génomes de rhizobia.
In case of nitrogen starvation, legume plants establish a symbiotic interaction with nitrogen-fixing soil bacteria called rhizobia. This interaction takes place in nodules where the symbionts are internalized and become bacteroids. Some legume clades also impose a differentiation process onto the bacteroids which become enlarged and polyploid, leading to elongated or spherical morphotypes. During my PhD work, I have studied bacteroid differentiation of Bradyrhizobium species in association with Aeschynomene spp.. These bacteroids display distinct differentiation levels depending on the plant host, and my analyses suggest that the most differentiated ones are also the most efficient. I investigated the bacterial factors potentially involved in the adaptations to differentiation and host-specificity, and related to the higher efficiency of the most differentiated bacteroids using global-omics approaches without a priori. The analyzed conditions were bacteroids of distinct morphotypes and free-living reference cultures. Activated functions under symbiotic conditions were identified, as well as host-specific ones. Functional analyses were performed on genes of interest. However, the bacterial mutants did not display drastic symbiotic phenotypes, showing the existence of complex gene networks leading to high resilience of rhizobial genomes.
Les bactéries endosymbiotiques Wolbachia se transmettent verticalement de la mère aux descendants mais également horizontalement d'un hôte arthropode à l'autre. Chez les isopodes terrestres, les ...Wolbachia montrent une diversité et une prévalence élevées et bouleversent la reproduction de leurs hôtes en féminisant les mâles ou en induisant des incompatibilités de croisements.Nous avons utilisé la diversité de ces souches de Wolbachia pour étudier leur plasticité en terme de changement d'hôte et l'évolution de leur virulence. Nos résultats montrent la grande plasticité des Wolbachia, s'installant chez tous les hôtes testés. Par ailleurs, un transfert horizontal est possible à partir de différentes sources tissulaires : ovaires et hémocytes ainsi que lors de la prédation d'un individu infecté. De plus, les transferts horizontaux aux échelles intra comme interspécifique peuvent modifier la nature des interactions entre Wolbachia et leurs hôtes et mener à une relation de type hôte-pathogène. Ce type d'interaction a été spécifiquement exploré pour comprendre les mécanismes impliqués dans les changements de virulence. Nous avons démontré que la réponse autophagique de l'hôte contre Wolbachia pouvait jouer un rôle majeur dans la nature pathogène de l'interaction. De plus, par une approche d'évolution expérimentale, nous montrons que Wolbachia peut devenir pathogène pour son hôte natif chez qui elle est habituellement peu virulente. En détournant les Wolbachia de leur voie de transmission verticale et en les transmettant uniquement de manière horizontale, elles deviennent beaucoup plus invasives et tuent leur hôte.
Wolbachia are basically endosymbiotic bacteria, vertically transmitted from mother to offspring. However, they can also pass horizontally from one arthropod host to another. Among terrestrial isopods, the Wolbachia exhibit high diversity and prevalence and disturb their host reproduction by feminizing males into females or inducing crossing incompatibilities.We took advantage of this diversity of Wolbachia in terrestrial isopods to study their plasticity through hosts switching and the evolution of their virulence. Our results highlighted their huge plasticity as they were capable of settling in all the tested terrestrial isopods hosts. These horizontal transfers can be performed with different tissues as a source of Wolbachia: Ovaries and hemocytes, but also when an uninfected host preyed an infected one. Moreover, horizontal transfers can modify the nature of the interaction between Wolbachia and their hosts moving towards a host-pathogen relationship. We further explored such conflicting interactions in order to investigate the reasons of the changes in virulence. We demonstrated that the host autophagic response against Wolbachia can turn against the host itself and making the symbiont a pathogen. Moreover, by using experimental evolution, we revealed that Wolbachia could increase in virulence against the native host it normally does not harm in a context of vertical transmission. When we disrupted the vertical transmission mode in order to exclusively transmit Wolbachia by horizontal contagious mode, they became much more invasive and rapidly killed their native host.
