More than 15 years of research have documented behavioural differences between callous-unemotional and non-callous subtypes of children with antisocial behaviour. Recent studies also suggest that ...children with callous-unemotional traits may be genetically vulnerable to antisocial behaviour, while those without callous-unemotional traits appear to have primarily environmental aetiology to their antisocial behaviour. Furthermore neurocognitive profiles differ between antisocial children with and without callous-unemotional traits. While the former group appears emotionally under-reactive, particularly to others' distress, the latter group may be emotionally over-reactive, particularly to perceived threat. In this review we provide an overview of the current evidence base with regard to callous-unemotional and non-callous subgroups of children with antisocial behaviour and discuss the implications of the current evidence base for prevention and intervention. This overview selectively focuses on recent advances in this area of research, as well as earlier studies where these help set the research context.
The current study aims to ascertain how different variants of callous-unemotional traits differ in their psychopathology, exposure to aggression and violence, and aggressive and violent behavior. If ...secondary/distressed variants (high in callous-unemotional traits and high in anxiety) and primary/traditional variants (high in callous-unemotional traits and low in anxiety) differ along these dimensions, it may speak to their different etiologies, treatment needs (e.g., trauma focused), and responsiveness to treatment. The current sample consisted of 799 adolescents from high schools (n = 419) and juvenile detention centers (n = 380). Participants were interviewed regarding their callous-unemotional traits, psychopathology, exposure to aggression and violence, and aggressive and violent behavior. Parents/guardians and teachers/staff members also reported on participants' callous-unemotional traits and aggressive and violent behavior. A model-based cluster analysis indicated that there were four clusters in the data set, based on callous-unemotional traits and anxiety: a nonvariant cluster, a primary/traditional callous-unemotional cluster, a secondary/distressed callous-unemotional cluster, and a "fearful" cluster. Secondary/distressed variants of psychopathy exhibited significantly greater symptoms of depression and psychoticism, more exposure to low level aggression and neighborhood violence, and more aggressive and violent behavior, as compared to the other clusters. Adolescents with callous-unemotional traits might not be a homogeneous group, but rather may differ in attitudes, behaviors, and exposure to risk, therefore differing in their treatment needs and responsiveness.

Accumulating research suggests that psychopathy can be disaggregated into low-anxious primary and high-anxious secondary variants, and this research may be important for understanding antisocial ...youths with callous-unemotional traits. Using model-based cluster analysis, the present study disaggregated 165 serious male adolescent offenders (M age = 16) with high scores on the Youth Psychopathic Traits Inventory into primary and secondary variants based on the presence of anxiety. The results indicated that the secondary, high-anxious variant was more likely to show a history of abuse and scored higher on measures of emotional and attentional problems. On a picture version of the dot-probe task, the low-anxious primary variant was not engaged by emotionally distressing pictures, whereas the high-anxious secondary variant was more attentive to such stimuli (Cohen d = 0.71). Although the two groups differed as hypothesized from one another, neither differed significantly in their emotional processing from a nonpsychopathic control group of offending youth (n = 208). These results are consistent with the possibility that the two variants of psychopathy, both of which were high on callous-unemotional traits, may have different etiological pathways, with the primary being more related to a deficit in the processing of distress cues in others and the secondary being more related to histories of abuse and emotional problems.
The influence of genetic variation on resting-state neural networks represents a burgeoning line of inquiry in psychiatric research. Monoamine oxidase A, an X-linked gene, is one example of a ...molecular target linked to brain activity in psychiatric illness. Monoamine oxidase A genetic variants, including the high and low variable nucleotide tandem repeat polymorphisms, have been shown to differentially affect brain functional connectivity in healthy humans. However, it is currently unknown whether these same polymorphisms influence resting-state brain activity in clinical conditions. Given its high burden on society and strong connection to violent behavior, antisocial personality disorder is a logical condition to study, since in vivo markers of monoamine oxidase A brain enzyme are reduced in key affect-modulating regions, and striatal levels of monoamine oxidase A show a relation with the functional connectivity of this same region.
We utilized monoamine oxidase A genotyping and seed-to-voxel-based functional connectivity to investigate the relationship between genotype and corticostriatal connectivity in 21 male participants with severe antisocial personality disorder and 19 male healthy controls.
Dorsal striatal connectivity to the frontal pole and anterior cingulate gyrus differentiated antisocial personality disorder subjects and healthy controls by monoamine oxidase A genotype. Furthermore, the linear relationship of proactive aggression to superior ventral striatal-angular gyrus functional connectivity differed by monoamine oxidase A genotype in the antisocial personality disorder groups.
These results suggest that monoamine oxidase A genotype may affect corticostriatal connectivity in antisocial personality disorder and that these functional connections may also underlie use of proactive aggression in a genotype-specific manner.
Psychopathy is a personality disorder that is strongly linked to criminal behavior. Using 18Ffallypride positron emission tomography and blood oxygen level-dependent functional magnetic resonance ...imaging, we found that impulsive-antisocial psychopathic traits selectively predicted nucleus accumbens dopamine release and reward anticipation-related neural activity in response to pharmacological and monetary reinforcers, respectively. These findings suggest that neurochemical and neurophysiological hyper-reactivity of the dopaminergic reward system may comprise a neural substrate for impulsive-antisocial behavior and substance abuse in psychopathy.

Borderline personality disorder (BPD) and antisocial personality disorder (ASPD) are the two most frequently diagnosed and researched DSM-5 personality disorders, and both are characterized by high ...levels of trait neuroticism. Fatty acid amide hydrolase (FAAH), an enzyme of the endocannabinoid system (ECS), has been linked to regulation of mood through modulation of anandamide, an endocannabinoid. We hypothesized that prefrontal cortex (PFC) FAAH binding would relate to trait neuroticism in personality disorders. Thirty-one individuals with personality disorders (20 with BPD and 11 with ASPD) completed the investigation. All participants completed the revised NEO Personality Inventory, which yields standardized scores (e.g., T scores) for the traits of neuroticism, openness, conscientiousness, agreeableness, and extraversion. All participants were medication free and were not utilizing illicit substances as determined by drug urinalysis. Additionally, none of the participants had a comorbid major depressive episode, bipolar disorder, psychotic disorder, or substance use disorder. Each participant underwent one
CCURB PET scan. Consistent with our hypothesis, neuroticism was positively correlated with PFC FAAH binding (r = 0.42, p = 0.021), controlling for genotype. Neuroticism was also positively correlated with dorsal putamen FAAH binding (r = 0.53, p = 0.0024), controlling for genotype. Elevated brain FAAH is an endophenotype for high neuroticism in BPD and ASPD. Novel pharmacological therapeutics that inhibit FAAH could emerge as potential new treatments for BPD and ASPD with high neuroticism.
Abstract Brain-imaging studies suggest that antisocial and violent behavior is associated with structural and functional deficits in the prefrontal cortex, but there is heterogeneity in findings and ...it is unclear whether findings apply to psychopaths, non-violent offenders, community-based samples, and studies employing psychiatric controls. A meta-analysis was conducted on 43 structural and functional imaging studies, and the results show significantly reduced prefrontal structure and function in antisocial individuals. Effect sizes were significant for both structural and functional studies. With minor exceptions, no statistically significant moderating effects of sample characteristics and methodological variables were observed. Findings were localized to the right orbitofrontal cortex, right anterior cingulate cortex, and left dorsolateral prefrontal cortex. Findings confirm the replicability of prefrontal structural and functional impairments in antisocial populations and highlight the involvement of orbitofrontal, dorsolateral frontal, and anterior cingulate cortex in antisocial behavior.

Emotional dysfunctions have long been associated with psychopathy. Yet, the extent to which these dysfunctions include problems in emotion regulation (ER) has only recently become clearer. In this study, we first reviewed theoretical and empirical literature on psychopathy and ER, and then examined associations between ER and psychopathy in four diverse samples from two countries (MTurk, college, community, and offender samples from the United States and Italy; total N = 1940). We employed Structural Equation Modeling (SEM) to examine latent associations between ER and measures of psychopathy and the dark triad, respectively. Results revealed a consistent pattern of moderate associations between psychopathy and poorer ER. These patterns were replicated across samples and psychopathy assessment, and held when including Machiavellianism and narcissism in the models. Replicating and extending recent studies, these findings provide incremental evidence that the emotional dysfunctions associated with psychopathy include problems in ER. Thus, we contend that prevention efforts in the general community, as well as clinical risk assessment and treatment planning in clinical (forensic) populations should include an emphasis on ER, as it may represent an important factor explaining some of the maladaptive correlates of psychopathic personality.
This study aimed to identify distinct subgroups of adults in a general population sample (N = 2,500; 52.6% females) based on their scores on three psychopathy dimensions. Using latent profile ...analysis, five groups were identified among males and females separately, including a psychopathic personality group. Multivariate analyses of variance showed that this latter group had higher levels of aggression, offending, substance use, attention-deficit/hyperactivity symptoms, internalizing problems, and maltreatment than most of the other groups. Associated features of males and females with a psychopathic personality were very similar; however, salient gender differences did emerge. Specifically, females with a psychopathic personality were more frequently exposed to sexual abuse, expressed more emotional difficulties, and engaged in higher levels of relational aggression. In conclusion, person-oriented analyses identified adults with a personality that looks like psychopathy, and furthered our understanding of gender similarities and differences in these adults.
Abstract Background The Dark Triad (DT) describes a set of three closely related personality traits, Machiavellianism, narcissism, and psychopathy. The aim of this study was to examine the ...associations between DT traits, sleep disturbances, anxiety sensitivity and intolerance of uncertainty. Method A total of 341 adults ( M = 29 years) completed a series of questionnaires related to the DT traits, sleep disturbances, anxiety sensitivity, and intolerance of uncertainty. Results A higher DT total score was associated with increased sleep disturbances, and higher scores for anxiety sensitivity and intolerance of uncertainty. In regression analyses Machiavellianism and psychopathy were predictors of sleep disturbances, anxiety sensitivity, and intolerance of uncertainty. Conclusions Results indicate that specific DT traits, namely Machiavellianism and psychopathy, are associated with sleep disturbances, anxiety sensitivity and intolerance of uncertainty in young adults.
