4OH and NaOH solutions for pH control. In this manner ammonium concentration was successfully controlled between inhibitory and growth limiting concentrations. In this fermentation, as in many industrial fermentations, the carbon nutrient was fed in an open loop, feedforward control scheme to maintain carbon-limited growth. However, such strategies are not robust and can fail if unplanned process perturbations occur as a result of sensor failures, feed system failures, and accumulation of inhibitory substances. In this fermentation, at-line NIR measurement of carbon nutrient (glycerol) and the inhibitory byproduct (acetate) provided a method for improving such open loop feed forward control by incorporating a feed back component. The improved control of ammonium, acetate and glycerol facilitated a biomass accumulation to 86gl
−1. This is believed to be the highest biomass concentration reported for a recombinant protein producing
E. coli fermentation with a complex medium feed. At-line NIR analysis was also used to determine and identify bioreactor faults by detecting a DO probe failure and a pH controller failure.
Les procédés chromatographiques multicolonnes continus se sont imposés comme technique séparative dans l’industrie pharmaceutique en raison de leur productivité élevée et de leur développement ...rapide. De nos jours, la modélisation, le dimensionnement et l’optimisation des procédés LMS et Varicol sont considérés comme établis. Le fonctionnement automatique optimisé et robuste de ces procédés reste cependant un sujet de recherche. Le plus souvent, les paramètres opératoires sont choisis en deçà du réglage optimal afin d’inclure une marge de robustesse. Ils sont ajustés manuellement par un opérateur expérimenté pour maintenir les produits à leurs spécifications de pureté. Le nombre élevé de séparations chromatographiques rend crucial le développement d’une application d’un contrôle avancé de ces procédés. Récemment, plusieurs méthodes de contrôle des procédés LMS ont été proposées. Ce travail de thèse présente une nouvelle approche de contrôle avancé. Cette commande ajuste les paramètres opératoires d’une séparation grâce à deux types de mesure : une mesure en ligne et une mesure hors ligne. Une validation expérimentale du schéma de contrôle a été effectuée sur un procédé Varicol. Les résultats présentés démontrent que la commande permet d’amener la pureté des produits à leurs spécifications, d’optimiser les performances du procédé et de répondre aux perturbations, tout en sécurisant la pureté du produit cible
Multicolumn continuous chromatographic processes became a key separation technology in the areas of pharmaceutical industry thanks to high productivity and short process development times. Today, modeling, design, and optimization of SMB and Varicol, are well established. However a robust and optimized operation of processes is still an open issue. The common practice is to operate processes under suboptimal operating conditions in order to gain the necessary robustness. The operating parameters are tuned manually by experienced operators in order to maintain the product specifications. Therefore, as chromatographic applications spread, process control problem becomes increasingly important. Recently, several control methodologies of SMB process have been proposed. This thesis work introduces a new advanced control system approach. This system adjusts operating parameters thanks to two kinds of measurements : an in-line measurement and an at-line measurement. The performance of this control scheme is demonstrated through several experiments on Varicol. The reported results aim to demonstrate that the controller is able to deliver the products within the specifications, to optimize the process performance and to answer to disturbances while protecting the key product
Under the expectation that the behavior of a spin glass (SG) system shows various aspects depending on the time- and space-scale of observations, we have studied the typical Ising spin glass ...Fe0.50Mn0.50TiO3 by measuring dc-magnetization, ac-susceptibility, Mössbauer spectrum, neutron scattering intensity and μ+SR time spectrum. We have demonstrated that the application of various methods with different time- and space-scale of observations to the same system is useful to clarify the property of the spin glass freezing, the Ising character and the de Almeida and Thouless-line.
Sample clarification is a common operation in biochemical analytical methods for removing interfering or unwanted particulates from an analyte sample. Filtration provides one option for removal of ...particulates. However, in many cases the loss of soluble protein due to filter adsorption is unacceptable and an alternative must be sought. In this paper a microcentrifuge designed to automatically sample, spin, deliver supernatant to an analyser and wash out solids from the bowl is described. The performance of the system is assessed in terms of its clarification efficiency and the time required to achieve satisfactory clarification. Additionally, the effects of different protein precipitating agents on yeast homogenate samples separated using the microcentrifuge are studied where the system is used to deliver supernatant to a flow injection analyser. The paper demonstrates that the microcentrifuge may be used to separate rapidly such samples on a time scale between 10–60 s depending upon the type and size of sample and be successfully used as a component of an at-line monitoring system.
Near-infrared spectrophotometric analysis is a rapid technique that typically uses the reflectance of a solid sample at several wavelengths to determine the sample's composition. A computerized ...modeling process is generally used to correct for background and sample-matrix interferences. The modeling process employs a training set of samples to, in effect, "teach" the computer to recognize relationships between minute spectral features and sample composition. The contents of the training-set samples must be determined initially by some other reference method before applying the near-IR technique. The model developed from near-IR spectra and reference values gives the sample composition using a number of linear equations. Each of these equations expresses a particular component concentration as a weighted sum of the signals observed at a number of near-IR wavelengths. Instruments used for near-IR spectrophotometry can be as simple as a filter photometer or a grating monochromator. The broad spectral peaks and highly correlated wavelength vectors generally limit the number of wavelengths used in the model. Little or no sample preparation is required by near-IR methods, and many solid samples can be directly analyzed. Near-IR spectrophotometry has found application in agriculture, industry, biology, medicine, and even satellite remote sensing.
Les industries alimentaires disposent de peu de capteurs instrumentaux rapides et peu coûteux permettant d'évaluer, en cours de transformation, les caractéristiques sensorielles des produits. Les ...évaluations réalisées par les opérateurs directement sur la ligne de fabrication sont ainsi souvent utilisées pour déterminer certaines propriétés des produits, suivre leur évolution au cours de la transformation et, le cas échéant, proposer une rétroaction sur ce procédé. Cet article présente une méthode de formalisation de ces évaluations. La mesure sensorielle descriptive classique a été adaptée afin de prendre en compte les spécificités de l'évaluation en ligne: c'est une mesure temps réel, réalisée généralement par un seul opérateur avec auto-entraînement et autocontrôle des performances, sur des produits qui ne sont pas, a priori, anonymes. Ces spécificités ont amené à développer le concept d'indicateur sensoriel et à proposer une grille de formalisation permettant de le décrire. Cette grille comporte sept éléments. Cinq proviennent de la démarche descriptive classique: nom, définition, mode opératoire, échelle de mesure, références. Deux éléments supplémentaires ont été introduits: une caractéristique d'espace pour tenir compte de la variabilité des produits et une caractéristique de temps qui prend en compte les modifications du produit au fur et à mesure de la transformation. Cette méthode a été appliquée sur une étape de la fabrication du saucisson sec, l'étuvage. Sept indicateurs sensoriels ont été identifiés et structurés sur la base de la connaissance détenue par un opérateur entraîné. Cette connaissance a été transmise vers un opérateur-observateur au cours de la formalisation. Une évaluation des performances a montré que ces deux opérateurs présentaient un bon accord entre eux. Il est ainsi possible de formaliser les évaluations réalisées sur la ligne de fabrication et de les transmettre à un nouvel opérateur. La mesure humaine en ligne peut être employée comme variable d'entrée dans des boucles de rétroaction
