Colostrum replacement products for use in goat kids are sourced from bovine colostrum and often used by producers to supplement or replace maternal colostrum to prevent infections. To compare the ...colostrum replacement products fed on-farm to caprine colostrum a cross-sectional study was undertaken. Ontario dairy goat producers were asked to collect first milking colostrum from their goats and samples of the reconstituted commercial replacement product currently in use. The frozen samples were thawed and submitted for testing of fat, protein and lactose content, IgG1 concentration and aerobic bacterial culture. Compared with caprine colostrum, the reconstituted replacement products were lower in protein (11.7%; P = 0.0007), and fat (4.6%; P < 0.0001) and higher in lactose (5.4%; P < 0.0001) on average. The average IgG1 concentration in goat colostrum (53.5 g/L; range: 16.6–1985.8) was significantly higher than in colostrum replacement products (33.7 g/L; range: 10.7–55.3) (P < 0.0001). The Brix cut-point for good quality goat colostrum (50 g/L) was calculated at 23% (sensitivity = 69.6%, specificity = 88.0%) for goat colostrum and 26% for the colostrum replacement product (sensitivity = 87.5%, specificity = 100%). The average aerobic count for goat colostrum was lower (2.95 log10 cfu/mL) than the colostrum replacement product samples that were cultured (3.85 log10 cfu/mL; P < 0.0001). Further investigation into colostrum replacement products, including on-farm storage of opened powdered product and mixing and storage of reconstituted product, is warranted. Variability in the levels of IgG1, aerobic bacterial growth and fat, protein and lactose content in colostrum replacement products also requires further exploration to determine their effects on kid health.

Colostrum management is the single most important management factor in determining calf health and survival. Additional benefits of good colostrum management include improved rate of gain and future ...productivity. Successful colostrum management requires producers to provide calves with a sufficient volume of clean, high-quality colostrum within the first few hours of life. This article reviews the process of colostrogenesis and colostrum composition, and discusses key components in developing a successful colostrum management program. In addition, the article discusses approaches for monitoring and proposes new goals for passive immunity in dairy herds.
Acquisition of high quality colostrum is an important factor influencing neonatal calf health. Many methods have been used to assess the Ig concentration of colostrum; however, improved, validated ...evaluation tools are needed. The aims of this study were to evaluate both optical and digital Brix refractometer instruments for the measurement of Ig concentration of colostrum as compared with the gold standard radial immunodiffusion assay laboratory assessment and to determine the correlation between Ig measurements taken from fresh and frozen colostrum samples for both Brix refractometer instruments. This research was completed using 288 colostrum samples from 3 different farms. It was concluded that the optical and digital Brix refractometers were highly correlated for both fresh and frozen samples (r=0.98 and r=0.97, respectively). Correlation between both refractometer instruments for fresh and frozen samples and the gold standard radial immunodiffusion assay were determined to be very similar, with a correlation coefficient between 0.71 and 0.74. Both instruments exhibited excellent test characteristics, indicating an appropriate cut-off point of 22% Brix score for the identification of good quality colostrum.
A calf should receive 10–12 % of its bodyweight in high quality colostrum ( >50 g/L immunoglobulin) shortly after birth in order to confer passive immunity, with calves ideally receiving their first ...feed of colostrum in the first 1–2 hours of life (Godden et al., 2019). It is recommended (not validated) that total counts for bacteria and coliforms in colostrum should not exceed 100,000 colony forming units per mL (CFU/mL) and 10,000 CFU/mL, respectively. This study was conducted between March and September 2023. Eighty-five colostrum samples from 5 commerical dairy farms in Dumfries and Galloway in Scotland were purposively selected. Samples were collected from various stages during the colostrum harvest, storage and feeding process to provide multiple samples with variable (low, medium and high) bacterial contamination. The objective of this study was to validate the diagnostic accuracy of the 3 M Petrifilm™ Coliform Count Plate (CCP) and Aerobic Count Plate (ACP) for colostrum bacteriology. The Petrifilm™ Aerobic Count Plates were compared to 5 % sheep blood agar (SBA) (total bacteria counts, TBC), and the Petrifilm™ Coliform Count Plates were compared to MacConkey plates (MAC) (total coliform counts, TCC) and test sensitivity and specificity were calculated.
Colostrum bacterial contamination was highly variable between farms (TBC median and interquartile range= 50,000CFU/mL and 546000CFU/mL; TCC median and interquartile range=100,750CFU/mL and 188,500CFU/mL). Overall correlation between the Petrifilms™ and reference plate measures was moderate (Spearman rho=0.38–0.47). The statistical accuracy of the Petrifilm™ systems and the test sensitivity were improved by lowering the bacterial cutpoint from 100,000CFU/mL to 91,000 CFU/mL for the aerobic counts plates and from 10,000CFU/mL to 1500 CFU/mL for the coliform count plates. The Petrifilm™ system could be used as a monitoring tool to provide quick and statistically accurate results for TBC and TCC but should not be seen as a comprehensive solution for poor colostrum hygiene.
•Petrifilms™ could provide quick and accurate results for monitoring bacterial contamination of bovine colostrum.•Petrifilms™ could conceivably be applied successfully in an in-house veterinary laboratory.•Petrifilm™ sensitivity was improved by lowering test cutpoints to 1500 CFU/mL for coliforms and 91000 CFU/mL for total aerobic bacteria.

Bovine milk and colostrum provide essential nutrients and immunologically active factors that are beneficial to a newborn calf. Milk-and-colostrum-derived exosomes are known as the most important for cellular communication. Exosomes also contain non-coding RNA, such as microRNA. However, there is limited information about exosomal miRNA derived from the milk and colostrum of Holstein and DAK cattle. This study aimed to identify and characterize the exosomal microRNA in the milk and colostrum of Holstein and Doğu Anadolu Kirmizisi (DAK) cows. For this purpose, total RNA isolation was carried out on the milk and colostrum samples that were collected from the Holstein and DAK cattle breeds. The RNA samples were subjected to RNA sequencing and the microRNAs were determined. Lastly, gene ontology analysis was performed for target genes. A total of 795 miRNAs that were expressed differently were identified. A total of 545 of these were known miRNAs and 260 were found to be novel miRNAs. In the functional enrichment analysis, the miRNAs expressed in Holstein milk were mostly associated with milk synthesis, and those in colostrum were mostly involved in the immunity pathways. It was also observed that the miRNAs expressed in DAK milk regulated milk fat and protein metabolism, and there were miRNAs that regulated immune pathways in the colostrum. In addition to this, many novel miRNAs were defined in DAK colostrum. When the target genes of exosomal miRNA in Holstein and DAK milk and colostrum were compared, it was suggested that the DAK breed had genes that were mostly associated with the immune system. As a result, the data obtained from this study will provide beneficial contributions to potential miRNA biomarker studies for milk yield and mastitis.
The objective of this study was to investigate the effects of heat treatment on colostral low-abundant proteins, IgG and IgA, insulin, and insulin-like growth factor I (IGF-I), as well as bacteria ...and somatic cells. First-milking colostrum samples >8 L and Brix % > 22.0 were harvested from 11 Holstein cows on a commercial dairy in New York State and split into 2 aliquots using single-use colostrum bags. One aliquot of each pair was cooled on ice immediately after harvest (raw, R; n = 11), and the other was heat-treated for 60 min at 60°C (heat, H; n = 11). All samples were analyzed for IgG and IgA via radial immunodiffusion assay and insulin and IGF-I concentrations by radioimmunoassay. Total bacterial counts and somatic cell counts (SCC) were determined using standard plate culture techniques and flow cytometry, respectively. Samples from a subset of 5 pairs (n = 10) were further analyzed by nano liquid chromatography-tandem mass spectroscopy, after ultracentrifugation at 100,000 × g for 60 min at 4°C to enrich the low-abundant protein whey fraction. Data were analyzed using either paired t-test or Wilcoxon signed-rank test or using an online software package to analyze proteomics data. Outcomes of proteomics analysis were fold change ≥1.5 between pairs, and paired t-tests with false discovery rate–adjusted P-value < 0.05. The median reduction of IgA concentrations was 8.5% (range: 0–38.0%) due to heat treatment, whereas IgG concentrations did not change due to treatment. Insulin concentrations decreased by a median of 22% (7–45%), and IGF-I decreased by 10% (0–18%) in H samples. Heat treatment was associated with a median reduction of SCC of 36% (0–90%) in paired samples, as well as a median reduction in total bacterial count of 93% (45–100%) in H versus R samples. Proteomics analysis identified a total of 328 unique proteins that were present in all 10 samples. Nine of the 25 proteins that decreased by at least 1.5-fold in H compared with R were identified as complement proteins. We conclude that heat treatment of colostrum is associated with a reduction in the concentration of bacterial counts and SCC, IgA, insulin, and IGF-I. In addition, proteomics analysis of colostral whey identified several complement components and other proteins that decreased in abundance due to heat treatment. Although IgG concentrations were unaffected and a reduction in bacterial counts was achieved, the change in several immunologically active proteins and growth factors may have biologically important effects on the developing immune system of the neonate fed heat-treated colostrum.

Our objectives were to determine the effect of oxytocin ...use during colostrum harvest on colostrum yield and IgG concentration in Holstein dairy cows on a commercial dairy in New York and to describe associations of cow characteristics with these outcomes. Animals were enrolled between July and October 2023 using a randomized block design, with day of enrollment as the unit of randomization. A median (range) of 10 (3–19) cows were enrolled per day. Treatments were (1) 40 IU oxytocin (OXY40), (2) 20 IU oxytocin (OXY20), and (3) an untreated control group (CNTR). Oxytocin was administered i.m. ∼45 s before unit attachment in a rotary parlor. Colostrum weight was measured using the colostrum bucket-embedded scale or a platform scale when the yield was less than the smallest bucket scale. The concentration of IgG (IgG) in colostrum was determined using radial immunodiffusion and used to calculate total IgG, and DM (%) was determined by oven drying. Individual cow characteristics such as parity, calf sex, weight, dystocia score, stillbirth, and milk production in wk 4 of lactation; additionally, for multiparous cows, dry period length and previous lactation dry-off linear SCS were collected. Data were analyzed separately for primiparous and multiparous cows using backward stepwise elimination. To produce final mixed effects ANOVA models, primiparous cows (n = 201) were randomized to 35.8% (n = 72) OXY40, 32.8% (n = 66) OXY20, and 31.3% (n = 63) CNTR. Multiparous groups (n = 435) were randomized to 34.7% (n = 151) OXY40, 29.7% (n = 129) OXY20, and 35.6% (n = 155) CNTR. The median (range) colostrum yield was 6.0 (0–20.6) kg and IgG was 98.5 (0.1–293.6) g/L in the study population. In primiparous cows, OXY40 had a higher colostrum yield (LSM 95% CI) of 5.4 (4.9–5.9) kg compared with both OXY20 (4.1 3.5–4.7 kg) and CNTR (3.8 3.3–4.3 kg). In multiparous cows, OXY40, OXY20, and CNTR did not differ in colostrum yield (5.9 5.3–6.5, 5.7 5.1–6.3, and 5.4 4.8–6.0 kg, respectively), but colostrum yield was greater in parity 2 compared with all other parities, cows giving birth to male calves, cows with the highest milk production at wk 4 of lactation, and cows with a dry period of >65 d. Oxytocin use did not affect IgG in either primiparous or multiparous cows, but IgG was highest in cows in parity ≥4 and lowest in cows dry >65 d. In summary, oxytocin use at 40 IU i.m. was associated with a higher colostrum yield in primiparous cows only and no change in IgG. Oxytocin use likely addressed disturbed milk ejection and therefore increased colostrum yield in heifers milked for the first time in a rotary parlor. This study confirms cow characteristics associated with colostrum production within a single herd.
The objective of this study was to characterize the quality of maternal colostrum (MC) fed to newborn dairy calves in the United States and identify the proportion of MC that meets industry standards ...for IgG concentration and total plate count (TPC). Samples of MC (n=827) were collected from 67 farms in 12 states between June and October 2010. Samples were collected from Holsteins (n=494), Jerseys (n=87), crossbred (n=7), and unidentified dairy cattle (n=239) from first (n=49), second (n=174), third or greater (n=128), and unknown (n=476) lactations. Samples were identified as fresh (n=196), refrigerated (n=152), or frozen (n=479) before collection, as well as whether the sample was from an individual cow (n=734) or pooled (n=93). Concentration of IgG in MC ranged from <1 to 200mg/mL, with a mean IgG concentration of 68.8mg/mL (SD=32.8). Almost 30% of MC contained <50mg of IgG/mL. The IgG concentration increased with parity (42.4, 68.6, and 95.9mg/mL in first, second, and third and later lactations, respectively). No differences in IgG concentration were observed among breeds or storage method; however, IgG was highest in samples collected in the Midwest and lowest in samples collected in the Southwest (79.7 vs. 64.3mg/mL). Total plate count of samples ranged from 3.0 to 6.8 log10 cfu/mL, with a mean of 4.9 log10 cfu/mL (SD=0.9) and was greater in samples collected in the Southeast compared with other regions of the country. Pooled samples had greater TPC than individual samples and refrigerated samples had greater TPC than frozen and fresh samples. Almost 43% of samples collected had TPC >100,000cfu/mL, 16.9% of the samples had >1million cfu/mL. Only 39.4% of the samples collected met industry recommendations for both IgG concentration and TPC. Almost 60% of MC on dairy farms is inadequate, and a large number of calves are at risk of failure of passive transfer or bacterial infections, or both. Also, the data indicate that regional differences exist in colostrum quality.
Refractometry using a Brix refractometer has been proposed as a means to estimate IgG concentration in bovine maternal colostrum (MC). The refractometer has advantages over other methods of ...estimating IgG concentration in that the Brix refractometer is inexpensive, readily available, less fragile, and less sensitive to variation in colostral temperature, season of the year and other factors. Samples of first-milking MC were collected from 7 dairy farms in Maine, New Hampshire, Vermont, and Connecticut (n=84) and 1 dairy farm in California (n=99). The MC was milked from the cow at 6.1±5.6h postparturition and a sample was evaluated for Brix percentage by using an optical refractometer. Two additional samples (30mL) were collected from the milk bucket, placed in vials, and frozen before analysis of total IgG by radial immunodiffusion (RID) using commercially available plates and by turbidimetric immunoassay (TIA). The second sample was analyzed for total bacterial counts and coliform counts at laboratories in New York (Northeast samples) and California (California samples). The Brix percentage (mean ± SD) was 23.8±3.5, IgG concentration measured by RID was 73.4±26.2g/L, and IgG concentration measured by TIA was 67.5±25.0g/L. The Brix percentage was highly correlated (r=0.75) with IgG analyzed by RID. The Brix percentage cut point to define high- or low-quality colostrum (50g of IgG/L measured by RID) that classified more samples correctly given the proportion of high- (86%) and low-quality (14%) samples in this study was 21%, which is slightly lower than other recent estimates of Brix measurements. At this cut point, the test sensitivity, specificity, positive and negative predictive values, and accuracy were 92.9, 65.5, 93.5, 63.3, and 88.5%, respectively. Measurement of IgG by TIA correlated with Brix (r=0.63) and RID (r=0.87); however, TIA and RID methods of IgG measurement were not consistent throughout the range of samples tested. We conclude that Brix measurement of total solids in fresh MC is an inexpensive, rapid, and satisfactorily accurate method of estimating IgG concentration. A cut point of 21% Brix to estimate samples of MC >50g/L was most appropriate for our data. Measurement of IgG in MC by TIA differed from measurement by RID.
Passive immunity in calves is evaluated or quantified by measuring serum or plasma IgG or serum total protein within the first 7 d of age. While these measurements inform about circulating ...concentrations of this important protein, they are also a proxy for evaluating all of the additional benefits of colostral ingestion. The current individual calf standard for categorizing dairy calves with successful passive transfer or failure of passive transfer of immunity are based on serum IgG concentrations of ≥10 and <10 g/L, respectively. This cutoff was based on higher mortality rates in calves with serum IgG <10 g/L. Mortality rates have decreased since 1991, but the percentage of calves with morbidity events has not changed over the same time period. Almost 90% of calves sampled in the USDA National Animal Health Monitoring System's Dairy 2014 study had successful passive immunity based on the dichotomous standard. Based on these observations, a group of calf experts were assembled to evaluate current data and determine if changes to the passive immunity standards were necessary to reduce morbidity and possibly mortality. In addition to the USDA National Animal Health Monitoring System's Dairy 2014 study, other peer-reviewed publications and personal experience were used to identify and evaluate potential standards. Four options were evaluated based on the observed statistical differences between categories. The proposed standard includes 4 serum IgG categories: excellent, good, fair, and poor with serum IgG levels of ≥25.0, 18.0–24.9, 10.0–17.9, and <10 g/L, respectively. At the herd level, we propose an achievable standard of >40, 30, 20, and <10% of calves in the excellent, good, fair, and poor categories, respectively. Because serum IgG concentrations are not practical for on-farm implementation, we provide corresponding serum total protein and %Brix values for use on farm. With one-third of heifer calves in 2014 already meeting the goal of ≥25 g/L serum IgG at 24 h of life, this achievable standard will require more refinement of colostrum management programs on many dairy farms. Implementation of the proposed standard should further reduce the risk of both mortality and morbidity in preweaned dairy calves, improving overall calf health and welfare.
