U terapijskom polju je izmjeren spektar plazma elektrode u području 540-940 nm (atomske
linije Ne I i O I) dok se kod izboja kroz zrak bilježi i spektar mikro-izboja u području
290-440 nm (molekularne linije N2 i N2+). Na uzorku tkiva su izmjerene vrijednosti
efektivnog napona (22-95 mV), i efektivne struje (0,22-0,95 mA) na osnovu kojih su
procijenjene vrijednosti gustoće struje (4-19 μA/mm2) kao i snage (5-65 μW) te predane
energije u ovisnosti o vremenu izlaganja (0,14-10,75 mJ kod trajanja zahvata 30-
120 sekundi). Kod oba modaliteta primjene, u frekvencijskom području 33 kHz, zabilježena je
jakost magnetskog polja (1-3,07 A/m) i gustoće magnetskog toka (1,25-3,86 μT). Prilikom
pojave mikro-izboja u zračnom međuprostoru, u frekvencijskom području iznad 40 MHz, se
bilježi magnetsko polje jakosti (5-49 mA/m) te gustoće magnetskog toka od (6,5- 62,7 nT).
Jakost električnog polja na udaljenosti od 10 mm od elektrode iznosi 248-586 V/m. Tijekom
provedbe zahvata zabilježene su značajne promjene u temperaturi površine tkiva te one
ovisno o intenzitetu i trajanju zahvata (30-120 sek) iznose od 2.6 do 12,2°C. Statistička
analiza rezultata ukazala je na značajnu razliku u vrijednostima fizičkih veličinama s obzirom
na dva modaliteta primjene.
Na osnovu navedenih dokaza se zaključuje da djelovanje ispitivanog uređaja ne bi trebalo
pripisivati isključivo ozonu, kao što je to dosad bio slučaj, već kumulativnom učinku svih,
ovim istraživanjem dokazanih, fizičkih i kemijskih čimbenika koji su tijekom zahvata prisutni
u terapijskom polju.- A high-frequency ozone generator Ozonix (Biozonix GmbH, Germany) is classified as a
medical ozone generator, thus its therapeutic effects have, so far, been attributed exclusively
to ozone effect. The operative principle of the device and ozone production through a
dielectric barrier discharge using glass electrodes filled with a noble gas requires presence of
additional physical and chemical factors, besides ozone, in the treatment field during the
procedure. The purpose of the study was to determine the physical and chemical properties in
the treatment field of the said device. For that purpose, measuring of emission spectra, electric
and magnetic values as well as changes in surface temperature on tissue samples, as functions
of the selected intensity and exposure time, was conducted, and two basic application
modalities (contact discharge and air gap discharge) were used.
Materials and Methods: Optical emission spectroscopy was used to record the spectrum of a
plasma electrode’s glow discharge, a micro discharge occurring in the aerial interspace (air
gap) between the electrode and the targeted surface and the spectrum of the discharge through
a liquid medium on the surface of the tissue sample. By measuring electromagnetic values
present in the treatment field, physical values of electric and magnetic fields were recorded, as
well as currents passing through the treatment field. Thermographic measurements were used
to record temperature changes at different intensities (10-100%) and exposure durations (30,
60, 90 and 120 seconds).
Results: In the treatment field, two main emission spectra were recorded. Spectral lines of
plasma electrode’s glow discharge were recorded in the optical range between 540 and
940 nm, which were identified as atomic lines of neon (Ne I) and oxygen atoms (O I). This
spectrum is observed in all treatment modalities. During the formation of micro-discharges in
the air gap between the electrode and the treated surface, spectral lines were recorded in the
optical range between 290 and 440 nm, which were identified as molecular lines of N2 and N2+.
On the tissue samples (pork skin), as a function of output intensity (10-100%), electrical
values were recorded by use of oscilloscope and low impedance (Uin = 100) probe: RMS
voltage (22-95 mV), RMS current (0,22- 0.95 mA). These values were used to calculate
current density (4-19 μA/mm2) as well as power (5-65 μW). Using the same duration periods
as the ones used in the examination of temperature change at the tissue surface (30-120 sec),
the values of committed (delivered) energy were estimated (0,14-10,75 mJ).
Magnetic fields were recorded at two frequency ranges. In the frequency range of 33 kHz,
magnetic field was recorded in both treatment modalities as a function of intensity (10-100%)
for direct contact discharge H(1,93 ± 0,59 A/m), B (2,43 ± 0,75 μT), f (32151 ± 985Hz) and
air gap discharge H (2,5 ± 0,52 A/m), B (3,14 ± 0,65 μT), f (34526 ± 833 Hz). Only in the
case of micro-discharge formation in the air gap were additional peaks of magnetic field
observed in a wide frequency range from 40 to above 400 MHz. Due to the low intensity,
magnetic field values were calculated only for 41.5 MHz peak, where H field was
0,032 ± 0,014 A/m and B field was 40 ± 17,4 nT. At 10 mm distance from plasma electrode
surface, electric field strength was recorded in the range from 248 to 586 V/m depending on
the selected intensity (10-100%). During the treatment, a mild increase of temperature on the
surface of the tissue samples was recorded (2,6-12,2°C) as a function of intensity (20-100%)
and exposure time (30-120 seconds).
Conclusion: The results of the conducted research confirm the presence of mildly ionized
cold atmospheric plasma in the treatment field during air gap discharge. In both treatment
modalities, the presence of light in visible red and near infrared spectra, electric field,
magnetic fields, the currents flowing through the tissue and mild heat generated on the surface
of the tissue sample were confirmed in the treatment field. Furthermore, a significant
difference was determined in the values of physical properties with respect to the application
modality, where the values of temperature increase and magnetic field strengths are
significantly higher in the air gap modality whilst the values of electric current (and other
derived values) are lower compared to direct contact modality.
According to the results of this research, it can be concluded that treatment effects with the
tested devices must be attributed not only to ozone, but to cumulative effects of all physical
and chemical factors, the presence of which was recorded in the treatment field during this

Zbog stila života proizvodnja različite vrste otpada u kućanstvu i industriji je u konstantnom porastu. Kako bi se smanjila količina otpada, koja svoj životni ciklus završi kao smeće na odlagalištima ...radeći ekološki problem sadašnjoj i budućim generacijama, važno je pravilno razvrstavanje i odlaganje kako bi se mogao koristiti kao sirovina. Plastika je nezamjenljivi materijal današnjice u područjima pakiranja, transporta, građevinarstva, medicine i elektronike. Zbog svojih svojstava i cijene koristi se u različite svrhe i praktički ne možemo zamisliti život bez nje što rezultira ogromnim količinama producirane plastike. Nesavjesno postupanje s plastičnim otpadom dovodi do onečišćenja okoliša, a posebice mora i oceana. Dugotrajna razgradnja plastičnih materijala je veliki problem, a dodatni problem je što njenom razgradnjom nastaje mikroplastika koja dospijeva u vodu, zemlju i zrak, a time i u prehrambeni lanac. Stoga je recikliranje najbolja opcija gospodarenja plastičnim otpadom. Oznaka otisnuta ili utisnuta na plastičnoj ambalaži daje informaciju o vrsti plastičnog materijala i mogućnosti reciklaže za određene svrhe. Mehaničkim postupcima plastični otpad se pretvara u reciklat koji se može koristiti za nove plastične proizvode. Plastični materijali zbog svojih svojstava i mogućnosti oblikovanja imaju gotovo neograničene mogućnosti te ih je moguće prilagoditi zahtjevima tržišta, a u današnje digitalno doba manualnu i tvorničku proizvodnju sve više zamjenjuje tehnologija 3D tiska usmjerena na izradu raznih proizvoda od polimernih materijala odnosno plastike. Ovaj rad obuhvaća pregled aktualnih mogućnosti u 3D tisku, te razmatra pristup pretvaranja plastičnog otpada u 3D filamente koji bi doprinjeli nužnom smanjenju plastičnog otpada.
Le processus de filamentation découle de la balance dynamique de plusieurs effets linéaires et non-linéaires, permettant l'auto-stabilisation du faisceau laser sur des distances de propagation ...significativement supérieures à celles prédites par la théorie des faisceaux gaussiens. Cette thèse s'attache à décrire ce phénomène dans les gaz rares en modèle dit de champ fort, par opposition au modèle usuel, utilisé dans le cadre de la théorie des perturbations et s'appuyant sur l'approximation du champ faible. L'influence des harmoniques, jusqu'ici négligée, est alors revisitée tant au niveau microscopique qu'à l'échelle d'une propagation. Il est montré, expérimentalement et par l'appui de simulations numériques, qu'il suffit d'une faible proportion de troisième harmonique pour que la dynamique de la filamentation soit fortement impactée, notamment à travers le processus d'ionisation. Egalement, la proximité d'une résonance peut mener à de fortes modifications du mécanisme d'auto-guidage: lorsqu'un champ fondamental centré à 400nm et sa troisième harmonique interagissent avec du krypton, il est montré que le processus de modulation de phase croisée est renforcé et qu'il peut alors endosser un rôle défocalisant de manière non-négligeable. Une autre situation - à 300nm dans le krypton - amène à un renforcement résonant de la filamentation, se traduisant par l'allongement de la longueur du filament par rapport au cas hors-résonance.
Filamentation originates from the dynamic balance between linear and nonlinear effects, allowing the laser beam autostabilization over distances significantly greater than those predicted by linear optics laws. The aim of this thesis is to describe this phenomenon on rare gases under the strong field model, contrary to the commonly used perturbation model based on the weak field approximation. The influence of harmonics, so far neglected, is then revisited both at the microscopic level and at a propagation scale. It is shown, experimentally and with the support of numerical simulations, that a weak proportion of third harmonic is sufficient to strongly impact the filamentation dynamics, especially through the ionization process. Also, the vicinity of a resonance can lead to strong modifications of the self-guiding mechanism: when a fundamental field centered at 400nm and its third harmonic interact together with krypton, it is shown that the cross-phase modulation process is enhanced and can participate to the beam defocusing. Another situation - at 300nm in krypton - leads to resonantly enhanced filamentation, i.e. to longer filaments compared to the non-resonant case.
Candida albicans est une levure pathogène opportuniste de l’homme. La capacité de C. albicans à changer de forme en réponse à des stimuli externes, passant d’une croissance bourgeonnante à ...filamenteuse, est associée à sa virulence. Cette morphogenèse requiert une réorganisation du cytosquelette d’actine et un trafic membranaire ciblé. Chez Saccharomyces cerevisiae, les petites protéines G de type Arf jouent un rôle important dans le trafic membranaire et la polarité cellulaire. Le rôle de ces protéines chez C. albicans est largement méconnu. C. albicans a 3 protéines Arf, Arf1-Arf3 et 2 Arf-like, Arl1 et Arl3. Nos résultats indiquent que seule Arf2 est nécessaire à la viabilité et à la résistance aux antifongiques, et qu’Arf2 et Arl1 sont critiques pour la croissance filamenteuse hyphale; le mutant arl1/arl1 en particulier forme des hyphes 2 fois plus courtes que la souche sauvage. Les mutants Δ/pTetARF2 et arl1/arl1 ont un défaut de virulence dramatique et ARL1 est particulièrement critique pour la candidose oropharyngée. Nos résultats indiquent que les défauts du mutant Δ/pTetARF2 seraient dus à une altération du Golgi, et ceux d’arl1/arl1 de l’incapacité de ce mutant à restreindre sa croissance à un site unique. Ce défaut de croissance polarisée du mutant arl1/arl1 n’est pas lié à la mislocalisation de son effecteur Imh1, ni à une misrégulation de la phosphatidylsérine flippase Drs2. Par contre, nos données suggèrent que le défaut de croissance hyphale de ce mutant résulterait d’une hypersécrétion. Cette étude nous a permis d’identifier Arf2 et Arl1 comme protéines clés du trafic membranaire, critiques pour la croissance filamenteuse et la virulence de C. albicans.
The human fungal pathogen Candida albicans switches from budding to filamentous growth. This dramatic morphogenesis is critical for its virulence and requires sustained polarized growth, via exocytosis and endocytosis, as well as reorganization of intracellular compartments. In the yeast Saccharomyces cerevisiae, Arf G-proteins and their regulators function at the interface of membrane traffic and cell polarity. The roles of this class of proteins during the transition to filamentous growth and virulence in C. albicans are largely unknown. In C. albicans there are 3 Arf proteins, Arf1-Arf3 and 2 Arf-like proteins, Arl1 and Arl3. Our results reveal that only Arf2 is required for viability and sensitivity to antifungal drugs and that both Arf2 and Arl1 are required for hyphal growth, with arl1/arl1 hyphal filaments being 2-fold shorter than that of the wild-type strain. Furthermore, both Δ/pTetARF2 and arl1/arl1 mutants have drastically reduced virulence, with ARL1 particularly critical for oropharyngeal candidiasis. We show that the defects in Δ/pTetARF2 is due to an alteration of Golgi integrity, while the defects in the arl1 mutant are likely to result from the inability of this mutant to restrict growth to a single site. Further analyses of the arl1/arl1 mutant revealed that this defect does not result from a misregulation of the GRIP-domain golgin coiled-coil tethering protein Imh1 nor of the phosphatidylserine flippase Drs2. Rather, our results suggest that the arl1/arl1 hyphal growth defect results from increased secretion. Together our work identifies Arf2 and Arl1 as key regulators of membrane traffic, critical for hyphal growth and virulence.
Les instabilités de type Weibel naissent si la distribution des vitesses du plasma présente une anisotropie. Elles entraînent la génération d’un champ magnétique dû à la formation de filaments de ...courant ainsi qu’une activité électrostatique importante. Ces phénomènes de base apparaissent dans de nombreuses situations, naturelles (vent solaire, jets relativistes) ou expérimentales (interaction laser-plasma) : les plasmas dans lesquels ils naissent peuvent être relativistes ou non, magnétisés ou non, collisionnels ou non, ce qui pose la question du choix du modèle à utiliser pour les décrire. La théorie cinétique est le cadre le plus complexe dans lequel nous travaillerons. De par sa complexité, il est intéressant de développer des modèles réduits. Un premier travail mené au cours de cette thèse est l’utilisation d’un modèle fluide incluant la dynamique du tenseur de pression pour modéliser la phase linéaire des instabilités de type Weibel. On discute le rôle essentiel joué par les composantes hors diagonale du tenseur dans la génération du champ magnétique, puis la capacité du modèle à reproduire quantitativement ou qualitativement les résultats cinétiques en introduisant la notion de limite hydrodynamique. La seconde partie de la thèse est ciblée sur le développement du code semi-lagrangien relativiste VLEM utilisant une méthode de décomposition de domaine : on présente les principales méthodes mathématiques utilisées dans le code, puis on aborde la problématique de la conservation de la charge à laquelle on apporte une réponse reposant sur une adaptation de la méthode d’Esirkepov. Le code est enfin validé grâce à plusieurs simulations d’instabilités de type Weibel
Weibel-type instabilities occurs when the velocity distribution function of the charged particles displays a pronounced anisotropy. A long-lasting magnetic field is generated due to the formation of current filaments, and it is accompanied by an important electrostatic activity. These ``basic’’ phenomena have been greatly investigated because of their involvement in many physical problems, natural (solar wind, relativistic jets) or experimental (laser-plasma interaction) : they occurs in plasmas which can be collisional or not, magnetised or not, relativistic or not. One needs to choose a suitable model for their description. The kinetic theory is the most complete and somewhat complex theoretical framework which we will consider. Due to its complexity, it may be interesting to develop reduced models. The first work realised during this thesis is the utilisation of a non-relativistic fluid description, including the dynamics of the pressure tensor, in order to model the linear Weibel-type instabilities. We put in evidence the effect of the non-diagonal components of the tensor on the magnetic field generation. We discuss the ability of the model to reproduce quantitatively or qualitatively the kinetic results by introducing the hydrodynamics limit. The second part of this thesis work is dedicated to the development of the relativistic semi-lagrangian code VLEM, using a domain decomposition scheme : we present the main mathematical tools used in the code, then we deal with the problem of the charge conservation and propose a solution for VLEM, based on an adaptation of the Esirkepov method. Finally, we validate the code through simulations of Weibel-type
L'origine de champs magnétiques observés dans les plasmas de laboratoire et d'astrophysique est l'un des problèmes récurrents en physique des plasmas. À cet égard, les instabilités de type Weibel ...sont considérées d'une grande importance. Ces instabilités ont pour origine une anisotropie de température (instabilité de Weibel) et des moments des électrons (instabilité de filamentation de courant). L'objectif principal de cette thèse est l'étude théorique et numérique de ces instabilités dans un plasma non collisionnel en régime relativiste. Le premier aspect de ce travail est l'étude du régime non-linéaire de ces instabilités et du rôle des effets cinétiques et relativistes sur la structure des champs électromagnétiques auto-cohérents. Dans ce cadre, un problème essentiel pour les applications et la théorie, concerne l'identification et l'analyse des structures cohérentes développées spontanément dans le régime non-linéaire sur des échelles cinétiques. Un deuxième aspect du travail est le développement de techniques analytiques et numériques pour l'étude des plasmas non collisionnels. Le modèle mathématique de référence, à la base des études des plasmas chauds, est le modèle Vlasov-Maxwell, où l'équation de Vlasov (théorie des champs moyens) est couplée aux équations de Maxwell de façon auto-cohérente. Un modèle unidimensionnel, le modèle multi-faisceaux, a également été introduit durant cette thèse. Basé sur une technique de réduction en dimension, il est à la fois un modèle analytique "simple" présentant l'avantage de pouvoir résoudre une équation de Vlasov 1D pour chaque faisceau de particules, et un modèle numérique moins coûteux qu'un modèle complet
The origin of magnetic fields observed in laboratory and astrophysical plasmas is one ofthe most challenging problems in plasma physics. In this respect, the Weibel type instabilities are considered of key importance. These instabilities are caused by a temperature anisotropy (Weibel instability) and electron momentum (current filamentation instability). The main objective of this thesis is the theoretical and numerical study of these instabilities in a collisionless plasma in the relativistic regime. The first aspect of this work is to study the nonlinear regime of these instabilities and the role of kinetic and relativistic effects on the structure of self-consistent electromagnetic fields. In this context, a key problem for the theory and applications, is the identification and analysis of coherent structures developed spontaneously in the nonlinear regime of kinetic scales. A second aspect of the work is the development of analytical and numerical techniques for the study of collisionless plasmas. A mathematical model of reference is the Vlasov-Maxwell model, where the Vlasov equation (mean field theory) is coupled to the Maxwell equations in a self-consistent way. A one-dimensional model, the multi-stream model, is also introduced. Based on a dimensional reduction technique, it is both an analytical model "simple" having the advantage of being able to solve a 1D Vlasov equation for each particle beam, and a numerical model less expensive than a complete model
Candida lusitaniae est une levure pathogène opportuniste émergente, responsable d’infections sévères chez les sujets immunodéprimés, et constitue un important modèle d’étude fonctionnelle de gènes ...par génétique inverse. Cette levure est souvent associée à des résistances aux antifongiques, d’où la nécessité de mieux comprendre l’interaction entre l’hôte et le pathogène, afin d’identifier de nouvelles cibles thérapeutiques potentielles. Les molécules signal sont décrites comme modulatrices de cette interaction. Au cours de ce travail, nous avons créé un mutant dpp3Δ, inactivé pour une pyrophosphate phosphatase, chez C. lusitaniae. La mutation a pour conséquence de modifier le taux secrété de PEA/tyrosol, des alcools aromatiques récemment décrits comme molécules signal chez les espèces Candida. Les levures du mutant dpp3Δ présentent aussi un défaut de pseudo-filamentation et de reproduction sexuée. Comparé à la souche sauvage, le mutant dpp3Δ module différemment la réponse macrophagique, notamment la production de NO et ROS, ainsi que la sécrétion de TNF-α et d’IL-10. Curieusement, la sécrétion d’IL-10 est modulée par les levures même sans contact physique avec les macrophages. Par ailleurs, nous avons observé des effets du traitement des levures par le PEA ou le tyrosol sur la modification des réponses immunes macrophagiques. Enfin, nous avons montré que le gène DPP3 chez C. lusitaniae était un facteur de virulence essentiel dans des modèles murins d’infection, et qu’il augmentait le taux de TNF-α secrété et la colonisation des cerveaux.
C. lusitaniae, an emerging Candida species often associated with antifungal resistance, is an important model to study gene function by reverse genetics. Understanding the host-pathogen interaction helps identifying new virulence factors and potentially new antifungal targets. Signaling molecules are often reported as modulators of this interaction. Here we created a dpp3Δ knocked-out mutant, lacking a pyrophosphate phosphatase, in C. lusitaniae. This mutation modified the ratio of secreted PEA/tyrosol, two newly reported signaling molecules in Candida species. Colony morphology of yeast cells was also altered as yeasts had a defect in pseudo-filamentation, and mating capacity was severely reduced. Compared to the wild-type strain, the dpp3Δ knocked-out mutant differently affected NO and ROS production by macrophages as well as TNF-α and IL-10 secretion. Interestingly IL-10 secretion was found to be modulated by C. lusitaniae without the need of a physical contact with the phagocytes. Moreover we elucidated the effects of PEA and tyrosol on yeast cells leading to modulations in macrophage immune responses. At last, the DPP3 gene was found to be an essential pathogenicity factor in mice models, leading to an increase of TNF-α secretion and brain colonization.
Les travaux de cette thèse ont eu pour but la conception de guides d'onde dans des verres de chalcogénure, par une technique de photo-inscription. Celle-ci consiste à focaliser un laser femtoseconde ...dans le matériau et de l'interaction lumière-matière, il en résulte une variation d'indice de réfraction (Δn) permanente. Cette technique est rapide, en une seule étape et ne nécéssite pas l'intervention de processus chimique. De plus, elle permet la réelle conception de dispositifs en trois dimensions. Deux verres ont été étudiés, le GeGaS et l'As₂S₃. Les phénomènes responsables de ces modifications ont été étudiés et décrits pour chacun des verres. Dans le premier verre, la propagation d'un faisceau sous la forme de filament a été mise en évidence. Du déplacement des charges, il découle une modification de la structure du réseau vitreux qui conduit à une variation d'indice positive. Cette As₂S₃ a été étudiée et il a été montré que son amplitude est contrôlable au travers du taux de répétition du laser ou de la vitesse de déplacement de l'échantillon, alors que son diamètre reste constant indépendamment des conditions expérimentales. Dans le deuxième verre, des effets d'origine thermique ont été mis en évidence. Pour ce verre d'As₂S₃, alors que les Δn obtenues classiquement sont négatives, l'utilisation d'une trajectoire hélicoïdale innovante a mené à la réalisation de guides d'onde. Après une analyse systématique des différents paramètres expérimentaux, une maîtrise de l'amplitude de Δn et du diamètre du guide a été obtenue. Ainsi, des guides d'ondes monomodes, avec des propriétés équivalentes à l'état de l'art, ont été réalisé en une seule étape.
This thesis aimed at the waveguide conception in chalcogenide glass, with a photo-writing technique. This one consists on a laser focus into the material, from the light-matter interaction it results a permanent refractive index variation (Δn). This technique is fast, single step and it doesn't require any chemical process intervention. Furthermore it really enables the three dimensional devices design. Two glasses were studied, one is the GeGaS and the other the As₂S₃. The responsible phenomena of these modifications was studied and described for each of the glasses. In the first one, the beam propagation on a filament form was highlighting. From the charge displacement, it ensues a modification of the glassy network structure which leads to a positive index variation. This Δn was studied and it was shown that its amplitude is controllable trhough the laser repetition rate or with the sample displacement velocity while its diameter remains constant independently of the experimental conditions. Into the second glass, effects of thermal origin were highlighted. For this As₂S₃ glass, while classical photo-writing Δn are negative, the use of an innovative helical trajectory led to the waveguide conception. After a systematic analysis of the experimental parameter influence, a control of the Δn amplitude and of the waveguide diameter was obtaines. So, monomode waveguides, with equivalent properties to the state-of-the-art, were realized in a single step.
