This paper brings a brief review of the human patatin-like phospholipase domain-containing protein (PNPLA) family. Even though it consists of only nine members, their physiological roles and ...mechanisms of their catalytic activity are not fully understood. However, the results of a number of knock-out and gain- or loss-of-function research models suggest that these enzymes have an important role in maintaining the homeostasis and integrity of organelle membranes, in cell growth, signalling, cell death, and the metabolism of lipids such as triacylglycerol, phospholipids, ceramides, and retinyl esters. Research has also revealed a connection between PNPLA family member mutations or irregular catalytic activity and the development of various diseases. Here we summarise important findings published so far and discuss their structure, localisation in the cell, distribution in the tissues, specificity for substrates, and their potential physiological role, especially in view of their potential as drug targets.
Postoje brojne klasifikacije glomerulonefritisa prema kojima se oni dijele na akutne i kronične, primarne i sekundarne, nasljedne i stečene, proliferativne i neproliferativne i dr. U kliničkoj praksi ...najčešće se služimo klasifikacijom prema patohistološkom nalazu. Za mnoge glomerulonefritise histološka slika, kao i klinička prezentacija, jako variraju. Nijedna podjela glomerulonefritisa nije utemeljena na razumijevanju patogeneze pojedinih oblika bolesti glomerula. Kako nova znanstvena otkrića rasvjetljavaju patogenetske mehanizme, tako se mijenjaju i klasifikacije pojedinih glomerulopatija. Najbolji primjer za to je membranoproliferativni glomerulonefritis.
Ovaj revijalni rad donosi pregled dosadašnjih spoznaja o porodici PNPLA (engl. patatin-like phospholipase domain-containing proteins) ljudskih enzima. Iako ovu porodicu čini samo 9 članova, najmanji ...sadrži 253 aminokiseline, a najveći više od 1360 aminokiselina, fiziološka uloga i mehanizam katalitičke aktivnosti nisu do sada potpuno razriješeni niti za jednoga. Međutim, rezultati brojnih tzv. knock-out, gain-of-function i loss-of-function modela istraživanja upućuju na važnu ulogu ovih enzima u mnogim biološkim procesima, uključujući održavanje homeostaze i integriteta stanične membrane, rast stanice, staničnu signalizaciju, staničnu smrt i metabolizam lipida kao triacilglicerola, fosfolipida, ceramida i retinil estera. Također, rezultati istraživanja upozoravaju na povezanost mutacija i nepravilne aktivnosti pojedinih članova s razvojem raznih bolesti. Radi boljeg razumijevanja PNPLA porodice enzima i naglaska na njihov potencijal kao mete razvoja novih lijekova, donosimo sveobuhvatni pregled do sada poznatih spoznaja koje uključuju strukturu, lokalizaciju u stanici, distribuciju u tkivima, specifičnost prema supstratima i potencijalnu fiziološku ulogu.

Cellular signaling pathways have important roles in cellular growth, differentiation, inflammatory response and apoptosis and in regulation of cellular responses under various chemical stimulators. Different proteins which belong to these pathways may be exposed to loss-of-function or gain-of-function mutations; this may lead to many clinical phenotypes including primarily cancer. In this review information about basic working principles of these pathways and diseases related to them are included.
Bu çalışmada Candida enfeksiyonlarının patogenezinde önem taşıdığı öne sürülen fosfolipaz aktivitesinin araştırılması amaçlanmıştır. Çeşitli klinik örneklerden (kan, beyin omurilik sıvısı, parasentez ...mayi, katater, bronkoalveoler lavaj, trakeal aspirat, karaciğer absesi) izole edilen 80 Candida albicans, 10 Candida tropicalis, 6 Candida glabrata, 6 Candida guilliermondii, 4 Candida parapsilosis, 2 Candida krusei, 1 Candida kefyr olmak üzere toplam 109 Candida susu kullanılmış ve suşların fosfolipaz aktivitesi modifiye plak yöntemi ile incelenmiştir. Fosfolipaz aktivitesi (Pz), koloni çapını koloni ile birlikte presipitasyon zonunun toplam çapına oranı olarak hesaplanmıştır. Sonuç olarak, C.albicans izolatlarının %61.3'ünde (49/80) fosfolipaz aktivitesi saptanırken, albicans dışı Candida türlerinde fosfolipaz aktivitesi izlenmemiştir. Fosfolipaz salgılayan suşların ortalama Pz katsayısı 0.805±0.08 olarak saptanmıştır. Fosfolipaz pozitifliği açısından suşların izole edildiği bölgelere göre istatistiksel olarak anlamlı bir farklılık belirlenememiştir (X2 =5.5; p=0.137).
In this study, the phosphoiipase activity that was suggested to be important in the pathogenesis of Candida infections, has been investigated. A total of 109 Candida strains (80 Candida albicans, 10 C.tropicalis, 6 C.glabrata, 6 C.guilliermondii, 4 C.parapsilosis, 2 C.krusei, 1 C.kefyr) which were isolated from various clinical specimens (blood, cerebrospinal fluid, parasynthesis specimens, catheter samples, bronchoalveolar lavage fluid, tracheal aspirate, liver abscess) have been included in the study. Phosphoiipase activities were evaluated by modified plate method. Phosphoiipase activity (Pz) was determined as the ratio of colony diameter to the diameter of precipitation zone with the colony. While 61.3% (49/80) of C.albicans strains were found to be positive for phosphoiipase activity, none of the non-albicans Candida isolates exhibited phosphoiipase activity. The mean Pz coefficient value of phosphoiipase secreting isolates was calcuted as 0.805+-0.08.There was no statistically significant difference between the phospholipase activity af the isolates according to their isolation sites (X2=5.5; p=0.137).
