Environmental (e)DNA metabarcoding holds great promise for biomonitoring and ecotoxicological applications. However, few studies have compared the performance of eDNA versus eRNA metabarcoding in ...assessing organismal response to marine pollution, in experimental conditions. Here, we performed a chromium (Cr)-spiked mesocosm experimental test on benthic foraminiferal community to investigate the effects on species diversity by analysing both eDNA and eRNA metabarcoding data across different Cr concentrations in the sediment. Foraminiferal diversity in the eRNA data showed a significant negative correlation with the Cr concentration in the sediment, while a positive response was observed in the eDNA data. The foraminiferal OTUs exhibited a higher turnover rate in eRNA than in the eDNA-derived community. Furthermore, in the eRNA samples, OTUs abundance was significantly affected by the Cr gradient in the sediment (Pseudo-R2 = 0.28, p = 0.05), while no significant trend was observed in the eDNA samples. The correlation between Cr concentration and foraminiferal diversity in eRNA datasets was stronger when the less abundant OTUs (<100 reads) were removed and the analyses were conducted exclusively on OTUs shared between eRNA and eDNA datasets. This indicates the importance of metabarcoding data filtering to capture ecological impacts, in addition to using the putatively active organisms in the eRNA dataset. The comparative analyses on foraminiferal diversity revealed that eRNA-based metabarcoding can better assess the response to heavy metal exposure in presence of subtle concentrations of the pollutant. Furthermore, our results suggest that to unlock the full potential for ecosystem assessment, eDNA and eRNA should be studied in parallel to control for potential sequence artifacts in routine ecosystem surveys.

Stress factors such as climate change and drought may switch the role of temperate peatlands from carbon dioxide (CO2) sinks to sources, leading to positive feedback to global climate change. Water ...level management has been regarded as an important climate change mitigation strategy as it can sustain the natural net CO2 sink function of a peatland. Little is known about how resilient peatlands are in the face of future climate change scenarios, as well as how effectively water level management can sustain the CO2 sink function to mitigate global warming. The authors assess the effect of climate change on CO2 exchange of south Swedish temperate peatlands, which were either unmanaged or subject to water level regulation. Climate chamber simulations were conducted using experimental peatland mesocosms exposed to current and future representative concentration pathway (RCP) climate scenarios (RCP 2.6, 4.5 and 8.5). The results showed that all managed and unmanaged systems under future climate scenarios could serve as CO2 sinks throughout the experimental period. However, the 2018 extreme drought caused the unmanaged mesocosms under the RCP 4.5 and RCP 8.5 switch from a net CO2 sink to a source during summer. Surprisingly, the unmanaged mesocosms under RCP 2.6 benefited from the warmer climate, and served as the best sink among the other unmanaged systems. Water level management had the greatest effect on the CO2 sink function under RCP 8.5 and RCP 4.5, which improved their CO2 sink capability up to six and two times, respectively. Under the current climate scenario, water level management had a negative effect on the CO2 sink function, and it had almost no effect under RCP 2.6. Therefore, the researchers conclude that water level management is necessary for RCP 8.5, beneficial for RCP 4.5 and unimportant for RCP 2.6 and the current climate.

Cyanobacterial blooms are an increasing threat to water quality and global water security caused by the nutrient enrichment of freshwaters. There is also a broad consensus that blooms are increasing ...with global warming, but the impacts of other concomitant environmental changes, such as an increase in extreme rainfall events, may affect this response. One of the potential effects of high rainfall events on phytoplankton communities is greater loss of biomass through hydraulic flushing. Here we used a shallow lake mesocosm experiment to test the combined effects of: warming (ambient vs. +4°C increase), high rainfall (flushing) events (no events vs. seasonal events) and nutrient loading (eutrophic vs. hypertrophic) on total phytoplankton chlorophyll‐a and cyanobacterial abundance and composition. Our hypotheses were that: (a) total phytoplankton and cyanobacterial abundance would be higher in heated mesocosms; (b) the stimulatory effects of warming on cyanobacterial abundance would be enhanced in higher nutrient mesocosms, resulting in a synergistic interaction; (c) the recovery of biomass from flushing induced losses would be quicker in heated and nutrient‐enriched treatments, and during the growing season. The results supported the first and, in part, the third hypotheses: total phytoplankton and cyanobacterial abundance increased in heated mesocosms with an increase in common bloom‐forming taxa—Microcystis spp. and Dolichospermum spp. Recovery from flushing was slowest in the winter, but unaffected by warming or higher nutrient loading. Contrary to the second hypothesis, an antagonistic interaction between warming and nutrient enrichment was detected for both cyanobacteria and chlorophyll‐a demonstrating that ecological surprises can occur, dependent on the environmental context. While this study highlights the clear need to mitigate against global warming, oversimplification of global change effects on cyanobacteria should be avoided; stressor gradients and seasonal effects should be considered as important factors shaping the response.
Cyanobacteria are expected to benefit from a warmer climate, especially in nutrient‐rich waters. However, other important climate change factors—more extreme rainfall events—could affect this response (e.g. loss through flushing). This mesocosm study tested the combined effects of warming, extreme rainfall events and nutrient loading on cyanobacterial abundance. Warming increased the abundance of bloom‐forming taxa, but in combination with very high nutrient loading resulted in a negative, not positive, interaction. The impact of extreme rainfall events was only apparent in the winter. Stressor gradients and season should be considered as important factors shaping the response to global change.
Submerged macrophytes are of key importance for the structure and functioning of shallow lakes and can be decisive for maintaining them in a clear water state. The ongoing climate change affects the ...macrophytes through changes in temperature and precipitation, causing variations in nutrient load, water level and light availability. To investigate how these factors jointly determine macrophyte dominance and growth, we conducted a highly standardized pan‐European experiment involving the installation of mesocosms in lakes. The experimental design consisted of mesotrophic and eutrophic nutrient conditions at 1 m (shallow) and 2 m (deep) depth along a latitudinal temperature gradient with average water temperatures ranging from 14.9 to 23.9°C (Sweden to Greece) and a natural drop in water levels in the warmest countries (Greece and Turkey). We determined percent plant volume inhabited (PVI) of submerged macrophytes on a monthly basis for 5 months and dry weight at the end of the experiment. Over the temperature gradient, PVI was highest in the shallow mesotrophic mesocosms followed by intermediate levels in the shallow eutrophic and deep mesotrophic mesocosms, and lowest levels in the deep eutrophic mesocosms. We identified three pathways along which water temperature likely affected PVI, exhibiting (a) a direct positive effect if light was not limiting; (b) an indirect positive effect due to an evaporation‐driven water level reduction, causing a nonlinear increase in mean available light; and (c) an indirect negative effect through algal growth and, thus, high light attenuation under eutrophic conditions. We conclude that high temperatures combined with a temperature‐mediated water level decrease can counterbalance the negative effects of eutrophic conditions on macrophytes by enhancing the light availability. While a water level reduction can promote macrophyte dominance, an extreme reduction will likely decrease macrophyte biomass and, consequently, their capacity to function as a carbon store and food source.
Macrophytes are central in the functioning of shallow lakes. Ongoing climate and land‐use change are altering simultaneously temperatures, nutrient runoff and water levels, and hence main drivers of macrophytes. In our large‐scale pan‐European experiment along a temperature gradient, we investigated interacting effect of temperature, nutrients and water level on the macrophyte growth. Our results suggested that positive temperature effects on macrophytes are highly contingent on nutrient‐status and water level. Particularly under eutrophic conditions evaporation‐driven water level change, impacting underwater light‐conditions, can act as a switch for whether macrophytes maintain dominance or are out‐competed by algae. Thus, water level change can compensate for negative eutrophication effects.
1. Biotic resistance represents an important natural barrier to potential invaders throughout the world, yet the underlying mechanisms that drive such resistance are still debated. In theory, native ...communities should repel both functionally similar invaders which compete for the same resources, and invaders which possess less competitive traits. However, environmental stress, trade-offs across vital rates and competition-induced plastic trait shifts may modify expected competitive outcomes, thereby influencing invasion dynamics. 2. In order to test these theoretical links between trait distributions and biotic resistance, we performed a mesocosm experiment with 25 non-native ornamental species invading native plant communities. Each non-native species was grown with and without the native community under two watering treatments (regular and reduced). We measured biotic resistance as the difference in performance of nonnative individuals grown with and without the community in terms of their survival, growth and reproduction. We quantified overall functional dissimilarity between non-native ornamental individuals and native communities based on the combination of plant height, specific leaf area and seed mass. Then, assuming each of these traits is also potentially linked to competitive ability, we measured the position of non-natives on trait hierarchies. While height is positively correlated with competitive ability for light interception, conservative leaf and seed characteristics provide greater tolerance to competition for other resources. Finally, we quantified plastic trait shifts of non-native individuals induced by competition. 3. Indeed, the native community repelled functionally similar individuals by lowering the invader's survival rate. Simultaneously, shorter ornamental individuals with larger specific leaf areas were less tolerant to biotic resistance from the community across vital rates, although the effect of trait hierarchies often depended on watering conditions. Finally, non-natives responded to competition by shifting their traits. Most importantly, individuals with more competitive traits were able to overcome biotic resistance also through competition-induced plastic trait shifts. 4. Synthesis. Our results highlight that both functional dissimilarity and trait hierarchies mediate biotic resistance to ornamental plant invaders. Nevertheless, environmental stress as well as opposing trends across vital rates are also influential. Furthermore, plastic trait shifts can reinforce potential invaders' competitive superiority, determining a positive feedback.
Dispersal success is crucial for the survival of species in metacommunities. Zooplankton species engage in dispersal through time (i.e., egg bank) and space (i.e., vectors) by means of resting eggs. ...However, dispersal to patches does not equate to successful colonization, as there is a clear distinction between dispersal rates and successful colonization. We performed a field mesocosm experiment assessing dispersal and colonization success of zooplankton from resting eggs or transport via directional wind/airborne and biotic vectors in the vicinity of three ponds. By using active vs. sterile pond sediments and mesh-covered vs. open mesocosms, we disentangled the two mechanisms of dispersal, i.e., from the egg bank vs. space. We found that for both rotifers and cladocerans, sediment type, mesh cover and duration of the experiment influenced species richness and species composition. The relative contribution of resting stages to dispersal and colonization success was substantial for both rotifers and cladocerans. However, wind/airborne dispersal was relatively weak for cladocerans when compared to rotifers, whereas biotic vectors contributed to dispersal success especially for cladocerans. Our study demonstrates that dispersal and colonization success of zooplankton species strongly depends on the dispersal mode and that different dispersal vectors can generate distinct community composition.
In the past decade, mesocosms have emerged as a useful tool for the environmental study of engineered nanomaterials (ENMs) as they can mimic the relevant exposure scenario of contamination. Herein, ...we analyzed the scientific outcomes of aquatic mesocosm experiments, with regard to their designs, the ENMs tested, and the end points investigated. Several mesocosm designs were consistently applied in the past decade to virtually mimic various contamination scenarios with regard to ecosystem setting as well as ENMs class, dose, and dosing. Statistical analyses were carried out with the literature data to identify the main parameters driving ENM distribution in the mesocosms and the potential risk posed to benthic and planktonic communities as well as global ecosystem responses. These analyses showed that at the end of the exposure, mesocosm size (water volume), experiment duration, and location indoor/outdoor had major roles in defining the ENMs/metal partitioning. Moreover, a higher exposure of the benthic communities is often observed but did not necessarily translate to a higher risk due to the lower hazard posed by transformed ENMs in the sediments (e.g., aggregated, sulfidized). However, planktonic organisms were generally exposed to lower concentrations of potentially more reactive and toxic ENM species. Hence, mesocosms can be complementary tools to existing standard operational procedures for regulatory purposes and environmental fate and risk assessment of ENMs. To date, the research was markedly unbalanced toward the investigation of metal-based ENMs compared to metalloid- and carbon-based ENMs but also nanoenabled products. Future studies are expected to fill this gap, with special regard to high production volume and potentially hazardous ENMs. Finally, to take full advantage of mesocosms, future studies must be carefully planned to incorporate interdisciplinary approaches and ensure that the large data sets produced are fully exploited.
Previous studies indicated that weathered conventional plastics and bioplastics pose ecotoxicological risks. Here, the effects of artificial and natural weathering on the ecotoxicity of three ...compostable bags and a conventional polyethylene (PE) bag are investigated. With that aim, a 21-day artificial indoor weathering experiment featuring UV light, UV-filtered light, and darkness was run simultaneously to a 120-day outdoor littoral mesocosm exposure featuring natural light, UV-filtered light, and shaded conditions. Acute toxicity of so-weathered plastic specimens was tested in vivo using the sensitive Paracentrotus lividus sea-urchin embryo test. PE was nontoxic from the beginning and did not gain toxicity due to UV weathering. In contrast, for bioplastics, dry artificial UV weathering increased toxicity in comparison to the dark control. Weathering in outdoor mesocosm led to a rapid loss of toxic properties due to leaching in rainwater. With a higher UV dosage, a plastic-type-dependent regain of toxicity was observed, most likely driven by enhanced availability or transformation of functional additives or due to bioplastic degradation products. PE showed moderate UV absorbance, while bioplastics showed high UV absorbance. This study highlights the potential of biodegradable plastics to pose enhanced ecotoxicological risk due to weathering under environmentally relevant conditions.
Plant community composition and functional traits respond to chronic drivers such as climate change and nitrogen (N) deposition. In contrast, pulse disturbances from ecosystem management can ...additionally change resources and conditions. Community responses to combined environmental changes may further depend on land‐use legacies. Disentangling the relative importance of these global change drivers is necessary to improve predictions of future plant communities. We performed a multifactor global change experiment to disentangle drivers of herbaceous plant community trajectories in a temperate deciduous forest. Communities of five species, assembled from a pool of 15 forest herb species with varying ecological strategies, were grown in 384 mesocosms on soils from ancient forest (forested at least since 1850) and postagricultural forest (forested since 1950) collected across Europe. Mesocosms were exposed to two‐level full‐factorial treatments of warming, light addition (representing changing forest management) and N enrichment. We measured plant height, specific leaf area (SLA) and species cover over the course of three growing seasons. Increasing light availability followed by warming reordered the species towards a taller herb community, with limited effects of N enrichment or the forest land‐use history. Two‐way interactions between treatments and incorporating intraspecific trait variation (ITV) did not yield additional inference on community height change. Contrastingly, community SLA differed when considering ITV along with species reordering, which highlights ITV’s importance for understanding leaf morphology responses to nutrient enrichment in dark conditions. Contrary to our expectations, we found limited evidence of land‐use legacies affecting community responses to environmental changes, perhaps because dispersal limitation was removed in the experimental design. These findings can improve predictions of community functional trait responses to global changes by acknowledging ITV, and subtle changes in light availability. Adaptive forest management to impending global change could benefit the restoration and conservation of understorey plant communities by reducing the light availability.
