MicroRNAs are a large group of non-coding nucleic acids, usually 20–22 nt long, which bind to regulatory sections of messenger RNA (mRNA) and inhibit gene expression. Endocrine disruptors such as those from the phthalate group imitate or block these hormonal effects, and our previous study showed a long-lasting decrease in plasma testosterone levels in rat offspring exposed to a mixture of three phthalates
and postnatally. These effects were also observed at the behavioural level. To shed more light on these findings, in this new study we compared testicular tissue morphology between control and phthalatetreated males and investigated possible persistent changes and sex differences in the expression of two hippocampal microRNAs – miR- 15b-5p and miR-34a-5p – participating in the transcription of steroidogenic genes. Histologically observed changes in testicular tissue morphology of phthalate-exposed males compared to control support testosterone drop observed in the previous study. At the microRNA level, we observed more significant changes in phthalate-treated females than in males. However, we are unable to relate these effects to the previously observed behavioural changes.

Eine ...bedienerfreundliche Methode namens miRNA‐Ziel‐RNA‐Affinitätsreinigung (miR‐TRAP) identifiziert direkt in vivo die Ziele der miRNA und könnte bei der Suche nach solchen Zielen in Krankheitsmodellen unter physiologischen Bedingungen hilfreich sein.

Nearly 97% of the human genome is the non-coding DNA, which varies from one species to another, and changes in these sequences are frequently noticed to manifest clinical and circumstantial ...malfunction. Numerous non-protein-coding genes are recently found to encode microRNAs, which are responsible for RNA-mediated gene silencing through RNA interference (RNAi)-like pathways. MicroRNAs (miRNAs), small single-stranded 17–25 nucleotide RNAs capable of interfering with intracellular messenger RNAs (mRNAs) that contain either complete or partial complementarity, are useful for the design of new therapies against cancer polymorphism and viral mutation. Currently over 1000 native miRNA species found in vertebrates and many more new miRNA homologs continue to be identified, however, most of their functions remain to be determined. In this book, many new perspectives of the miRNA research are reviewed and discussed, including their roles in stem cell maintenance, embryonic development, tissue differentiation, adult physiology, disease pathology, cancer research, viral infection, genetic engineering in plants, and utility in cosmetic applications. These new findings may not only provide significant insight into the various mechanisms of miRNAs but also offer a great opportunity in developing new therapeutic interventions.
Mikroribonukleinske kiseline (miRNA) velika su skupina nekodirajućih nukleinskih kiselina, obično dugih 20 – 22 nt, koje se vežu za regulacijske dijelove glasničke RNA (mRNA) i inhibiraju ekspresiju ...gena. Aktivnost genoma usto reguliraju i hormoni. Endokrini disruptori, poput onih iz skupine ftalata, oponašaju ili sprječavaju djelovanje hormona, a naše je ranije istraživanje upozorilo na dugoročni pad razina testosterona u mladunaca štakora izloženih mješavini triju ftalata in utero i nakon poroda. Ti su učinci zamijećeni i na razini ponašanja. Cilj je ovog novog ispitivanja bio rasvijetliti te prijašnje rezultate tako što smo u njemu usporedili morfologiju tkiva testisa kontrolne i skupine izložene ftalatima te istražili moguće stalne promjene i razlike između spolova u ekspresiji dviju miRNA u hipokampusu – miR-15b-5p i miR-34a-5p – koji sudjeluju u transkripciji gena odgovornih za steroidogenezu. Histološki utvrđene promjene u tkivu testisa izloženih mužjaka u odnosu na kontrolu potvrdile su morfološku pozadinu pada razina testosterona iz ranijeg ispitivanja. Na razini miRNA uočene su značajnije promjene u izloženih ženki nego u mužjaka, no ne možemo povezati te učinke s ranije zamijećenim promjenama u ponašanju.
MikroRNA (miRNA) jsou krátké nekódující RNA, které regulují genovou expresi na posttranskripční úrovni. Zúčastňují se řady kritických biologických procesů včetně onkogeneze. V patogenezi karcinomu ...z renálních buněk (renal cell carcinoma – RCC) mohou působit jako onkogeny i jako tumor supresory. Prostřednictvím cílových proteinů různých signálních drah ovlivňují apoptózu, buněčný růst, migraci, invazi, proliferaci, formaci buněčných kolonií či angiogenezi a jsou přímo zapojeny do procesů hypoxie a epiteliálně-mezenchymální proměny. Odlišně exprimované hladiny miRNA umožňují rozlišení zdravé a nádorové tkáně ledviny a dokonce jednotlivých podtypů RCC. Cirkulující a močové miRNA jsou potenciálními biomarkery pro neinvazivní diagnostiku RCC a sledování relapsu onemocnění. Navíc je prokázána prognostická hodnota některých miRNA, a to jak z hlediska identifikace vysoce rizikových primárních tumorů, tak z hlediska odpovědi na biologickou léčbu u metastatických nádorů. Cílem článku je poskytnout přehlednou a co nejaktuálnější informaci o roli miRNA v patogenezi RCC a miRNA využitelných pro diagnostiku a stanovení prognózy RCC.
MicroRNAs (miRNA/miRNAs) are short non-coding RNAs that regulate gene expression at post-transcriptional level. They are involved in a number of critical biological processes including carcinogenesis. In the pathogenesis of renal cell carcinoma (RCC) they can act both as oncogenes and tumor suppressors. They regulate apoptosis, cell growth, migration, invasion, proliferation, colony formation or angiogenesis through target proteins involved in several signaling pathways and they are directly involved in mechanisms of hypoxia and epithelial-to-mesenchymal transition. Differentially expressed miRNAs can differentiate tumor tissue from healthy renal tissue and even different RCC subtypes. Circulating and urinary miRNAs could become biomarkers for non-invasive diagnosis or detection of the relapse of RCC. Moreover, prognostic value of several miRNAs has been shown. They may help identify high-risk primary tumors and estimate treatment response to biological treatment in metastatic tumors. The aim of the article is to provide comprehensive and the most up-to-date information about the role of miRNAs in the pathogenesis of RCC and their diagnostic/prognostic value.

Karcinom pankreatu je nádorové onemocnění se špatnou prognózou. V minulosti byl pozorován jeho zvýšený výskyt u pacientů s diabetes mellitus. Nicméně dosud není zcela jasné, jestli je diabetes ...rizikovým faktorem nádorové transformace nebo jestli vzniká až druhotně v souvislosti s nádorem. Klíčem úspěchu onkologické léčby je časná detekce karcinomu k provedení pankreatektomie – dosud jediné možné kurativní terapie. K tomu je potřeba časných diagnostických markerů. Pancreatic cancer is a malignancy with a poor prognosis. Its increased incidence was observed in diabetic patients in the past. However, there is no evidence, whether diabetes is a risk factor for neoplastic transformation, or if it arises secondarily as a result of the tumor. The key is an early detection of cancer to perform pancreatectomy – the only possible curative therapy yet. This requires early diagnostic markers. Malé regulační molekuly, tzv. mikroRNA, se podílejí na regulaci všech základních životních procesů v buňkách. Jsou regulátory exprese genů na posttranskripční úrovni. Ovlivňují tuto expresi vazbou na ...cílové mRNA, která vede k blokaci translace a štěpení mRNA. Nejnovější výzkumy ukázaly, že jejich výskyt není omezen jen na buňky, ale mikroRNA se vyskytují také v tělních tekutinách. Vzhledem k tomu, že diferenciální exprese určitých mikroRNA je často asociovaná s mnoha patologickými procesy, detekce deregulovaných mikroRNA v diagnosticky relevantních vzorcích (např. krvi, plazmě/séru, moči) se může stát základem nových diagnostických postupů. U gynekologických nádorových onemocnění je nalezení nových diagnostických biomarkerů důležitým cílem současných výzkumů. V tomto článku naleznete přehled cirkulujících a extracelulárních mikroRNA, které byly nejnověji nalezeny deregulovány u karcinomu ovaria, endometria a cervixu, především ve vzorcích krve, séra/plazmy a moči.
Small, non-coding molecules of RNA, microRNAs, have been shown to participate in regulation of key biological processes in the cells. They regulate the expression at post-transcriptional level by binding to target mRNA, resulting in mRNA degradation or translation inhibition. The most recent investigations revealed that microRNAs may occur not only in the cells, but also in many body fluids. Differential expression of particular microRNAs is often associated with many pathological states. Therefore, the detection of deregulated microRNAs in diagnostically relevant samples such as blood, plasma/serum, or urine may become the basis for the establishment of novel diagnostic procedures. Finding alternative diagnostic biomarkers for gynecological cancers is the goal of many recent investigations. We present here the survey of novel circulating and extracellular microRNAs which have been found deregulated in ovarian cancer, endometrial cancer and cervical cancer, particularly in blood, plasma/serum and urine samples.
