Tumor necrosis factor-alpha (TNF-alpha) is a polypeptide implicated in patophysiology of inflammatory process inhibition of tumor growth, and development of septic shock. In the project we measured the concentration of both cytokines in gingival fluid, gingiva and sera of patients with periodontal disease and beagle dogs with experimentally induced periodontitis. The level of bacterial plaque was estimated by Plaque Index (PI). Gingival inflammation was assessed by Gingival Index (GI) and periodontal destruction was evaluated by measuring probing depth (PD) and epithelial attachment loss (EAL). Gingival fluid was collected from ten patients and fourteen beagle dogs by paper strips and microcapillary tubes. Gingival tissue samples were obtained from nine patientswho needed periodontal surgery treatment. The sera of seven dogs was analysed before and nine months after induction of experimental periodontitis.The level of TGF-beta1 and TNF-alpha in crevicular fluid, supernant of gingival tissue and sera was measured by ELISA kit. Results showed that the concentration of TGF-beta1 is significantly higher in fluid samples and gingival tissue in patients taken from the sites with deeper pockets. The level of TGF-beta1 in dogsć fluid samples was increased during pocket formation up to 3 mm and decreased in deeper pockets. The concentrationof TGF-beta1 was also decreased in sera of dogs nine months afterthe start of experimental periodontitis.
