Paramagnetic NMR probes provide valuable long‐range structural information on proteins and protein complexes. A new, stable, two‐armed lanthanoid probe is reported that can be attached to a protein site‐specifically via chemically inert thioether linkages. A two‐armed probe: α‐Bromoacetamides are functionalised in a DOTA‐like structure to form a new double‐arm paramagnetic probe (see structure). This probe can be site‐specifically attached to proteins and the ligation products are stable under reducing conditions. This probe creates a new avenue for studying biomolecules using paramagnetic NMR spectroscopy.
