    Background. The reticulocyte is a clinically important indirect indicator of erythropoietic activity in patients. However, determination of the reticulocyte count using a light microscope has low ... accurracy and reproducibility rates. Recently, automated reticulocyte counters have been developed, which allow for a more rapid and accurate counting of reticulocytes. Methods. In our study, we compared the following methods for reticulocyte count determination: the microscopic brilliant cresyl blue methodand flow cytometric EPICS Profile (Coulter) method using thiazole orange(TO). The purpose of the study was following: to select the most suitable TO concentration to be used with the EPICS Profile cytometer. The following TO concentrations were used: 0,2 mg/L, 0.1 mg/L, 0.07 mg/L 0.05 mg/L. Results and conclusions. The most appropriate TO concentration for counting reticulocyte by the EPICS Profile is 0,1 mg/L. The concentration was select according to the following criteria: good agreement of the results obtained by microscopy with the results yielded by the EPICS Profile cytometer, absence of statistically significant differences between the results, and agreement in the normal ranges. There was a good correlation between the methods studied. The correlation coefficent(r) ranged from 0,82 and 0,87. The mean intra-assay coefficients of variation for determination by light microscopy and EPICS Profile cytometer was 27.5% and 8.4% respectively.
