The underdetermined blind source separation method based on sparse component analysis in the time–frequency domain is introduced to estimate the modal parameters in this study. This study attempts to ...reveal the relationship with the modal parameter identification methods based on the time–frequency representation. A five-degree-of-freedom mass-spring-damper system and a real experiment conducted on a TC4 titanium-alloy column are used to confirm the proposed method. Furthermore the proposed method is applied to extract the time-varying modal parameters of the column in the temperature-varying environment. The extracted results evidently show the thermal effect on the modal parameters.

LncRNA (long non-coding RNA) and mRNA form a competitive endogenous RNA (ceRNA) network by competitively binding to common miRNAs. This network regulates various processes of plant growth and ...development at the post-transcriptional level. Somatic embryogenesis is an effective means of plant virus-free rapid propagation, germplasm conservation, and genetic improvement, which is also a typical process to study the ceRNA regulatory network during cell development. Garlic is a typical asexual reproductive vegetable. Somatic cell culture is an effective means of virus-free rapid propagation in garlic. However, the ceRNA regulatory network of somatic embryogenesis remains unclear in garlic. In order to clarify the regulatory role of the ceRNA network in garlic somatic embryogenesis, we constructed lncRNA and miRNA libraries of four important stages (explant stage: EX; callus stage: AC; embryogenic callus stage: EC; globular embryo stage: GE) in the somatic embryogenesis of garlic. It was found that 44 lncRNAs could be used as precursors of 34 miRNAs, 1511 lncRNAs were predicted to be potential targets of 144 miRNAs, and 45 lncRNAs could be used as eTMs of 29 miRNAs. By constructing a ceRNA network with miRNA as the core, 144 miRNAs may bind to 1511 lncRNAs and 12,208 mRNAs. In the DE lncRNA-DE miRNA-DE mRNA network of adjacent stages of somatic embryo development (EX-VS-CA, CA-VS-EC, EC-VS-GE), by KEGG enrichment of adjacent stage DE mRNA, plant hormone signal transduction, butyric acid metabolism, and C5-branched dibasic acid metabolism were significantly enriched during somatic embryogenesis. Since plant hormones play an important role in somatic embryogenesis, further analysis of plant hormone signal transduction pathways revealed that the auxin pathway-related ceRNA network (lncRNAs-miR393s-
) may play a role in the whole stage of somatic embryogenesis. Further verification by RT-qPCR revealed that the lncRNA125175-miR393h-
network plays a major role in the network and may affect the occurrence of somatic embryos by regulating the auxin signaling pathway and changing the sensitivity of cells to auxin. Our results lay the foundation for studying the role of the ceRNA network in the somatic embryogenesis of garlic.
Seedlessness is one of the important quality and economic traits favored by grapevine consumers, which are mainly affected by phytohormones, especially gibberellin (GA). GA is widely utilized in ...seedless berry production and could effectively induce grape seed embryo abortion. However, the molecular mechanism underlying this process, like the role of RNA silencing in the biosynthesis pathway of GA remains elusive. Here, Gibberellin 3-β dioxygenase2 (GA3ox2) as the last key enzyme in GA biosynthesis was predicated as a potential target gene for miR3633a, and two of them were identified as a GA response in grape berries. We also analyzed the promoter regions of genes encoding GA biosynthesis and found the hormone-responsive elements to regulate grape growth and development. The cleavage interaction between VvmiR3633a and VvGA3ox2 was validated by RLM-RACE and the transient co-transformation technique in tobacco in vivo. Interestingly, during GA-induced grape seed embryo abortion, exogenous GA promoted the expression of VvmiR3633a, thereby mainly repressing the level of VvGA3ox2 in seed embryos. We also observed a negative correlation between down-regulated VvGA20ox2/VvGA3ox2 and up-regulated VvGA2ox3/VvGA2ox1, of which GA inactivation was greater than GA synthesis, inhibited active GA content, accompanied by the reduction of VvSOD and VvCAT expression levels and enzymatic activities. These series of changes might be the main causes of grape seed embryo abortion. In conclusion, we have preliminarily drawn a schematic mode of GA-mediated VvmiR3633a and related genes regulatory network during grape seed abortion induced by exogenous GA. Our findings provide novel insights into the GA-responsive roles of the VvmiR3633a-VvGA3ox2 module in the modulation of grape seed-embryo abortion, which has implications for the molecular breeding of high-quality seedless grape berries.
Grape is highly sensitive to gibberellin (GA), which is crucial during seed and berry development (SBD) either by itself or by interacting with other hormones, such as auxin, Abscisic acid (ABA), and ...Cytokinin (CK). However, no systematic analysis of GA metabolic and signal transduction (MST) pathway has been undertaken in grapevine.
In this study, total endogenous GA
content significantly decreased during SBD, and a total of 48 known genes in GA metabolic (GAM; 31) and signal transduction (ST; 17) pathways were identified in this process. In the GAM pathway, out of 31 genes, VvGA20ox1-1, VvGA3ox4-1, and VvGA2ox1-1 may be the major factors interacting at the green-berry stage (GBS) accompanied with higher accumulation rate. GA biosynthesis was greater than GA inactivation at GBS, confirming the importance of seeds in GA synthesis. The visible correlation between endogenous GA
content and gene expression profiles suggested that the transcriptional regulation of GA biosynthesis pathway genes was a key mechanism of GA accumulation at the stone-hardening stage (SHS). Interestingly, we observed a negative feedback regulation between VvGA3oxs-VvGAI1-4, VvGA2oxs-VvGAI1-4, and VvGID1B-VvGAI1-4 in maintaining the balance of GA
content in berries. Moreover, 11 miRNAs may be involved in the modulation of GA MST pathway by mediating their target genes, such as VvGA3ox, VvGID1B, and VvGAMYB. Many genes in auxin, ABA, and CK MST pathways were further identified and found to have a special pattern in the berry, and the crosstalk between GA and these hormones may modulate the complex process during SBD through the interaction gene network of the multihormone pathway. Lastly, based on the expression characterization of multihormone MST pathway genes, a proposed model of the GA-mediated multihormone regulatory network during SBD was proposed.
Our results provided novel insights into GA-mediated regulatory networks during SBD in grape. The complexity of GA-mediated multihormone ST in SBD was also elucidated, thereby providing valuable information for future functional characterizations of specific genes in grape.
Exogenous GA is widely used to efficiently induce grape seedless berry development for significantly improving berry quality. Recently, we found that VvmiR166s are important regulators of response to ...GA in grapes, but its roles in GA-induced seedless grape berry development remain elusive. Here, the precise sequences of VvmiR166s and their targets VvREV, VvHB15 and VvHOX32 were determined in grape cv. ‘Rosario Bianco’, and the cleavage interactions of VvmiR166s-VvHB15/VvHOX32/VvREV modules and the variations in their cleavage roles were confirmed in grape berries. Exogenous GA treatment significantly induced a change in their expression correlations from positive to negative between VvmiR166s and their target genes at the seeds during the stone-hardening stages (32 DAF–46 DAF) in grape berries, indicating exogenous GA change action modes of VvmiR166s on their targets in this process, in which exogenous GA mainly enhanced the negative regulatory roles of VvmiR166s on VvHB15 among all three VvmiR166s-target pairs. The transient OE-VvmiR166a-h/OE-VvHB15 in tobacco confirmed that out of the VvmiR166 family, VvmiR166h/a/b might be the main factors in modulating lignin synthesis through inhibiting VvHB15, of which VvmiR166h-VvHB15-NtPAL4/NtCCR1/NtCCR2/NtCCoAMT5/NtCOMT1 and VvmiR166a/b-VvHB15-NtCAD1 are the potential key regulatory modules in lignin synthesis. Together with the GA-induced expression modes of VvmiR166s-VvHB15 and genes related to lignin synthesis in grape berries, we revealed that GA might repress lignin synthesis mainly by repressing VvCAD1/VvCCR2/VvPAL2/VvPAL3/Vv4CL/VvLac7 levels via mediating VvmiR166s-VvHB15 modules in GA-induced grape seedless berries. Our findings present a novel insight into the roles of VvmiR66s that are responsive to GA in repressing the lignin synthesis of grape seedless berries, with different lignin-synthesis-enzyme-dependent action pathways in diverse plants, which have important implications for the molecular breeding of high-quality seedless grape berries.
In order to screen candidate aquaporin genes involved in resisting osmotic stress, we analyzed the physiological responses and the expression levels of aquaporin genes in garlic under drought and ...salt stress with ‘Er Shuizao’ as plant material. Different physiological indicators were detected under drought and salt stress treatments. RT-qPCR was used to detect the expression levels of the candidate aquaporin genes in specific tissues. Finally, we screened AsPIP1-3 as a candidate gene and analyzed its function. The results showed that the relative water content and chlorophyll content of leaves decreased, the O2− production rate increased, and H2O2 accumulated in garlic under drought and salt stress. The activities of SOD, POD, and CAT enzymes first increased and then decreased in garlic. The content of soluble sugar and proline increased to maintain cell osmotic balance, and the content of MDA and relative conductivity continued to increase. Most aquaporin gene expression first increased and then decreased in garlic under drought and salt stress. AsPIP1-3 gene expression is up-regulated under drought and salt stress in garlic. The relative expression was the highest on the 6th day of stress, being related to antioxidant enzyme activity and osmotic regulation. The consistent changes in gene expressions and physiological responses indicated that AsPIP1-3 played a role in resisting garlic osmotic stress. AsPIP1-3 was located on the cell membrane, being consistent with the predicted results of subcellular localization. The germination rate and root length of transgenic Arabidopsis under drought stress were significantly different from the wild type. Drought stress reduced the ROS accumulation of transgenic Arabidopsis, and the antioxidant enzyme activity was significantly higher than the wild type. The relative conductivity and MDA content significantly decreased, and the proline content increased under drought stress. The expression level of the genes related to drought stress response (AtRD22, AtP5CS, AtABF3, and AtLEA) significantly increased. The overexpression of AsPIP1-3 genes improved the drought tolerance of transgenic Arabidopsis plants, showing that AsPIP1-3 proteins enhanced drought tolerance. Our study laid a foundation for exploring the regulatory mechanism of garlic to abiotic stress.
The regulation of solid-state emission based on cocrystal engineering is an emerging strategy for developing next-generation luminescent materials. Here, three luminescent cocrystals (AA-DITFB, ...AA-TFTPA and AA-TCNB) of 9-anthraldehyde (9-AA) are reported, which exhibit a broad range of green-to-red emission. Tuning the photophysical properties of 9-AA via cocrystals is based on different mechanisms. Compared with single-component crystals, the difference in the emission properties of AA-DITFB and AA-TFTPA originates from the π–π interaction between chromophores. As for AA-TCNB, its prominent red-shifted emission is the result of the charge-transfer interaction, which is confirmed by infrared spectroscopy, “hole-electron” analysis and charge-transfer spectroscopy. This work not only reveals the relationship between molecular structure and fluorescent properties, but also proposes a strategy to develop multicolor luminescent systems with tunable efficiency and lifetime.
Thermosalient (TS) crystals have gained considerable attention due to their potential applications in various fields, including in actuators, sensors, energy harvesting, and artificial muscles. ...Herein, co-crystallization was employed to construct TS crystals by forming a twisted angle between aromatic rings. Two multicomponent trimethoprim (TMP) TS cocrystals, TMP-25HBA and TMP-OA, were obtained. Differential scanning calorimetry (DSC) and variable-temperature powder X-ray diffraction (VT-PXRD) revealed that TMP-OA undergoes a solid-to-solid phase transition, while TMP-25HBA does not exhibit any phase transition. To the best of our knowledge, TMP-25HBA is the first multicomponent TS crystal without phase transition. The TS effect of both crystals is a result of the unit cell’s anisotropic expansion.
This article presents a time-varying modal parameter identification method based on the novel information criterion (NIC) algorithm and a post-process method for time-varying modal parameter ...estimation. In the practical application of the time-varying modal parameter identification algorithm, the identified results contain both real modal parameters and aberrant ones caused by the measurement noise. In order to improve the quality of the identified results as well as sifting and validating the real modal parameters, a post-process procedure based on density-based spatial clustering of applications with noise (DBSCAN) algorithm is introduced. The efficiency of the proposed approach is first verified through a numerical simulation of a cantilever Euler-Bernoulli beam with a time-varying mass. Then the proposed approach is experimentally demonstrated by composite sandwich structure in a time-varying high temperature environment. The identified results illustrate that the proposed approach can obtain real modal frequencies in low signal-to-noise ratio (SNR) scenarios.
Sweating is an important purification process involved in the layer melt crystallization technique. Modeling of the sweating process remains difficult due to the complicated association of mass ...balance, heat balance, and fluid flow through a dynamic crystal layer. A model of sweating was proposed in this work, attempting to give a facile prediction of the dynamic sweat mass and concentration based on the initial crystal layer structure other than empirical correlations. Heat balance equations were taken as the governing equations to calculate the crystal layer temperature and residual melt concentration. A curved capillary bundle model that obeys fractal distribution was adapted to describe the dynamic porous crystal layer. By combining mass balance, the nonlinear variation of melted mass with sweating time was estimated. Subsequently, para-xylene was used as the model material to validate the sweating model. A quasiequilibrium between solid and liquid was found at a slow sweating rate range in this work, which verified the model assumption and greatly simplify the calculation. It revealed that the melting amount of the crystal layer and the imbibition melt increased nonlinearly with the increase of temperature. The curved capillary bundle model was found to be able to give an accurate estimation of sweat mass according to the experimental data. The model has accurate predictions on the S-shaped curve of sweat mass and sweat concentration compared to the experimental sweating results.
