The North American Project to Evaluate Soil Health Measurements was initiated with the objective to identify widely applicable soil health measurements for evaluation of agricultural management ...practices intended to improve soil health. More than 20 indicators were chosen for assessment across 120 long‐term agricultural research sites spanning from north‐central Canada to southern Mexico. The indicators being evaluated include common standard measures of soil, but also newer techniques of visible and near‐infrared reflectance spectroscopy, a smart phone app, and metagenomics. The aim of using consistent sampling and analytical protocols across selected sites was to provide a database of soil health indicator results that can be used to better understand how land use and management has affected the condition of soil ecosystem provisioning for agricultural biomass production and water resources, as well as nutrient and C cycling. The objective of this paper is to provide documentation of the overall design, and methods being employed to identify soil health indicators sensitive across agricultural management practices, pedologies, and geographies.
The present review explores the concept of learning within the context of neurorehabilitation after spinal cord injury (SCI). The aim of physical therapy and neurorehabilitation is to bring about a ...lasting change in function—to encourage learning. Traditionally, it was assumed that the adult spinal cord is hardwired—immutable and incapable of learning. Research has shown that neurons within the lower (lumbosacral) spinal cord can support learning after communication with the brain has been disrupted by means of a thoracic transection. Noxious stimulation can sensitize nociceptive circuits within the spinal cord, engaging signal pathways analogous to those implicated in brain-dependent learning and memory. After a spinal contusion injury, pain input can fuel hemorrhage, increase the area of tissue loss (secondary injury), and undermine long-term recovery. Neurons within the spinal cord are sensitive to environmental relations. This learning has a metaplastic effect that counters neural over-excitation and promotes adaptive learning through an up-regulation of brain-derived neurotrophic factor (BDNF). Exposure to rhythmic stimulation, treadmill training, and cycling also enhances the expression of BDNF and counters the development of nociceptive sensitization. SCI appears to enable plastic potential within the spinal cord by down-regulating the Cl− co-transporter KCC2, which reduces GABAergic inhibition. This enables learning, but also fuels over-excitation and nociceptive sensitization. Pairing epidural stimulation with activation of motor pathways also promotes recovery after SCI. Stimulating motoneurons in response to activity within the motor cortex, or a targeted muscle, has a similar effect. It is suggested that a neurofunctionalist approach can foster the discovery of processes that impact spinal function and how they may be harnessed to foster recovery after SCI.

Background
Blood collection from donors on testosterone therapy (TT) is restricted to red blood cell (RBC) concentrates to avoid patient exposure to supraphysiological testosterone (T). The objective ...of this study was to identify TT‐related changes in RBC characteristics relevant to transfusion effectiveness in patients.
Study Design
This was a two‐part study with cohorts of patients and blood donors on TT. In part 1, we conducted longitudinal evaluation of RBCs collected before and at three time points after initiation of T. RBC assays included storage and oxidative hemolysis, membrane deformability (elongation index), and oximetry. In part 2, we evaluated the fate of transfused RBCs from TT donors in immunodeficient mice and by retrospective analyses of NIH's vein‐to‐vein databases.
Results
TT increased oxidative hemolysis (1.45‐fold change) and decreased RBC membrane deformability. Plasma free testosterone was positively correlated with oxidative hemolysis (r = .552) and negatively correlated with the elongation index (r = −.472). Stored and gamma‐irradiated RBCs from TT donors had lower posttransfusion recovery in mice compared to controls (41.6 ± 12 vs. 55.3 ± 20.5%). Recipients of RBCs from male donors taking T had 25% lower hemoglobin increments compared to recipients of RBCs from non‐TT male donors, and had increased incidence (OR, 1.80) of requiring additional RBC transfusions within 48 h of the index transfusion event.
Conclusions
TT is associated with altered RBC characteristics and transfusion effectiveness. These results suggest that clinical utilization of TT RBCs may be less effective in recipients who benefit from longer RBC survival, such as chronically transfused patients.
The European X‐ray Free‐Electron Laser (FEL) became the first operational high‐repetition‐rate hard X‐ray FEL with first lasing in May 2017. Biological structure determination has already benefitted ...from the unique properties and capabilities of X‐ray FELs, predominantly through the development and application of serial crystallography. The possibility of now performing such experiments at data rates more than an order of magnitude greater than previous X‐ray FELs enables not only a higher rate of discovery but also new classes of experiments previously not feasible at lower data rates. One example is time‐resolved experiments requiring a higher number of time steps for interpretation, or structure determination from samples with low hit rates in conventional X‐ray FEL serial crystallography. Following first lasing at the European XFEL, initial commissioning and operation occurred at two scientific instruments, one of which is the Single Particles, Clusters and Biomolecules and Serial Femtosecond Crystallography (SPB/SFX) instrument. This instrument provides a photon energy range, focal spot sizes and diagnostic tools necessary for structure determination of biological specimens. The instrumentation explicitly addresses serial crystallography and the developing single particle imaging method as well as other forward‐scattering and diffraction techniques. This paper describes the major science cases of SPB/SFX and its initial instrumentation – in particular its optical systems, available sample delivery methods, 2D detectors, supporting optical laser systems and key diagnostic components. The present capabilities of the instrument will be reviewed and a brief outlook of its future capabilities is also described.

Phospholipid fatty acid analysis (PLFA) provides an easy to use and robust measure of changing soil microbial condition. The method provides data on both the quantity and composition of the soil ...microbial community- critical knowledge because the community is an important component of soil health. However, it is challenging for new researchers to know how to process data, how to interpret the results, and to know its effectiveness in evaluating soil health management. We set out to address these challenges using the North American Project to Evaluate Soil Health Measurements PLFA dataset. The dataset is comprised of results of over 1800 agricultural soil samples from a range of environmental regions and management practices. Using this dataset, we identified that quantifying soil PLFA biomass through summing the biomarkers from C14:0 to C20:0 yielded greater biomass results than summing specific biomarkers and was almost identical to summing C9:0 to C20:0. We utilized microbial biomass with common and novel biomarker ratios to evaluate the response of the soil microbial community to changes in the environment or changes in management practice. These ratios were based on commonly used genotypic categorization (e.g., Gram-positive to Gram-negative) or based on chemical structure with chemical ratios of either universal characteristics (e.g., unsaturated to saturated) or targeted phenotypic biomarkers known to shift with changing growth conditions (e.g., a15:0 and a17:0 to i15:0 and i17:0). One novel phenotypic ratio, the adaptation response ratio (ARR), significantly correlated with mean annual temperature and soil pH across grassland reference plots, a wheat climate transect, an intensive vegetable agricultural dataset, and a wheat-corn dataset from Mexico. Neither the common genotypic ratio nor ratios based on chemical structure demonstrated clear and consistent trends with environmental conditions. Targeted phenotypic biomarker ratios and microbial biomass also detected significant differences with soil health management practices of decreased tillage, cover crops, organic nutrients, residue retention, and rotation diversity. Therefore, with standardized microbial biomass and biomarker calculations that significantly vary with environmental conditions and management practices, these results support a wider understanding and adoption of the PLFA method in soil health management.
IntroductionBack and neck pain are the leading causes of disability worldwide. Doctors of chiropractic (DCs) are trained to manage these common conditions and can provide non-pharmacological ...treatment aligned with international clinical practice guidelines. Although DCs practice in over 90 countries, chiropractic care is rarely available within integrated healthcare delivery systems. A lack of DCs in private practice, particularly in low-income communities, may also limit access to chiropractic care. Improving collaboration between medical providers and community-based DCs, or embedding DCs in medical settings such as hospitals or community health centres, will improve access to evidence-based care for musculoskeletal conditions.Methods and analysesThis scoping review will map studies of DCs working with or within integrated healthcare delivery systems. We will use the recommended six-step approach for scoping reviews. We will search three electronic data bases including Medline, Embase and Web of Science. Two investigators will independently review all titles and abstracts to identify relevant records, screen the full-text articles of potentially admissible records, and systematically extract data from selected articles. We will include studies published in English from 1998 to 2020 describing medical settings that have established formal relationships with community-based DCs (eg, shared medical record) or where DCs practice in medical settings. Data extraction and reporting will be guided by the Proctor Conceptual Model for Implementation Research, which has three domains: clinical intervention, implementation strategies and outcome measurement. Stakeholders from diverse clinical fields will offer feedback on the implications of our findings via a web-based survey.Ethics and disseminationEthics approval will not be obtained for this review of published and publicly accessible data, but will be obtained for the web-based survey. Our results will be disseminated through conference presentations and a peer-reviewed publication. Our findings will inform implementation strategies that support the adoption of chiropractic care within integrated healthcare delivery systems.
Physical activity programs are viable contexts to foster life skills. Researchers contend that programs must be structured to deliberately teach life skills and teach how such skills can be ...transferred. The purpose of this study is to understand female youths' perceptions of life skills transfer from participation in a physical activity-based life skills program. Interviews were conducted with eight youth. Results indicated that youth learned intrapersonal (i.e., emotional regulation, focus, goal setting), interpersonal (i.e., respect, responsibility, social skills), and physical activity skills and applied these skills in other life domains. Leaders' practical strategies used to explicitly teach transferable skills are discussed.
Escherichia coli O157:H7 fecal shedding in feedlot cattle is common and is a public health concern due to the risk of foodborne transmission that can result in severe, or even fatal, disease in ...people. Despite a large body of research, few practical and cost-effective farm-level interventions have been identified. In this study, a randomized controlled trial was conducted to assess the effect of reducing the level of water in automatically refilling water-troughs on fecal shedding of E. coli O157:H7 in feedlot cattle. Pens in a feedlot in the Texas Panhandle were randomly allocated as control (total number: 17) or intervention (total number: 18) pens. Fecal samples (2,759 in total) were collected both at baseline and three weeks after the intervention, and tested for the presence of E. coli O157:H7 using immunomagnetic bead separation and selective culture. There was a strong statistical association between sampling date and the likelihood of a fecal sample testing positive for E. coli O157:H7. Pen was also a strong predictor of fecal prevalence. Despite accounting for this high level of clustering, a statistically significant association between reduced water levels in the trough and increased prevalence of E. coli O157:H7 in the feces was observed (Odds Ratio = 1.6; 95% Confidence Interval: 1.2-2.0; Likelihood Ratio Test: p = 0.02). This is the first time that such an association has been reported, and suggests that increasing water-trough levels may be effective in reducing shedding of E. coli O157:H7 in cattle feces, although further work would be needed to test this hypothesis. Controlling E. coli O157:H7 fecal shedding at the pre-harvest level may lead to a reduced burden of human foodborne illness attributed to this pathogen in beef.

Youth leadership programming has become an increasingly common context to foster basic psychological needs and promote youth development. The purpose of this qualitative study was to explore strategies involved in fostering youth needs support within six leadership programs. Two leaders and 30 youth participated in semi-structured interviews to better understand the strategies used to foster needs support. Findings revealed that leaders were able to foster a sense of relatedness among youth through building trusting adult-youth relationships and nurturing an inclusive environment. Maximizing choice and negotiating youth voice helped to foster youth’s autonomy. Finally, creating a task-oriented climate and providing intentional opportunities for skill-building helped to foster youth’s competence. Findings suggest that training for leaders is critical in understanding what, and how strategies should be employed to help foster youth needs support in leadership programming. Limitations and future directions are outlined.
