Seismic assessment of existing masonry structures requires a numerical model able to both reproduce their nonlinear behaviour and account for the different sources of uncertainties; the latter have ...to be dealt with since the unavoidable lack of knowledge on the input parameters (material properties, geometry, boundary conditions, etc.) has a relevant effect on the reliability of the seismic response provided by the numerical approaches. The steadily increasing necessity of combining different sources of information/knowledge makes the Bayesian approach an appealing technique, not yet fully investigated for historic masonry constructions. In fact, while the Bayesian paradigm is currently employed to solve inverse problems in several sectors of the structural engineering domain, only a few studies pay attention to its effectiveness for parameter identification on historic masonry structures. This study combines a Bayesian framework with probabilistic structural analyses: starting from the Bayesian finite element model updating by using experimental data it provides the definition of robust seismic fragility curves for non-isolated masonry towers. A comparison between this method and the standard deterministic approach illustrates its benefits. This article is part of the theme issue 'Environmental loading of heritage structures'.
The new Italian building code, published in 2018 MIT in NTC 2018: D.M. del Ministero delle Infrastrutture e dei trasporti del 17/01/2018. Aggiornamento delle Norme Tecniche per le Costruzioni (in ...Italian), 2018, explicitly refers to the Italian “Guidelines for the assessment and mitigation of the seismic risk of the cultural heritage” PCM in DPCM 2011: Direttiva del Presidente del Consiglio dei Ministri per valutazione e riduzione del rischio sismico del patrimonio culturale con riferimento alle norme tecniche per le costruzioni, G.U. n. 47 (in Italian), 2011 as a reliable source of guidance that can be employed for the vulnerability assessment of heritage buildings under seismic loads. According to these guidelines, three evaluation levels are introduced to analyse and assess the seismic capacity of historic masonry structures, namely: (1) simplified global static analyses; (2) kinematic analyses based on local collapse mechanisms, (3) detailed global analyses. Because of the complexity and the large variety of existing masonry typologies, which makes it particularly problematic to adopt a unique procedure for all existing structures, the guidelines provide different simplified analysis approaches for different structural configurations, e.g. churches, palaces, towers. Among the existing typologies of masonry structures there considered, this work aims to deepen validity, effectiveness and scope of application of the Italian guidelines with respect to heritage masonry towers. The three evaluation levels proposed by the guidelines are here compared by discussing the seismic risk assessment of a representative masonry tower: the Cugnanesi tower located in San Gimignano (Italy). The results show that global failure modes due to local stress concentrations cannot be identified if only simplified static and kinematic analyses are performed. Detailed global analyses are in fact generally needed for a reliable prediction of the seismic performance of such structures.
The recent developments of micro-electro-mechanical systems and wireless sensor networks allow today the use of low-cost and small-size sensors for continuous monitoring of civil structures. Both ...these features are very important for the low impact of the sensor grid in heritage structures, ensuring a low-cost and sustainable dynamic monitoring system. Over the last 20 years the use of sensor networks for continuous monitoring has received a growing interest. Anyway, still numerous questions remain opened about the sensitivity of measurement devices, the optimization of number and positioning of sensors, the energy efficiency of the network, and the development of algorithms for real-time data analysis. This paper, based on the aforementioned motivations, discusses about a monitoring system made of micro-electro-mechanical sensors connected through a wireless network. The architecture of the wireless sensor network and the automatized procedure proposed for the continuous processing of the recorded signals are discussed and described with reference to an explicative masonry tower case study. It is believed that the proposed technologies can provide an economical and relatively non-invasive tool for real-time structural monitoring and that, moreover, the availability of large amounts of data from actual measurements can give effective information on the structural behaviour of historic constructions.

The effectiveness of new shear test methods for evaluating the face-bonding quality of Cross-Laminated Timber (CLT) panels was examined by comparing experimental data and numerical modelling. The common characteristic of the specimens was the loading with angle of 45° with respect to the wood grain, in order to avoid rolling shear during test. In addition, the sampling methodology along the panel was investigated, as well as the relation between shear and delamination tests, and the possibility of coupling them using the same specimen. The results demonstrated that all the proposed shear test methods were effective for evaluating the quality of bonding among layers in CLT panels; however, the practical applicability of the methods led to elect the most suitable for inclusion in technical standards. Shear and delamination results proved not to be correlated, and the results showed that the size of the specimen is a crucial factor in determining the outcomes of delamination tests. Therefore, while it is feasible to propose the coupling of accelerated aging procedures with shear tests, the size of the samples need to be higher than the one tested here.
This paper reports the results of a ground penetrating radar (GPR) survey of the ground-floor of Academia Gallery (Florence, Italy) where the Michelangelo’s David is exhibited to the public. The ...equipment used was a step-frequency GPR operating in the 100 MHz-1 GHz band, named ORFEUS. The survey covered an area of 13 m × 7.3 m, and the scans were performed along two orthogonal directions. Acquisitions in the same direction were separated by 0.25 m from each other. The GPR was able to confirm the underground structure, as it can be deducted by planimetry and historical documentation. In particular, the radar clearly detected the air-conditioning ducts under the floor and an approximately circular foundation below the basement of the statue.
Oxidative stress can alter the expression level of many microRNAs (miRNAs), but how these changes are integrated and related to oxidative stress responses is poorly understood. In this article, we ...addressed this question by using in silico tools. We reviewed the literature for miRNAs whose expression is altered upon oxidative stress damage and used them in combination with various databases and software to predict common gene targets of oxidative stress-modulated miRNAs and affected pathways. Furthermore, we identified miRNAs that simultaneously target the predicted oxidative stress-modulated miRNA gene targets. This generated a list of novel candidate miRNAs potentially involved in oxidative stress responses. By literature search and grouping of pathways and cellular responses, we could classify these candidate miRNAs and their targets into a larger scheme related to oxidative stress responses. To further exemplify the potential of our approach in free radical research, we used our explorative tools in combination with ingenuity pathway analysis to successfully identify new candidate miRNAs involved in the ubiquitination process, a master regulator of cellular responses to oxidative stress and proteostasis. Lastly, we demonstrate that our approach may also be useful to identify novel candidate connections between oxidative stress-related miRNAs and autophagy. In summary, our results indicate novel and important aspects with regard to the integrated biological roles of oxidative stress-modulated miRNAs and demonstrate how this type of in silico approach can be useful as a starting point to generate hypotheses and guide further research on the interrelation between miRNA-based gene regulation, oxidative stress signaling pathways, and autophagy.
Recent Italian earthquakes have underlined the need for wide monitoring and safety assessment of architectonical heritage. This has emerged also from requirements of the new Italian Technical ...Recommendations for buildings. Within this subject the paper investigates the seismic vulnerability of a specific monumental masonry building: the Vicarious Palace (Palazzo del Vicario) in Pescia, a small town near Florence. The structural behavior of the Palace was investigated using a finite element model in which the non-linearities of the masonry were considered by proper constitutive assumptions. The seismic behavior was evaluated by the pushover method, according to the Italian Technical Recommendations. The results were compared with the ones obtained by a simplified approach based on the kinematic theorem of limit analysis. Comparisons of the expected seismic demand vs the seismic capacity of the Palace confirm the weakness of this type of building to suffer extensive damage under earthquakes, as frequently observed in similar construction typologies. Additionally, the comprehension of the structural behavior under seismic loading allows the identification of a proper retrofitting strategy.
The evaluation of the effects of pollution (e.g., Hg pollution) is a difficult task and relies mostly on biomonitoring based on bioindicators. The application of biomarkers may represent a ...complementary or alternative approach in environmental biomonitoring. Mercury is known to pose a significant health hazard due to its ability to cross cellular membranes, bioaccumulate, and biomagnify. In the present research, the effects of short-term (i.e., 24 h) Hg exposure in the symbiont-bearing benthic foraminiferal species Amphistegina lessonii are evaluated using several biomarkers (i.e., proteins and enzymes). Mercury leads to significant changes in the biochemistry of cells. Its effects are mainly associated with oxidative stress (i.e., production of reactive oxygen species: ROS), depletion of glutathione (GSH), and alteration of protein synthesis. Specifically, our findings reveal that exposure to Hg leads to the consumption of GSH by GPx and GST for the scavenging of ROS and the activation of antioxidant-related enzymes, including SOD and GSH-enzymes (GST, GSR, GPx, and Se-GPx), that are directly related to a defense mechanism against ROS. The Hg exposure also activates the MAPK (e.g., p-p38) and HSP (e.g., HSP 70) pathways. The observed biochemical alterations associated with Hg exposure may represent effective and reliable proxies (i.e., biomarkers) for the evaluation of stress in A. lessonii and lead to a possible application for the detection of early warning signs of environmental stress in biomonitoring.
This paper summarizes the results of a two-day dynamic monitoring of Michelangelo's David subject to environmental loads (city traffic and pedestrian loading induced by tourists visiting the ...Accademia Gallery). The monitoring was carried out by a no-contact technique using an interferometric radar, whose effectiveness in measuring the resonant frequencies of structures and historic monuments has proved over the last years through numerous monitoring activities. Owing to the dynamic behavior of the measurement system (radar and tripod), an accelerometer has been installed on the radar head to filter out the movement component of the measuring instrument from the measurement of the David's displacement. Measurements were carried out in the presence and absence of visitors, to assess their influence on the dynamic behavior of the statue. A numerical model of the statue was employed to evaluate the experimental results.
An emerging problem for human health is the exposure to non-regulated mineral fibers with an asbestos-like crystal habit, particularly fibrous zeolites. This study aimed to determine if and how ...selected fibrous zeolites (erionite, mesolite, and thomsonite) induce toxicity effects on two different in vitro cellular models, the adherent murine hippocampal (HT22) and human immortalized T lymphocyte (Jurkat) cell lines. Before proceeding with the cellular tests, the three zeolite samples were investigated using scanning electron microscopy–energy-dispersive spectroscopy and X-ray powder diffraction techniques. The cells were treated with 0.1 µM and 1 µM of fibrous erionite, mesolite, and thomsonite for 12, 24, and 48 h. Results showed a cytotoxic effect of erionite in both cellular models and revealed different toxic behaviors of the mesolite and thomsonite fibers, suggesting other potential mechanisms of action. The outcome of this study would be a first step for further research on fine biochemical interactions of zeolite fibers with cells and future in vivo investigations.
