This study describes the purification of Bixa orellana L. leaf lectin (BoLL) and the evaluation of its antimicrobial activity and in vivo toxicity. BoLL was isolated from saline extract of leaves ...through protein precipitation with ammonium sulfate (60% saturation) followed by ion exchange (CM-Sephadex) and size exclusion (Sephadex G-75) chromatographies. The lectin was characterized by polyacrylamide gel electrophoresis under denaturing conditions (SDS-PAGE), stability towards heating and pH variation, as well as influence of ions on its hemagglutinating activity (HA). HA inhibition assay by different carbohydrates and glycoproteins was also performed. The antimicrobial activity of BoLL was investigated against pathogens for humans and plants. In vivo acute toxicity in mice was assessed using a dose of 100 mg/kg per os or i.p. BoLL was isolated with high specific HA (purification fold: 53.89) and showed a single 19-kDa polypeptide band in SDS-PAGE. BoLL SHA increased in the presence of 20 mM Mg2+ ions, was resistant to heating at 100 °C, and stable at acidic pH range. HA inhibition was detected especially by glycoproteins, but also by monosaccharides. BoLL showed bacteriostatic and bactericidal effects against Escherichia coli minimum inhibitory (MIC) and bactericidal (MBC) concentrations of 400 and 800 μg/mL, respectively and Staphylococcus aureus (MIC and MBC: 400 μg/mL), as well as bacteriostatic effect against Bacillus megaterium and Micrococcus luteus (MIC: 18.2 and 800 μg/mL, respectively). No changes were observed in hematological, biochemical, and behavioral parameters in the acute toxicity test. However, animals treated with BoLL at 100 mg/kg per os or i.p. showed histopathological alterations in the liver, spleen, and kidneys. In conclusion, our data add new biotechnological value for B. orellana leaves, since they contain a thermostable lectin with antibacterial activity, which stimulate future studies aiming at its biomedical application. However, caution should be considered in future studies due to some signs of toxicity in mice.
•A protocol for purification of a lectin from Bixa orellana leaf (BoLL) was defined.•BoLL showed a single 19-kDa polypeptide band in SDS-PAGE.•Hemagglutinating activity of BoLL increased in the presence of Mg2+ ions, was resistant to heating at 100 °C, and stable at acidic pH range.•BoLL showed bacteriostatic and bactericidal effects•BoLL (100 mg/kg per os or i.p.) did not cause the death of Swiss mice.
Low areal bone mineral density (BMD), increased fracture risk and altered bone remodeling have been described among stone formers (SFs), but the magnitude of these findings differs by age, sex, ...menopausal status and urinary calcium (uCa). This study aimed to investigate volumetric BMD (vBMD), bone microarchitecture and biomechanical properties by high-resolution peripheral quantitative computed tomography (HR-pQCT) and finite element analysis (FEA) in young SFs, irrespective of calciuria, further distinguishing trabecular from cortical compartments.
HR-pQCT/FEA was performed at the distal tibia (DT) and distal radius (DR) in 106 SFs (57 males and 49 premenopausal females; median age 37 years) and compared with 106 non-SFs (NSFs) retrieved from an existing database, matched for age, sex and body mass index (BMI). Biochemical/hormonal serum and urinary parameters were obtained from SFs.
SFs exhibited significantly lower trabecular number (TbN) and higher trabecular separation (TbSp) than NSFs at both anatomical sites and lower cortical porosity in the DR. In a subgroup analysis separated by sex, female SFs presented significantly lower TbvBMD, relative bone volume fraction (BV/TV) and TbN and higher TbSp than NSFs at both sites, while male SFs showed significantly lower stiffness and failure load. Multivariate analysis showed TbN to be independently associated with sex and BMI at both sites and with uCa at the DR.
The present findings suggest that bone disease represents an early event among SFs, associated at least in part with calcium excretion and mainly characterized by trabecular bone microarchitecture impairment, especially among women, but with reduced bone strength parameters in men.
Brunsting-Perry type pemphigoid (BPP) is a rare subepidermal blistering disease and a cause of secondary cicatricial alopecia. It was originally described by Brunsting and Perry in 1957 as a rare ...variant of cicatricial pemphigoid, characterized by bullous lesions limited to the head, neck, scalp, and upper trunk with mild or no mucosal involvement. We report 2 cases of BPP cicatricial alopecia with histopathology of subepidermal blister formation, different clinical presentation, and different salt-split test results. One patient had features of bullous pemphigoid (BP) with important oral mucosal involvement (not yet reported in the literature), and the second patient had typical features of epidermolysis bullosa acquisita (EBA). The secondary cicatricial alopecia may be due to different antigens associated with either BP or EBA. The phenomenon of epitope spreading could explain the association between 2 distinctive bullous diseases in the same patient, justifying the divergent findings of the immunofluorescence. The specific target antigen of BPP is yet to be defined.
Diabetes mellitus (DM) and thyroid dysfunction (TD) are the two most common endocrine disorders in clinical practice. The unrecognized TD may adversely affect the metabolic control and add more risk ...to an already predisposing scenario for cardiovascular diseases. The objective of this study was to investigate the prevalence of TD in patients with type 1 and type 2 diabetes mellitus (T1DM and T2DM).
This is an observational cross-sectional study. Three hundred eighty-six (386) patients with T1DM or T2DM that regularly attended the outpatient clinic of the Diabetes unit, Hospital Universitário Pedro Ernesto, participated in the study. All patients underwent a clinical and laboratory evaluation. Thyroid dysfunction was classified as clinical hypothyroidism (C-Hypo) if TSH > 4.20 μUI/mL and FT4 < 0.93 ng/dL; Subclinical hypothyroidism (SC-Hypo) if TSH > 4.20 μUI/ml and FT4 ranged from 0.93 to 1.7 ng/dL; Subclinical hyperthyroidism (SC-Hyper) if TSH < 0.27 μUI/ml and FT4 in the normal range (0.93 and 1.7 ng/dL) and Clinical hyperthyroidism (C-Hyper) if TSH < 0.27 μUI/ml and FT4 > 1.7 μUI/mL. Autoimmunity were diagnosed when anti-TPO levels were greater than 34 IU/mL. The positive autoimmunity was not considered as a criterion of thyroid dysfunction.
The prevalence of TD in all diabetic patients was 14,7%. In patients who had not or denied prior TD the frequency of TD was 13%. The most frequently TD was subclinical hypothyroidism, in 13% of patients with T1DM and in 12% of patients with T2DM. The prevalence of anti-TPO antibodies was 10.8%. Forty-four (11.2%) new cases of TD were diagnosed during the clinical evaluation. The forty-nine patients with prior TD, 50% with T1DM and 76% with T2DM were with normal TSH levels.
We conclude that screening for thyroid disease among patients with diabetes mellitus should be routinely performed considering the prevalence of new cases diagnosed and the possible aggravation the classical risk factors such as hypertension and dyslipidemia, arising from an undiagnosed thyroid dysfunction.
