Polo-like kinase 1 (Plk1) is a conserved serine/threonine kinase that plays an essential role in regulating the many processes involved in mitotic entry and progression. In humans, Plk1 is expressed ...primarily during late G(2) and M phases and, in conjunction with Cdk1/cyclin B1, acts as master regulatory kinases for the myriad protein substrates involved in mitosis. Plk1 overexpression is strongly associated with cancer and has been correlated with poor prognosis in a broad range of human tumor types. We have identified a potent, selective, reversible, ATP-competitive inhibitor of Plk1, GSK461364A, capable of inhibiting cell growth of most proliferating cancer cell lines tested. We observe distinct cell cycle effects of GSK461364A depending on the dose used. The predominant phenotype for cells treated with GSK461364A is prometaphase arrest with characteristic collapsed polar polo spindle. At high concentrations, GSK461364A delays mitotic entry in G(2) followed by gradual progression into terminal mitosis; in some cell lines, this correlates with decreased apoptosis. Cell culture growth inhibition by GSK461364A can be cytostatic or cytotoxic but leads to tumor regression in xenograft tumor models under proper dose scheduling. Finally, we describe pharmacodynamic biomarkers of GSK461364A activity (pHH3 and Plk1) that are currently being evaluated in human cancer clinical trials.
This paper presents a geologic and mineralogical investigation of asbestiform amphibole from blueschist in the Diablo Range, northern California. The analysis of fibers in air samples shows that the ...dominant blueschist amphibole composition ranges from glaucophane to subordinate winchite. In outcrop, blueschist amphibole exhibits a velvety luster, and its occurrence ranges from crosscutting veins to highly deformed foliated and lineated tectonites. TEM and SEM photographs reveal a highly fibrous habit typically associated with asbestiform amphiboles. Dimensional analysis reveals a mean fiber width of 0.27 μm, and lengths and aspect ratios are shorter than reported for commercially exploitable asbestos, with a mean length of 2.8 μm and mean aspect ratio of 11.5. The data are consistent with other research showing that the width population of fibers, and not length or aspect ratio, is the key indicator of the asbestiform habit. The blueschist data are compared to non-asbestiform hornblende amphibole in granitic rocks. The fiber concentration of the hornblende (458 MFG) is low compared to the blueschist amphibole (107,880 MFG) and chrysotile in serpentinite (196,066 MFG), indicating that under similar conditions, the exposure potential from cleaved particles could be several orders of magnitude lower than from asbestiform particles. The asbestiform habit of glaucophane is present at two other locations in the Franciscan Complex, suggesting that asbestiform blueschist amphibole may be characteristic within blueschist terranes, and rock formations containing asbestiform amphibole may be more common and widespread than previously assumed.

The roles of divalent metal ions in DNA cleavage by the
EcoRV endonuclease were studied by using Co
2+ or Mn
2+ as substitutes for the natural cofactor Mg
2+. In steady-state experiments with a 12 bp ...oligonucleotide substrate, Co
2+ yielded a similar turnover rate to that with Mg
2+, but Mn
2+ gave a slower rate. Single turnovers of
EcoRV on this substrate were analysed by stopped-flow and quench-flow methods, to determine the rates for the formation of the ternary enzyme-DNA-metal complex, the hydrolysis of the phosphodiester bonds and the dissociation of the cleaved DNA. With Co
2+, all three steps had similar rates to those with Mg
2+. In contrast, Mn
2+ gave a faster rate for phosphodiester hydrolysis than either Mg
2+ or Co
2+, but a slower rate for product dissociation, thus accounting for its low turnover rate. Single turnovers on plasmids also yielded faster rates for substrate hydrolysis with Mn
2+ compared to Mg
2+ and Co
2+. Since Mn
2+ gave the most rapid rates for the hydrolytic step, despite being less electronegative than Co
2+, the function of the metal ion at the active site of
EcoRV cannot be just the polarisation of the scissile phosphate. Moreover, the minimal scheme for the Co
2+-catalysed reaction requires two metal ions for DNA cleavage. The metal ions seem to be involved in the precise positioning of both the substrate and the water that acts as the attacking nucleophile and in activating that water molecule. A model is presented to account for how two metal ions might fulfil these functions.
An (r,z,k)-mixed graph G has every vertex with undirected degree r, directed in- and out-degree z, and diameter k. In this paper, we study the case r=z=1, proposing some new constructions of ...(1,1,k)-mixed graphs with a large number of vertices N. Our study is based on computer techniques for small values of k and the use of graphs on alphabets for general k. In the former case, the constructions are either Cayley or lift graphs. In the latter case, some infinite families of (1,1,k)-mixed graphs are proposed with diameter of the order of 2log2N.
Relational loss, death, and mourning are all part of life and human relationships. Protracted and compounded grief occurs when something interferes with the normal grieving process. The psychotherapy ...of grief is described and two primary relational approaches are emphasized: face-to-face psychotherapy and the use of internal imagery via the empty-chair technique. In both approaches, the aim is to provide a balanced expression of affect, including emotional pain, anger, resentment, appreciation, and love. Several case examples are provided.
