Bean anthracnose is a seedborne disease of common bean (Phaseolus vulgaris) caused by the fungal pathogen Colletotrichum lindemuthianum. Using seed that did not test positive for the pathogen has ...been proven to be an effective strategy for bean anthracnose control. To quantify the extent of anthracnose seed infection, a real‐time PCR‐based diagnostic assay was developed for detecting C. lindemuthianum in seeds of the commercial bean class navy bean. The ribosomal DNA (rDNA) region consisting of part of the18S rDNA, 5.8S rDNA, internal transcribed spacers (ITS) 1, 2 and part of the 28S rDNA of seven races of C. lindemuthianum, 21 isolates of Colletotrichum species and nine other bean pathogens were sequenced with the universal primer set ITS5/ITS4. Based on the aligned sequence matrix, one primer set and a probe were designed for a SYBR Green dye assay and a TaqMan MGB (minor groove binder) assay. The primer set was demonstrated to be specific for C. lindemuthianum and showed a high sensitivity for the target pathogen. The detection limit of both assays was 5 fg of C. lindemuthianum genomic DNA. To explore the correlation between the lesion area and the DNA amount of C. lindemuthianum in bean seed, seeds of the navy bean cultivar Navigator with lesions of different sizes, as well as symptomless seeds, were used in both real‐time PCR assays.
The last decade has witnessed great interest in research on content-based image retrieval (CBIR). In 2009, Lin et al. proposed a smart CBIR system based on colour and texture feature. Their system ...has a high detection rate except the cases where image objects have similar shapes. To enhance the detection rate a shape-based image feature called object-moment is proposed in this paper. Object-moment uses the moment of force to compute the object edge feature by calculating the distance from each edge pixel to the axis, and adding them up as a feature. Besides, we integrate the colour features (NSOM, CSOM) and the texture features (CCM, DBPSP) to enhance image detection rate and simplify computation of image retrieval. A series of analyses and comparisons are performed in our experiments to demonstrate that our proposed method improves the retrieval accuracy significantly.
The 3-stage Clos network is the most-studied switching network. However, exact conditions on the strictly nonblocking multicast three-stage Clos network under various models are difficult to get, due ...to the messy detail and because each case employs a different argument. Hwang and Liaw (2000) made the latest attempt and pointed out errors in previous attempts. However, they made errors, too. In this paper, we propose a unifying approach to study those models systematically (which also applies to some wide-sense nonblocking (WSNB) networks). We also propose a new routing algorithm, and use the unifying approach to derive a necessary and sufficient condition for WSNB.
We find that less than 0.01 monolayer of S can enhance surface self-diffusion on Cu(111) by several orders of magnitude. The measured dependence of two-dimensional island decay rates on S coverage ...(theta(S)) is consistent with the proposal that Cu3S3 clusters are responsible for the enhancement. Unexpectedly, the decay and ripening are diffusion limited with very low and very high theta(S) but not for intermediate theta(S). To explain this result we propose that surface mass transport in the intermediate region is limited by the rate of reaction to form Cu3S3 clusters on the terraces.
We have studied the dislocation structures that occur in films of Ag, Au, and Ag
0.5Au
0.5 alloy on a Ru(0
0
0
1) substrate. Monolayer (ML) films form herringbone phases while films two or more ...layers thick contain triangular patterns of dislocations. We use scanning tunneling microscopy (STM) and low-energy electron diffraction (LEED) to determine how the film composition affects the structure and periodicity of these ordered structures. One layer of Ag forms two different herringbone phases depending on the exact Ag coverage and temperature. Low-energy electron microscopy (LEEM) establishes that a reversible, first-order phase transition occurs between these two phases at a certain temperature. We critically compare our 1
ML Ag structures to conflicting results from an X-ray scattering study H. Zajonz et al., Phys. Rev. B 67 (2003) 155417. Unlike Ag, the herringbone phases of Au and AgAu alloy are independent of the exact film coverage. For two layer films in all three systems, none of the dislocations in the triangular networks thread into the second film layer. In all three systems, the in-plane atomic spacing of the second film layer is nearly the same as in the bulk. Film composition does, however, affect the details of the two layer structures. Ag and Au films form interconnected networks of dislocations, which we refer to as “trigons.” In 2
ML AgAu alloy, the dislocations form a different triangular network that shares features of both trigon and moiré structures. Yet another well-ordered structure, with square symmetry, forms at the boundaries of translational trigon domains in 2
ML Ag films but not in Au films.
Using low-energy electron microscopy, we show that de-wetting of Ag and Cu films on Ru(0
0
0
1) occurs by 3-D islands migrating across step edges in the “downhill” direction. We have observed islands ...thicken by more than 50 atomic layers in this manner. The island migration allows 3-D growth to occur in a way that avoids the nucleation barrier associated with forming 2-D islands on top of 3-D islands. Indeed, without substrate steps this nucleation barrier is not surmounted, and no 3-D islands are formed in the films studied.
A cDNA library was constructed from field pea leaves infected by the downy mildew pathogen, Peronospora viciae f. sp. pisi, using a suppression subtractive hybridisation approach. The library ...consists of 399 expressed sequence tags, from which 207 unisequences were obtained after sequence assembly. Of the unisequences, six were shown to be of Peronospora viciae f. sp. pisi origin. The remaining unisequences were subjected to gene ontology analysis and their functions were predicted in silico. Eleven of these unisequences (representing 24 clones) shared significant sequence similarities with Arabidopsis genes known to be involved in downy mildew resistance, including the well‐characterised genes RPP5, RPP6 and RPP27. Expression analysis of five selected unisequences by real‐time PCR indicated that all five were up‐regulated during downy mildew pathogenesis, suggesting a significant role for these genes in the host response to downy mildew infection.
