Functional ingredients for human health have recently become the focus of research. One such potentially versatile therapeutic component is fucose-containing sulfated polysaccharides (FCSPs), ...referred to as fucoidans. The exploitation of marine brown algae provides a rich source of FCSPs because of their role as a structural component of the cell wall. Fucoidans are characterized by a sulfated fucose backbone. However, the structural characterization of FCSPs is impeded by their structural diversity, molecular weight, and complexity. The extraction and purification conditions significantly influence the yield and structural alterations. Inflammation is the preliminary response to potentially injurious inducements, and it is of the utmost importance for modulation in the proper direction. Improper manipulation and/or continuous stimuli could have detrimental effects in the long run. The web of immune responses mediated through multiple modulatory/cell signaling components can be addressed through functional ingredients, benefiting patients with no side effects. In this review, we attempted to address the involvement of FCSPs in the stimulation/downregulation of immune response cell signaling. The structural complexity and its foremost influential factor, extraction techniques, have also attracted attention, with concise details on the structural implications of bioactivity.

Abstract We present an observational study of wind acceleration based on four low-ionization broad absorption line (BAL) quasars (J0136, J1238, J1259, and J1344). J0136 and J1344 (group 1) are ...radio-quiet and show large BAL-velocity shifts as opposed to stable line-locking associated absorption lines (AALs). Notably, J1344 displays a linear relation between BAL-velocity shift and time interval over three consecutive epochs, characteristic of compelling evidence for BAL acceleration. J1238 and J1259 (group 2) exhibit small BAL-velocity shifts along with steep-spectrum, weak radio emission at 3.0 and 1.4 GHz. All four quasars have spectral energy distributions (SEDs) with a peak at λ rest ∼ 10 μ m, suggesting a link between the BAL acceleration and hot dust emission. The group-2 quasars are redder than group-1 quasars and have a steeper rise at 1 μ m < λ rest < 3 μ m in their SEDs. All but J1238 exhibit a steep rise followed by a plateau-like time evolution in BAL-velocity shift. Our investigations, combined with previous studies of BAL acceleration, indicate that (1) the coupling process between the BALs and the interstellar medium (ISM) is one of the major avenues for the origin of quasar reddening and patchy obscuration, (2) AAL outflows are ubiquitous and likely signify large-scale remnants of BAL winds coupled to the ISM, and (3) wind deceleration that is closely linked to the BAL–ISM coupling process may produce weak radio emission in otherwise radio-quiet quasars.

The remote sensing of Earth surface changes is an active research field aimed at the development of methods and data products needed by scientists, resource managers, and policymakers. Fire is a ...major cause of surface change and occurs in most vegetation zones across the world. The identification and delineation of fire-affected areas, also known as burned areas or fire scars, may be considered a change detection problem. Remote sensing algorithms developed to map fire-affected areas are difficult to implement reliably over large areas because of variations in both the surface state and those imposed by the sensing system. The availability of robustly calibrated, atmospherically corrected, cloud-screened, geolocated data provided by the latest generation of moderate resolution remote sensing systems allows for major advances in satellite mapping of fire-affected area. This paper describes an algorithm developed to map fire-affected areas at a global scale using Moderate Resolution Imaging Spectroradiometer (MODIS) surface reflectance time series data. The algorithm is developed from the recently published Bi-Directional Reflectance Model-Based Expectation change detection approach and maps at 500 m the location and approximate day of burning. Improvements made to the algorithm for systematic global implementation are presented and the algorithm performance is demonstrated for southern African, Australian, South American, and Boreal fire regimes. The algorithm does not use training data but rather applies a wavelength independent threshold and spectral constraints defined by the noise characteristics of the reflectance data and knowledge of the spectral behavior of burned vegetation and spectrally confusing changes that are not associated with burning. Temporal constraints are applied capitalizing on the spectral persistence of fire-affected areas. Differences between mapped fire-affected areas and cumulative MODIS active fire detections are illustrated and discussed for each fire regime. The results reveal a coherent spatio-temporal mapping of fire-affected area and indicate that the algorithm shows potential for global application.
The limited availability of treatments for many infectious diseases highlights the need for new treatments, particularly for viral infections. Natural compounds from seaweed are attracting increasing ...attention for the treatment of various viral diseases, and thousands of novel compounds have been isolated for the development of pharmaceutical products. Seaweed is a rich source of natural bioactive compounds, including polysaccharides. The discovery of algal polysaccharides with antiviral activity has significantly increased in the past few decades. Furthermore, unique polysaccharides isolated from seaweeds, such as carrageenan, alginates, fucoidans, galactans, laminarians, and ulvans, have been shown to act against viral infections. The antiviral mechanisms of these agents are based on their inhibition of DNA or RNA synthesis, viral entry, and viral replication. In this article, we review and provide an inclusive description of the antiviral activities of algal polysaccharides. Additionally, we discuss the challenges and opportunities for developing polysaccharide-based antiviral therapies, including issues related to drug delivery and formulation. Finally, this review highlights the need for further research for fully understanding the potential of seaweed polysaccharides as a source of antiviral agents and for developing effective treatments for viral diseases.

This was witnessed to be achieved via fucoidan acting on mediators such as iNOS and COX-2 including pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β), with dose dependent down-regulation. Further, the effect is exhibited by the suppression of TLR mediated MyD88, IKK complex, ultimately hindering NF-κB and MAPK activation, proposing its therapeutic applications in inflammation related disorders. The research findings provide an insight in relation to the sustainable utilization of fucoidan from marine brown algae
as a potent anti-inflammatory agent in the nutritional, pharmaceutical, and cosmeceutical sectors.
Fish head byproducts derived from surimi processing contribute about 15% of the total body weight, which are beneficial to health because they contain essential nutrients. In this study, olive ...flounder (OF) was the target species in order to maximize the byproduct utilization. In RAW 264.7 macrophages, the seven hydrolysates from OF head byproducts were examined for their inhibitory potential against inflammation and the oxidative stress induced by lipopolysaccharides (LPS). The pepsin hydrolysate (OFH-PH) demonstrated strong anti-inflammatory activity via the down-regulation of NO production, with an IC50 value of 299.82 ± 4.18 µg/mL. We evaluated the inhibitory potential of pro-inflammatory cytokines and PGE2 to confirm these findings. Additionally, iNOS and COX-2 protein expressions were confirmed using western blotting. Furthermore, the results from the in vivo zebrafish model demonstrated that OFH-PH decreased the LPS-elevated heart rate, NO production, cell death, and intracellular ROS level, while increasing the survival percentage. Hence, the obtained results of this study serve as a platform for future research and provide insight into the mediation of inflammatory disorders. These results suggest that OFH-PH has the potential to be utilized as a nutraceutical and functional food ingredient.
Activation of human telomerase reverse transcriptase (hTERT) is necessary for limitless replication in tumorigenesis. Whereas hTERT is transcriptionally silenced in normal cells, most tumor cells ...reactivate hTERT expression by alleviating transcriptional repression through diverse genetic and epigenetic mechanisms. Transcription-activating hTERT promoter mutations have been found to occur at high frequencies in multiple cancer types. These mutations have been shown to form new transcription factor binding sites that drive hTERT expression, but this model cannot fully account for differences in wild-type (WT) and mutant promoter activation and has not yet enabled a selective therapeutic strategy. Here, we demonstrate a novel mechanism by which promoter mutations activate hTERT transcription, which also sheds light on a unique therapeutic opportunity. Promoter mutations occur in a core promoter region that forms tertiary structures consisting of a pair of G-quadruplexes involved in transcriptional silencing. We show that promoter mutations exert a detrimental effect on the folding of one of these G-quadruplexes, resulting in a nonfunctional silencer element that alleviates transcriptional repression. We have also identified a small drug-like pharmacological chaperone (pharmacoperone) molecule, GTC365, that acts at an early step in the G-quadruplex folding pathway to redirect mutant promoter G-quadruplex misfolding, partially reinstate the correct folding pathway, and reduce hTERT activity through transcriptional repression. This transcription-mediated repression produces cancer cell death through multiple routes including both induction of apoptosis through inhibition of hTERT’s role in regulating apoptosis-related proteins and induction of senescence by decreasing telomerase activity and telomere length. We demonstrate the selective therapeutic potential of this strategy in melanoma cells that overexpress hTERT.
This paper studies the dynamics of a spinning three-body tethered satellite formation by a flexible multibody dynamics method. The tethered system is specially designed to constitute a Space Infrared ...Interferometric Telescope for a space exploration mission. Two sub-satellites are deployed from the main-satellite by using long flexible tethers and moving along trajectories of Archimedes spirals. Both of the large displacement and large deformation of the variable-length tethers are described by utilizing the Absolute Nodal Coordinate Formulation method in a framework of Arbitrary Lagrange-Euler (ANCF-ALE) description. The Natural Coordinate Formulation (NCF) method is used for describing the overall rigid motions of the three satellites. Considering the mass flow of tether element, the dynamic equations of tether element are derived by using the D'Alembert's principle. The governing dynamic equations of the system are obtained with a method of Lagrange multipliers. The tether deployment is achieved by regulating deployed velocity and applied jet forces. Numerical case studies are presented for investigating the parametric influences on the deployment dynamics of the tethered system by comparisons.
