Spinal fusion is still considered to be an important option for treating various spinal disorders. To induce solid spinal fusion, osteoinductive and/or osteoconductive agents have been widely adopted. Autogenous LF, however, has never been seriously considered as a carrier for ex vivo osteoinductive gene therapy for spinal fusion.
In vitro experiment: Degenerated human LF was harvested and cultured. Type 5 adenovirus lacZ (Ad/lacZ) and BMP-2 construct (Ad/BMP-2) were produced. LF cell cultures were then exposed to Ad/BMP-2. Expressions of osteocalcin and BMP-2 mRNA were analyzed by reverse transcriptase-polymerase chain reaction (RT-PCR). Western blot analysis was performed to detect osteocalcin protein. Alkaline phosphatase and von Kossa stains were used to detect osteogenic markers and bone nodule formation, respectively. In vivo experiment: Human LF tissues treated with Ad/lacZ, Ad/BMP-2, and saline were implanted into the subcutaneous tissue of nude mice. After 4 weeks, nude mice were radiographed and killed. Implanted LF tissues were harvested and histologically stained.
LF cell cultures with Ad/BMP-2 revealed strong expression of BMP-2 and osteocalcin mRNA in RT-PCR and osteocalcin protein in western blot analysis. LF cell culture with saline showed baseline expression of BMP-2, osteocalcin mRNA, and osteocalcin protein, respectively. Furthermore, LF cell culture with Ad/BMP-2 demonstrated the expression of alkaline phosphatase and bone nodule formation in the aforementioned histochemical stain. LF tissues with Ad/BMP-2 revealed de novo osteogenesis in nude mice, whereas LF with Ad/lacZ or saline showed only remaining LF tissue without sign of bone formation.
Human LF cells transduced with Ad/BMP-2 exhibited the expression of osteogenic phenotype and bone nodule formation. Additionally, genetically modified human LF with BMP-2 cDNA clearly demonstrated de novo osteogenesis, which supports the concept that biologically modified LF can be a substitute for autogenous bone graft in spinal fusion surgery.
본 연구는 자일로올리고당의 과학적이고 체계적인 표준 화된 시험법을 마련하여 다양한 제형의 제품에 적용하고 자 하였다. 최적화된 시험법을 마련하기 위해 초음파 처 리 시간, 산 가수분해 시간 및 농도를 검토하여 전처리 방 법을 비교 평가하였으며, HPLC-UVD를 이용하여 시료 중 의 자일로올리고당을 분석하였다. 분석 시 1-phenyl-3- ...methyl-5-pyrazoline (PMP)으로 유도체화하고, photo diode array (PDA)가 장착된 high performance liquid chromatography (HPLC) (Nanospace SI-2)를 사용하였으며, 칼럼은 Cadenza C18 (4.6 × 250 mm, 3 μm)이었으며, 이동상은 pH를 6.0으 로 맞춘 20 mM 인산완충용액과 아세토니트릴을 78:22 비 율로 혼합하여 사용하였고, 0.5 mL/min 유속으로 254 nm 로 하여 분석하였다. 건강기능식품 등 시험법 마련 표준 절차에 관한 가이드라인에 따라 밸리데이션을 수행하고, 표준화된 시험법을 이용하여 유통 중인 건강기능식품 대 상 품목에 대해 시험법 적용 여부를 확인하였다. 표준화 된 시험법은 자일로올리고당을 함유한 건강기능식품 품질 관리에 대한 신뢰성을 더 높일 것으로 본다.
This study aimed to develop a scientifically and systematically standardized xylooligosaccharide analytical method that can be applied to products with various formulations. The analysis method was conducted using HPLC with Cadenza C18 column, involving pre-column derivatization with 1-phenyl-3-methyl-5-pyrazoline (PMP) and UV detection at 254 nm. The xylooligosaccharide content was analyzed by converting xylooligosaccharide into xylose through acid hydrolysis. The pre-treated methods were compared and evaluated by varying sonication time, acid hydrolysis time, and concentration. Optimal equipment conditions were achieved with a mobile phase consisting of 20 mM potassium phosphate buffer (pH 6)-acetonitrile (78:22, v/v) through isocratic elution at a flow rate of 0.5 mL/min (254 nm). Furthermore, we validated the advanced standardized analysis method to support the suitability of the proposed analytical procedure such as specificity, linearity, detection limits (LOD), quantitative limits (LOQ), accuracy, and precision. The standardized analysis method is now in use for monitoring relevant health-functional food products available in the market. Our results have demonstrated that the standardized analysis method is expected to enhance the reliability of quality control for healthy functional foods containing xylooligosaccharide.
In this paper, we have evaluated select gate lateral coupling (SGLC) cell by using a 4kbits (kb) macro. Using large data cell VT distributions, tail bit behavior can be analyzed. The test chip is ...fabricated on a 90 nm high voltage CMOS process without any additional steps. Thanks to the unique lateral coupling operation method, the size of the SGLC cell is comparable to SRAM of the same technology node resulting in the smallest single-poly non-volatile memory (NVM) cell size of 1.34 μm 2 at 90 nm technology node. However, because of the high aspect ratio of unit cell geometry of 3.28 : 1, it was difficult to arrange word-line and bit-line decoder efficiently. For the 4-kb SGLC cell macro, by structuring the top and bottom side located cell addressing decoders, we achieved efficient decoder architecture and further memory density extension capability. Through the 150 ºC retention bake test, we analyzed the possible retention weak cells that may be generated by the silicide-FG structure. As a result, over 10-years lifetime for code storage applications is verified without tail bit occurrences.
Worker health centers and health zones provide occupational healthcare services to workers in underserved areas of industrial health. Health zones cannot counsel workers on occupational disease ...prevention; worker health center personnel have to provide such services through in-person visits. Video teleconsultation can increase the accessibility of high-quality services to workers who visit health zones and improve professional personnel’s efficiency. This study aimed to develop a system and an infrastructure that could facilitate video teleconsultation services between worker health centers and health zones, and analyze the services’ operation status. A video teleconsultation process based on the role of local and remote institutions was designed. Based on the designed services, a web system supporting teleconsultation and an integrated gateway guaranteeing the interface with medical equipment in the web environment was developed. After analyzing the working environment, an infrastructure consisting of networks, video equipment, medical equipment, and servers necessary for teleconsultation was designed. Statistical results from the developed system and a satisfaction survey conducted by the Korea Occupational Safety and Health Agency were used to analyze the video teleconsultation services’ operation status. A system, gateway, and hardware infrastructure within 21 worker health centers and 21 health zones has been operating this service since January 1, 2018. Over two years, the total number of teleconsultation service users was 4979, and the total number of questionnaire respondents was 1270. The total number of service activities was 6388 and total satisfaction was rated as 4.58 (± 0.59) out of 5. In this study, video teleconsultation services were evaluated as being very satisfactory. In particular, service accessibility and local medical personnel service were rated highly, demonstrating that the developed system and infrastructure were well designed according to the goals of the project.
We develop a novel method to measure six geometric errors (two position-independent geometric errors and four position-dependent geometric errors) of a rotary manipulator; we use two displacement ...sensors and one measurement target to this end. To enhance measurement accuracy, the method initially removes setup errors that distort the sensor data and then models the geometric errors using an nth-order polynomial equation exhibiting C1-continuity. The data are used to define the relative positions of the reference and measurement co-ordinate systems using a homogeneous transform matrix. The geometric errors of a rotary manipulator are measured using the least squares method to exploit the linear relationship between the errors and the measurement target profile. We validate our method via simulation and assess measuring uncertainties with consideration of measurement noise.
To reveal the origins of diffuse H-alpha emissions observed around the Herbig star MWC 1080, we have performed a high-resolution near-infrared (NIR) spectroscopic observation using the Immersion ...GRating INfrared Spectrograph (IGRINS). In the NIR H and K bands, we detected various emission lines (six hydrogen Brackett lines, seven H2 lines, and an Fe II line) and compared their spatial locations with the optical (H-alpha and S II) and radio (13CO and CS) line maps. The shock-induced H2 and Fe II lines indicate the presence of multiple outflows, consisting of at least three, associated young stars in this region. The kinematics of H2 and Fe II near the northeast (NE) cavity edge supports that the NE main outflow from MWC 1080A is the blueshifted one with a low inclination angle. The H2 and Fe II lines near the southeast molecular region newly reveal that additional highly-blueshifted outflows originate from other young stars. The fluorescent H2 lines were found to trace photodissociation regions formed on the cylindrical surfaces of the main outflow cavity, which are expanding outward with a velocity of about 10-15 km/s. For the H-alpha emission, we identify its components associated with two stellar outflows and two young stars in addition to the dominant component of MWC 1080A scattered by dust. We also report a few faint H-alpha features located ~0.4 pc away in the southwest direction from MWC 1080A, which lie near the axes of the NE main outflow and one of the newly-identified outflows.
As the prevalence of hypertension in the elderly population is increasing, information regarding the characteristics, optimal blood pressure targets, and special considerations for elderly ...hypertensive patients is needed to improve clinical outcomes. Various factors should be considered when managing hypertension in elderly patients, and there are many controversial issues and conflicting results related to the optimal treatment of hypertension in the elderly. In this review, we provide an overview of the epidemiology and characteristics of elderly hypertensive patients and discuss the optimal treatment of hypertension in elderly people.
