Comprehensive phenotypic profiling of heterogeneous circulating tumor cells (CTCs) at single‐cell resolution has great importance for cancer management. Herein, a novel spectrally combined encoding ...(SCE) strategy was proposed for multiplex biomarker profiling of single CTCs using a multifunctional nanosphere‐mediated microfluidic platform. Different cellular biomarkers uniquely labeled by multifunctional nanosphere barcodes, possessing identical magnetic tags and distinct optical signatures, enabled isolation of heterogeneous CTCs with over 91.6 % efficiency and in situ SCE of phenotypes. By further trapping individual CTCs in ordered microstructures on chip, composite single‐cell spectral signatures were conveniently and efficiently obtained, allowing reliable spectral‐readout for multiplex biomarker profiling. This SCE strategy exhibited great potential in multiplex profiling of heterogeneous CTC phenotypes, offering new avenues for cancer study and precise medicine.

Compressed sensing (CS) has the potential to reduce magnetic resonance (MR) data acquisition time. In order for CS-based imaging schemes to be effective, the signal of interest should be sparse or ...compressible in a known representation, and the measurement scheme should have good mathematical properties with respect to this representation. While MR images are often compressible, the second requirement is often only weakly satisfied with respect to commonly used Fourier encoding schemes. This paper investigates the use of random encoding for CS-MRI, in an effort to emulate the "universal" encoding schemes suggested by the theoretical CS literature. This random encoding is achieved experimentally with tailored spatially-selective radio-frequency (RF) pulses. Both simulation and experimental studies were conducted to investigate the imaging properties of this new scheme with respect to Fourier schemes. Results indicate that random encoding has the potential to outperform conventional encoding in certain scenarios. However, our study also indicates that random encoding fails to satisfy theoretical sufficient conditions for stable and accurate CS reconstruction in many scenarios of interest. Therefore, there is still no general theoretical performance guarantee for CS-MRI, with or without random encoding, and CS-based methods should be developed and validated carefully in the context of specific applications.
Humidity is one of the most important indicators affecting human health. Here, a pair of covalent organic frameworks (COFs) of positional isomers (p‐COF and o‐COF) for indoor humidity regulation is ...reported. Although p‐COF and o‐COF have the same sql topology and pore size, they exhibit different water adsorption behaviors due to the subtle differences in water adsorption sites. Particularly, o‐COF exhibits a steep adsorption isotherm in the range of 45–65% RH with a hysteresis loop, which is perfectly suitable for indoor humidity regulation. In the laboratory experiment, when the humidity of the external environment is 20–75% RH, o‐COF can control the humidity of the room in the range of 45–60% RH. o‐COF has shown great potential as a dual humidification/dehumidification adsorbent for indoor humidity regulation.

Both laboratory experiment and real environment experiment confirm that o‐COF is a new generation of indoor humidity regulation material.
Emerging evidence indicates that myeloma overexpressed (MYEOV) is an oncogene and plays crucial roles in multiple human cancers. However, its roles in the development of pancreatic ductal ...adenocarcinoma (PDAC) remain elusive. Here, we provide evidence of essential roles of MYEOV in the development and progression of PDAC. In tumor specimens derived from pancreatic cancer patients, MYEOV was overexpressed and associated with poor prognosis. In addition, MYEOV expression in PDAC was upregulated through promoter hypomethylation. MYEOV depletion impaired metastatic ability and proliferation of PDAC cells both in vitro and in vivo, whereas its overexpression had the opposite effect. Mechanistic investigations revealed that MYEOV interacted with SRY-Box Transcription Factor 9 (SOX9), a well-known oncogenic transcription factor in PDAC. This interaction occurred mainly in the nuclei of PDAC cells and increased transcriptional activity of SOX9. Furthermore, MYEOV promoted the expression of Hairy and enhancer of split homolog-1 (HES1), a SOX9 target gene, by enhancing SOX9 DNA-binding ability to the HES1 enhancer without affecting the protein level and subcellular localization of SOX9. HES1 knockdown partly abrogated the oncogenic effect of MYEOV. Our findings suggest that MYEOV could be a potential prognostic biomarker and therapeutic target for PDAC.
Dyslipidemia in pregnancy are associated with gestational diabetes mellitus (GDM), preeclampsia, preterm birth and other adverse outcomes, which has been extensively studied in western countries. ...However, similar studies have rarely been conducted in Asian countries. Our study was aimed at investigating the associations between maternal dyslipidemia and adverse pregnancy outcomes among Chinese population.
Data were derived from 934 pairs of non-diabetic mothers and neonates between 2010 and 2011. Serum blood samples were assayed for fasting total cholesterol (TC), triglycerides (TG), high-density lipoprotein-cholesterol (HDL-C), and low-density lipoprotein-cholesterol (LDL-C) concentrations during the first, second and third trimesters. The present study explored the associations between maternal lipid profile and pregnancy complications and perinatal outcomes. The pregnancy complications included GDM, preeclampsia and intrahepatic cholestasis of pregnancy (ICP); the perinatal outcomes included preterm birth, small/large for gestational age (SGA/LGA) infants and macrosomia. Odds ratios (ORs) and 95% confidence intervals (95% CIs) were calculated and adjusted via stepwise logistic regression analysis. Optimal cut-off points were determined by ROC curve analysis.
After adjustments for confounders, every unit elevation in third-trimester TG concentration was associated with increased risk for GDM (OR = 1.37, 95% CI: 1.18-1.58), preeclampsia (OR = 1.50, 95% CI: 1.16-1.93), ICP (OR = 1.28, 95% CI: 1.09-1.51), LGA (OR = 1.13, 95% CI: 1.02-1.26), macrosomia (OR = 1.19, 95% CI: 1.02-1.39) and decreased risk for SGA (OR = 0.63, 95% CI: 0.40-0.99); every unit increase in HDL-C concentration was associated with decreased risk for GDM and macrosomia, especially during the second trimester (GDM: OR = 0.10, 95% CI: 0.03-0.31; macrosomia: OR = 0.25, 95% CI: 0.09-0.73). The optimal cut-off points for third-trimester TG predicting GDM, preeclampsia, ICP, LGA and SGA were separately ≥ 3.871, 3.528, 3.177, 3.534 and ≤ 2.530 mmol/L. The optimal cut-off points for third-trimester HDL-C identifying GDM, macrosomia and SGA were respectively ≤ 1.712, 1.817 and ≥ 2.238 mmol/L.
Among Chinese population, maternal high TG in late pregnancy was independently associated with increased risk of GDM, preeclampsia, ICP, LGA, macrosomia and decreased risk of SGA. Relative low maternal HDL-C during pregnancy was significantly associated with increased risk of GDM and macrosomia; whereas relative high HDL-C was a protective factor for both of them.
Impaired activation and regulation of the extinction of inflammatory cells and molecules in injured neuronal tissues are key factors in the development of epilepsy. SerpinA3N is mainly associated ...with the acute phase response and inflammatory response. In our current study, transcriptomics analysis, proteomics analysis, and Western blotting showed that the expression level of Serpin clade A member 3N (SerpinA3N) is significantly increased in the hippocampus of mice with kainic acid (KA)-induced temporal lobe epilepsy, and this molecule is mainly expressed in astrocytes. Notably, in vivo studies using gain- and loss-of-function approaches revealed that SerpinA3N in astrocytes promoted the release of proinflammatory factors and aggravated seizures. Mechanistically, RNA sequencing and Western blotting showed that SerpinA3N promoted KA-induced neuroinflammation by activating the NF-κB signaling pathway. In addition, co-immunoprecipitation revealed that SerpinA3N interacts with ryanodine receptor type 2 (RYR2) and promotes RYR2 phosphorylation. Overall, our study reveals a novel SerpinA3N-mediated mechanism in seizure-induced neuroinflammation and provides a new target for developing neuroinflammation-based strategies to reduce seizure-induced brain injury.
Ascorbic acid (AA), as one of the most important vitamins, participates in various physiological reactions in the human body and is implicated with many diseases. Therefore, the development of ...effective methods for monitoring the AA level in living systems is of great significance. Up to date, various technologies have been developed for the detection of AA. However, few methods can realize the direct detection of endogenous AA in living cells. In this work, we for the first time reported that near-infrared (NIR) graphene quantum dots (GQD) possessed good two-photon fluorescence properties with a NIR emission at 660 nm upon exciting with 810 nm femtosecond pulses and a two-photon (TP) excitation action cross-section (δΦ) of 25.12 GM. They were then employed to construct a TP nanoprobe for detection and bioimaging of endogenous AA in living cells. In this nanosystem, NIR GQDs (NGs), which exhibited lower fluorescence background in living system to afford improved fluorescence imaging resolution, were acted as fluorescence reporters. Also CoOOH nanoflakes were chosen as fluorescence quenchers by forming on the surface of NGs. Once AA was introduced, CoOOH was reduced to Co2+, which resulted in a “turn-on” fluorescence signal of NGs. The proposed nanoprobe demonstrated high sensitivity toward AA, with the observed LOD of 270 nM. It also showed high selectivity to AA with excellent photostability. Moreover, the nanoprobe was successfully used for TP imaging of endogenous AA in living cells as well as deep tissue imaging.
Spin waves can be used as information carriers with low energy dissipation. The excitation and propagation of spin waves along reconfigurable magnonic circuits is the subject of much interest in the ...field of magnonic applications. Here we experimentally demonstrate an effective excitation of spin waves in reconfigurable magnetic textures at frequencies as high as 15 GHz and wavelengths as short as 80 nm from Ni80Fe20 (Py) nanodisk–film hybrid structures. Most importantly, we demonstrate these spin wave modes, which were previously confined within a nanodisk, can now couple to and propagate along a nanochannel formed by magnetic domain walls at zero magnetic bias field. The tunable high-frequency, short-wavelength, and propagating spin waves may play a vital role in energy efficient and programmable magnonic devices at the nanoscale.
Guide Basin is a potential site for enhanced geothermal system development in China. The optimal strategy is needed to maximize heat production potential and provide support for the future ...stimulation design. In this work, sensitivity analysis of influence factors on water flow path, production water temperature, and flow impedance is performed on the basis of hydraulic–thermal model of Guide Basin. The factors include reservoir parameters (length, width, height, permeability, and temperature) and operation parameters (injection rate, injection temperature, open hole length, and height difference of open hole). Results indicate that the main affecting factors of water flow path are stimulated reservoir permeability and injection rate. The most significant influencing factor of flow impedance is stimulated reservoir permeability. Reservoir geometry has the greatest influence on production water temperature. The stable period of production water temperature is mainly affected by reservoir geometry and the height difference of open hole. In the theoretically optimal strategy, the electrical powers of 150, 400, and 450 kg/s are 12.3, 32.7–33.3, and 36.2–37.6 MW, respectively. In the practically optimal strategy, the maximum injection rate is 80 kg/s and electrical power ranges from 0 to 1.25 MW. For future applications, the shallow reservoirs with high temperature are expected.
