Aromatic ketones were synthesized from alkylbenzene derivatives through a novel method that employs cobalt(II) perchlorate as catalyst and oxone as oxidant. The reaction smoothly proceeded at room temperature with yields reaching 96%.
Genetic variants and lifestyle factors have been associated with gastric cancer risk, but the extent to which an increased genetic risk can be offset by a healthy lifestyle remains unknown. We aimed ...to establish a genetic risk model for gastric cancer and assess the benefits of adhering to a healthy lifestyle in individuals with a high genetic risk.
In this meta-analysis and prospective cohort study, we first did a fixed-effects meta-analysis of the association between genetic variants and gastric cancer in six independent genome-wide association studies (GWAS) with a case-control study design. These GWAS comprised 21 168 Han Chinese individuals, of whom 10 254 had gastric cancer and 10 914 geographically matched controls did not. Using summary statistics from the meta-analysis, we constructed five polygenic risk scores in a range of thresholds (p=5 × 10−4 p=5 × 10−5 p=5 × 10−6 p=5 × 10−7, and p=5 × 10−8) for gastric cancer. We then applied these scores to an independent, prospective, nationwide cohort of 100 220 individuals from the China Kadoorie Biobank (CKB), with more than 10 years of follow-up. The relative and absolute risk of incident gastric cancer associated with healthy lifestyle factors (defined as not smoking, never consuming alcohol, the low consumption of preserved foods, and the frequent intake of fresh fruits and vegetables), was assessed and stratified by genetic risk (low quintile 1 of the polygenic risk score, intermediate quintile 2–4 of the polygenic risk score, and high quintile 5 of the polygenic risk score). Individuals with a favourable lifestyle were considered as those who adopted all four healthy lifestyle factors, those with an intermediate lifestyle adopted two or three factors, and those with an unfavourable lifestyle adopted none or one factor.
The polygenic risk score derived from 112 single-nucleotide polymorphisms (p<5 × 10−5) showed the strongest association with gastric cancer risk (p=7·56 × 10−10). When this polygenic risk score was applied to the CKB cohort, we found that there was a significant increase in the relative risk of incident gastric cancer across the quintiles of the polygenic risk score (ptrend<0·0001). Compared with individuals who had a low genetic risk, those with an intermediate genetic risk (hazard ratio HR 1·54 95% CI 1·22–1·94, p=2·67 × 10−4) and a high genetic risk (2·08 1·61–2·69, p<0·0001) had a greater risk of gastric cancer. A similar increase in the relative risk of incident gastric cancer was observed across the lifestyle categories (ptrend<0·0001), with a higher risk of gastric cancer in those with an unfavourable lifestyle than those with a favourable lifestyle (2·03 1·46–2·83, p<0·0001). Participants with a high genetic risk and a favourable lifestyle had a lower risk of gastric cancer than those with a high genetic risk and an unfavourable lifestyle (0·53 0·29–0·99, p=0·048), with an absolute risk reduction of 1·12% (95% CI 0·62–1·56).
Chinese individuals at an increased risk of incident gastric cancer could be identified by use of our newly developed polygenic risk score. Compared with individuals at a high genetic risk who adopt an unhealthy lifestyle, those who adopt a healthy lifestyle could substantially reduce their risk of incident gastric cancer.

Bamboo is a material with excellent development prospects. It is increasingly used in furniture, decoration, building, and bridge construction. In this study, Furfurylated bamboo bundles and ...phenol-formaldehyde resin were used to make bamboo-scrimber composites (BSCs) via molding-recombination and hot-pressing processes. The effects of the impregnation mode, furfuryl-alcohol concentration, and curing temperature on the various physical-mechanical properties and durability of the composites were evaluated. Scanning-electron microscopy (SEM) was used to observe the microstructural differences. Fourier-transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) were employed to investigate changes in the chemical constituents. The heat resistance was also investigated using thermogravimetric analysis. The results showed that the density of the furfurylated BSC increased by up to 22% compared with that of the BSC-C with the same paving mode. The furfurylated BSCs had lower moisture contents: the average moisture content of the furfurylated BSCs was 25~50% lower than that of the BSC-C. In addition, the furfurylated BSCs showed better dimensional stability and durability, since the decay-resistance grade of the BSCs was raised from decay resistance (class II) to strong decay resistance (class I). In terms of the mechanical properties, the furfurylation had a slight negative effect on the mechanical strength of the BSCs, and the modulus of rupture (MOR) and horizontal shear strength (HSS) of the BSCs were increased to a certain extent under most of the treatment conditions. In particular, the highest HSS for indoor use and MOR of the furfurylated BSCs increased by 21% and 9% compared with those of the untreated BSCs, respectively. The SEM results indicated that the FA resin effectively filled in the bamboo-cell cavities and vessels, and the modified bamboo-parenchyma cells were compressed more tightly and evenly. The FTIR and XPS spectroscopy showed that the hydroxyl group of carboxylic acid of the bamboo-cell-wall component reacted with that of the furan ring, and the cellulose and hemicellulose underwent acid hydrolysis to a certain extent after the furfurylation. Overall, the present study highlights the potential of furfurylation as a modification method to enhance BSC products. The purpose of this study was to screen differentially expressed genes in PCOS using gene chip data and investigate the biological functions of these DEGs in PCOS. Additionally, the study aimed to ...analyze the potential clinical significance of these genes using clinical data. In this study, we first screened the DEGs related to PCOS by using the gene chip data (GSE5090) from GEO database. Target gene prediction software was used to predict the target genes for these DEGs, and their functional enrichment was analysed. Subsequently, the STRING online tool and Cytoscape software were utilized to identify key genes by constructing protein-protein interaction networks (PPI). In the analysis of the GSE5090 dataset, seventeen differentially expressed genes (DEGs) were identified. Functional enrichment analysis revealed that these DEGs are predominantly associated with biological functions related to polycystic ovary syndrome (PCOS). Moreover, the tissue-specific expression analysis highlighted immune system markers, with a notable difference observed in 18 of these markers, accounting for 20.5% of the total. By constructing PPI networks and key gene regulatory networks, a total of three genes (RPL13, LEP, and ANXA1) were identified as key genes. In addition, the column-line graphical model performed well in predicting the risk of PCOS. Using ROC curves, the model proved to be effective in diagnosis. This study represents the first application of a bioinformatics approach to identify and confirm high expression levels of RPL13, LEP, and ANXA1 in patients with Polycystic Ovary Syndrome (PCOS). These key genes-RPL13, LEP, and ANXA1-may present viable targets for therapeutic interventions in PCOS, underscoring their potential clinical importance.
To estimate the compressive strength of cement-based materials with mining waste, the dataset based on a series of experimental studies was constructed. The support vector machine (SVM), decision ...tree (DT), and random forest (RF) models were developed and compared. The beetle antennae search (BAS) algorithm was employed to tune the hyperparameters of the developed machine learning models. The predictive performances of the three models were compared by the evaluation of the values of correlation coefficient (R) and root mean square error (RMSE). The results showed that the BAS algorithm can effectively tune these artificial intelligence models. The SVM model can obtain the minimum RMSE, while the BAS algorithm is inefficient in DT and RF models. The SVM, DT, and RF models can be used to predict the compressive strength of cement-based materials using solid mining waste as aggregate effectively and accurately, with high R values and lower RMSE values. The RF algorithm can obtain the highest value of R and the lowest value of RMSE, demonstrating the highest accuracy. The solid mining waste to cement ratio is the most important variable to affect the compressive strength. Curing time was also an important parameter in the compressive strength of cemented materials, followed by the water-solid ratio of mining waste and fine sand ratio.

PubMed, Cochrane Library, EMBASE, and Web of Science databases were searched for studies published before 20 August 2021. Results were analyzed using Review Manager 5.2 software. We used Trial Sequential Analysis (TSA) software to avoid false-positive results caused by random errors.
We included 7 studies in this systematic review and meta-analysis. The results of the meta-analysis showed that compared with placebo, the subjective sleep quality of patients with PD significantly improved after melatonin treatment (MD = -2.19, 95% CI: -3.53 to -0.86,
= 0.001). In the systematic review, we qualitatively analyzed the efficacy of melatonin on the objective sleep quality of patients with PD, and the results showed that melatonin exerted a positive effect with good safety and tolerability. However, there was no significant improvement in excessive daytime sleepiness assessed by the Epworth Sleepiness Scale (ESS).
We found that melatonin can significantly improve the subjective and objective sleep quality of patients with PD with good safety and tolerability. Melatonin could be considered an effective treatment for insomnia in patients with PD.
Telomeres play a key role in the maintenance of chromosome integrity and stability, and telomere shortening is involved in initiation and progression of malignancies. A series of epidemiological ...studies have examined the association between shortened telomeres and risk of cancers, but the findings remain conflicting.
A dataset composed of 11,255 cases and 13,101 controls from 21 publications was included in a meta-analysis to evaluate the association between overall cancer risk or cancer-specific risk and the relative telomere length. Heterogeneity among studies and their publication bias were further assessed by the χ(2)-based Q statistic test and Egger's test, respectively.
The results showed that shorter telomeres were significantly associated with cancer risk (OR = 1.35, 95% CI = 1.14-1.60), compared with longer telomeres. In the stratified analysis by tumor type, the association remained significant in subgroups of bladder cancer (OR = 1.84, 95% CI = 1.38-2.44), lung cancer (OR = 2.39, 95% CI = 1.18-4.88), smoking-related cancers (OR = 2.25, 95% CI = 1.83-2.78), cancers in the digestive system (OR = 1.69, 95% CI = 1.53-1.87) and the urogenital system (OR = 1.73, 95% CI = 1.12-2.67). Furthermore, the results also indicated that the association between the relative telomere length and overall cancer risk was statistically significant in studies of Caucasian subjects, Asian subjects, retrospective designs, hospital-based controls and smaller sample sizes. Funnel plot and Egger's test suggested that there was no publication bias in the current meta-analysis (P = 0.532).
The results of this meta-analysis suggest that the presence of shortened telomeres may be a marker for susceptibility to human cancer, but single larger, well-design prospective studies are warranted to confirm these findings.
Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of paratuberculosis. As a potential zoonotic pathogen, MAP also seriously threatens human health and social security. At ...present, long non-coding RNA (lncRNA) has attracted wide attention as an useful biomarker in various diseases. Therefore, our study analyzed the lncRNA expression profiles and lncRNA-mRNA regulatory network of MAP infected bovine monocytes-macrophages and uninfected bovine cells by high-throughput sequencing. A total of 4641 differentially expressed lncRNAs genes were identified, including 3111 up-regulated genes and 1530 down-regulated genes. In addition, lncRNA-mRNA interaction analysis was performed to predict the target genes of lncRNA. Among them, after MAP infection, 86 lncRNAs targeted to mRNA, of which only 6 genes were significantly different. The results of Gene Ontology (GO) enrichment analysis showed that the differentially expressed genes significantly enriched in functional groups were related to immune regulation. Multiple signal pathways including NF-κB, NOD-like receptor, Cytokine-cytokine receptor, Toll-like receptor signaling pathway, Chemokine signaling pathway, and other important biochemical, metabolic and signal transduction pathways were enriched in Kyoto Encyclopedia of Genes and Genomes (KEGG). In this study, analysis of macrophage transcriptomes in response to MAP infection is expected to provide key information to deeply understand role of the pathogen in initiating an inappropriate and persistent infection in susceptible hosts and molecular mechanisms that might underlie the early phases of paratuberculosis.
Recent evidence indicates that small noncoding RNA molecules known as microRNAs (miRNAs) can function as tumor suppressors and oncogenes. Mutation, misexpression, and altered mature miRNA processing ...are implicated in carcinogenesis and tumor progression. Because SNPs in pre-miRNAs could alter miRNA processing, expression, and/or binding to target mRNA, we conducted a systematic survey of common pre-miRNA SNPs and their surrounding regions and evaluated in detail the association of 4 of these SNPs with the survival of individuals with non-small cell lung cancer (NSCLC). When we assumed that disease susceptibility was inherited as a recessive phenotype, we found that the rs11614913 SNP in hsa-mir-196a2 was associated with survival in individuals with NSCLC. Specifically, survival was significantly decreased in individuals who were homozygous CC at SNP rs11614913. In the genotype-phenotype correlation analysis of 23 human lung cancer tissue samples, rs11614913 CC was associated with a statistically significant increase in mature hsa-mir-196a expression but not with changes in levels of the precursor, suggesting enhanced processing of the pre-miRNA to its mature form. Furthermore, binding assays revealed that the rs11614913 SNP can affect binding of mature hsa-mir-196a2-3p to its target mRNA. Therefore, the rs11614913 SNP in hsa-mir-196a2 may be a prognostic biomarker for NSCLC. Further characterization of miRNA SNPs may open new avenues for the study of cancer and therapeutic interventions.
It has been demonstrated that there are abundant stable microRNAs (miRNAs) in plasma/serum, which can be detected and are potentially disease specific. However, the lack of suitable endogenous ...controls for serum miRNA detection is the restriction for the widely usage of this kind of biomarkers and for the between-laboratory comparison of the findings. We first systematically screened for endogenous control miRNAs (ECMs) by testing 10 pooling samples (using both Solexa sequencing and TaqMan low density array) and 50 individual samples (using quantitative reverse transcription-PCR) of different cancer traits and healthy controls. Then we assessed serum miRNAs used as potential biomarkers for breast cancer risk prediction based on a two-stage case-control analysis, including 48 breast cancer patients and 48 controls for the discovery stage and 76 breast cancer patients and 76 controls for validation. We identified two candidate ECMs (miRNA-191 and miRNA-484). Normalized by the two ECMs, we found four miRNAs (miR-16, miR-25, miR-222 and miR-324-3p) that were consistently differentially expressed between breast cancer cases and controls. The area under the receiver operating characteristic curve is 0.954 for the four-miRNA signature in the discovery stage (sensitivity = 0.917 and specificity = 0.896) and 0.928 in the validation stage (sensitivity = 0.921 and specificity = 0.934). In conclusion, the four-miRNA signature from serum may serve as a non-invasive prediction biomarker for breast cancer. Furthermore, we proposed the combination of miRNA-484 and miRNA-191 as endogenous control for serum miRNA detection, at least for most common cancers.
