Previous studies have suggested that solar‐induced chlorophyll fluorescence (SIF) is correlated with Gross Primary Production (GPP). However, it remains unclear to what extent this relationship is ...due to absorbed photosynthetically active radiation (APAR) and/or light use efficiency (LUE). Here we present the first time series of near‐surface measurement of canopy‐scale SIF at 760 nm in temperate deciduous forests. SIF correlated with GPP estimated with eddy covariance at diurnal and seasonal scales (r2 = 0.82 and 0.73, respectively), as well as with APAR diurnally and seasonally (r2 = 0.90 and 0.80, respectively). SIF/APAR is significantly positively correlated with LUE and is higher during cloudy days than sunny days. Weekly tower‐based SIF agreed with SIF from the Global Ozone Monitoring Experiment‐2 (r2 = 0.82). Our results provide ground‐based evidence that SIF is directly related to both APAR and LUE and thus GPP, and confirm that satellite SIF can be used as a proxy for GPP.

Dysbiosis of gut microbiota is linked to the development of colorectal cancer (CRC). However, microbiota-based stratification of CRC tissue and how this relates to clinicomolecular characteristics ...and prognosis remains to be clarified.
Tumor and normal mucosa from 423 patients with stage I to IV CRC were profiled by bacterial 16S rRNA gene sequencing. Tumors were characterized for microsatellite instability (MSI), CpG island methylator phenotype (CIMP), APC, BRAF, KRAS, PIK3CA, FBXW7, SMAD4, and TP53 mutations, subsets for chromosome instability (CIN), mutation signatures, and consensus molecular subtypes (CMS). Microbial clusters were validated in an independent cohort of 293 stage II/III tumors.
Tumors reproducibly stratified into 3 oncomicrobial community subtypes (OCSs) with distinguishing features: OCS1 (Fusobacterium/oral pathogens, proteolytic, 21%), right-sided, high-grade, MSI-high, CIMP-positive, CMS1, BRAF V600E, and FBXW7 mutated; OCS2 (Firmicutes/Bacteroidetes, saccharolytic, 44%), and OCS3 (Escherichia/Pseudescherichia/Shigella, fatty acid β-oxidation, 35%) both left-sided and exhibiting CIN. OCS1 was associated with MSI-related mutation signatures (SBS15, SBS20, ID2, and ID7) and OCS2 and OCS3 with SBS18 related to damage by reactive oxygen species. Among stage II/III patients, OCS1 and OCS3 both had poorer overall survival compared with OCS2 for microsatellite stable tumors (multivariate hazard ratio HR, 1.85; 95% confidence interval CI, 1.15–2.99; P = .012; and HR, 1.52; 95% CI 1.01–2.29; P = .044, respectively) and left-sided tumors (multivariate HR, 2.66; 95% CI, 1.45–4.86; P = .002; and HR, 1.76; 95% CI, 1.03–3.02; P = .039, respectively).
OCS classification stratified CRCs into 3 distinct subgroups with different clinicomolecular features and outcomes. Our findings provide a framework for a microbiota-based stratification of CRC to refine prognostication and to inform the development of microbiota-targeted interventions.

Purpose
To assess the changes in intrapulpal temperature between electric high‐speed handpieces of different coolant functions (‘Water Jet’ and ‘Water Spray’), coolant port designs (1‐ and 4‐port), ...suction use, and bur and tooth types using an experimental in vitro setup.
Materials and methods
Forty‐four extracted anterior and posterior teeth were collected. A total of 18 groove cuts (n = 18/coolant port spray design, bur and tooth type group) and 12 groove cuts (n = 12/tooth type and suction use) were completed, with a total of 264 groove cuts. Real‐time temperature and duration were recorded at 1‐s intervals throughout the preparation process using a thermocouple and digital data logger setup (GFX Data Logger Series and EL USB‐TC; Lascar Electronics Inc., USA), and the data retrieved using EasyLog Software (EasyLog USB; Lascar Electronics Inc., USA). Statistical analysis was performed (SPSS V.27) for the change in temperature using the analysis of variance and post hoc analysis.
Results
The majority of the specimen cuts, regardless of tooth (anterior or posterior) and bur (diamond or carbide) types, handpiece coolant port design, and suction use showed an overall decreasing trend in intrapulpal temperature. No cuts caused a mean temperature change that reached the critical temperature of 42.5°C or resulted in an overall increase in intrapulpal temperature when the 60‐s duration was completed.
Conclusions
The tested electric handpieces efficiently reduced intrapulpal temperature, with the majority displaying a decreasing trend. A greater decrease in intrapulpal temperature was observed in canines compared to premolars; carbide burs compared to diamond; and with no suction preparations compared to when suction was used.
To measure polishability of occlusal splint materials manufactured by various methods.
Seven occlusal splint materials manufactured by four different methods – Heat cured (Vertex Rapid simplified ...Clear), CAD-milled (Ceramill a-splint), Vacuum-formed (Proform splint) and 3D-printed (Freeprint Ortho, KeySplint Soft, DentaClear and FreePrint Splint 2.0) were tested for gloss, roughness, and surface hardness and elastic modulus. For all groups, the tests were repeated with the materials polished with three different polishing burs, pumice and high shine. All polishing procedures were standardised by applying the force of 1 N for 1 min at the set speed. 3D printed materials were further tested with additional specimens manufactured at different printing angles of 0°, 45° and 90°. Data was statistically analysed using ANOVA (SPSS Version 26) and MatLab (R2020a). Polished surfaces of each specimen were analysed under scanning electron microscope.
Vacuum-formed materials showed the highest polishability (80.61 ± 0.98 GU) with no statistical significance to heat-cured or CAD-milled (p = 1.00). Pumice and high shine polish significantly improved the gloss for all groups. The mean gloss and surface roughness for all 3D-printed materials ranged from 75.24 ± 25.05 GU to 0.18 ± 0.21 GU and 2.73 ± 3.18 μm to 0.06 ± 0.01 μm, which was significantly lower (p < 0.001) than heat-cured, CAD-milled and vacuum-formed materials. The highest hardness (0.40 ± 0.009 GPa), elastic modulus (6.06 ± 1.49 GPa) and gloss were found when materials were 3D-printed at 45°, with the lowest surface roughness.
Statistically significant differences in polishability were found among the different occlusal splint materials. The polishability and surface hardness of 3D-printed occlusal splint materials was influenced by the print angle. The 0° 3D-printed occlusal splint materials produced the highest gloss and the lowest surface roughness pre-polished, indicating that no polishing is required. While the 3D-printed occlusal splint materials at 45oand 90° required polishing with burs, pumice and high shine to reduce the surface roughness, there were layering structures created during printing.
Medical digital twins are computational disease models for drug discovery and treatment. Unresolved problems include how to organize and prioritize between disease-associated changes in digital ...twins, on cellulome- and genome-wide scales. We present a dynamic framework that can be used to model such changes and thereby prioritize upstream regulators (URs) for biomarker- and drug discovery.
We started with seasonal allergic rhinitis (SAR) as a disease model, by analyses of in vitro allergen-stimulated peripheral blood mononuclear cells (PBMC) from SAR patients. Time-series a single-cell RNA-sequencing (scRNA-seq) data of these cells were used to construct multicellular network models (MNMs) at each time point of molecular interactions between cell types. We hypothesized that predicted molecular interactions between cell types in the MNMs could be traced to find an UR gene, at an early time point. We performed bioinformatic and functional studies of the MNMs to develop a scalable framework to prioritize UR genes. This framework was tested on a single-cell and bulk-profiling data from SAR and other inflammatory diseases.
Our scRNA-seq-based time-series MNMs of SAR showed thousands of differentially expressed genes (DEGs) across multiple cell types, which varied between time points. Instead of a single-UR gene in each MNM, we found multiple URs dispersed across the cell types. Thus, at each time point, the MNMs formed multi-directional networks. The absence of linear hierarchies and time-dependent variations in MNMs complicated the prioritization of URs. For example, the expression and functions of Th2 cytokines, which are approved drug targets in allergies, varied across cell types, and time points. Our analyses of bulk- and single-cell data from other inflammatory diseases also revealed multi-directional networks that showed stage-dependent variations. We therefore developed a quantitative approach to prioritize URs: we ranked the URs based on their predicted effects on downstream target cells. Experimental and bioinformatic analyses supported that this kind of ranking is a tractable approach for prioritizing URs.
We present a scalable framework for modeling dynamic changes in digital twins, on cellulome- and genome-wide scales, to prioritize UR genes for biomarker and drug discovery.
Background: Experimental and observational studies have suggested that folate may play dual roles in colorectal cancer risk depending on the timing and dose.
Objective: We examined the latency ...between folate intake and the incidence of colorectal cancer.
Design: We prospectively examined associations between folate intake assessed every 2 to 4 y by using validated food-frequency questionnaires and risk of colorectal cancer and adenoma in the Nurses’ Health Study and Health Professionals Follow-Up Study, which included 2299 incident colorectal cancers and 5655 colorectal adenomas from 1980 to 2004.
Results: There was an association between total folate intake 12–16 y before diagnosis and lower risk of colorectal cancer (relative risk: 0.69; 95% CI: 0.51, 0.94; ≥800 compared with <250 μg folate/d), but there was no association between intake in the recent past and colorectal cancer risk. Long- and short-term intakes of total folate were associated with a lower risk of colorectal adenoma, with a strong association with intake 4–8 y before diagnosis (odds ratio: 0.68; 95% CI: 0.60, 0.78; ≥800 compared with <250 μg folate/d). The current use of multivitamins for >15 y, but not a shorter duration of use, was associated with lower risk of colorectal cancer; and a shorter duration of use was related to lower risk of adenoma. We did not observe an adverse effect of total folate or synthetic folic acid on risk of colorectal cancer or adenoma even during the folic acid fortification era.
Conclusion: Folate intake is inversely associated with risk of colorectal cancer only during early preadenoma stages.
Purpose
To systematically review studies on various materials and methods used for wear testing of occlusal devices and their antagonists in vitro and in vivo.
Methods
An electronic search in OVID, ...Web of Science, PubMed and Scopus was conducted using the following terms (MeSH words) with any synonyms and closed terms: “Splint*” OR “occlusal splint*” OR “night guard” OR “occlusal device” OR “occlusal devices” OR “deprogrammer” OR “bite splint” OR “bite plane” OR “orthotic appliance*” OR “orthotic devices” AND “wear” OR “two‐body wear” OR “three‐body wear” OR “tooth wear” OR “wear measurement*” OR “wear behaviour” OR “wear behavior” OR “abrasion” AND “Polymethyl Methacrylate” OR “PMMA” OR “acrylic resin*” OR “dental material*” OR “dental enamel” OR “CAD” OR “CAM” OR “PEEK” OR “material* testing”. Database search was limited to English‐language publications and published between 2001 and 1st of September 2021. A further hand search was done to ensure all materials were captured.
Results
After the removal of duplicates, 115 studies were identified, and 11 were chosen for review. Studies showed that the lowest volumetric loss was observed in PEEK occlusal device materials, whereas heat‐cure, CAD‐milled, and 3D printed occlusal device materials had no significant difference in wear. Vacuum‐formed materials showed the highest wear among all groups. Testing parameters were found to be inconsistent across all studies.
Conclusion
There is a need for standardization of in vitro and in vivo wear measurement and testing protocols as this study revealed a wide variety of testing protocols which potentially could influence the outcome. Polishing procedures are required for the material. Limited studies are available on 3D printed occlusal device materials and would therefore require further investigation, especially on printing build angles and settings. Further clinical studies would be advantageous to provide guidance on the selection of the best occlusal device material that would last the longest without remake.
Purpose
To perform a systematic review that provides an overview of the current literature on fatigue behavior of removable partial denture (RPD) clasp materials based on different retentive areas.
...Materials and methods
The review followed the Preferred Reporting Items for Systematic Review and Meta‐Analyses (PRISMA) guidelines. Electronic searches were done via PubMed, Scopus, and OVID for studies reporting on RPD clasps and the fatigue failure of clasp materials. Inclusion criteria were English language with full text and in vitro studies only. Exclusion criteria were studies that did not assess the fatigue of RPD clasp materials. A quality assessment and selection of full‐text articles were performed according to Consolidated Standards of Reporting Trials criteria.
Results
A total of 182 articles were initially identified and screened. After applying inclusion and exclusion criteria, 15 articles were selected for the final analysis. Seven of the included studies utilized a vertical insertion/removal testing approach. Ten studies used the constant deflection test. Three studies used untapered specimens, and 12 studies used tapered specimens. Ten studies performed post‐test analysis.
Conclusions
Cobalt‐chromium (CoCr) is the strongest material in terms of fatigue in relation to the undercut depth and the modern, digitally manufactured RPD clasp materials also exhibit comparable fatigue behavior. Recent RPD clasp materials such as polyetheretherketone (PEEK) or laser sintered CoCr, however, require further study in terms of their fatigue behavior. In order to improve the quality of future studies, a standardized and calibrated fatigue testing method is needed with standardized specimen size and shape, which will reduce the risk of bias and enable meta‐analysis for bulk comparison between studies.
It remains unknown whether increased risk with low levels of vitamin D is present for colon and/or rectal cancer. To investigate the association between circulating vitamin D levels and colon and ...rectal cancer, we examined the associations between plasma levels of 25-hydroxyvitamin D 25(OH)D and 1,25-dihydroxyvitamin D 1,25(OH)2D and colon and rectal cancer in the Physicians' Health Study and then conducted a meta-analysis of eight prospective studies of circulating levels of 25(OH)D and colon and rectal cancers, including the Physicians' Health Study. Study-specific ORs and 95% CIs were pooled by using a random-effects model. A total of 1,822 colon and 868 rectal cancers were included in the meta-analysis. We observed a significant inverse association for colorectal cancer (OR = 0.66; 95% CI, 0.54-0.81), comparing top versus bottom quantiles of circulating 25(OH)D levels. The inverse association was stronger for rectal cancer (OR = 0.50 for top versus bottom quantiles; 95% CI, 0.28-0.88) than colon cancer (OR = 0.77; 95% CI, 0.56-1.07; P value for difference between colon and rectal cancer = 0.20). These data suggest an inverse association between circulating 25(OH)D levels and colorectal cancer, with a stronger association for rectal cancer.
Non-coding RNAs have been drawing increasing attention in recent years as functional data suggest that they play important roles in key cellular processes. N-BLR is a primate-specific long non-coding ...RNA that modulates the epithelial-to-mesenchymal transition, facilitates cell migration, and increases colorectal cancer invasion.
We performed multivariate analyses of data from two independent cohorts of colorectal cancer patients and show that the abundance of N-BLR is associated with tumor stage, invasion potential, and overall patient survival. Through in vitro and in vivo experiments we found that N-BLR facilitates migration primarily via crosstalk with E-cadherin and ZEB1. We showed that this crosstalk is mediated by a pyknon, a short ~20 nucleotide-long DNA motif contained in the N-BLR transcript and is targeted by members of the miR-200 family. In light of these findings, we used a microarray to investigate the expression patterns of other pyknon-containing genomic loci. We found multiple such loci that are differentially transcribed between healthy and diseased tissues in colorectal cancer and chronic lymphocytic leukemia. Moreover, we identified several new loci whose expression correlates with the colorectal cancer patients' overall survival.
The primate-specific N-BLR is a novel molecular contributor to the complex mechanisms that underlie metastasis in colorectal cancer and a potential novel biomarker for this disease. The presence of a functional pyknon within N-BLR and the related finding that many more pyknon-containing genomic loci in the human genome exhibit tissue-specific and disease-specific expression suggests the possibility of an alternative class of biomarkers and therapeutic targets that are primate-specific.
