A lab-scale anammox up-flow anaerobic sludge blanket (UASB) reactor was run to investigate the influence of temperature on anammox performance and community structure. The anammox system had a higher substrate tolerance at 13 °C than at 18 °C. The adverse effects caused by the use of a lower temperature (8 °C) could be restored. The nitrogen removal rate (NRR) decreased with decreasing in situ specific anammox activity (SAA). Interestingly, the ex situ SAA acclimated at 23 °C, when exposed to ex situ temperatures of 33 and 28 °C, was higher than for those acclimated at 33 and 28 °C. No shift was observed in the optimum temperature for ex situ SAA in the whole lowering process of anammox UASB. More extracellular polymeric substances were produced in response to cooler conditions (18 °C and 13 °C). Ca. Kuenenia became much more abundant (55.18% of the microbial community) and had a competitive advantage over other anammox bacteria (AnAOB) at 13 °C.
To assess the reliability and validity of Patient Health Questionnaire-9 (PHQ-9) for patients with major depressive disorder (MDD) and to assess the feasibility of its use in psychiatric hospitals in ...China.
One hundred nine outpatients or inpatients with MDD who qualified the Diagnostic and Statistical Manual of Mental Disorders, fourth edition (DSM-IV) criteria completed PHQ-9 and Hamilton Depression Scale (HAMD-17). Two weeks after the initial evaluation, 54 randomly selected patients underwent repeat assessment using PHQ-9. For validity analysis, the construct validity and criterion validity were assessed. The internal concordance coefficient and the test-retest correlation coefficients were used for reliability analysis. The correlation between total score and scores for each item and the correlation between scores for various items were evaluated using Pearson correlation coefficient.
Principal components factor analysis showed good construct validity of the PHQ-9. PHQ-9 total score showed a positive correlation with HAMD-17 total score (r = 0.610, P < 0.001). With HAMD as the standard, PHQ-9 depression scores of 7, 15, and 21 points were used as cut-offs for mild, moderate, and severe depression, respectively. Consistency assessment was conducted between the depression severity as assessed by PHQ-9 and HAMD (Kappa = 0.229, P < 0.001). Intraclass correlation coefficient between PHQ-9 total score and HAMD total score was 0.594 (95% confidence interval, 0.456-0.704, P < 0.001). The Cronbach's α coefficient of PHQ-9 was 0.892. Correlation coefficients between each item score and the total score ranged from 0.567-0.789 (P < 0.01); the correlation coefficient between various item scores ranged from 0.233-0.747. The test-retest correlation coefficient for total score was 0.737.
PHQ-9 showed good reliability and validity, and high adaptability for patients with MDD in psychiatric hospital. It is a simple, rapid, effective, and reliable tool for screening and evaluation of the severity of depression.
The aims were to identify the possible influencing factors of health-related quality of life (HRQoL) and its domain-specific scores in patients with coronary heart disease (CHD). A total of 1247 ...patients with CHD from the Henan Rural Cohort Study (n = 39,259) were included in this study. The Chinese version of the European Quality of Life Five Dimension Five level scale (EQ-5D-5L) and Visual Analogue Scale (VAS) were used to evaluate HRQoL in patients with CHD. Tobit regression, generalized linear models and binary logistic regression were applied to determine the potential factors influencing the EQ-5D utility, as well as each domain, and the VAS. CHD patients had lower per capita monthly actual income, and higher rates of diabetes mellitus, stroke, anxiety and poor sleep quality, which significantly decreased EQ-5D index and VAS scores. In addition, sex, older age, education, not having a spouse, ever drinking alcohol, a high-fat diet, physical activity, hypertension and depression affected the various domain-specific EQ-5D scores in CHD patients. CHD patients in rural areas have a lower HRQoL. Factors associated with the EQ-5D index, including each domain, and the VAS need attention. CHD patients in rural areas need to be managed systematically.
The insights into how genome topology couples with epigenetic states to govern the function and identity of the corneal epithelium are poorly understood. Here, we generate a high-resolution Hi-C ...interaction map of human limbal stem/progenitor cells (LSCs) and show that chromatin multi-hierarchical organisation is coupled to gene expression. By integrating Hi-C, epigenome and transcriptome data, we characterize the comprehensive 3D epigenomic landscapes of LSCs. We find that super-silencers mediate gene repression associated with corneal development, differentiation and disease via chromatin looping and/or proximity. Super-enhancer (SE) interaction analysis identified a set of SE interactive hubs that contribute to LSC-specific gene activation. These active and inactive element-anchored loop networks occur within the cohesin-occupied CTCF-CTCF loops. We further reveal a coordinated regulatory network of core transcription factors based on SE-promoter interactions. Our results provide detailed insights into the genome organization principle for epigenetic regulation of gene expression in stratified epithelia.
Hematological malignancies possess a distinctive immunologic microenvironment compared with solid tumors. Here, using an established computational algorithm (CIBERSORT), we systematically analyzed ...the overall distribution of 22 tumor-infiltrating leukocyte (TIL) populations in more than 2000 bone marrow (BM) samples from 5 major hematological malignancies and healthy controls. Focusing on significantly altered TILs in acute myeloid leukemia (AML), we found that patients with AML exhibited increased frequencies of M2 macrophages, compared to either healthy controls or the other four malignancies. High infiltration of M2 macrophages was associated with poor outcome in AML. Further analysis revealed that CD206, a M2 marker gene, could faithfully reflect variation in M2 fractions and was more highly expressed in AML than normal controls. High CD206 expression predicted inferior overall survival (OS) and event-free survival (EFS) in two independent AML cohorts. Among 175 patients with intermediate-risk cytogenetics, the survival still differed greatly between low and high CD206 expressers (OS; P < .0001; 3-year rates, 56% v 32%; EFS; P < .001; 3-year rates, 47% v 25%). When analyzed in a meta-analysis, CD206 as a continuous variable showed superior predictive performance than classical prognosticators in AML (BAALC, ERG, EVI1, MN1, and WT1). In summary, M2 macrophages are preferentially enriched in AML. The M2 marker CD206 may serve as a new prognostic marker in AML.
The study of population spatialization has provided important basic data for urban planning, development, environment and other issues. With the development of urbanization, urban residential ...buildings are getting higher and higher, and the difference between urban and rural population density is getting larger and larger. At present, most population spatial studies adopt the grid scale, and the population in buildings is evenly divided into various grids, which will lead to the neglect of the population distribution in vertical space, and the authenticity is not strong. In order to improve the accuracy of the population distribution, this paper studied the spatial distribution of population at the building scale, combined the digital surface model (DSM) and the digital elevation model (DEM) to calculate the floor of buildings, and proposed a new index based on the total floor area of residential buildings, called residential population index (RPI). RPI is directly related to the number of people a building can accommodate, so it can effectively estimate the population of both urban and rural areas even if the structure of urban and rural buildings is very different. In addition, this paper combined remote sensing monitoring data with geographic big data and adopted principal component regression (PCR) method to construct RPI prediction model to obtain building-scale population distribution data of Qingdao in 2018, providing ideas for population spatialization research. Through field sampling survey and overall assessment, the results were basically consistent with the actual residential situation. The average error with field survey samples is 14.5%. The R2 is 0.643 and the urbanization rate is 69.7%, which are all higher than WorldPop data set. Therefore, this method can reflect the specific distribution of urban resident population, enhance the heterogeneity and complexity of population distribution, and the estimated results have important reference significance for urban management, urban resource allocation, environmental protection and other fields.
The air pollutant diffusion trend prediction plays an important role in the environment protection. The approaches in the existing studies rarely consider spatial and temporal information ...sequentially or cannot use spatiotemporal structure information effectively. In this paper, we propose a new deep learning-based air pollutant diffusion trend prediction model, called GATBL-Learning. It is an end-to-end neural network model consisting of Graph Attention Network (GAT) and Encoder-Decoder model. Since each city in the region will set as a node, GAT can extract spatial information from them by aggregation operation. Then Encoder-Decoder model, consisting of Bi-LSTM as Encoder and LSTM as Decoder, mines time series features and outputs the data containing spatiotemporal features. In addition, we also observe that the changes of air pollutants show seasonality discipline and training the seasonal model can improve the model adaptability. The results demonstrate the proposed prediction model is effective and superior.
In the past few years, with the rapid development of computer execution capabilities, target recognition strategies based on convolutional neural networks have become mainstream algorithms in the ...field of object detection. However, due to the blurred background and dim light, the object detection task in the night environment still faces greater visual challenges. This article is strongly inspired by DCGAN (Deep Convolution Generative Adversarial Networks). We use night images as input, generate virtual target scenes similar to the daytime environment through game training of generators and discriminators; and to obtain high-precision detection results, we combine the currently very advanced Faster R-CNN (Region-based Convolution Neural Networks) target detection system, through deep convolution feature fusion and multi-scale ROI (Region Of Interest) pooling. A series of experimental results show that our method achieves an mAP of 82.6% in the detection of its own night scene dataset, which is significantly higher than the original Faster R-CNN alone of 80.4%. Therefore, our method can meet the actual needs of target detection tasks in night scene. We sincerely hope that our approach will contribute to future research.
Adult stem cell identity, plasticity, and homeostasis are precisely orchestrated by lineage-restricted epigenetic and transcriptional regulatory networks. Here, by integrating super-enhancer and ...chromatin accessibility landscapes, we delineate core transcription regulatory circuitries (CRCs) of limbal stem/progenitor cells (LSCs) and find that RUNX1 and SMAD3 are required for maintenance of corneal epithelial identity and homeostasis. RUNX1 or SMAD3 depletion inhibits PAX6 and induces LSCs to differentiate into epidermal-like epithelial cells. RUNX1, PAX6, and SMAD3 (RPS) interact with each other and synergistically establish a CRC to govern the lineage-specific cis-regulatory atlas. Moreover, RUNX1 shapes LSC chromatin architecture via modulating H3K27ac deposition. Disturbance of RPS cooperation results in cell identity switching and dysfunction of the corneal epithelium, which is strongly linked to various human corneal diseases. Our work highlights CRC TF cooperativity for establishment of stem cell identity and lineage commitment, and provides comprehensive regulatory principles for human stratified epithelial homeostasis and pathogenesis.
As one of the potential energy crops, Arundo donax Linn. is a renewable source for the production of biofuels and bioproducts. In the present study, milled wood lignin (MWL) and alkaline lignin (AL) ...from stems and foliage of A. donax were isolated and characterized by FT-IR spectroscopy, UV spectroscopy, GPC, 31P NMR, 2D HSQC NMR, and DFRC. The results indicated that both stem and foliage lignins were HGS type lignins. The semiquantitative HSQC spectra analysis demonstrated a predominance of β-O-4′ aryl ether linkages (71–82%), followed by β-β′, β-5′, β-1′, and α,β-diaryl ethers linkages in the lignins. Compared to stem lignins, foliage lignins had less β-O-4′ alkyl-aryl ethers, lower weight-average molecular weight, less phenolic OH, more H units, and lower S/G ratio. Moreover, tricin was found to incorporate into the foliage lignins (higher content of condensed G units) in significant amounts and might be alkaline-stable.
