Friend leukemia integration 1 (FLI1) is an ETS transcription factor family member. Here, we identified cg11017065 as the most hyper‐methylated cytosine and guanine (CpG) in colorectal cancer (CRC), ...which belongs to the FLI1 gene. Moreover, integrated bioinformatics prediction and analysis of our cohort showed that FLI1 expression was downregulated and DNA methylation was elevated in CRC. Bioinformatics prediction also indicated that patients overexpressing FLI1 had higher survival rates than those with low FLI1 expression. CRC cells with ectopic expression of FLI1 had reduced invasion, migration, cloning ability and increased apoptosis. Furthermore, DNA‐methyltransferase 3b (DNMT3b) was found to be significantly overexpressed in CRC, and low DNMT3b expression predicted a prolonged survival. DNMT3b bound to the FLI1 promoter. Inhibition of DNMT3b increased FLI1 expression and inhibited the malignant phenotype of CRC cells. Inhibition of FLI1 reversed the phenotypic modulation by DNMT3b depletion in vitro and in vivo. In conclusion, our data indicate that DNMT3b potentiates CRC cell proliferation, migration, and invasion through downregulating FLI1.
This study aimed to investigate the effects of psychological nursing care (PNC) on anxiety relief in perioperative lung cancer (LC) patients.
We searched the Cochrane Library, PubMed, Embase, CNKI, ...CBM, and Wangfang electronic databases from inception to May 1, 2022. Eligible randomized controlled trials (RCTs) investigating the effects and safety of PNC on anxiety relief in perioperative LC patients. Anxiety was the primary outcome measure. The secondary outcomes were depression, length of hospital stay, and the occurrence of adverse events.
Six eligible RCTs with 494 patients were included in this study. Compared with routine nursing care, PNC showed better outcomes in terms of anxiety relief (mean difference MD = -13.24; random 95% confidence interval (CI), -18.28 to -8.20; P<.001), depression decrease (MD = -11.84; random 95% CI, -18.67 to -5.01; P < .001), and length of hospital stay (MD = -2.6; fixed 95% CI, -3.13 to -2.07; P < .001). No data on adverse events were pooled because only 1 trial reported this outcome.
This study showed that PNC may benefit more than routine nursing care for patients with LC in anxiety, depression, and length of hospital stay. High-quality RCTs are needed to validate the current findings in the future.
Abstract
With the study of human diseases and biological processes increasing, a large number of non-coding variants have been identified and facilitated. The rapid accumulation of genetic and ...epigenomic information has resulted in an urgent need to collect and process data to explore the regulation of non-coding variants. Here, we developed a comprehensive variation annotation database for human (VARAdb, http://www.licpathway.net/VARAdb/), which specifically considers non-coding variants. VARAdb provides annotation information for 577,283,813 variations and novel variants, prioritizes variations based on scores using nine annotation categories, and supports pathway downstream analysis. Importantly, VARAdb integrates a large amount of genetic and epigenomic data into five annotation sections, which include ‘Variation information’, ‘Regulatory information’, ‘Related genes’, ‘Chromatin accessibility’ and ‘Chromatin interaction’. The detailed annotation information consists of motif changes, risk SNPs, LD SNPs, eQTLs, clinical variant-drug-gene pairs, sequence conservation, somatic mutations, enhancers, super enhancers, promoters, transcription factors, chromatin states, histone modifications, chromatin accessibility regions and chromatin interactions. This database is a user-friendly interface to query, browse and visualize variations and related annotation information. VARAdb is a useful resource for selecting potential functional variations and interpreting their effects on human diseases and biological processes.
Research Findings: The present study analyzes whether child factors are linked to variations in teacher cognitive activation (CA) among individual children in math-related activities within Chinese ...kindergarten classrooms. The findings show that teachers tend to provide direct CA with high concreteness and that child engagement with tasks is significantly positively associated with the ratio of teacher CA to individual children. Moreover, a child's math ability is significantly negatively associated with both the ratio and overall contingency degree of CA that a child experiences. Practice or Policy: These results suggest that teachers should improve their knowledge and competence to adapt their teaching behaviors to child characteristics to provide children with high-quality and equitable math learning opportunities and that children should be taught to highly engage to obtain more proactive reactions from their learning environments.
We aimed to demonstrate the tolerability and feasibility and the effect of remote ischemic post-conditioning on cognitive functioning in patients with post-stroke cognitive impairment.
This was a ...single-center, randomized, outcome-blinded, placebo-controlled trial, randomized 1:1 to receive 4 cycles of remote ischemic post-conditioning or a sham procedure for 7 days. The primary outcome measure was tolerability and feasibility of remote ischemic post-conditioning. Secondary outcomes to measure the neurological function with national institute of health stroke scale and the cognitive impairment with Montreal Cognitive Assessment scale and Alzheimer's disease assessment scale-cognitive (at baseline, 90 days, 180 days).
48 patients (24 RIPC and 24 Control) were recruited. remote ischemic post-conditioning was well tolerated with 90 out of 96 cycles completed in full. 4 patients experienced vascular events in the control group: 3 cerebrovascular and 1 cardiovascular event versus only 2 cerebrovascular events in the RIPC group. We showed the similar result in the neurological function with national institute of health stroke scale score with no statistically significant differences between RIPC and control group at baseline (P = 0.796) and 90 days (P = 0.401) and 180 days (P = 0.695). But compare with baseline, it was significantly difference in the control and RIPC group at 90 days (P < 0.05) and 180 days (P < 0.05). The comparison of Montreal Cognitive Assessment scale between two groups both showed that P > 0.05 at baseline which was no statistical difference, but P < 0.05 at 90 days and 180 days which were significant statistical difference. The comparison of Alzheimer's disease assessment scale-cognitive between two groups showed that P > 0.05 at baseline (P = 0.955) and 90 days (P = 0.138) was no statistical difference, but P = 0.005<0.05 at 180 days was significant statistical difference.
The remote ischemic post-conditioning for post-stroke cognitive impairment was well tolerated, safe and feasible. The remote ischemic post-conditioning may improve neurological and cognitive outcomes in patients with post-stroke cognitive impairment. A larger trial is warranted. The present study examined the interactive effects of organizational resources on classroom quality of 90 Chinese kindergartens using principal and teacher questionnaires and observational ratings of classroom practices. The results revealed that the average monthly teacher salary, child-teacher ratio and instructional management had significant direct associations with classroom quality, and the three-year average fiscal funding had a significantly positive association with instructional management and significant indirect association with classroom quality via the mediation of instructional management. The findings suggested that the allocation and organization of resources for teachers and instructional management in support of teachers and teaching are important for improving classroom quality, and that kindergarten leaders play a critical role in effectively transforming available resources into quality classroom practices.
In China, preschool curriculum has undergone reform and profound changes. Much remains unknown, however, regarding preschool teachers' teaching beliefs after 30 years of curriculum reform and ...adaptation. This study aimed to address the issue and investigate teachers' beliefs concerning teaching mathematics. Twelve preschool teachers in China participated in the study: they watched videos of mathematics lessons conducted in the US and were interviewed with questions regarding the lessons. Qualitative analysis was then applied; themes were searched from the participants' responses. The study revealed that Chinese preschool teachers acknowledged the features of (a) active learning, (b) involving every child, and (c) connecting lessons with everyday activities. However, they believed that their own teaching would be more purposeful: they would choose a specific content focus, use a series of steps, and make explicit summary at the end. The study also revealed Chinese preschool teachers' solid specialized content knowledge in mathematics.
Compared with the former policies on early childhood education, the policies recently issued in mainland China clearly defined early childhood education as an integral part of education and social ...public welfare and stipulated the responsibilities of the government in its development, shifting the developmental orientation to promoting social equity. In implementing the new policies, the developmental goals that the local governments set appear divorced from reality, and the developmental paths collide with the current macro social and economic system. This means that the development mode that many local governments exploited is not sustainable. It is suggested that the government should design developmental goals according to actual conditions, reform the public revenue and tax system, and the ECE funding system and introduce a new way to provide public ECE service.
Research Findings: An effective quality rating system will lead to quality assurance and improvement of early childhood education programs. Many studies have been conducted to examine the validity of ...quality rating instruments in Western countries, but few have been done in China. The present study investigated the effectiveness of the Beijing Kindergarten Quality Rating System (BKQRS) by conducting onsite observations in 5 Demonstration, 5 R1C1, and 4 R2C2 kindergartens using the Chinese Kindergarten Environment Rating Scale (CKERS) as the criterion. Results indicated that none of the 3 types of kindergartens performed well on the CKERS. Although Demonstration and R1C1 kindergartens performed better than R2C2 kindergartens, there was no significant difference between Demonstration and R1C1 kindergartens. Results indicated that the BKQRS failed to gauge the quality levels of various kindergartens as accurately as was expected. Practice or Policy: Suggestions for improvement are discussed, and implications for other cultural contexts-such as insights for developing culturally appropriate quality assurance practices-are addressed.
