Unsupervised machine learning (ML) techniques are applied to the characterization of the adsorption of rare earth elements (REEs) by zeolites in continuous flow. The successful application of ...principal component analysis (PCA) and K-Means algorithms from ML allowed for a wide range assessment of the adsorption results. This global approach permits the evaluation of the different stages of the sorption cycles and their optimization and improvement. The results from ML are also used for the definition of a regression model to estimate other REEs' recoveries based on the known values of the tested REEs. Overall, it was possible to remove more than 70% of all REEs from aqueous solutions during the adsorption assays and to recover over 80% of the REEs entrapped on the zeolites using an optimized desorption cycle.
Nanocomplexes systems made up natural poylymers have pharmacotechnical advantages such as increase of water solubility and a decrease of drugs toxicity. Amphotericin B (AmB) is a drug apply as ...anti-leishmanial and anti-fungal, however it has low water solubility and high toxicity, limiting its therapeutic application. With this in mind, the present study aimed to produce nanocomplexes composed by alginate (Alg), a natural polymer, with AmB covered by nanocrystals from bacterial cellulose (CNC). For this reason, the nanocomplexes were produced utilizing sodium alginate, amphotericin B in a borate buffer (pH 11.0). The CNC was obtained by enzymatic hydrolysis of the bacterial cellulose. To CNC cover the nanocomplexes 1 ml of the nanocomplexes was added into 1 ml of 0.01% CNC suspension. The results showed an ionic adsorption of the CNC into the Alg-AmB nanocomplexes surface. This phenomena was confirmed by an increase in the particle size and PDI decrease. Besides, nanocomplexes samples covered by CNC showed uniformity. The amorphous inclusion of AmB complex into the polysaccharide chain network in both formulations. AmB in the nanocomplexes was in supper-aggregated form and showed good biocompatibility, being significantly less cytotoxic in vitro against kidney cells and significantly less hemolytic compared to the free-drug. The in vitro toxicity results indicated the Alg-AmB nanocomplexes can be considered a non-toxic alternative to improve the AmB therapeutic effect. All process to obtain nanocomplexes and it coat was conduce without organic solvents, can be considered a green process, and allowed to obtain water soluble particles. Furthermore, CNC covering the nanocomplexes brought additional protection to the system can contribut advancement in the pharmaceutical.
Propolis, a natural product made by bees with resins and balsams, is known for its complex chemical composition and remarkable bioactivities. In this study, propolis extraction was studied seeking ...extracts with strong bioactivities using less orthodox solvents, with some derived from apiary products. For that, a propolis sample collected from Gerês apiary in 2018 (G18) was extracted by maceration with six different solvents: absolute ethanol, ethanol/water (7:3), honey brandy, mead, propylene glycol and water. The solvent influence on the chemical composition and antioxidant and antimicrobial activities of the extracts was investigated. Antioxidant potential was assessed by the DPPH free-radical-scavenging assay and the antimicrobial activity by the agar dilution method. Chemical composition of the extracts was determined in vitro by three colorimetric assays: total ortho-diphenols, phenolics and flavonoids contents and the LC-MS technique. To our knowledge, this is the first time that solvents such as honey brandy and mead have been studied. Honey brandy showed considerable potential to extract propolis active compounds able to inhibit the growth of bacteria such as the methicillin-sensitive Staphylococcus aureus and Propionibacterium acnes (MIC values of 100 and 200 µg/mL, respectively) and the fungi Candida albicans and Saccharomyces cerevisiae (MIC = 500 µg/mL, for both). Mead extracts displayed high antioxidant capacity (EC50 = 1.63 ± 0.27 µg/mL) and great activity against resistant bacteria such as the methicillin-resistant Staphylococcus aureus and Escherichia coli (MIC = 750 µg/mL, for both). The production of such solvents made from beehive products further promotes a diversification of apiary products and the exploration of new applications using eco-friendly solutions.
The decentralized production of bioelectricity as well as the bioremediation of contaminated sediments might be achieved by the incorporation of an anode into anaerobic sediments and a cathode ...suspended in the water column. In this context, a sediment microbial fuel cell microcosm was carried out using different configurations of electrodes and types of materials (carbon and stainless steel). The results showed a long-term continuous production of electricity (>300 days), with a maximum voltage of approximately 100 mV reached after ~30 days of operation. A twofold increase of voltage was noticed with a twofold increase of surface area (~30 mV to ~60 mV vs. 40 cm2 to 80 cm2), while a threefold increase was obtained after the substitution of a carbon anode by one of stainless steel (~20 mV to ~65 mV vs. 40 cm2 to 812 cm2). Cyclic voltammetry was used to evaluate sediment bacteria electroactivity and to determine the kinetic parameters of redox reactions. The voltammetric results showed that redox processes were limited by the diffusion step and corresponded to a quasi-reversible electron charge transfer. These results are encouraging and give important information for the further optimization of sediment microbial fuel cell performance towards the long-term operation of sediment microbial fuel cell devices.

Physicochemical conditions of human digestion were simulated in vitro, and its impact on the chemical composition of GBE was investigated by electrospray ionization-mass spectrometry. The persistence of bioactivity was investigated by subjecting GBE and the in vitro digested extract (DGBE) to the same methodology. Results

Conclusion
The chemical composition of GBE was affected by in vitro digestion, but the antioxidant and antigenotoxic activities persisted. Therefore,
G. biloba
extract may be suitable for use as food additive and contribute to a healthy colon.

Peroxidases can be used in decolorization processes and the treatment of textiles effluents. This study evaluates the potential of the turnip peroxidase enzyme in the decolorization of the phthalocyanine textile dye Reactive Blue 21 (RB21). Some factors such as pH, the amount of H2O2 and the enzyme were evaluated in order to determine the optimum conditions for the enzyme performance. The reaction products formed during the decolorization of the RB21 dye were analyzed by high-performance liquid chromatography–mass spectrometry coupling (LC–ESI/MS). LC–ESI/MS analysis showed that the decolorization of the dye RB21 by turnip peroxidase is due to the breaking up of the chromatogenous system. The tests for toxicity towards lettuce seeds showed an increase of the toxicity after enzymatic treatment of the dye. This study verifies the viability of the use of the turnip peroxidase enzyme in the biodegradation of textile dyes.
Advanced urothelial bladder cancer (UBC) patients are tagged by a dismal prognosis and high mortality rates, mostly due to their poor response to standard-of-care platinum-based therapy. Mediators of ...chemoresistance are not fully elucidated. This work aimed to study the metabolic profile of advanced UBC, in the context of cisplatin resistance. Three isogenic pairs of parental cell lines (T24, HT1376 and KU1919) and the matching cisplatin-resistant (R) sublines were used. A set of functional assays was used to perform a metabolic screening on the cells. In comparison to the parental sublines, a tendency was observed towards an exacerbated glycolytic metabolism in the cisplatin-resistant T24 and HT1376 cells; this glycolytic phenotype was particularly evident for the HT1376/HT1376R pair, for which the cisplatin resistance ratio was higher. HT1376R cells showed decreased basal respiration and oxygen consumption associated with ATP production; in accordance, the extracellular acidification rate was also higher in the resistant subline. Glycolytic rate assay confirmed that these cells presented higher basal glycolysis, with an increase in proton efflux. While the results of real-time metabolomics seem to substantiate the manifestation of the Warburg phenotype in HT1376R cells, a shift towards distinct metabolic pathways involving lactate uptake, lipid biosynthesis and glutamate metabolism occurred with time. On the other hand, KU1919R cells seem to engage in a metabolic rewiring, recovering their preference for oxidative phosphorylation. In conclusion, cisplatin-resistant UBC cells seem to display deep metabolic alterations surpassing the Warburg effect, which likely depend on the molecular signature of each cell line.
This work reports the synthesis of nanocomposite photocatalytic materials based on nitrogen-doped TiO2nano, SiO2 and different percentages of HY zeolite (0, 12, 25 and 50%). These materials were ...characterized by using Fourier transformed infrared spectroscopy, X-ray diffraction, N2 adsorption–desorption, UV-vis diffuse reflectance spectroscopy and scanning electron microscopy. The nanocomposites, which presented an energy band gap of about 3.03eV, were immobilized on cotton fabric and their self-cleaning properties were investigated by decolourization of rhodamine B (RhB) dye in aqueous solution under simulated solar irradiation. The fabrics coated with the photocatalysts, containing and not containing zeolites, showed the same RhB decolourization (about 95%) after 5h, excluding the situation where a large amount of HY (50%) was used in the nanocomposites. However, results obtained from high performance liquid chromatography analysis depicted that in the presence of the HY zeolite a more effective RhB degradation was achieved. In fact, even after the use of five consecutive cycles, the RhB decolourization remained high (about 85%). Generally, the photodegradation of RhB solution in the presence of cotton fabrics functionalized with TiO2nano, TiO2nano-SiO2 and TiO2nano-SiO2-0.25 HY resulted in the formation of products that exhibited a similar cytotoxic effect when compared to the untreated RhB solution and subjected to the same tested concentrations and incubation times.

Borkh.) are two of the most popular fruits worldwide. The phenolic compounds they offer are associated with human health benefits due to their antioxidant properties. Since ...these fruits' by-products are not yet fully exploited, it is important to characterize them, especially in terms of their antioxidant properties. The aim of this study was to determine the antioxidant properties of old traditional cultivars, six regional pear cultivars and five regional apple cultivars grown in the Alcobaça region (Portugal). Antioxidant capacity assays were used to evaluate the antioxidant properties. Generally, the antioxidant capacity, total phenolics content (TPC), and total flavonoids content (TFC) of fruit byproducts (both seeds and peels) were higher than the corresponding mesocarp, indicating their potential as sources of beneficial antioxidant compounds. Moreover, a UHPLC-ToF-MS method was optimized and validated in order to quantify 21 distinct phenolics in these fruit samples. The analytical method's suitability for quantifying phenolic compounds was demonstrated by an evaluation of linearity, limit of detection, limit of quantification, precision and accuracy. This method was used to determine the phenolic composition of samples of regional (local) cultivars. The phenolics in the fruit samples with the highest concentrations were phlorizin and chlorogenic acid. Principal component analysis (PCA) was used to separate distinct fruit species while emphasizing their similarities and differences.
Fatty acids are produced by eukaryotes like baker's yeast Saccharomyces cerevisiae mainly using a large multifunctional type I fatty acid synthase (FASI) where seven catalytic steps and a carrier ...domain are shared between one or two protein subunits. While this system may offer efficiency in catalysis, only a narrow range of fatty acids are produced. Prokaryotes, chloroplasts and mitochondria rely instead on a FAS type II (FASII) where each catalytic step is carried out by a monofunctional enzyme encoded by a separate gene. FASII is more flexible and capable of producing a wider range of fatty acid structures, such as the direct production of unsaturated fatty acids. An efficient FASII in the preferred industrial organism S. cerevisiae could provide a platform for developing sustainable production of specialized fatty acids. We functionally replaced either yeast FASI genes (FAS1 or FAS2) with a FASII consisting of nine genes from Escherichia coli (acpP, acpS and fab -A, -B, -D, -F, -G, -H, -Z) as well as three from Arabidopsis thaliana (MOD1, FATA1 and FATB). The genes were expressed from an autonomously replicating multicopy vector assembled using the Yeast Pathway Kit for in-vivo assembly in yeast. Two rounds of adaptation led to a strain with a maximum growth rate (μmax) of 0.19 h−1 without exogenous fatty acids, twice the growth rate previously reported for a comparable strain. Additional copies of the MOD1 or fabH genes resulted in cultures with higher final cell densities and three times higher lipid content compared to the control.
