Numerous studies have evaluated laboratory methods to quantify prolamin proteins in dry corn; however, the utility of methods to quantify functional prolamins, which impede starch digestibility, in ...high-moisture corn (HMC) is less defined. As a result, a common rapid turbidimetric (rTM) laboratory procedure was modified (rapid Bradford method, rBM), extracting buffer-soluble proteins before prolamin solubilization in an effort to better quantify functional prolamins in HMC. Twenty samples of dry and HMC were evaluated by rTM and rBM procedures. Prolamin concentration in dry corn, as estimated by rTM or rBM methods, ranged from 6.12 to 2.20g/100 g of DM or 5.46 to 2.41g/100 g of DM, respectively. Dry corn mean prolamin concentrations, as estimated by rTM or rBM methods, were similar at 3.65 or 3.66g/100 g of DM. Prolamin concentration in HMC, as estimated by rTM, ranged from 4.99 to 3.24g/100 g of DM, with a mean prolamin concentration of 4.19g/100 g of DM, but estimation of prolamins in HMC by the rBM method resulted in lower mean (4.19 vs. 3.24g/100 g of DM) prolamin concentration. Prolamin concentration in dry and HMC measured by rTM was negatively related to peak absolute rates (PAR; mL/0.1 g of DM) of in vitro gas production. However, relationships between rTM prolamin concentration and PAR were not homogeneous and were different between dry and HMC. Prolamin proteins as determined by rBM were likewise negatively related to PAR, but corn type did not influence rBM prolamin concentration by PAR relationships. Data suggest that the rBM method defined more similar functional prolamin proteins, which impede starch degradability, in dry and HMC.
Kernel processing increases starch digestibility in whole-plant corn silage (WPCS). Corn silage processing score (CSPS), the percentage of starch passing through a 4.75-mm sieve, is widely used to ...assess degree of kernel breakage in WPCS. However, the geometric mean particle size (GMPS) of the kernel-fraction that passes through the 4.75-mm sieve has not been well described. Therefore, the objectives of this study were (1) to evaluate particle size distribution and digestibility of kernels cut in varied particle sizes; (2) to propose a method to measure GMPS in WPCS kernels; and (3) to evaluate the relationship between CSPS and GMPS of the kernel fraction in WPCS. Composite samples of unfermented, dried kernels from 110 corn hybrids commonly used for silage production were kept whole (WH) or manually cut in 2, 4, 8, 16, 32 or 64 pieces (2P, 4P, 8P, 16P, 32P, and 64P, respectively). Dry sieving to determine GMPS, surface area, and particle size distribution using 9 sieves with nominal square apertures of 9.50, 6.70, 4.75, 3.35, 2.36, 1.70, 1.18, and 0.59mm and pan, as well as ruminal in situ dry matter (DM) digestibilities were performed for each kernel particle number treatment. Incubation times were 0, 3, 6, 12, and 24 h. The ruminal in situ DM disappearance of unfermented kernels increased with the reduction in particle size of corn kernels. Kernels kept whole had the lowest ruminal DM disappearance for all time points with maximum DM disappearance of 6.9% at 24 h and the greatest disappearance was observed for 64P, followed by 32P and 16P. Samples of WPCS (n=80) from 3 studies representing varied theoretical length of cut settings and processor types and settings were also evaluated. Each WPCS sample was divided in 2 and then dried at 60°C for 48 h. The CSPS was determined in duplicate on 1 of the split samples, whereas on the other split sample the kernel and stover fractions were separated using a hydrodynamic separation procedure. After separation, the kernel fraction was redried at 60°C for 48 h in a forced-air oven and dry sieved to determine GMPS and surface area. Linear relationships between CSPS from WPCS (n=80) and kernel fraction GMPS, surface area, and proportion passing through the 4.75-mm screen were poor. Strong quadratic relationships between proportion of kernel fraction passing through the 4.75-mm screen and kernel fraction GMPS and surface area were observed. These findings suggest that hydrodynamic separation and dry sieving of the kernel fraction may provide a better assessment of kernel breakage in WPCS than CSPS.

A spectral cascade-transport model has been developed and applied to turbulent channel flows (Reτ=550, 950, and 2000 based on friction velocity, uτ; or Reδ=8500; 14,800 and 31,000, based on the mean velocity and channel half-width). This model is an extension of a spectral model previously developed for homogeneous single and two-phase decay of isotropic turbulence and uniform shear flows; and a spectral turbulence model for wall-bounded flows without resolving the boundary layer. Data from direct numerical simulation (DNS) of turbulent channel flow was used to help develop this model and to assess its performance in the 1D direction across the channel width. The resultant spectral model is capable of predicting the mean velocity, turbulent kinetic energy and energy spectrum distributions for single-phase wall-bounded flows all the way to the wall, where the model source terms have been developed to account for the wall influence. The model has been implemented into the 3D multiphase CFD code NPHASE-CMFD and the latest results are within reasonable error of the 1D predictions.
The primary objective of this study was to determine the effect of site of semen deposition on fertilization rate and embryo quality in superovulated cows. The hypothesis was that deposition of semen ...into the uterine horns would increase the fertilization rate compared with deposition of semen into the uterine body. The secondary objective was to evaluate the effect of uterine environment on fertilization rate and embryo quality. It was hypothesized that subclinical endometritis at the onset of superstimulation would decrease the fertilization rates and embryo quality. In experiment 1, 17 superovulated heifers were randomly assigned to receive artificial insemination (AI) into the uterine body or uterine horns. The total number of fertilized structures and fertilization rate from superovulated heifers was increased (P = 0.04 and P = 0.02, respectively) when semen was deposited into the uterine horns compared with the uterine body. Other embryo characteristics did not differ based on the site of semen deposition. In experiment 2, 14 lactating dairy cows were superovulated twice and were randomly assigned to receive AI into the uterine body or deep into the uterine horns using a crossover design. Neither fertilization rate nor any other embryo characteristics were improved when semen was placed deep into the uterine horns compared with the uterine body. In experiment 3, 72 superovulated lactating dairy cows were randomly assigned to receive AI into the uterine body or uterine horns. Before initiation of superstimulatory treatments, an endometrial cytology sample was collected from each cow. Ova/embryos were collected by a nonsurgical technique at 70 ± 3 days in milk. Similar to experiment 2, neither fertilization rate nor any other embryo characteristics differed based on the site of semen deposition in experiment 3. The percentage of cows with subclinical endometritis did not differ between treatments. Interestingly, there was a tendency (P = 0.09) for a reduction in embryo recovery rate and a reduction (P = 0.01) in the fertilization rate for cows with subclinical endometritis. In conclusion, deposition of semen into the uterine horns rather than into the uterine body did not improve the fertilization rate or embryo quality in superovulated cows. Subclinical endometritis decreased the fertilization rate in superovulated cows.
Vitreousness and Stenvert kernel hardness characteristics in corn germplasm have been related to in situ and in vivo starch digestibility in lactating dairy cows. Because of the tedious nature of ...determining vitreousness by manual dissection or conducting Stenvert hardness measurements, it is challenging to screen corn germplasm for animal performance potential. The resistance of corn to grinding is typically measured in a Stenvert mill and quantified by grinding time and column height. The baseline shift (BLS) of near-infrared reflectance spectroscopy, which is a normal artifact of grinding-induced particle size differentiation, was calculated and evaluated as an alternative measure of grinding resistance. Stenvert grind time, column height, BLS, and ruminal DM degradability (RDMD) of 31 diverse corn germplasm harvested in triplicate at 2 maturities were made. After grinding, total and 6 partial (100nm) BLS were calculated Σ xi+xj…, where xi=log10(1/R) at the ith nanometer and R=reflectance and compared with vitreousness, Stenvert grinding time, column height, or RDMD of corns using correlation and regression procedures. Total and all partial BLS were correlated with vitreousness, Stenvert measures, and RDMD. Relationships between BLS and vitreousness, Stenvert measures, or RDMD were stronger for partial BLS at shorter near-infrared wavelengths (1,080–1,180nm) than at longer wavelengths (i.e., 2,280–2,380nm) or total near-infrared BLS (1,000–2,498nm). The partial BLS from 1,080 to 1,180nm and vitreousness were better related to RDMD of corn germplasm than Stenvert grind time or column height.
Whole-plant corn was harvested at early dent, quarter milkline, two-thirds milkline, and black layer stages to evaluate the effects of maturity on intake, digestion, and milk production when corn was ...fed as silage in the diet. Twenty multiparous Holstein cows were used in a replicated experiment with a 4×4 Latin square design with 28-d periods. Diets containing 50% forage (67% corn silage and 33% alfalfa silage) and 50% concentrate (dry matter basis) were fed as total mixed rations. Moisture contents were 69.9, 67.6, 64.9, and 58.0% for silages from corn harvested at early dent, quarter milkline, two-thirds milkline, and black layer stages, respectively. Intakes of dry matter were similar across the four treatments and ranged from 3.73 to 3.79% of body weight. Milk production was highest (33.4 kg/d) for cows fed silage from corn harvested at the two-thirds milkline stage and lowest (32.4 kg/d) for cows fed silage from corn harvested at the early dent stage. Milk protein production was highest for cows fed silage from corn harvested at the two-thirds milkline stage (1.17 vs. 1.12 to 1.13 kg/d). Apparent total tract digestion of dry matter, organic matter, crude protein, acid detergent fiber, and starch was lowest for cows fed silage from corn harvested at the black layer stage. Although starch intake was similar for cows fed silage from corn harvested at the two-thirds milkline stage and for cows fed silage from corn harvested at the black layer stage (9 kg/d), intake of digestible starch was 0.4 kg/d lower for cows fed silage from corn harvested at the black layer stage. The optimum stage for corn that was ensiled was two-thirds milkline with some flexibility between quarter and two-thirds milkline.
Theory and observation indicate that changes in the rate of primary production can alter the balance between the bottom-up influences of plants and resources and the top-down regulation of herbivores ...and predators on ecosystem structure and function. The Exploitation Ecosystem Hypothesis (EEH) posited that as aboveground net primary productivity (ANPP) increases, the additional biomass should support higher trophic levels. We developed an extension of EEH to include the impacts of increases in ANPP on belowground consumers in a similar manner as aboveground, but indirectly through changes in the allocation of photosynthate to roots. We tested our predictions for plants aboveground and for phytophagous nematodes and their predators belowground in two common arctic tundra plant communities subjected to 11 years of increased soil nutrient availability and/or exclusion of mammalian herbivores. The less productive dry heath (DH) community met the predictions of EEH aboveground, with the greatest ANPP and plant biomass in the fertilized plots protected from herbivory. A palatable grass increased in fertilized plots while dwarf evergreen shrubs and lichens declined. Belowground, phytophagous nematodes also responded as predicted, achieving greater biomass in the higher ANPP plots, whereas predator biomass tended to be lower in those same plots (although not significantly). In the higher productivity moist acidic tussock (MAT) community, aboveground responses were quite different. Herbivores stimulated ANPP and biomass in both ambient and enriched soil nutrient plots; maximum ANPP occurred in fertilized plots exposed to herbivory. Fertilized plots became dominated by dwarf birch (a deciduous shrub) and cloudberry (a perennial forb); under ambient conditions these two species coexist with sedges, evergreen dwarf shrubs, and
Sphagnum
mosses. Phytophagous nematodes did not respond significantly to changes in ANPP, although predator biomass was greatest in control plots. The contrasting results of these two arctic tundra plant communities suggest that the predictions of EEH may hold for very low ANPP communities, but that other factors, including competition and shifts in vegetation composition toward less palatable species, may confound predicted responses to changes in productivity in higher ANPP communities such as the MAT studied here.
Multiple metabolic and hormonal factors can affect the success of protocols for ovarian superstimulation. In this study, the effect of acute feed restriction and increased LH content in the ...superstimulatory FSH preparation on numbers of ovulations, fertilization, and embryo quality in lactating dairy cows was evaluated. Two experiments were performed using a Latin square design with treatments arranged as a 2×2 factorial: feed restriction (FR; 25% reduction in dry matter intake) compared with ad libitum (AL) feeding, combined with high (H) versus low (L) LH in the last 4 injections of the superstimulatory protocol. As expected, FR decreased circulating insulin concentrations (26.7 vs. 46.0μU/mL). Two analyses were performed: one that evaluated the complete Latin square in experiment 2 and a second that evaluated only the first periods of experiments 1 and 2. For both analyses, follicle numbers, ovulation rates, and corpora lutea on d 7 were not different. In the first period analysis of experiments 1 and 2, we observed an interaction between feed allowance and amount of LH on fertilization rates, percentage of embryos or oocytes that were quality 1 and 2 embryos, and number of embryos or oocytes that were degenerate. Fertilization rates were greater for the AL-L (89.4%) and FR-H (80.1%) treatments compared with the AL-H (47.9%) and FR-L (59.9%) treatments. Similarly, the proportion of total embryos or oocytes designated as quality 1 and 2 embryos was greater for AL-L (76.7%) and FR-H (73.4%) treatments compared with AL-H (35.6%) and FR-L (47.3%) treatments. In addition, the number of degenerate embryos was decreased for AL-L (1.3) and FR-H (0.4) treatments compared with the AL-H (2.6) and FR-L (2.3) treatments. Thus, cows with either too low (FR-L) or too high (AL-H) insulin and LH stimulation had lesser embryo production after superstimulation because of reduced fertilization rate and increased percentage of degenerate embryos. Therefore, interaction of the gonadotropin content of the superstimulatory preparation with the nutritional program of the donor cow needs to be considered to optimize success of ovarian superstimulatory protocols.
Prior to genomic selection on a trait, a reference population needs to be established to link marker genotypes with phenotypes. For costly and difficult-to-measure traits, international collaboration ...and sharing of data between disciplines may be necessary. Our aim was to characterize the combining of data from nutrition studies carried out under similar climate and management conditions to estimate genetic parameters for feed efficiency. Furthermore, we postulated that data from the experimental cohorts within these studies can be used to estimate the net energy of lactation (NEL) densities of diets, which can provide estimates of energy intakes for use in the calculation of the feed efficiency metric, residual feed intake (RFI), and potentially reduce the effect of variation in energy density of diets. Individual feed intakes and corresponding production and body measurements were obtained from 13 Midwestern nutrition experiments. Two measures of RFI were considered, RFIMcal and RFIkg, which involved the regression of NEL intake (Mcal/d) or dry matter intake (DMI; kg/d) on 3 expenditures: milk energy, energy gained or lost in body weight change, and energy for maintenance. In total, 677 records from 600 lactating cows between 50 and 275 d in milk were used. Cows were divided into 46 cohorts based on dietary or nondietary treatments as dictated by the nutrition experiments. The realized NEL densities of the diets (Mcal/kg of DMI) were estimated for each cohort by totaling the average daily energy used in the 3 expenditures for cohort members and dividing by the cohort’s total average daily DMI. The NEL intake for each cow was then calculated by multiplying her DMI by her cohort’s realized energy density. Mean energy density was 1.58 Mcal/kg. Heritability estimates for RFIkg, and RFIMcal in a single-trait animal model did not differ at 0.04 for both measures. Information about realized energy density could be useful in standardizing intake data from different climate conditions or management systems, as well as investigating potential genotype by diet interactions.
The objective of this study was to investigate the effects of tallow and choice white grease (CWG) fed at 0, 2, and 4% of the diet dry matter (DM) on rumen fermentation and performance of dairy cows ...when corn silage is the sole forage source. Fifteen midlactation Holstein cows were used in a replicated 5×5 Latin square design with 21-d periods. Treatments were 0% fat (control), 2% tallow, 2% CWG, 4% tallow, and 4% CWG (DM basis). The forage:concentrate ratio was 50:50, and diets were formulated to contain 18% crude protein and 32% neutral detergent fiber (DM basis). Cows were allowed ad libitum consumption of diets fed twice daily as total mixed rations. Cows fed supplemental fat had lower DM intake and produced less milk and milk fat than cows fed the control diet. Feeding 4% fat reduced milk production and milk fat yield relative to feeding 2% fat. Treatments had little effect on the concentration of trans-octadecenoic acids in milk fat. Total trans fatty acids were poorly related to changes in milk fat percentage. Ruminal pH and total volatile fatty acids concentration were not affected by supplemental fat. The acetate:propionate ratio, NH3-N, and numbers of protozoa in the rumen were significantly decreased when fat was added to the diets. Source of dietary fat did not affect rumen parameters. There was no treatment effect on in situ corn silage DM and neutral detergent fiber disappearance. Including fat in corn silage-based diets had negative effects on milk production and rumen fermentation regardless of the source or level of supplemental fat.
