Digital soil mapping has been widely used to develop statistical models of the relationships between environmental variables and soil attributes. This study aimed at determining and mapping the ...spatial distribution of the variability in soil chemical properties of the agricultural floodplain lands of the Bara district in Nepal. The study was carried out in 23 Village Development Committees with 12,516 ha total area, in the southern part of the Bara district. A total of 109 surface soil samples (0 to 15 cm depth) were collected and analyzed for pH, organic matter (OM), nitrogen (N), phosphorus (P, expressed as P2O5), potassium (K, expressed as K2O), zinc (Zn), and boron (B) status. Descriptive statistics showed that most of the measured soil chemical variables (other than pH and P2O5) were skewed and non-normally distributed and logarithmic transformation was then applied. A geostatistical tool, kriging, was used in ArcGIS to interpolate measured values for those variables and several digital map layers were developed based on each soil chemical property. Geostatistical interpolation identified a moderate spatial variability for pH, OM, N, P2O5, and a weak spatial variability for K2O, Zn, and B, depending upon the use of amendments, fertilizing methods, and tillage, along with the inherent characteristics of each variable. Exponential (pH, OM, N, and Zn), Spherical (K2O and B), and Gaussian (P2O5) models were fitted to the semivariograms of the soil variables. These maps allow farmers to assess existing farm soils, thus allowing them to make easier and more efficient management decisions and maintain the sustainability of productivity.
Supernumerary mini-chromosomes-a unique type of genomic structural variation-have been implicated in the emergence of virulence traits in plant pathogenic fungi. However, the mechanisms that ...facilitate the emergence and maintenance of mini-chromosomes across fungi remain poorly understood. In the blast fungus Magnaporthe oryzae (Syn. Pyricularia oryzae), mini-chromosomes have been first described in the early 1990s but, until very recently, have been overlooked in genomic studies. Here we investigated structural variation in four isolates of the blast fungus M. oryzae from different grass hosts and analyzed the sequences of mini-chromosomes in the rice, foxtail millet and goosegrass isolates. The mini-chromosomes of these isolates turned out to be highly diverse with distinct sequence composition. They are enriched in repetitive elements and have lower gene density than core-chromosomes. We identified several virulence-related genes in the mini-chromosome of the rice isolate, including the virulence-related polyketide synthase Ace1 and two variants of the effector gene AVR-Pik. Macrosynteny analyses around these loci revealed structural rearrangements, including inter-chromosomal translocations between core- and mini-chromosomes. Our findings provide evidence that mini-chromosomes emerge from structural rearrangements and segmental duplication of core-chromosomes and might contribute to adaptive evolution of the blast fungus.
Western corn rootworm, Diabrotica virgifera virgifera LeConte, has evolved resistance to transgenic maize, Zea maize L., that produces the insecticidal protein Cry3Bb1, which is derived from the ...bacterium Bacillus thuringiensis. We hypothesized that the level of Cry3Bb1 resistance in populations of western corn rootworm could be influenced by farming practices. To test this hypothesis, we evaluated the effect of field history on resistance to Cry3Bb1 maize by western corn rootworm. In 2013 and 2014, rootworm adults were collected from the four types of maize fields: 1) current problem fields, 2) past problem fields, 3) rotated maize fields, and 4) continuous maize fields. Those field populations along with seven Bt-susceptible control populations were tested for Cry3Bb1 resistance with both plant-based and diet-based bioassays. All field populations were resistant to Cry3Bb1 regardless of field history, however, some variation in the degree of resistance was found. For all categories of field populations, larval survivorship on Cry3Bb1 maize was significantly higher than control populations, and did not differ from survival on non-Bt maize. Evidence of resistance to Cry3Bb1 maize in plant-based bioassays was further supported by diet-based bioassays and we found a positive relationship between LC50 values from diet-based bioassays and the larval survivorship in plant-based bioassays. This study provides evidence of Cry3Bb1 resistance throughout the agricultural landscape studied, irrespective of the field history, and highlights the need for improved resistance management approaches, such as better use of integrated pest management to better delay pest resistance.

HIV testing is an entry point to access HIV care and prevention services. Building Healthy Online Communities developed a website ( TakeMeHome.org ) where participants can order HIV home test kits. ...The purpose of this study was to analyze the costs and impact of the TakeMeHome program.
We estimated the costs of TakeMeHome across all participating jurisdictions for the first year of the program. We estimated program costs using purchase orders and invoices, contracts, and allocation of staff time, and the costs included website design, participant recruitment, administration and overhead, HIV self-test kits, and shipping and handling. Primary outcomes of the analysis were total program cost, cost per HIV test, and cost per new HIV diagnosis.
The TakeMeHome program distributed 5323 HIV self-tests to 4859 participants over a 12-month period. The total program cost over this period was $314,870. The cost per HIV test delivered was estimated at $59, and the cost per person tested was $65. The program identified 18 confirmed new HIV diagnoses (0.6% positivity) verified with surveillance data in 7 health jurisdictions at $169,890. The cost per confirmed new HIV diagnosis was estimated at $9440.
The TakeMeHome program delivered HIV self-testing at a reasonable cost, and the program may be a cost-effective use of HIV prevention resources. The public-private partnership can be an effective mechanism to validate HIV diagnoses identified with self-testing and provide HIV prevention and linkage to care services.
Leishmaniasis is one of the World's most problematic diseases in developing countries. Traditional medicines to treat leishmaniasis have serious side effects, as well as significant parasite ...resistance problems. In this work, two alkaloids 1 and 2 were obtained from Corydalis govaniana Wall and seven alkaloids 3-9, were obtained from Erythrina verna. The structures of the compounds were confirmed by mass spectrometry and 1D- and 2D-NMR spectroscopy. The leishmanicidal activity of compounds 1-9 against Leishmania amazonensis was tested on promastigote forms and cytotoxicity against J774 (macrophage cell line) was assessed in vitro. Compound 1 showed potent activity (IC50 = 0.18 µg/mL), compared with the standard amphotericin B (IC50 = 0.20 µg/mL). The spirocyclic erythrina-alkaloids showed lower leishmanicidal activity than dibenzoquinolizine type alkaloids.
Genus Corydalis is a rich source of isoquinoline alkaloids reported to having potential bioactivities. Corydalis chaerophylla collected from Nepal at an altitude of 2400–4800 m was extracted using ...hexane, methanol and chloroform as solvents. The resulting hexane, methanol and chloroform extracts were subjected to LC–DAD‐MSn analysis to yield fifteen different alkaloids. To assess any potential pharmacological properties, antimicrobial activity against two Gram‐positive, two Gram‐negative bacterial strains and one fungal strain was assessed, revealing significant inhibitive action of the methanol and chloroform extracts. Of the extracts obtained using chloroform contained the highest content of phenolic compounds at 113 mg GAE/g, while the highest total flavonoid content was found for the hexane extract with a value of 46.45 mg QE/g. The chloroform extract also exhibited a considerable antioxidant activity at IC50 value, 261.5±3 μg/mL, for the DPPH assay. Conversely, the methanol extract exhibited the highest LC50 value for Brine Shrimp cytotoxicity at 196±3 μg/mL being least potential for the test. The methanol extract was found to be the most active against α‐amylase inhibition with an IC50 of 51.52±2 μg/mL. In an in vivo acute oral toxicity study against mice, methanol and chloroform extracts presented harmful effects with 1000.36 mg/kg BW and 515 mg/kg BW for LD50, respectively. By analyzing all the results of the solvents used, the chloroform extract was found to be the most active, a feature that will be used in future isolation procedures and other pharmacological tests.
This paper aims to analyze the effects of biogas and electricity based cooking on energy use and greenhouse gas (GHG) as well as local air pollutant emissions during 2010–2050 in the case of Nepal, ...which is highly dependent on traditional biomass (mainly fuelwood) for cooking. The country is rich in hydropower resources. A long-term bottom-up energy system model has been developed using Asia-Pacific Integrated Model/Enduse (AIM/Enduse) model for the analysis. The study developed a business as usual (BAU) scenario and three alternative cooking scenarios. Three alternative scenarios, named as “CL”, “CM” and “CH” scenarios; consider low, medium and high level of penetrations of electric- and biogas-based cooking options, respectively. The changes in energy use and electricity generation in the BAU and alternative scenarios have been compared. Fuelwood consumption in the residential sector in 2050 when compared to the BAU would decrease by 12.5% in CL, 19.0% in CM and 24.2% in CH scenarios; and liquefied petroleum gas (LPG) consumption would decrease by 12.8% in CL, 16.3% in CM and 19.6% in CH scenarios. The electricity generation requirement in 2050 would increase by 9.4% in CL, 13.9% in CM and 17.0% in CH scenarios. Finally, the assessment of GHG and local pollutant emissions shows the decrease in all gases in CL, CM and CH scenarios when compared to the BAU.

The evolution of resistance and cross-resistance threaten the sustainability of genetically engineered crops that produce insecticidal toxins derived from the bacterium Bacillus thuringiensis (Bt). ...Western corn rootworm, Diabrotica virgifera virgifera LeConte, is a serious pest of maize and has been managed with Bt maize since 2003. We conducted laboratory bioassays with maize hybrids producing Bt toxins Cry3Bb1, mCry3A, eCry3.1Ab, and Cry34/35Ab1, which represent all commercialized Bt toxins for management of western corn rootworm. We tested populations from fields where severe injury to Cry3Bb1 maize was observed, and populations that had never been exposed to Bt maize. Consistent with past studies, bioassays indicated that field populations were resistant to Cry3Bb1 maize and mCry3A maize, and that cross-resistance was present between these two types of Bt maize. Additionally, bioassays revealed resistance to eCry3.1Ab maize and cross-resistance among Cry3Bb1, mCry3A and eCry3.1Ab. However, no resistance or cross-resistance was detected for Cry34/35Ab1 maize. This broad-spectrum resistance illustrates the potential for insect pests to develop resistance rapidly to multiple Bt toxins when structural similarities are present among toxins, and raises concerns about the long-term durability of Bt crops for management of some insect pests.
Cultural heritage is a reflection of a community’s adopted ways of life. It comprises customs, behaviors, settings, physical objects, artistic expression, and ideals. As an impact of the Earthquake ...in 2015, the heritage building suffered catastrophic damage. This research examined the Thimi community’s approach to urban heritage construction through conservation, reconstruction, renovation, and restoration, and shares problems, success stories, and lessons learned in the cases of Phalcha (Rest House) and Dyo (Temple). The Thimi Municipality provided all required technical and financial support for these renovation and reconstruction activities. Conducting a depth literature review and understanding the process of both qualitative and quantitative methods of rebuilding, renovating, and restoring the urban cultural heritage structure, this research uses a case study approach. Locals, consultants, and technical employees involved in the rehabilitation and planning work provided most of the materials for all case studies. This study finds that the residents of Thimi feel pride in having their heritage structures restored by local community participation. Despite the local community efforts, there are a few shortcomings with the financial, technical, social, and administrative constructions and understanding of customary practices. The Madhyapur Thimi Municipality has developed measures to support communities willing to lead urban heritage conservation.
