Plastic, as a form of marine litter, is found in varying quantities and sizes around the globe from surface waters to deep-sea sediments. Identifying patterns of microplastic distribution will ...benefit an understanding of the scale of their potential effect on the environment and organisms. As sea ice extent is reducing in the Arctic, heightened shipping and fishing activity may increase marine pollution in the area. Microplastics may enter the region following ocean transport and local input, although baseline contamination measurements are still required. Here we present the first study of microplastics in Arctic waters, south and southwest of Svalbard, Norway. Microplastics were found in surface (top 16 cm) and sub-surface (6 m depth) samples using two independent techniques. Origins and pathways bringing microplastic to the Arctic remain unclear. Particle composition (95% fibres) suggests they may either result from the breakdown of larger items (transported over large distances by prevailing currents, or derived from local vessel activity), or input in sewage and wastewater from coastal areas. Concurrent observations of high zooplankton abundance suggest a high probability for marine biota to encounter microplastics and a potential for trophic interactions. Further research is required to understand the effects of microplastic-biota interaction within this productive environment.
Zooplankton plays a pivotal role in marine ecosystems and the characterisation of its biodiversity still represents a challenge for marine ecologists. In this study, mesozooplankton composition from ...46 samples collected in summer along the western Adriatic Sea, was retrieved by DNA metabarcoding analysis. For the first time, the highly variable fragments of the mtDNA COI and the V9 region of 18S rRNA genes were used in a combined matrix to compile an inventory of mesozooplankton at basin scale. The number of sequences retrieved after quality filtering were 824,148 and 223,273 for COI and 18S (V9), respectively. The taxonomical assignment against reference sequences, using 95% (for COI) and 97% (for 18S) similarity thresholds, recovered 234 taxa. NMDS plots and cluster analysis divided coastal from offshore samples and the most representative species of these clusters were distributed according to the dominant surface current pattern of the Adriatic for the summer period. For selected sampling sites, mesozooplankton species were also identified under a stereo microscope providing insights on the strength and weakness of the two approaches. In addition, DNA metabarcoding was shown to be helpful for the monitoring of non-indigenous marine metazoans and spawning areas of commercial fish species. We defined pros and cons of applying this approach at basin scale and the benefits of combining the datasets from two genetic markers.
Malaria is still a devastating disease with 228 million cases globally and 405,000 lethal outcomes in 2018, mainly in children under five years of age. The threat of emerging malaria strains ...resistant to currently available drugs has made the search for novel drug targets compelling. The process by which Plasmodium falciparum parasites invade the host cell has been widely studied, but only a few erythrocyte proteins involved in this process have been identified so far. The erythrocyte protein Rac1 is a GTPase that plays an important role in host cell invasion by many intracellular pathogens. Here we show that Rac1 is recruited in proximity to the site of parasite entry during P. falciparum invasion process and that subsequently localizes to the parasitophorous vacuole membrane. We also suggest that this GTPase may be involved in erythrocyte invasion by P. falciparum, by testing the effect of specific Rac1 inhibitory compounds. Finally, we suggest a secondary role of the erythrocyte GTPase also in parasite intracellular development. We here characterize a new erythrocyte protein potentially involved in P. falciparum invasion of the host cell and propose the human GTPase Rac1 as a novel and promising antimalarial drug target.
Anchovy and sardine in the Mediterranean are known to share the same habitat and, consequently, to interact with one another. These two sympatric pelagic species are planktivorous and consume a wide ...range of planktonic prey items during all their developmental stages, potentially overlapping their ecological niches, although the feeding interactions between these species have been poorly investigated. Here we compare the dietary habits of the juvenile phases of anchovy and sardine during different seasons in the northwestern Mediterranean Sea, through analysis of their stomach contents and of their feeding-related anatomical characteristics. In this study we show that juveniles of anchovy and sardine do not compete for food, and we describe significant dietary differences between anchovy and sardine due to their different alimentary tract morphology.

The yeast Saccharomyces cerevisiae is a reference model system and one of the widely used microorganisms in many biotechnological processes. In industrial yeast applications, combined strategies aim ...to maximize biomass/product yield, with the fed-batch culture being one of the most frequently used. Flow cytometry (FCM) is widely applied in biotechnological processes and represents a key methodology to monitor cell population dynamics. We propose here an application of FCM in the analysis of yeast cell cycle along the time course of a typical S. cerevisiae fed-batch culture. We used two different dyes, SYTOX Green and SYBR Green, with the aim to better define each stage of cell cycle during S. cerevisiae fed-batch culture. The results provide novel insights in the use of FCM cell cycle analysis for the real-time monitoring of S. cerevisiae bioprocesses.
Monitoring the spreading of marine invasive species represents one of the most relevant challenges for marine scientists in order to understand their impact on the environment. In recent years, ...citizen science is becoming more and more involved in research programs, especially taking advantage of new digital technologies. Here, we present the results obtained in the first 20 months (from 12 July 2019 to 8 March 2021) since launching avvistAPP. This new app was conceived to track the spreading of the invasive ctenophore Mnemiopsis leidyi in the Adriatic Sea; it was also designed to collect sightings of 18 additional marine taxa (ctenophores, jellyfish, sea turtles, dolphins, salps and noble pen shell). A total of 1224 sightings were recorded, of which 530 referred to Mnemiopsis, followed by the scyphozoan jellyfish Rhizostoma pulmo (22%), Cotylorhiza tuberculata (11%) and Aurelia spp. (8%). avvistAPP produced data confirming the presence of Mnemiopsis (often in abundances > 20 individuals m−2) along almost the entire Italian coast in the summer of 2019 and 2020.
Epithelial-derived alarmins (IL-33, TSLP, and IL-25) play an upstream role in the pathogenesis of asthma. Basophil-derived cytokines are a pivotal component of allergic inflammation. We evaluated the
...effects of IL-33, TSLP, and IL-25, alone and in combination with IL-3 on purified peripheral blood human basophils (hBaso) and bone marrow-derived mouse basophils (mBaso) in modulating the production of IL-4, IL-13, CXCL8 or the mouse CXCL8 equivalents CXCL1 and CXCL2. IL-3 and IL-33, but not TSLP and IL-25, concentration-dependently induced IL-4, IL-13, and CXCL8 release from hBaso. IL-3 synergistically potentiated the release of cytokines induced by IL-33 from hBaso. In mBaso, IL-3 and IL-33 rapidly induced IL-4 and IL-13 mRNA expression and protein release. IL-33, but not IL-3, induced CXCL2 and CXCL1 from mBaso. Differently from hBaso, TSLP induced IL-4, IL-13, CXCL1 and CXCL2 mRNA expression and protein release from mBaso. IL-25 had no effect on IL-4, IL-13, and CXCL1/CXCL2 mRNA expression and protein release even in the presence of IL-3. No synergism was observed between IL-3 and either IL-25 or TSLP. IL-3 inhibited both TSLP- and IL-33-induced CXCL1 and CXCL2 release from mBaso. Our results highlight some similarities and marked differences between the effects of IL-3 and alarmins on the release of cytokines from human and mouse basophils.
Food availability is thought to exert a bottom-up control on the population dynamics of small pelagic fish; therefore, studies on trophic ecology are essential to improve their management. Sardina ...pilchardus is one of the most important commercial species in the Adriatic Sea, yet there is little information on its diet in this area. Adult sardines were caught in the Gulf of Trieste (northern Adriatic) from spring 2006 to winter 2007. Experimental catches conducted over 24-h cycles in May, June and July showed that the sardines foraged mainly in the late afternoon. A total of 96 adult sardines were analysed: the number of prey varied from a minimum of 305 to a maximum of 3318 prey/stomach, with an overall mean of 1259 ± 884 prey/stomach. Prey items were identified to the lowest possible taxonomical level, counted and measured at the stereo-microscope. Overall, sardines fed on a wide range of planktonic organisms (87 prey items from 17 μm to 18.4 mm were identified), with copepods being the most abundant prey (56%) and phytoplankton never exceeding 10% of the prey. Copepods of the Clauso-Paracalanidae group and of the genus Oncaea were by far the most important prey. The carbon content of prey items was indirectly estimated from prey dry mass or body volume. Almost all carbon uptake relied on a few groups of zooplankton. Ivlev’s selectivity index showed that sardines positively selected small preys (small copepods < 1 mm size), but also larger preys (such as teleost eggs, decapod larvae and chaetognaths), confirming their adaptive feeding capacity.
Immune cell-derived extracellular vesicles (EV) affect tumor progression and hold promise for therapeutic applications. Eosinophils are major effectors in Th2-related pathologies recently implied in ...cancer. Here, we evaluated the anti-tumor activities of eosinophil-derived EV following activation with the alarmin IL-33. We demonstrate that IL-33-activated mouse and human eosinophils produce higher quantities of EV with respect to eosinophils stimulated with IL-5. Following incorporation of EV from IL-33-activated eosinophils (Eo33-EV), but not EV from IL-5-treated eosinophils (Eo5-EV), mouse and human tumor cells increased the expression of cyclin-dependent kinase inhibitor (CDKI)-related genes resulting in cell cycle arrest in G0/G1, reduced proliferation and inhibited tumor spheroid formation. Moreover, tumor cells incorporating Eo33-EV acquired an epithelial-like phenotype characterized by E-Cadherin up-regulation, N-Cadherin downregulation, reduced cell elongation and migratory extent in vitro, and impaired capacity to metastasize to lungs when injected in syngeneic mice. RNA sequencing revealed distinct mRNA signatures in Eo33-EV and Eo5-EV with increased presence of tumor suppressor genes and enrichment in pathways related to epithelial phenotypes and negative regulation of cellular processes in Eo33-EV compared to Eo5-EV. Our studies underscore novel IL-33-stimulated anticancer activities of eosinophils through EV-mediated reprogramming of tumor cells opening perspectives on the use of eosinophil-derived EV in cancer therapy.
