High-energy photons from the Crab Nebula
The Crab Nebula contains a pulsar that excites the surrounding gas to emit high-energy radiation. The combination of the pulsar's youth and nearby location ...makes the nebula the brightest gamma-ray source in the sky. The LHAASO Collaboration report observations of this source at energies of tera– to peta–electron volts, extending the spectrum of this prototypical object. They combine these data with observations at lower energies to model the physics of the emission process. The multiwave-length data can be explained by a combination of synchrotron radiation and inverse Compton scattering.

Detection of the Crab Nebula at peta–electron volt energies constrains the gamma-ray emission mechanism.
The Crab Nebula is a bright source of gamma rays powered by the Crab Pulsar’s rotational energy through the formation and termination of a relativistic electron-positron wind. We report the detection of gamma rays from this source with energies from 5 × 10
−4
to 1.1 peta–electron volts with a spectrum showing gradual steepening over three energy decades. The ultrahigh-energy photons imply the presence of a peta–electron volt electron accelerator (a pevatron) in the nebula, with an acceleration rate exceeding 15% of the theoretical limit. We constrain the pevatron’s size between 0.025 and 0.1 parsecs and the magnetic field to ≈110 microgauss. The production rate of peta–electron volt electrons, 2.5 × 10

ergs per second, constitutes 0.5% of the pulsar spin-down luminosity, although we cannot exclude a contribution of peta–electron volt protons to the production of the highest-energy gamma rays.
In Alzheimer's disease (AD), neurodegenerative signals such as amyloid-beta (Aβ) and the precursors of neurotrophins, outbalance neurotrophic signals, causing synaptic dysfunction and ...neurodegeneration. The neurotrophin receptor p75 (p75NTR) is a receptor of Aβ and mediates Aβ-induced neurodegenerative signals. The shedding of its ectodomain from the cell surface is physiologically regulated; however, the function of the diffusible p75NTR ectodomain (p75ECD) after shedding remains largely not known. Here, we show that p75ECD levels in cerebrospinal fluid and in the brains of Alzheimer's patients and amyloid-beta precursor protein (APP)/PS1 transgenic mice were significantly reduced, due to inhibition of the sheddase-tumor necrosis factor-alpha-converting enzyme by Aβ. Restoration of p75ECD to the normal level by brain delivery of the gene encoding human p75ECD before or after Aβ deposition in the brain of APP/PS1 mice reversed the behavioral deficits and AD-type pathologies, such as Aβ deposit, apoptotic events, neuroinflammation, Tau phosphorylation and loss of dendritic spine, neuronal structures and synaptic proteins. Furthermore, p75ECD can also reduce amyloidogenesis by suppressing β-secretase expression and activities. Our data demonstrate that p75ECD is a physiologically neuroprotective molecule against Aβ toxicity and would be a novel therapeutic target and biomarker for AD.
The diffuse Galactic γ-ray emission, mainly produced via interactions between cosmic rays and the interstellar medium and/or radiation field, is a very important probe of the distribution, ...propagation, and interaction of cosmic rays in the Milky Way. In this Letter, we report the measurements of diffuse γ rays from the Galactic plane between 10 TeV and 1 PeV energies, with the square kilometer array of the Large High Altitude Air Shower Observatory (LHAASO). Diffuse emissions from the inner (15°<l<125°, |b|<5°) and outer (125°<l<235°, |b|<5°) Galactic plane are detected with 29.1σ and 12.7σ significance, respectively. The outer Galactic plane diffuse emission is detected for the first time in the very- to ultra-high-energy domain (E>10 TeV). The energy spectrum in the inner Galaxy regions can be described by a power-law function with an index of -2.99±0.04, which is different from the curved spectrum as expected from hadronic interactions between locally measured cosmic rays and the line-of-sight integrated gas content. Furthermore, the measured flux is higher by a factor of ∼3 than the prediction. A similar spectrum with an index of -2.99±0.07 is found in the outer Galaxy region, and the absolute flux for 10≲E≲60 TeV is again higher than the prediction for hadronic cosmic ray interactions. The latitude distributions of the diffuse emission are consistent with the gas distribution, while the longitude distributions show clear deviation from the gas distribution. The LHAASO measurements imply that either additional emission sources exist or cosmic ray intensities have spatial variations.
High tumor mutational burden (TMB-H) is correlated with enhanced objective response rate (ORR) and progression-free survival (PFS) for certain cancers receiving immunotherapy. This study aimed to ...investigate the safety and efficacy of toripalimab, a humanized programmed death-1 (PD-1) antibody, in advanced gastric cancer (AGC), and the predictive survival benefit of TMB and PD-L1.
We reported on the AGC cohort of phase Ib/II trial evaluating the safety and activity of toripalimab in patients with AGC, oesophageal squamous cell carcinoma, nasopharyngeal carcinoma and head and neck squamous cell carcinoma. In cohort 1, 58 chemo-refractory AGC patients received toripalimab (3 mg/kg d1, Q2W) as a monotherapy. In cohort 2, 18 chemotherapy-naive AGC patients received toripalimab (360 mg d1, Q3W) with oxaliplatin 130 mg/m2 qd, d1, capecitabine 1000 mg/m2 b.i.d., d1–d14, Q3W as first-line treatment. Primary end point was ORR. Biomarkers such as PD-L1 and TMB were evaluated for correlation with clinical efficacy.
In cohort 1, the ORR was 12.1% and the disease control rate (DCR) was 39.7%. Median PFS was 1.9 months and median OS was 4.8 months. The TMB-H group showed significant superior OS than the TMB-L group 14.6 versus 4.0 months, HR = 0.48 (96% CI 0.24–0.96), P = 0.038, while PD-L1 overexpression did not correlate with significant survival benefit. A 77.6% of patients experienced at least one treatment-related adverse event (TRAE), and 22.4% of patients experienced a grade 3 or higher TRAE. In cohort 2, the ORR was 66.7% and the DCR was 88.9%. A 94.4% of patients experienced at least one TRAE and 38.9% of patients experienced grade 3 or higher TRAEs.
Toripalimab has demonstrated a manageable safety profile and promising antitumor activity in AGC patients, especially in combination with XELOX. High TMB may be a predictive marker for OS of AGC patients receiving toripalimab as a single agent.

Abstract Radio observations of pulsars offer a potential method to probe the intricate microstructure in the turbulent interstellar medium. Here we report on a high-resolution dynamic spectral ...analysis of the “swooshing pulsar” B0919+06 observed with the Five-hundred-meter Aperture Spherical radio Telescope over multiple epochs and with the ultrawideband receiver on the Parkes radio telescope. For all observations, the dynamic scintillation spectra, two-dimensional autocovariance functions, and secondary spectra are presented. At 1250 MHz, the decorrelation bandwidth, diffraction timescale, and the drift rate are determined to be Δ ν d = 25.89 ± 7.55 MHz, Δ τ d = 14.42 ± 3.98 minutes, and dt / d ν = 0.07 ± 0.14 minutes MHz −1 , respectively. The frequency dependencies of the scintillation parameters exhibit single power-law spectral behaviors, indicating that the electron density fluctuations in the interstellar medium approximately follow the Kolmogorov spectrum. The secondary spectra exhibit two distinct parabolic arcs with well-determined curvatures of 0.002 and 0.02 s 3 for the outer and inner arcs, respectively. The locations of the scattering screens are approximately determined to be 157.3 and 726.0 pc, respectively, from the pulsar for isotropic scattering. The inner scintillation arc is present contemporaneously over a wide frequency range, indicating that the scintillation arc is a broadband phenomenon. The arc curvature scales with observing frequency as a power law with an index of −2.05 ± 0.05, which implies that the scattering screen spans a physical distance from 689.7 to 883.3 pc from the pulsar.
Abstract
We have carried out a detailed study of polarimetric individual pulse emission from the pulsar J1701−3726 (B1658−37), observed at 1369 MHz using the Parkes 64 m radio telescope. The ...single-pulse sequences reveal the presence of the three major emission phenomena of pulse nulling, mode changing, and subpulse drifting. Trimodal distribution of the pulse energy is present, implying one population of nulls and two others of emission in the phase window. The mean flux density of the normal mode is almost two times that of the abnormal mode. Our data show that, for PSR J1701−3726, 64% of the time was spent in the normal mode and 12% was in the abnormal mode. The single pulses show the presence of two distinct periodic modulations using a fluctuation spectral analysis. About 24% of the nulls are found to create alternating bunches of nulls and bursts in a quasiperiodic manner with a longer periodicity of 48 ± 4 rotational periods. Additionally, the pulsar presents a steady even–odd modulated feature with a stationary longitude within the pulse window. The ramifications for constraining the viewing geometry and understanding the radio emission mechanisms are discussed.
This study presents an "Earth observation-based" method for estimating root zone storage capacity – a critical, yet uncertain parameter in hydrological and land surface modelling. By assuming that ...vegetation optimises its root zone storage capacity to bridge critical dry periods, we were able to use state-of-the-art satellite-based evaporation data computed with independent energy balance equations to derive gridded root zone storage capacity at global scale. This approach does not require soil or vegetation information, is model independent, and is in principle scale independent. In contrast to a traditional look-up table approach, our method captures the variability in root zone storage capacity within land cover types, including in rainforests where direct measurements of root depths otherwise are scarce. Implementing the estimated root zone storage capacity in the global hydrological model STEAM (Simple Terrestrial Evaporation to Atmosphere Model) improved evaporation simulation overall, and in particular during the least evaporating months in sub-humid to humid regions with moderate to high seasonality. Our results suggest that several forest types are able to create a large storage to buffer for severe droughts (with a very long return period), in contrast to, for example, savannahs and woody savannahs (medium length return period), as well as grasslands, shrublands, and croplands (very short return period). The presented method to estimate root zone storage capacity eliminates the need for poor resolution soil and rooting depth data that form a limitation for achieving progress in the global land surface modelling community.

We report the discovery of an ultrahigh-energy (UHE) gamma-ray source, LHAASO J2108+5157, by analyzing the LHAASO-KM2A data of 308.33 live days. A significant excess of gamma ray–induced ...showers is observed in both energy bands of 25−100 and >100 TeV with 9.5
σ
and 8.5
σ
, respectively. This source is not significantly favored as an extended source with an angular extension smaller than the point-spread function of KM2A. The measured energy spectrum from 20 to 200 TeV can be approximately described by a power-law function with an index of −2.83 ± 0.18
stat
. A harder spectrum is demanded at lower energies considering the flux upper limit set by Fermi-LAT observations. The position of the gamma-ray emission is correlated with a giant molecular cloud, which favors a hadronic origin. No obvious counterparts have been found, and deeper multiwavelength observations will help to cast new light on this intriguing UHE source.
Layered Ti-Al metal composite (LMC) fabricated by hot-pressing and hot-rolling process displays higher ductility than that of both components. In this paper, a combination of digital image ...correlation (DIC) and X-ray tomography revealed that strain delocalization and constrained crack distribution are the origin of extraordinary tensile ductility. Strain delocalization was derived from the transfer of strain partitioning between Ti and Al layer, which relieved effectively the strain localization of LMC. Furthermore, the extensive cracks of LMC were restricted in the interface due to constraint effect. Layered architecture constrained the distribution of cracks and significantly relieved the strain localization. Meanwhile, the transfer of strain partitioning and constrained crack distribution were believed to inhibit the strain localization of Ti and change the deformation mechanisms of Ti. Our finding enriches current understanding about simultaneously improving the strength and ductility by structural design.
