This article aims to study the theoretical informatization heritage of Xi Jinping’s socialist thought with Chinese characteristics in the new era. The article first uses web crawler technology to ...obtain data, followed by a multimodal fusion model based on the self-attention mechanism to analyze the data. The study results show that the Kinetics400 dataset processed by this method achieves 94.2% in top-1 video classification accuracy. In socialist core values textual content classification, the model has strong predictive ability, with most of the categories being on the diagonal, indicating high accuracy. The conclusion points out that the method proposed in this paper effectively promotes the digital dissemination of Xi Jinping’s socialist thought with Chinese characteristics in the new era, and provides new perspectives and technical support for the theory informatization inheritance.
Triboelectric nanogenerators (TENGs) are attracting more and more attention since they can convert various mechanical energies into electric energy. However, in traditional TENGs for harvesting ...rotation energy, most of the contacts between two triboelectric materials are rigid-to-rigid contact with very large friction force, which limits their practical application. Here, we report an ultra-low-friction triboelectric–electromagnetic hybrid nanogenerator (NG). A freestanding mode TENG and a rotating electromagnetic generator (EMG) are integrated together to realize the complementary individual merits. The very soft and elastic contact between the two triboelectric materials in the TENG results into very small friction force. The influences of the type and the dimensions of the dielectric material on the performance of the TENG are studied systematically from theory to experiments. The results indicate that the open-circuit voltage and the transfer charge of the TENG increase with the rotation speed, which is very different from a traditional rotary TENG and is due to the increase of the contact area. The optimized TENG has a maximal load voltage of 65 V and maximal load power per unit mass of 438.9 mW/kg under a speed rotation of 1000 rpm, while the EMG has a maximal load voltage of 7 V and maximal load power density of 181 mW/kg. This demonstration shows that the hybrid NG can power a humidity/temperature sensor by converting wind energy into electric energy when the wind speed is 5.7 m/s. Meanwhile, it can be used as a self-powered wind speed sensor to detect wind speed as low as 3.5 m/s.
The epithelial cell-adhesion molecule (EpCAM) is hyperglycosylated in carcinoma tissue and the oncogenic function of EpCAM primarily depends on the degree of glycosylation. Inhibiting EpCAM ...glycosylation is expected to have an inhibitory effect on cancer. We analyzed the relationship of BAP31 with 84 kinds of tumor-associated antigens and found that BAP31 is positively correlated with the protein level of EpCAM. Triple mutations of EpCAM N76/111/198A, which are no longer modified by glycosylation, were constructed to determine whether BAP31 has an effect on the glycosylation of EpCAM. Plasmids containing different C-termini of BAP31 were constructed to identify the regions of BAP31 that affects EpCAM glycosylation. Antibodies against BAP31 (165–205) were screened from a human phage single-domain antibody library and the effect of the antibody (VH-F12) on EpCAM glycosylation and anticancer was investigated. BAP31 increases protein levels of EpCAM by promoting its glycosylation. The amino acid region from 165 to 205 in BAP31 plays an important role in regulating the glycosylation of EpCAM. The antibody VH-F12 significantly inhibited glycosylation of EpCAM which, subsequently, reduced the adhesion of gastric cancer cells, inducing cytotoxic autophagy, inhibiting the AKT-PI3K-mTOR signaling pathway, and, finally, resulting in proliferation inhibition both in vitro and in vivo. Finally, we clarified that BAP31 plays a key role in promoting N-glycosylation of EpCAM by affecting the Sec61 translocation channels. Altogether, these data implied that BAP31 regulates the N-glycosylation of EpCAM and may represent a potential therapeutic target for cancer therapy.
Harvesting ambient mechanical energy is a key technology for realizing self-powered electronics. With advantages of stability and durability, a liquid–solid-based triboelectric nanogenerator (TENG) ...has recently drawn much attention. However, the impacts of liquid properties on the TENG performance and the related working principle are still unclear. We assembled herein a U-tube TENG based on the liquid–solid mode and applied 11 liquids to study the effects of liquid properties on the TENG output performance. The results confirmed that the key factors influencing the output are polarity, dielectric constant, and affinity to fluorinated ethylene propylene (FEP). Among the 11 liquids, the pure water-based U-tube TENG exhibited the best output with an open-circuit voltage (

) of 81.7 V and a short-circuit current (

) of 0.26 μA for the shaking mode (0.5 Hz), which can further increase to 93.0 V and 0.48 μA, respectively, for the horizontal shifting mode (1.25 Hz). The U-tube TENG can be utilized as a self-powered concentration sensor (component concentration or metalion concentration) for an aqueous solution with an accuracy higher than 92%. . We successfully lighted up 60 LEDs and powered a temperature–humidity meter. Given its high output performance, the water-FEP U-tube TENG is a very promising approach for harvesting water-wave energy for self-powered electronics.
Increased stemness is causally linked to the development of chemoresistance in cancers. B-cell receptor-associated protein 31 (BAP31) has been identified to play an oncogenic role in many types of ...cancer. However, the role of BAP31 in 5-fluorouracil (5-FU) chemosensitivity and stemness of colorectal cancer (CRC) is still unknown. The aim of this study was to investigate the biological function and molecular mechanism of BAP31 in regulating 5-FU chemosensitivity and stemness. The correlation between BAP31 expression and 5-FU chemosensitivity was examined using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide and colony formation assays. Cancer stemness was analyzed using tumor sphere formation and Western blot assays. Western blot and immunofluorescence analyses of the knockdown cell lines were performed to explore the possible mechanisms. Finally, we investigated the function of BAP31 by constructing xenograft nude mouse models in vivo. In this study, we demonstrated that BAP31 was increased in CRC cells, and knockdown of BAP31 reduced the half maximal inhibitory concentration (IC50) of 5-FU, while this effect was reversed by overexpression of BAP31. In addition, knockdown of BAP31 substantially reduced the stemness of CRC cells in vitro. Consistently, knockdown of BAP31 significantly suppressed the tumorigenicity and stemness of CRC in vivo. The functional study further suggested that knockdown of BAP31 downregulated galectin-3 to inhibit the accumulation of β-catenin, which in turn repressed the transcription of downstream target genes (c-MYC, SOX2) of the Wnt/β-catenin signaling pathway. Knockdown of BAP31 reduced stemness by inhibiting the Wnt/β-catenin signaling pathway to increase 5-FU chemosensitivity. Importantly, intrabodies against BAP31 suppressed tumor growth and enhanced the antitumor effects of 5-FU in vivo. Therefore, using intrabodies against BAP31 may be a strategy for improving the antitumor effect of 5-FU in CRC.
Glucagon-like peptide-1 (GLP-1) is a novel antidiabetic agent used in clinical practice. Recently, it was reported to exert a renoprotective effect in the human kidney-2 cells and kidneys of diabetic ...rats, which was induced by one type of GLP-1 analog, liraglutide, in the presence of high glucose. However, most of the previous findings mainly focused on its indirect effect in inhibiting the advanced glycation end products. Here, besides glycemic control, we also demonstrated a stimulatory role of liraglutide in promoting autophagy and relieving oxidative stress in Zucker diabetic fatty rats. The renoprotective effect of liraglutide has been demonstrated by significantly decreasing urinary albumin (
< 0.01) and ameliorating renal pathological changes (
< 0.001) in vivo. Besides that, proliferation of human epithelial kidney cell line HKC-8 and human embryonic kidney-293 cells has increased after treating with exendin-4, a GLP-1 receptor (GLP-1R) agonist. Moreover, GLP-1 could positively improve the progression of autophagy in vivo and in vitro through regulating the autophagy-related protein light chain 3 and p62 via AMP-activated protein kinase (AMPK)-mammalian target of rapamycin (mTOR) signaling pathway. Simultaneously, it could reverse NF-erythroid 2-related factor 2 (NRF2) translocation into the nuclei and suppress oxidative stress. In terms of mechanism, the renoprotective effect of GLP-1 would be exerted via the GLP-1R-AMPK-mTOR-autophagy-reactive oxygen species signaling axis. The present study not only illustrates the renoprotective effect of GLP-1 in diabetic kidney disease (DKD) rats, but also for the first time elucidates the underlying mechanism that is independent of controlling glucose, which implies that GLP-1 might be a novel therapeutic strategy for the prevention and treatment of DKD.
In order to alleviate the relationship between mining and roadway, the 3204 working face and the 3206 roadway in Shanxi Taitou coal mine are taken as an example, and the width of mining and the ...support parameters of mining while reversing mining and excavation under dynamic pressure are optimized. The research includes field investigations, theoretical analysis, numerical simulation, and field tests. Based on the characteristics of roof fracture and the distribution of coal pillar stresses that determine the coal pillar is 18.7 m wide, the control scheme of mining while reversing mining and excavation was developed; the stress of coal pillar and the characteristics of roadway deformation and failure are summarized. By means of FLAC3D numerical simulation software, the influence of coal pillar widths and different mining positions on the stability of roadway surrounding rock are discussed. The asymmetric support structure of trapezoidal roadway is proposed as the core support, and the support scheme of dense bolts and anchor cables is proposed. The support of the 3206 return airway is composed of bolt, anchor cable, and anchor mesh, combined with M-shaped steel belt and steel beam. Through the research on the current situation of roadway support, the support scheme is optimized to make the 3206 return airway meet the production requirements, which provides a new breakthrough for roadway support under dynamic pressure in deep mines in China.
A parallel high-resolution underwater target detection network is proposed to address the problems of complex underwater scenes and limited target feature extraction capability. First, a ...high-resolution network (HRNet), a lighter high-resolution human posture estimation network, is used to improve the target feature representation and effectively reduce the semantic information lost in the image during sampling. Then, the attention module (A-CBAM) is improved to capture complex feature distributions by modeling the two-dimensional space in the activation function stage through the introduction of the flexible rectified linear units (FReLU) activation function to achieve pixel-level spatial information modeling capability. Feature enhancement in the spatial and channel dimensions is performed to improve understanding of fuzzy targets and small target objects and to better capture irregular and detailed object layouts. Finally, a receptive field augmentation module (RFAM) is constructed to obtain sufficient semantic information and rich detail information to further enhance the robustness and discrimination of features and improve the detection capability of the model for multi-scale underwater targets. Experimental results show that the method achieves 81.17%, 77.02%, and 82.9% mean average precision (mAP) on three publicly available datasets, specifically underwater robot professional contest (URPC2020, URPC2018) and pattern analysis, statistical modeling, and computational learning visual object classes (PASCAL VOC2007), respectively, demonstrating the effectiveness of the proposed network.
Ribosome stalls can result in ribosome collisions that elicit quality control responses, one function of which is to prevent ribosome frameshifting, an activity that entails the interaction of the ...conserved yeast protein Mbf1 with uS3 on colliding ribosomes. However, the full spectrum of factors that mediate frameshifting during ribosome collisions is unknown. To delineate such factors in the yeast
, we used genetic selections for mutants that affect frameshifting from a known ribosome stall site, CGA codon repeats. We show that the general translation elongation factor eEF3 and the integrated stress response (ISR) pathway components Gcn1 and Gcn20 modulate frameshifting in opposing manners. We found a mutant form of eEF3 that specifically suppressed frameshifting, but not translation inhibition by CGA codons. Thus, we infer that frameshifting at collided ribosomes requires eEF3, which facilitates tRNA-mRNA translocation and E-site tRNA release in yeast and other single cell organisms. In contrast, we found that removal of either Gcn1 or Gcn20, which bind collided ribosomes with Mbf1, increased frameshifting. Thus, we conclude that frameshifting is suppressed by Gcn1 and Gcn20, although these effects are not mediated primarily through activation of the ISR. Furthermore, we examined the relationship between eEF3-mediated frameshifting and other quality control mechanisms, finding that Mbf1 requires either Hel2 or Gcn1 to suppress frameshifting with wild-type eEF3. Thus, these results provide evidence of a direct link between translation elongation and frameshifting at collided ribosomes, as well as evidence that frameshifting is constrained by quality control mechanisms that act on collided ribosomes.
Reading frame maintenance is critical for accurate translation. We show that the conserved eukaryotic/archaeal protein Mbf1 acts with ribosomal proteins Rps3/uS3 and eukaryotic Asc1/RACK1 to prevent ...frameshifting at inhibitory CGA-CGA codon pairs in the yeast
. Mutations in
that allow frameshifting implicate eukaryotic conserved residues near the mRNA entry site. Mbf1 and Rps3 cooperate to maintain the reading frame of stalled ribosomes, while Asc1 also mediates distinct events that result in recruitment of the ribosome quality control complex and mRNA decay. Frameshifting occurs through a +1 shift with a CGA codon in the P site and involves competition between codons entering the A site, implying that the wobble interaction of the P site codon destabilizes translation elongation. Thus, eukaryotes have evolved unique mechanisms involving both a universally conserved ribosome component and two eukaryotic-specific proteins to maintain the reading frame at ribosome stalls.
