Neurofilament light chain (NfL) has emerged as a biomarker of neuroaxonal damage in several neurologic conditions. With increasing availability of fourth-generation immunoassays detecting NfL in ...blood, aspects of pre-analytical stability of this biomarker remain unanswered. This study investigated NfL concentrations in serum and plasma samples of 32 patients with neurological diagnoses using state of the art Simoa technology. We tested the effect of delayed freezing of up to 7 days and statistically determined stability and validity of measured concentrations. We found concentrations of NfL in serum and plasma to remain stable at room temperature when processing of samples is delayed up to 7 days (serum: mean absolute difference 0.9 pg/mL, intraindividual variation 1.2%; plasma: mean absolute difference 0.5 pg/mL, intraindividual variation 1.3%). Consistency of these results was nearly perfect for serum and excellent for plasma (intraclass correlation coefficients 0.99 and 0.94, respectively). In conclusion, the soluble serum and plasma NfL concentration remains stable when unprocessed blood samples are stored up to 7 days at room temperature. This information is essential for ensuring reliable study protocols, for example, when shipment of fresh samples is needed.
Abstract
The improvement of experimental models for disorders requires a constant approximation towards the dysregulated tissue. In psychiatry, where an impairment of neuronal structure and function ...is assumed to play a major role in disease mechanisms and symptom development, this approximation is an ongoing process implicating various fields. These include genetic, animal, and post-mortem studies. To test hypotheses generated through these studies, in vitro models using non-neuronal cells such as fibroblasts and lymphocytes have been developed. For brain network disorders, cells with neuronal signatures would, however, represent a more adequate tissue. Considering the limited accessibility of brain tissue, research has thus turned towards neurons generated from induced pluripotent stem cells as well as directly induced neurons, cerebral organoids, and olfactory neuroepithelium. Regarding the increasing importance and amount of research using these neuronal cells, this review aims to provide an overview of all these models to make sense of the current literature. The development of each model system and its use as a model for the various psychiatric disorder categories will be laid out. Also, advantages and limitations of each model will be discussed, including a reflection on implications and future perspectives.

With a heterogeneous uptake and a high test-retest reliability, this radioligand offers promise to deepen our understanding of the GluN2B-containing NMDAR in the pathophysiology and treatment of neuropsychiatric disease such as Alzheimer disease and major depression. Additionally, it could help in the selection of appropriate doses of GluN2B-targeting drugs.
In August 1998, severe forest fires occurred in many parts of Canada, especially in the Northwest Territories. In the week from August 5 to 11, more than 1000 different fires burned >1 × 106 ha of ...boreal forest, the highest 1‐week sum ever reported throughout the 1990s. In this study we can unambigously show for the first time that these fires caused pronounced large‐scale haze layers above Europe and that they influenced concentrations of carbon monoxide and other trace gases at the surface station Mace Head in Ireland over a period of weeks. Transport took place across several thousands of kilometers. An example of such an event, in which a pronounced aerosol layer was observed at an altitude of 3–6 km over Germany during August 1998, is investigated in detail. Backward trajectories ending at the measured aerosol layer are calculated and shown to have their origin in the forest fire region. Simulations with a particle dispersion model reveal how a substantial amount of forest fire emissions was transported across the Atlantic. The resulting aerosol lamina over Europe is captured well by the model. In addition, the model demonstrates that the forest fire emissions polluted large regions over Europe during the second half of August 1998. Surface measurements at Mace Head are compared to the model results for an anthropogenic and a forest fire carbon monoxide tracer, respectively. While wet deposition removed considerable amounts of aerosol during its transport, forest fire carbon monoxide reached Europe in copious amounts. It is estimated that during August 1998, 32%, 10%, and 58% of the carbon monoxide enhancement over the background level at Mace Head were caused by European and North American anthropogenic emissions and forest fire emissions, respectively.

Testing object oriented software is even for human beings a challenging task. Automating it is therefore very helpful for developers but by no means trivial. In this paper we present AIana, an ...approach for automatically generating complex objects used as test input data that satisfy a given precondition in terms of Design by Contract™ specification. AIana transforms the existing Design by Contract™ specification of the parameter type and the precondition of the method under test to PDDL (plan domain description language). Based on it, existing AI planners can be used to create a plan, i.e., a method sequence that transforms the object to the goal state. The goal state is given by the precondition of the method under test. AIana is evaluated on two case studies: a student developed stack based calculator, and a real-world event based application developed by our industry partner. AIana outperforms a random approach significantly in terms of methods tested and line coverage on both case studies.
