Here, ...we report that activation of TRPV4 induced massive exocytosis in both melanoma A375 cell and heterologous expression systems. We show here that, upon application of TRPV4-specific agonists, prominent vesicle priming from endoplasmic reticulum (ER) was observed, followed by morphological changes of mitochondrial crista may lead to cell ferroptosis. We further identified interactions between TRPV4 and folding/vesicle trafficking proteins, which were triggered by calcium entry through activated TRPV4. This interplay, in turn, enhanced TRPV4-mediated activation of folding and vesicle trafficking proteins to promote exocytosis. Our study revealed a signaling mechanism underlying stimulus-triggered exocytosis in melanoma and highlighted the role of cellular sensor TRPV4 ion channel in mediating ferroptosis.
The present study aimed to assess the correlation of fibroblast growth factor (FGF)-23 expression with clinical characteristics, then further explore its value in predicting 2-year in-stent ...restenosis (ISR) risk in coronary heart disease (CHD) patients underwent percutaneous coronary intervention (PCI) with drug-eluting stent (DES).
In this prospective, single-center, observational study, totally 214 CHD patients treated by PCI with DES were consecutively recruited, and peripheral blood samples were collected prior to PCI with DES for serum samples isolation. Following, FGF-23 level in the serum samples was detected via enzyme linked-immuno-sorbent Assay. The follow-up coronary angiography was performed at 1 year and 2 years after PCI or if suspected ISR symptoms occurred.
FGF-23 was positively correlated with fasting blood-glucose, insulin resistance, serum creatinine, serum uric acid, LDL-C, high-sensitivity C-reactive protein, cardiac troponin I and N-terminal-proB-type natriuretic peptide, while was negatively associated with HDL-C and left ventricular ejection fraction (all P < 0.01). Furthermore, FGF-23 was positively correlated with hypercholesteremia, hyperuricemia and family history of CAD (all P < 0.05). However, it did not correlate with other chronic complications, biochemical indexes, lesion features or PCI parameters (all P > 0.05). Moreover, FGF-23 level was higher in 2-year ISR patients (n = 38) compared to 2-year non-ISR patients (n = 176) (P < 0.001), and receiver operating characteristic curve indicated that FGF-23 was of good value in predicting 2-year ISR risk (AUC 0.828, 95% CI 0.761-0.896).
FGF-23 correlates with endocrine and metabolism dysregulation, worse cardiac and renal function, inflammation level, stenosis degree of target lesion, and serves as an independent risk factor for 2-year ISR risk in CHD patients underwent PCI with DES.
With the massive application of IoT and sensor technologies, the study of lightweight ciphers has become an important research topic. In this paper, an effective lightweight LZUC (lightweight Zu ...Chongzhi) cipher based on chaotic system is proposed to improve the traditional ZUC algorithm. In this method, a further algorithm is designed for the process of integrating chaos into the lightweighting of ZUC. For the first time, this design introduces the logistic chaotic system into both the LFSR (linear feedback shift register) and nonlinear F-function of the cryptographic algorithm. The improved LZUC algorithm not only achieves a certain effect in lightweighting, but also has good statistical properties and security of the output sequence. To verify the performance of the LZUC cipher, we performed NIST statistical tests and information entropy analysis on its output key streams and discussed the typical attacks on the algorithm’s resistance to weak key analysis, guess–determination analysis, time–stored data trade-off analysis, and algebraic analysis. In addition, we completed the design of an image security system using the LZUC cipher. Histogram analysis and correlation analysis are used to analyze both plaintext and ciphertext data. At the end of the article, the plaintext and ciphertext images displayed by LCD can be further visualized to verify the encryption effectiveness of the LZUC cipher.
The Awulale Iron Metallogenic Belt (AIMB) has developed many medium–large iron deposits, of which the Zhibo iron deposit is selected as the research object in this paper. The Zhibo deposit’s ore ...primarily consists of magnetite as the main mineral, accompanied by extensive epidotization. The mineral assemblage includes diopside, albite, actinolite, epidote, chlorite, K-feldspar, quartz, calcite, chalcopyrite, and pyrite. Magnetite is classified into two groups based on sulfide content and mineral assemblage (MagI for sulfide-free and MagII for sulfide-rich ores). Two-stage mineralization of magnetite has been identified based on mineral assemblages and paragenesis, including the magmatic stage MagI and hydrothermal stage MagII. Mag I shows inhomogeneous backscattered electron (BSE) textures and consists of BSE-light and -dark domains (Mag I-L and MagI-D). Seven subtypes of magnetite have been recognized in this deposit. MagI-L and MagI-D have formed in the magmatic stage and show BSE images in light and dark colors, respectively. MagI-L is anhedral to subhedral and is inclusion-free. MagI-D has mainly replaced MagI-L along fractures and contains inclusions and pores. MagII has formed in the hydrothermal stage and is characterized by coupled dissolution–reprecipitation (DRP) textures. It can be divided into five sub-generations, that is, MagII-1, MagII-2, MagII-3, MagII-L, and MagII-D. MagII-1, MagII-2, and MagII-3 comprise the core–mantle–rim texture, while MagII-L and MagII-D comprise the core–rim texture. MagII-1 is BSE-light and is enriched with inclusions and pores. MagII-2 has partly replaced MagII-1 and exhibits oscillatory zoning under BSE imaging. It also contains inclusions. BSE-light MagII-3 occurs as overgrowth along MagII-2 margins and is inclusion-free. MagI magnetite is enriched with V, Cr, and Ni, whereas MagII is enriched with W, Ta, Nb, Sr, Sb, Sn, Y, Zr, Mg, Al, and Ti, indicating a decreased temperature of magnetite formation. MagI-L crystallizes from the original magma, while MagI-D is formed from the residual magma enriched with incompatible elements. MagII crystallizes from later multiple hydrothermal activities through the dissolution of early magnetite and the re-precipitation of later magnetite or from MagI-D which has later undergone a hydrothermal overprinting process. According to the texture and chemical composition of magnetite from the Zhibo deposit, we suggest that the Zhibo iron deposit was formed from the initial magmatic origin and then underwent a hydrothermal overprinting process.
Suicidal ideation is a critical early stage in the progression towards suicidal be havior. Prior research has established links between sleep quality, impulsivity, and suicidal tendencies, yet the ...interaction among these factors has been less explored. This study aims to explore the mediating role of impulsivity in the relationship between sleep quality and suicidal ideation in adolescents.
Employing a cross-sectional study design, 6,974 questionnaires were distributed,including the Socio-demographic Characteristics Questionnaire, Barratt Impulsiveness Scale, the Positive and Negative Suicide Ideation Inventory,and the Pittsburgh Sleep Quality Index Scale. The participants were high school and middle school students from 33 schools in northeastern Sichuan, China, selected through random cluster sampling.
Of these 6,786 questionnaires were analyzed. The participant distribution included 47.2% male and 52.8% female students, with 68.3% from junior schools and 31.7% from senior schools. The prevalence of suicidal ideation was found to be 13.6%. The analysis, which involved correlation analysis and the construction of a structural equation model, revealed that sleep quality had a significant positive effect on impulsivity (β:0.289,p < 0.05), and impulsivity, in turn, had a positive impact on suicidal ideation (β:0.355,p < 0.05).Moreover, sleep quality was directly linked to suicidal ideation (β:0.208,p < 0.05). Thus, sleep quality affects suicidal ideation both directly and indirectly through impulsivity.
The results of this study suggest that both sleep quality and impulsivity are significant direct influencers of suicidal ideation among adolescents in the region studied, with impulsivity also playing an indirect role in the relationship between sleep quality and suicidal ideation.
Measurement of laser intensity inside a femtosecond laser filament is a challenging task. In this work, we suggest a simple way to characterize laser peak intensity inside the filament in air. It is ...based on the signal ratio measurement of two nitrogen fluorescence lines, namely, 391 nm and 337 nm. Because of distinct excitation mechanisms, the signals of the two fluorescence lines increase with the laser intensity at different orders of nonlinearity. An empirical formula has been deduced according to which laser peak intensity could be simply determined by the fluorescence ratio.
Skyrise greenery, including green roofs and vertical gardens, has emerged as an indispensable tool for sustainable urban planning with multiple ecological and economic benefits. A bibliometric ...analysis was used to provide a systematic review of the functions associated with skyrise greenery in urban landscapes. Key research tools, including the “Bibliometrix” R package and “CiteSpace” 6.2 R4, highlight the depth and breadth of the literature covering skyrise greenery. In 2000–2022, a total of 1474 original journal articles were retrieved. Over this period, there was an exponential increase in the number of publications, reflecting both enhanced knowledge and increasing concerns regarding climate change, the urban heat island, and urbanization. Of the total, ~58% of the articles originated from China, followed by the USA, Italy, Australia, and Canada. The research themes, such as urban heat islands, carbon sequestration, hydrology, and air quality, have been identified as the frontier in this fields. Furthermore, researchers from developed countries contributed the most publications to this domain, while developing countries, such as China, play an increasing role in the design and performance evaluation of vertical greenery. Key benefits identified in vertical green systems (e.g., green roofs and walls) include thermal regulation, sustainable water management, air-quality improvement, noise reduction, and biodiversity enhancement. In addition, several potential future research prospectives are highlighted. This review provides a comprehensive insight into exploring the pivotal role of skyrise greenery in shaping sustainable, resilient urban futures, coupled with sustainable urban planning.
Sperm detection performance is particularly critical for sperm motility tracking. However, there are a large number of non-sperm objects, sperm occlusion and poorly detailed texture features in semen ...images, which directly affect the accuracy of sperm detection. To solve the problem of false detection and missed detection in sperm detection, a multi-sperm target detection model, Yolov5s-SA, with an SA attention mechanism is proposed based on the YOLOv5s algorithm. Firstly, a depthwise, separable convolution structure is used to replace the partial convolution of the backbone network, which can ensure stable precision and reduce the number of model parameters. Secondly, a new multi-scale feature fusion module is designed to enhance the perception of feature information to supplement the positional information and high-resolution of the deep feature map. Finally, the SA attention mechanism is integrated into the neck network before the output of the feature map to enhance the correlation between the feature map channels and improve the fine-grained feature fusion ability of YOLOv5s. Experimental results show that compared with various YOLO algorithms, the proposed algorithm improves the detection accuracy and speed to a certain extent. Compared with the YOLOv3, YOLOv3-spp, YOLOv5s and YOLOv5m models, the average accuracy increases by 18.1%, 15.2%, 6.9% and 1.9%, respectively. It can effectively reduce the missed detection of occluded sperm and achieve lightweight and efficient multi-sperm target detection.
Abstract
Background
Among rural Chinese patients with non-communicable diseases (NCDs), low socioeconomic status increases the risk of developing NCDs and associated financial burdens in paying for ...medicines and treatments. Despite the chronic disease medicine reimbursement policy of the local government in Nantong City, China, various barriers prevent patients from registering for and benefitting from the policy. This study aims to develop a behavior science-based intervention program for promoting the adoption of the policy and to evaluate the effectiveness of the program compared with usual practices.
Methods
Barriers and opportunities affecting stakeholders in adopting the policy were identified through contextual research and summarized through behavior mapping. The intervention is designed to target these barriers and opportunities through behavior science theories and will be evaluated through a 6-month cluster randomized controlled trial in Tongzhou District, Nantong, China. A total of 30 villages from two townships are randomized in a 1:1 ratio to either the intervention or the control arm (usual practices). Village doctors in the intervention arm (1) receive systematic training on policy details, registration procedures, and intervention protocol, (2) promote the policy and encourage registration, (3) follow up with patients in the first, third, and sixth months after the intervention, and (4) receive financial incentives based on performance. The primary outcome is policy registration rate and the secondary outcomes include the number of patients registering for the policy, medical costs saved, frequency of village doctor visits, and health measures such as blood pressure and glucose levels.
Discussion
This study is one of very few that aims to promote adoption of NCDs outpatient medication reimbursement policies, and the first study to evaluate the impact of these policies on patients’ financial and physical wellbeing in China. The simple, feasible, and scalable intervention is designed based on the theories of behavior science and is applicable to similar low-income regions nationwide where outpatient medical costs remain a financial burden for patients.
