Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) is the causative agent of the pandemic disease COVID-19, which is so far without efficacious treatment. The discovery of therapy reagents ...for treating COVID-19 are urgently needed, and the structures of the potential drug-target proteins in the viral life cycle are particularly important. SARS-CoV-2, a member of the Orthocoronavirinae subfamily containing the largest RNA genome, encodes 29 proteins including nonstructural, structural and accessory proteins which are involved in viral adsorption, entry and uncoating, nucleic acid replication and transcription, assembly and release, etc. These proteins individually act as a partner of the replication machinery or involved in forming the complexes with host cellular factors to participate in the essential physiological activities. This review summarizes the representative structures and typically potential therapy agents that target SARS-CoV-2 or some critical proteins for viral pathogenesis, providing insights into the mechanisms underlying viral infection, prevention of infection, and treatment. Indeed, these studies open the door for COVID therapies, leading to ways to prevent and treat COVID-19, especially, treatment of the disease caused by the viral variants are imperative.

Although numerous studies have investigated the relationships between hydrological drought (HD) and meteorological drought (MD) in different regions, few studies have examined the influence of reservoir operations during drought periods on such relationships, particularly at multiple time scales. This study presents a useful framework to examine the influences of reservoir operation rules during drought periods on the multi-scale correlations between HD and MD. Two standardized drought indices (Standardized Streamflow Index (SSI) and Standardized Precipitation Index (SPI)) with different timescales (1, 3, 6, 12, and 24 months) were used to compare the pre-reservoir and post-reservoir periods. Also, a simple algorithm was proposed to examine the difference value (Dvalue) between the inflow and outflow under different reservoir operation rules and it was used to evaluate the impacts of the reservoir operation rules during the drought on the correlation of the HD and MD. Monthly streamflow and precipitation records from 1960 to 2015 and monthly inflow and outflow records of a large reservoir (Xinfengjiang reservoir) during 1974–2009 in the basin of the Dongjiang River in south China were used to demonstrate the applicability of the analysis methodology. The results indicate that (1) the reservoir operations during the post-reservoir period (1974–2015) exerted a significant influence (positive impact) on the evolution of the short-term (1 and 3 months) HD by reducing the drought duration and alleviating its magnitude and altered the linear correlation between the HD and MD compared to the pre-reservoir period (1960–1972). However, the effects on the long-term (e.g., 12 and 24 months) HD and its correlation with the MD were not apparent. (2) The impacts of the reservoir operation under the drought condition on the relationship between the HD and MD were clearly different from those under normal rules. This study provides additional information for policy-makers to regarding reservoir management during a drought.
Volatile methyl siloxanes (VMS), which are considered to be the most troublesome impurities in current biogas-cleaning technologies, need to be removed. In this study, we fabricated a series of ...Fe3O4–urea-modified reduced graphene-oxide aerogels (Fe3O4–urea–rGOAs) by using industrial-grade graphene oxide as the raw material. A fixed-bed dynamic adsorption setup was built, and the adsorption properties of the Fe3O4–urea–rGOAs for hexamethyldisiloxane (L2, as a VMS model pollutant) were studied. The properties of the as-prepared samples were investigated by employing various characterization techniques (SEM, TEM, FTIR, XRD, Raman spectroscopy, and N2 adsorption/desorption techniques). The results showed that the Fe3O4–urea–rGOA–0.4 had a high specific surface area (188 m2 g−1), large porous texture (0.77 cm3 g−1), and the theoretical maximum adsorption capacity for L2 (146.5 mg g−1). The adsorption capacity considerably increased with a decrease in the bed temperature of the adsorbents, as well as with an increase in the inlet concentration of L2. More importantly, the spent Fe3O4–urea–rGOA adsorbent could be readily regenerated and showed an excellent adsorption performance. Thus, the proposed Fe3O4–urea–rGOAs are promising adsorbents for removing the VMS in biogas.
Volatile methyl siloxanes (VMS) must be removed because the formation of silica in the combustion process seriously affects the resource utilization of biogas. Herein, a series of APTMS ...((3–aminopropyl)trimethoxysilane)-modified activated porous carbon (APC) adsorbents (named APTMS@APC) were prepared for VMS efficient removal. The as-prepared adsorbents were characterized using SEM, FTIR, Raman, X-ray diffraction analyses, and N2 adsorption/desorption. The results showed that the surface modification with APTMS enhanced the hydrophobicity of APC with the water contact angle increasing from 74.3° (hydrophilic) to 127.1° (hydrophobic), and meanwhile improved its texture properties with the SBET increasing from 981 to 1274 m2 g−1. The maximum breakthrough adsorption capacity of APTMS@APC for hexamethyldisiloxane (L2, model pollutant) was 360.1 mg g−1. Effects of an inlet L2 concentration (31.04–83.82 mg L−1) and a bed temperature (0–50 °C) on the removal of L2 were investigated. Meanwhile, after five adsorption–desorption cycles, the APTMS@APC demonstrated a superior cycling performance. This indicated that the hydrophobic APTMS@APC has a great significance to remove VMS.
In this study, an inland reservoir water quality parameters’ inversion model was developed using a back propagation (BP) neural network to conduct reservoir eutrophication evaluation, according to ...multi-temporal remote sensing images and field observations. The inversion model based on the BP neural network (the BP inversion model) was applied to a large inland reservoir in Jiangmen city, South China, according to the field observations of five water quality parameters, namely, Chlorophyl-a (Chl-a), Secchi Depth (SD), total phosphorus (TP), total nitrogen (TN), and Permanganate of Chemical Oxygen Demand (CODMn), and twelve periods of Landsat8 satellite remote sensing images. The reservoir eutrophication was evaluated. The accuracy of the BP inversion model for each water parameter was compared with that of the linear inversion model, and the BP inversion models of two parameters (i.e., Chl-a and CODMn) with larger fluctuation range were superior to the two multiple linear inversion models due to the ability of improving the generalization of the BP neural network. The Dashahe Reservoir was basically in the state of mesotrophication and light eutrophication. The area of light eutrophication accounted for larger proportions in spring and autumn, and the reservoir inflow was the main source of nutrient salts.
A novel newsvendor model-based framework for regional industrial water resources allocation that considers uncertainties in water supply and demand was proposed in this study. This framework ...generates optimal water allocation schemes while minimizing total costs. The total cost of water allocation consists of the allocated water cost, the opportunity loss for not meeting water demand, and the loss of the penalty for exceeding water demand. The uncertainties in water demand and supply are expressed by cumulative distribution functions. The optimal water allocation for each water use sector is determined by the water price, the unit loss of the penalty and opportunity loss, and the cumulative distribution functions. The model was then applied to monthly water allocation for domestic, industrial, and agricultural water use in two counties of Huizhou City, China, whose water supply mainly depends on Baipenzhu Reservoir. The water demand for each water use sector and the monthly reservoir inflow showed good fits with the uniform and P-III distributions, respectively. The water demand satisfied ratio for each water use sector was stable and increased for the optimal water allocation scheme from the newsvendor model-based framework, and the costs were lower compared with the actual water allocation scheme. The novel framework is characterized by less severe water shortages, lower costs, and greater similarity to actual water use compared with the traditional deterministic multi-objective analysis model, and demonstrates strong robustness in the advantages of lower released surplus water and higher water demand satisfied ratio. This novel framework yields the optimal water allocation for each water use sector by integrating the properties of the market (i.e., determining the opportunity loss for not meeting water demand) with the government (i.e., determining the water price and the loss of the penalty for exceeding water demand) under the strictest water resources management systems.
Methanobactins (Mbns) are a family of copper-binding peptides involved in copper uptake by methanotrophs, and are potential therapeutic agents for treating diseases characterized by disordered copper ...accumulation. Mbns are produced via modification of MbnA precursor peptides at cysteine residues catalyzed by the core biosynthetic machinery containing MbnB, an iron-dependent enzyme, and MbnC. However, mechanistic details underlying the catalysis of the MbnBC holoenzyme remain unclear. Here, we present crystal structures of MbnABC complexes from two distinct species, revealing that the leader peptide of the substrate MbnA binds MbnC for recruitment of the MbnBC holoenzyme, while the core peptide of MbnA resides in the catalytic cavity created by the MbnB-MbnC interaction which harbors a unique tri-iron cluster. Ligation of the substrate sulfhydryl group to the tri-iron center achieves a dioxygen-dependent reaction for oxazolone-thioamide installation. Structural analysis of the MbnABC complexes together with functional investigation of MbnB variants identified a conserved catalytic aspartate residue as a general base required for MbnBC-mediated MbnA modification. Together, our study reveals the similar architecture and function of MbnBC complexes from different species, demonstrating an evolutionarily conserved catalytic mechanism of the MbnBC holoenzymes.
To expand the applications of graphene-based materials to biogas purification, a series of reduced graphene oxide aerogels (rGOAs) were prepared from industrial grade graphene oxide using a simple ...hydrothermal method. The influences of the hydrothermal preparation temperature on the textural properties, hydrophobicity and physisorption behavior of the rGOAs were investigated using a range of physical and spectroscopic techniques. The results showed that the rGOAs had a macro-porous three-dimensional network structure. Raising the hydrothermal treatment temperature reduced the number of oxygen-containing groups, whereas the specific surface area (
), micropore volume (
) and water contact angle values of the rGOAs all increased. The dynamic adsorption properties of the rGOAs towards hexamethyldisiloxane (L2) increased with increasing hydrothermal treatment temperature and the breakthrough adsorption capacity showed a significant linear association with

and contact angle. There was a significant negative association between the breakthrough time and inlet concentration of L2, and the relationship could be reliably predicted with a simple empirical formula. L2 adsorption also increased with decreasing bed temperature. Saturated rGOAs were readily regenerated by a brief heat-treatment at 100 °C. This study has demonstrated the potential of novel rGOA for applications using adsorbents to remove siloxanes from biogas.
Sentiment analysis is an important topic in natural language processing (NLP) and text classifications. The existing algorithms of lexicon-based sentiment classification can deal with small corpus ...datasets or simple semantic texts. With the growth of text corpus data, word embedding methods have been gaining more attention. However, the single static word vector obtained by these methods can not accurately express the semantic information of the text. To optimize the word vector, we propose a text sentiment classification model using the double word embedding methods (DWE), which combines two models, GloVe and Word2vec, to represent the text to form a combinatory input of dual channels of convolution neural network (CNN). Based on the word vector fine-tuning strategy, the initial word vector is continuously learned and adjusted to find the CNN sentiment classification model with better combination input than a single vector representation. Experiment results show that DWE can effectively improve the accuracy of sentiment classification, which reaches 94.8%.
Abstract
Since 2011, China has implemented its most stringent water management system to effectively protect water resources and guarantee socioeconomic development. More basin-scale water division ...schemes have been developed to act as references for basin-scale water resources management. Water dispatching during dry periods is an effective way to guarantee the water supply for the river basin, and is also an important component of basin-scale water resources management. Given this, the present study proposes a framework for the water dispatching of river basins during dry periods under the most stringent water management system in China. This framework mainly consists of the analysis and forecasting of rainfall and inflow, the dispatching requirements for the main water users, major reservoirs, and sections, as well as safeguard measures. The Jian River Basin in South China is presented as a case study. The total discharge of the Gaozhou Reservoir in 2017 was 25 million m3 more than the target discharge specified in the water dispatching scheme, and the total water storage utilization ratio during the dispatch period was 4.7% higher than the target utilization ratio. These factors demonstrate the effectiveness and applicability of the proposed framework.
