Adrenomedullin (ADM) exerts anti-oxidant, anti-inflammatory and anti-apoptotic effects in Leydig cells. However, the role and mechanism of ADM in the pyroptosis of Leydig cells are poorly understood. ...This study first showed the protective effects of ADM on the pyroptosis and biological functions of Leydig cells exposed to lipopolysaccharide (LPS) by promoting autophagy. Primary rat Leydig cells were treated with various concentrations of LPS and ADM, together with or without N-acetyl-L-cysteine (NAC) or 3-methyladenine (3-MA). Cell proliferation was detected through CCK-8 and BrdU incorporation assays, and ROS level was measured with the DCFDA assay. Real-time PCR, western blot, immunofluorescence, transmission electron microscopy, TUNEL and flow cytometry were performed to examine ADM's effect on the pyroptosis, autophagy and steroidogenic enzymes of Leydig cells and AMPK/mTOR signalling. Like NAC, ADM dose-dependently reduced LPS-induced cytotoxicity and ROS overproduction. ADM also dose-dependently ameliorated LPS-induced pyroptosis by reversing the increased expression of NLRP3, ASC, caspase-1, IL-1β, IL-18, GSDMD, caspase-3, caspase-7, TUNEL-positive and PI and active caspase-1 double-stained positive rate, DNA fragmentation and LDH concentration, which could be rescued via co-incubation with 3-MA. ADM dose-dependently increased autophagy in LPS-induced Leydig cells, as confirmed by the increased expression of LC3-I/II, Beclin-1 and ATG-5; decreased expression of p62 and autophagosomes formation; and increased LC3-II/LC3-I ratio. However, co-treatment with 3-MA evidently decreased autophagy. Furthermore, ADM dose-dependently rescued the expression of steroidogenic enzymes, including StAR, P450scc, 3β-HSD and CYP17, and testosterone production in LPS-induced Leydig cells. Like rapamycin, ADM dose-dependently enhanced AMPK phosphorylation but reduced mTOR phosphorylation in LPS-induced Leydig cells, which could be rescued via co-incubation with 3-MA. In addition, pyroptosis was further decreased, and autophagy was further promoted in LPS-induced Leydig cells upon co-treatment with ADM and rapamycin. ADM may protect the steroidogenic functions of Leydig cells against pyroptosis by activating autophagy via the ROS-AMPK-mTOR axis.
Ammonia (NH3) emissions from fertilized soils to the atmosphere and the subsequent deposition to land surface exert adverse effects on biogeochemical nitrogen (N) cycling. The region‐ and ...crop‐specific emission factors (EFs) of N fertilizer for NH3 are poorly developed and therefore the global estimate of soil NH3 emissions from agricultural N fertilizer application is constrained. Here we quantified the region‐ and crop‐specific NH3 EFs of N fertilizer by compiling data from 324 worldwide manipulative studies and focused to map the global soil NH3 emissions from agricultural N fertilizer application. Globally, the NH3 EFs averaged 12.56% and 14.12% for synthetic N fertilizer and manure, respectively. Regionally, south‐eastern Asia had the highest NH3 EFs of synthetic N fertilizer (19.48%) and Europe had the lowest (6%), which might have been associated with the regional discrepancy in the form and rate of N fertilizer use and management practices in agricultural production. Global agricultural NH3 emissions from the use of synthetic N fertilizer and manure in 2014 were estimated to be 12.32 and 3.79 Tg N/year, respectively. China (4.20 Tg N/year) followed by India (2.37 Tg N/year) and America (1.05 Tg N/year) together contributed to over 60% of the total global agricultural NH3 emissions from the use of synthetic N fertilizer. For crop‐specific emissions, the NH3 EFs averaged 11.13%–13.95% for the three main staple crops (i.e., maize, wheat, and rice), together accounting for 72% of synthetic N fertilizer‐induced NH3 emissions from croplands in the world and 70% in China. The region‐ and crop‐specific NH3 EFs of N fertilizer established in this study offer references to update the default EF in the IPCC Tier 1 guideline. This work also provides an insight into the spatial variation of soil‐derived NH3 emissions from the use of synthetic N fertilizer in agriculture at the global and regional scales.

The tumor microenvironment (TME) is an essential intrinsic portion of hepatocellular carcinoma (HCC) for the regulation of its origination, development, invasion, and metastasis. As emerging ...components of the tumor-host interaction, exosomes are increasingly recognized as professional carriers of information in TME and as pivotal molecular entities involved in tumorigenic microenvironment setup. However, much remains unknown about the role of the exosome communication system within TME in the development and progression of HCC. In this review, we focus on the roles and probable mechanisms of TME in HCC and show the exosome-based immune regulation in TME to promote HCC. Multiple processes are involved in HCC, including tumor survival, growth, angiogenesis, invasion, and metastasis. We also discuss the specific roles of exosomes in HCC processes by molding hospitable TME for HCC, such as providing energy, transmitting protumor signals, and evading inhibitory signals. In addition, exosomes induce angiogenesis by changing the biological characteristics of endothelial cells and directly regulating proangiogenic and propermeability factors. Furthermore, exosomes may lead to HCC metastatic invasion by epithelial-mesenchymal transformation, extracellular matrix degradation, and vascular leakage. Finally, we summarize the therapeutic usage of exosomes in the HCC microenvironment and attempt to provide a theoretical reference for modern antitumor agents designed to target these mechanisms.

Experimental Approach
D1 receptor knockout (D1R−/−) mice, adeno‐associated viral 9‐short hairpin RNA carrying D2 receptor (AAV9‐shD2R)‐treated mice, 6‐hydroxydopamine treated (6‐OHDA) rats and primary enteric glial cells (EGCs) culture were used. Incubation fluid from colonic submucosal plexus and longitudinal muscle myenteric plexus were collected for GDNF and ACh measurements.
Key Results
D2 receptor‐immunoreactivity (IR), but not D1 receptor‐IR, was observed on EGCs. Both D1 receptor‐IR and D2 receptor‐IR were co‐localized on cholinergic neurons. Low concentrations of dopamine induced colonic GDNF secretion in a concentration‐dependent manner, which was mimicked by the D1 receptor agonist SKF38393, inhibited by TTX and atropine and eliminated in D1R−/− mice. SKF38393‐induced colonic ACh release was absent in D1R−/− mice. High concentrations of dopamine suppressed colonic GDNF secretion, which was mimicked by the D2 receptor agonist quinpirole, and absent in AAV‐shD2R‐treated mice. Quinpirole decreased GDNF secretion by reducing intracellular Ca2+ levels in primary cultured EGCs. Carbachol ( ACh analogue) promoted the release of GDNF. Quinpirole inhibited colonic ACh release, which was eliminated in the AAV9‐shD2R‐treated mice. 6‐OHDA treated rats with low ACh and high dopamine content showed decreased GDNF content and increased mucosal permeability in the colon.
Conclusion and Implications
Low concentrations of dopamine promote colonic GDNF secretion via D1 receptors on cholinergic neurons, whereas high concentrations of dopamine inhibit GDNF secretion via D2 receptors on EGCs and/or cholinergic neurons.
The continuous increase of nitrous oxide (N ₂O) abundance in the atmosphere is a global concern. Multiple pathways of N ₂O production occur in soil, but their significance and dependence on oxygen (O ...₂) availability and nitrogen (N) fertilizer source are poorly understood. We examined N ₂O and nitric oxide (NO) production under 21%, 3%, 1%, 0.5%, and 0% (vol/vol) O ₂ concentrations following urea or ammonium sulfate (NH ₄) ₂SO ₄ additions in loam, clay loam, and sandy loam soils that also contained ample nitrate. The contribution of the ammonia (NH ₃) oxidation pathways (nitrifier nitrification, nitrifier denitrification, and nitrification-coupled denitrification) and heterotrophic denitrification (HD) to N ₂O production was determined in 36-h incubations in microcosms by ¹⁵N- ¹⁸O isotope and NH ₃ oxidation inhibition (by 0.01% acetylene) methods. Nitrous oxide and NO production via NH ₃ oxidation pathways increased as O ₂ concentrations decreased from 21% to 0.5%. At low (0.5% and 3%) O ₂ concentrations, nitrifier denitrification contributed between 34% and 66%, and HD between 34% and 50% of total N ₂O production. Heterotrophic denitrification was responsible for all N ₂O production at 0% O ₂. Nitrifier denitrification was the main source of N ₂O production from ammonical fertilizer under low O ₂ concentrations with urea producing more N ₂O than (NH ₄) ₂SO ₄ additions. These findings challenge established thought attributing N ₂O emissions from soils with high water content to HD due to presumably low O ₂ availability. Our results imply that management practices that increase soil aeration, e.g., reducing compaction and enhancing soil structure, together with careful selection of fertilizer sources and/or nitrification inhibitors, could decrease N ₂O production in agricultural soils.
Thirty new, highly oxygenated and stereogenic 14‐membered macrocyclic diterpenoids, papyrifuranols A‐Z (1–26) and AA‐AD (27–30), and eight known analogs have been isolated from Boswellia papyrifera ...resins. All the structures were characterized by detailed spectral analyses, quantum calculations, X‐ray diffraction, and modified Mosher's methods. Notably, six previously reported structures were revised. Our study points out misleading factors of macrocyclic cembranoid (CB) representation in the past seven decades by analyzing of 25 X‐ray structures, lending a hand for the innately challenging structure identification of such flexible macrocyclic CBs and avoiding following the tracks of an overturned cart during future structure characterization and total synthesis. Biosynthetic conversions of all the isolates are proposed, and wound healing bioassays reveal that papyrifuranols N−P could significantly stimulate the proliferation and differentiation of umbilical cord mesenchymal stem cells.
Over the past seven decades several cembranoids have been reported, but uncertainty still exists about their structures. In this study, we isolated 38 macrocyclic cembranoids from B. papyrifera. Our 25 single‐crystal analysis uncovered misleading factors in the previously determined stereochemistries, and this led to six structures being revised. In addition, we propose a biosynthetic pathway and briefly discuss SARs.
Metal-organic frameworks (MOFs) are well known for their tunable structure and porosity. Many studies have shown they are promising for various important applications, for which their performance can ...be further enhanced by encapsulating functional species, such as luminescent guest molecules, within the frameworks. Although numerous MOFs are luminescent, very few emit white light and their quantum yield is usually low. Here we report a strategy to achieve efficient white-light emission by encapsulating an iridium complex in the MOF cavity. A mesoporous blue-emitting MOF is prepared as host to encapsulate a yellow-emitting iridium complex, Ir(ppy)2(bpy)(+). The resultant composites emit bright white light with good colour quality (for example, Commission International de I'Eclairage coordinates, colour-rendering index and correlated colour temperature of (0.31, 0.33), 84.5 and 5409 K, respectively), and high quantum yield up to 115 °C. This strategy may open new perspectives for developing high-performance energy-saving solid-state lighting materials.
Inadvertent intraoperative hypothermia is frequent during open surgeries; however, few studies on hypothermia during laparoscopic abdominal surgery have been reported. We aimed to investigate the ...incidence and risk factors for hypothermia in patients undergoing laparoscopic abdominal surgery. This single-center prospective cohort observational study involved patients undergoing laparoscopic surgery between October 2018 and June 2019. Data on core body temperature and potential variables were collected. A multivariate logistic regression analysis was performed to identify the risk factors associated with hypothermia. A Cox regression analysis was used to verify the sensitivity of the results. In total, 690 patients were included in the analysis, of whom 200 (29.0%, 95% CI: 26%-32%) had a core temperature < 36°C. The core temperature decreased over time, and the incident hypothermia increased gradually. In the multivariate logistic regression analysis, age (OR = 1.017, 95% CI: 1.000-1.034, P = 0.050), BMI (OR = 0.938, 95% CI: 0.880-1.000; P = 0.049), baseline body temperature (OR = 0.025, 95% CI: 0.010-0.060; P < 0.001), volume of irrigation fluids (OR = 1.001, 95% CI: 1.000-1.001, P = 0.001), volume of urine (OR = 1.001, 95% CI: 1.000-1.003, P = 0.070), and duration of surgery (OR = 1.010, 95% CI: 1.006-1.015, P < 0.001) were significantly associated with hypothermia. In the Cox analysis, variables in the final model were age, BMI, baseline body temperature, volume of irrigation fluids, blood loss, and duration of surgery. Inadvertent intraoperative hypothermia is evident in patients undergoing laparoscopic surgeries. Age, BMI, baseline body temperature, volume of irrigation fluids, and duration of surgery are significantly associated with intraoperative hypothermia.
