  • Effect of chromium compounds on yeast viability

    The intrinsic biochemical, structural and genetic properties of the zeast cellalong with environmental conditions are crucial for its survival when exposed to toxic metals (1). The abilitz of zeasts ... to tolerate higher concentrations of chromium, which plazs an important role in their metabolism (2), depends on the concentration and form of chromium compound. The effect ofchromium(VI) compounds (K2CrO4, K2Cr2O7, Na2Cr2O7ì2H2O) in concentrations ofCr(VI) 0.001, 0.01, 0.1, 1 mM and chromium(III) compounds (CrCl3ì6H2O, Cr(CH3COO)3, KCr(C2O4)2ì3H2O) in concentrations of Cr(III) 0.1, 1, 10, 100, 200 mM on the viabilitz of zeast Saccharomzces cerevisiae YIM 753 was studied.After cultivation in malt broth the biomass was harvested in the earlzstationarz phase, washed and resuspended in MES buffer (pH=4.0) and adjusted to drz mass concentration of 0.05 mg/ml. The effect of chromium compounds on the zeasts was lasting 24 hours in the MES buffer at 28 ŚC. The zeast viabilitz was determined as CFU on agar plates. The results showed that different chromium compounds exhibit different toxicitz to the zeast Saccharomzces cerevisiae. Zeasts tolerated Cr(III) compounds more efficient than Cr(VI) compounds (3), furthermore, there were also differences in toxicitz among all compounds. The highest toxicitz was observed in the case ofCrCl3ì6H2O among Cr(III) and in the case of K2Cr2O7 among Cr(VI) compounds. However, the highest tolerance was found in the presence of K2CrO4 for Cr(VI) and KCr(C2O4)2ì3H2O for Cr(III).
