    Distinctive granules which serve as a depot for diverse presynthesized multifunctional mediators are a hallmark of eosinophils and other granulocytes. A major component of eosinophil crystalloid ... granules are cationic proteins which exhibit characteristic staining properties. Compelling evidence suggests that these proteins, notably major basic protein 1 (MBP-1), can also be found extracellularly in association with mitochondrial DNA, forming so-called eosinophil extracellular traps (EETs). Aggregates of MBP-1 exert unselective toxicity toward not only pathogens but also host cells, thereby taking part in eosinophil-mediated immunopathologies. The role of mitochondrial DNA, which is believed to be the scaffold of EETs, has not yet been fully explained. Here, we aim to evaluate the effect of isolated bacterial, human genomic, and mitochondrial DNA on MBP-1-mediated toxicity toward E. coli and human epithelial BEAS-2B cells. Our results provide evidence that non- or low-methylated DNA such as bacterial and mitochondrial DNA greatly enhance MBP-1-mediated toxicity. Moreover, demethylated human genomic DNA, compared to its methylated analogue, also increases BEAS-2B and E. coli cell death, implying that DNA-mediated increased MBP-1-mediated toxicity is dependent on its methylation. Evaluation of known cell death signalling pathways and molecular mechanisms reveals that MBP-1 induces rapid cell death, the type of which has yet to be determined. The use of punicalagin, a membrane-stabilizing reagent, reverses MBP-1-mediated toxicity, suggesting membrane permeabilization as a key feature of MBP-1-induced cell death.
